1.Observation on clinical effects of three different treatment regimens for conservative treatment of ectopic pregnancy

Objective To explore the therapeutic efficacy of three different treatment regimens for conservative treatment of ec‐topic pregnancy(EP) .Methods Clinical data of 198 cases of patients with EP who were conservatively treated in this hospital from June 2012 to June 2014 were retrospectively analysed .All patients were divided into group A (39 cases) ,group B(92 cases) and group C(67 cases) ,which were treated with single‐dose of methotrexate(MTX) ,MTX combined with mifepristone and multiple‐dose of MTX respectively .The curative rate and time for human chorionic gonadotropin beta‐subunit(β‐HCG) dropping to normal were observed and recorded .Results The total curative rates in three groups were statistically different (P<0 .05) ,additionally , group C had the highest curative rate(86 .57% ) ,followed by group B with 84 .78% and group A with 66 .67% .In the three groups , for patients with serum level of β‐HCG 1 500-3 000 mIU/mL or >3 000 mIU/mL ,the curative rates were statistically different (P<0 .05) .Conclusion Single‐dose of MTX has relative low curative rate .It is recommend to use combination regimens or multi‐ple‐dose of MTX regimens ,and for patients with high level of β‐HCG multiple‐dose of MTX regimens might be the best choice .

3.Near-infrared diffuse correlation spectroscopy for early diagnosis of chronic periodontitis

Objective:To study the feasibility of the early diagnosis of chronic periodontitis using near-infrared diffuse correlation spectroscopy(DCS).Methods:30 subjects were inclded.The relative gingival blood flow(GBF) at the labial,mesial and distal healthy gingiva(the control) or the gingiva with periodontitis of maxillary anterior teeth was respectively measured by DCS before periodontal treatment,a week and a month after treatment.Results:The GBF of healthy gingiva group,gingivitis group and periodontitis group was 129.73 ± 10.70,95.51 ± 11.83 and 67.84 ± 13.05 respectively.Periodontal indexes were decreased and the relative GBF increased significantly 1 week and 1 month after treatment.Gingival index and sulcus bleeding index were respectively correlated to relative GBF(r =-0.902,r =-0.893).Conclusion:Near-infrared diffuse correlation spectroscopy can non-invasively detect the relative gingival blood flow in different stages of chronic periodontitis,it may be a method of early diagnosis of chronic periodontitis.
4.Study on isolating iufluenza viruses from MDCK cell

Objective To summarize the experience of isolating influenza virus from MDCK cell , and to optimize the isolation method for influenza isolation and raise the ability of influenza surveillance. Methods Sixty-one cases of influenza viruses positive specimens identified with PCR method by the influenza network laboratory in Xianyang CDC were used to infect the MDCK cells. The influenza viruses-infected MDCK cells were collected and characterized by hemagglutination test and hemagglutination inhibition assay. Meanwhile , different concentrations of glutamine liquid were added to the medium to investigate the effect of glutamine on the growth of MDCK cells. Results Twenty five flu strains were isolated from 61 cases influenza viruses positive specimens , of which 18 were A (H3N2) subtype strains and 7 were A (H1N1) subtype strains. The titer of HA was higher than 1∶16, and the titer of HI was higher than 1 ∶ 640. The proliferation rate of MDCK cells was low, and the cell activity was also decreased at 48 h after incubation with glutamine concentration lower than 0.1%. Conclusions The influenza viruses were isolated from the MDCK cells. The glutamine concentration is strongly associated with the activity of MDCK cells and cell apoptosis.
5.Expression of PDGF and growth of VSMC after mechanical injury and exposure to autologous serum

Objective To investigate the growth and expression of PDGF of VSMCs in response to stimulation of autologous serum and mechanical injury. Methods An vitro model simulating the condition as possible as that after PTA.PDGF of every medium sample from every group was detected by ELISA,and the values of MTT of every cellular sample was measured by MTT to show the growth and proliferation of every group. Results After stimulation by autologous serum and mechanical injury,SMCs of the experimental group showed the value of MTT increasing,but SMCs in control group reached on 3rd day.At the same time,the expression of PDGF also increased gradually,obtaining peak gradually up to peak on day 4/5 nearly 2.0-fold as much as that of SMCs in the control group. Conclusions After on the 5th day,stimulation with autologous serum and mechanical injury,VSMCs of rabbit showed the stronger ability of growth/proliferation,and autocrine of PDGF also increased gradually,reaching peak on 4-5 d,probobly simulating to those in vivo.

7.Feasibility study of adipose-derived mesenchymal stem cells in repairing traum-atic femoral neck fracture

Objective:To explore the feasibility of ADSCs in the clinical treatment of traumatic femoral neck fracture.Methods: After the intervention by ADSCs,femoral neck fracture repair was observed by hematoxylin-eosin staining.The vascular endothelial growth factor expression levels at the fracture segment were quantitatively measured by immunohistochemical staining,RT-qPCR was utilized to detect the mRNA formation in callus tissue.Results: In the study group,we observed less fracture repair than in the sham surgery group but more than in the blank group.Conclusion: ADSCs administration can promote osseous tissue and osteoblast repair,thereby contributing to the healing of traumatic femoral neck fracture in rats.

Objective To study the effect of nursing intervention for elderly patients with chronic obstructive pulmonary disease with oxygen treatment.Methods A total of 125 elderly obstructive pulmonary disease(COPD) patients with different nursing methods in our hospital were divided into observation group and control group.After given corresponding nursing care, patients''satisfaction of two groups were evaluated, and relevant influencing factors was analyzed.Results Nursing satisfaction in the observation group was obviously higher than that of the control group;Course of the disease, complications, bad feelings, and mechanical damage were influencing factors of oxygen efficacy for elderly patients with chronic obstructive pulmonary disease.Conclusion Nursing intervention can improve oxygen effect for senile chronic obstructive pulmonary disease patients, and improve quality of life, and correct cognition and compliance of patients.

10.The study on the rolling circle amplification for detecting hepatitis B virus covalently closed circular DNA

Objective To set up the rolling circle amplification (RCA) system for detecting hepatitis B virus (HBV) covalently closed circular DNA (cccDNA), and to evaluate the specificity and sensitivity of this system. Methods Plasmids containing full-length of wild-type HBV genome were treated with restriction enzyme and T4 DNA ligase, and then were concentrated. The DNA fragments were recovered by the nucleic acid purification kit and severed as standard HBV cccDNA. Total DNA was extracted from hepatic tissues of seven chronic hepatitis B patients. RCA method was used to amplify genomes from tissue samples. Standard HBV cccDNA, 3.2 kb liner HBV DNA, normal hepatic tissue samples and 15 serum samples of patients with chronic HBV infection were used as controls to determine the specificity of RCA. Ten-fold serial dilutions of standard HBV cccDNA were used for determining the sensitivity. Results The standard HBV cccDNA was successfully constructed and could be detected by RCA method. HBV cccDNA could be amplified from 2 mg hepatic tissue samples at least of HBV infected patients, and could be detected as low as 1 ×102 copy/μL. cccDNA was not detected in 3.2 kb liner HBV DNA, normal hepatic tissue samples and 15 serum samples of chronic HBV infected patients. Conclusion RCA method can be used for rapid and simple detection of HBV cccDNA with high specificity and sensitivity.
