1.Research of UVA in affecting Cx43 protein among rabbit lens epithelia cells

Objective:To probe the mechanism of inhibition in gap junction-mediated intercellular communication (GJIC) of rabbit lens epithelia cells(RLECs) irradiated by UVA. Methods:RLECs of 2~3 generation were divided into normal control group and ultraviolet irradiation group (The UV intensity was 0.2 mW/cm2 under a UV lamp with fixed position 50 cm, RLECs were exposed to ultraviolet lamp at different times: 5,10,15 minutes). Western blot techniques were employed to detect the protein levels of Cx43 irradiated by UVA. By indirect immunofluorescence histochemistry we analyzed the localization of Cx43 in the RLECs. Results:In UVA irradiation group and normal control group,there was expression of Cx43. With the increasing intensity,the expression of Cx43 was decreasing. The immunnostaining of Cx43 in RLECs revealed that the internalization of Cx43 from membrance to plasma and nuclear was found in UVA irradiated RLECs. Conclusion: Cell molecule level study proved UVA irradiation can reduce GJIC of RLECs,which wis associated with perturbations in localization and number of Cx43 among RLECs.

3.Recognition of walking stance phase and swing phase based on moving window.

Wearing transfemoral prosthesis is the only way to complete daily physical activity for amputees. Motion pattern recognition is important for the control of prosthesis, especially in the recognizing swing phase and stance phase. In this paper, it is reported that surface electromyography (sEMG) signal is used in swing and stance phase recognition. sEMG signal of related muscles was sampled by Infiniti of a Canadian company. The sEMG signal was then filtered by weighted filtering window and analyzed by height permitted window. The starting time of stance phase and swing phase is determined through analyzing special muscles. The sEMG signal of rectus femoris was used in stance phase recognition and sEMG signal of tibialis anterior is used in swing phase recognition. In a certain tolerating range, the double windows theory, including weighted filtering window and height permitted window, can reach a high accuracy rate. Through experiments, the real walking consciousness of the people was reflected by sEMG signal of related muscles. Using related muscles to recognize swing and stance phase is reachable. The theory used in this paper is useful for analyzing sEMG signal and actual prosthesis control.

4.Expression of Tim-3 and its relationship with Th17/Treg cytokine in the peripheral blood and airway in asthmatic murine

Objective To detect the expression of T cell immunoglobulin mucin 3 (Tim-3) and its relationship with Th17/Treg cytokine isolated from peripheral blood mononuclear cells (PBMCs) and broncho-alveolar lavage fluids (BALF) cells in the asthmatic murine model and to investigate the role of Tim-3 in the occurrence and development of the asthmatic inflammation. Methods The asthmatic murine model was established by the method of ovalbumin (OVA) injection and inhalation. The mice were randomly divided into 2 groups: the normal control and the asthma group. PBMCs and BALF cells in each group were collected. Expression of Tim-3 mRNA was detected by the real-time PCR assay. Levels of CD4+IL-17+ and CD4+CD25+FoxP3+, reflecting expressions of Th17 and Treg respectively , were detected by flow cytometry. The correlationbetween Tim-3 and Th17/Treg level was analyzed. Results Contrast with the normal control group, the expression of Tim-3 mRNA in PBMCs and BALF cells in the asthma group increased significantly (P < 0.01). OVA sensitization and challenges resulted in the increased CD4+IL-17 production and the decreased CD4+CD25+Foxp3+in PBMCs and BALF cells compared with those in the normal control group (both P < 0.05). The ratio of Th17/Treg significantly increased in asthma group (P < 0.05). Tim-3 mRNA expression in PBMCs and BALF cells was positively correlated with CD4+IL-17+ level and Th17/Treg ratio , respectively , and was negatively correlated with CD4+CD25+Foxp3+ level. Conclusions Tim-3 mRNA level in PBMCs and BALF cells in the asthma group was increased , indicating that Tim-3 might take part in the occurrence and development of asthmatic inflammation and has a very close relationship with the Th17/Treg imbalance.
5.Content Comparison of Polysaccharide and Protein in Male and Female Trichosanthis Radix

