Objective To detect the expression of insulin-like growth factor Ⅱ mRNA-binding protein 3 (IMP3) and p16 in cervical carcinoma and investigate their clinical significance.Methods Immunohistochemical Envision method was used