Objective:To compare the contents of polysaccharide and protein in male and female Trichosanthis Radix to provide reasonable explanation for that male plant was better. Methods:Polysaccharides in Trichosanthis Radix were obtained by water-extrac-tion and ethanol-precipitation method, and the content was determined by anthrone-sulfuric acid method. Tichosanthin ( TCS) was ob-tained by hydrochloric acid extraction and acetone precipitation. Based on the light absorption of Coomassie brilliant blue G-250 com-bined with proteins, the content of TCS was detected. Results: The polysaccharides content in male and female Trichosanthis Radix was 2. 628% and 3. 237%, TCS content was 0. 193% and 0. 280%, respectively. Conclusion: The contents of polysaccharides and TCS in male plant are higher than those in female plant. The male plant is a better source for Trichosanthis Radix.
6.Content Comparison of Amino Acid in Female and Male Plants of Trichosanthes Kirilowii

Objective:To compare the content of amino acid in female and male plants of Trichosanthes kirilowii. Methods:Total protein was hydrolyzed by hydrochloric acid, and then determined by HPLC. Results:The percentage of amino acid in female and male plants of Trichosanthes kirilowii was 7. 35% and 7. 04%, respectively. Conclusion:The method provides a new idea for the determi-nation of amino acid in the root of Trichosanthes kirilowii. The total amino acid content in the root of male plants is lower than that of fe-male plants, while the content of alkaline amino acid is opposite. The content of histidine in male plants is about twice as much as that in female plants. The essential amino acid content has no significant difference.
7.Application value of detection on stathmin, p16 and Ki-67 in the cervical intractable cases

Objective To investigate the clinical application value of stathmin, p16 and Ki-67 in the cervical intractable cases. Methods Immunohistochemical method was used to detect the expressions of stathmin, p16 and Ki-67 in surgical specimens of 288 cervical intractable cases, including 30 cases of cervical benign changes, 70 cases of cervical intraepithelial neoplasia (CIN)Ⅰ, 78 cases of CINⅡ, 85 cases of CINⅢand 25 cases of squamous cell carcinoma (SCC, as control group). The application value of stathmin, p16 and Ki-67 in the cervical cases were analyzed. Results The positive expression rates of Ki-67 of cervical benign changes and CINⅠwere 20.0 % (6/30) and 54.3 % (38/70) (χ2 = 3.29, P> 0.05). The expression rates of Ki-67 in CINⅡ, CINⅢ and SCC were all 100.0 %, and compared with the cervix benign changes, the differences were statistically significant (χ2= 112, P< 0.05). The expression rates of p16 in cervical benign changes and CINⅠwere 6.7 % (2/30) and 91.4 % (64/70), and there was significantly statistical difference (χ2=50.64, P<0.05). However, the expression rates of p16 in CINⅡ, CINⅢand SCC were all 100.0%, and compared with the cervix benign changes, the differences were statistically significant (χ2= 7.18, P< 0.01). The expression rates of stathmin in cervical benign changes, CINⅠ, CIN Ⅱ, CINⅢ and SCC were 3.3 %(1/30), 5.7 % (4/70), 23.1 % (18/78), 77.0 % (67/87) and 100.0 % (25/25), respectively, and there was no statistic difference in cervical benign changes, CINⅠand CINⅡ (χ2=0.68, P>0.05), but the expression rates in CINⅢ and SCC were higher than those in cervical benign change, CINⅠand CIN Ⅱ(P< 0.01). The positive expressions of stathmin, p16 and Ki-67 in each group of CIN were positively correlated (r= 0.412, P< 0.05). Conclusions Combined detection of p16 and Ki-67 can assist in the differential diagnosis of cervical intractable cases, and provide objective indicators for the classification and accurate diagnosis of CIN. Combined detection of p16 and stathmin may help to identify high-grade, low-grade CIN and cervix benign changes for the reduction of over-treatment.
8.Effects of Catalpol on Apoptosis andβ-Tubulin of Rabbit Sinoatrial Node Cells in Vitro Injured by Simulated Ischemia Reperfusion

ObjectiveTo observe the effects of catalpol, the effective component of Rehmanniae Radix, on atrionector cell apoptosis andβ-tubulin in vitro rabbit injured by simulated ischemia reperfusion;To explore the mechanism of treating sick sinus syndrome.MethodsAtrionector cells were collected from newborn rabbits. Cells were divided into 5 groups:normal group, model group, catalpol high, medium and low dose groups. Anoxia and aglycaemia were established to simulate ischemia. Atrionector cellular damage models were established by recovering the supply of oxygen and sugar. Normal control group and model group were given the same volume of culture medium, while catalpol high, medium and low dose groups were given medicine with relevant concentrations (100, 20, 10μg/mL, respectively). ELISA, FCM, laser scanning confocal microscopy were used to observe apoptosis rate andβ-tubulin of atrionector in each group.Results Apoptosis rate of the model group was obviously higher than the normal group (P<0.01), andβ-tubulin cleavage was obvious. Apoptosis rate in catalpol high, medium and low dose groups were significantly lower than that of the model group (P<0.01);β-tubulin structure were significantly more complete compared with the model group;the fluorescence intensity was significantly higher than that of model group (P<0.01).Conclusion Catalpol can inhibit atrionector cellular apoptosis caused by simulated ischemia reperfusion. Its protective effects on atrionectorβ-tubulin may be the mechanism of the treatment of Rehmanniae Radix for sick sinus syndrome.
9.Effect of ferulic acid on the apoptosis andβ-tubulin of rabbit sinoatrial node cell in vitro injured by low glucose and hypoxic

Objective To observe the effects of ferulic acid on cell apoptosis andβ-tubulinin vitro of rabbit sinoatrial node injured by hypoxia and low glucose, and to explore its mechanism.Methods Model cells were established on sinoatrial node cells from newborn rabbit by performing deprivation of oxygen and glucose. Then the model cells were divided into 5 groups: a normal control group, a model group, ferulic acid high, medium and low dose group. The normal control group and the model group were treated with equal volume of culture medium, and the three volume ferulic acid groups were treated with ferulic acid of 100, 20, and 10μg/ml respectively. Flow cytometry, laser scanning confocal microscopy were used to observe the sinoatrial node apoptosis rate and cytoskeleton proteinβ-tubulin in each group.Results Apoptosis rate of the model group was obviously higher than the normal group (56.95% ± 11.99%vs. 31.45% ± 6.32%,P<0.01), whileβ-tubulin cleavage was significantly lower than the normal group (5.141 ± 0.218vs. 8.035 ± 0.762,P<0.01). Apoptosis rate of ferulic acid high, medium dose group were significantly lower than the model group (24.85% ± 6.34%, 26.70% ± 9.84%vs. 56.95% ± 11.99%,P<0.01),β-tubulin structure were significantly more complete compared with the model group (7.927 ± 0.357, 5.961 ± 0.351vs. 5.141 ± 0.218,P<0.01).Conclusions Ferulic acid can suppress apoptosis of sinoatrial node cells caused by low glucose and oxygen. Protecting the sinoatrial node cell skeleton protein ofβ-tubulin may be one of the mechanisms.
10.Efficacy of preoperative femoral nerve block for preemptive analgesia in elderly patients undergoing total knee arthroplasty under general anesthesia

Objective To evaluate the efficacy of preoperative femoral nerve block on preemptive analgesia in the elderly patients undergoing total knee arthroplasty under general anesthesia.Methods Sixty ASA physical status Ⅰ-Ⅲ patients of both sexes,aged 65-75 yr,scheduled for elective total knee arthroplasty,were randomly divided into 3 groups (n =20 each) using a random number table:control group (group Ⅰ),preoperative femoral nerve block group (group Ⅱ) and postoperative femoral nerve block group (group Ⅲ).Single-injection femoral nerve block was performed with 0.375% ropivacaine 20 ml injected around the nerve under thc guidance of ultrasound immediately before induction of anesthesia and at the end of surgery in Ⅱ and Ⅲ groups,respectively.Patient-controlled intravenous analgesia (PCIA) with sufentanil (background infusion 2 ml/h,bolus dose 2 ml,lockout interval 15 min) was used until 2 days after surgery,and VAS score was maintained ≤ 3.When VAS score>3,flurbiprofen axetil 50 mg was injected intravenously as rescue medication.The consumption of sufentanil per hour,requirement for rescue medication,and development of adverse reactions were recorded within 24 h after surgery.Sufentanil-sparing degree was calculated.Results Compared with group Ⅰ,the consumption of sufentanil per hour,requirement for rescue medication and the incidence of nausea and vomiting were significantly decreased within 24 h after surgery in Ⅱ and Ⅲ groups.The consumption of sufentanil per hour,requirement for rescue medication and the incidence of nausea and vomiting were significantly lower within 24 h after surgery in group Ⅱ than in group Ⅲ.The consumption of sufentanil per hour was decreased by 35% in group Ⅱ as compared with group Ⅰ,and decreased by 18% in group Ⅱ as compared with group Ⅲ.Conclusion Preoperative femoral nerve block can provide good preemptive analgesia in the elderly patients undergoing total knee arthroplasty under general anesthesia.
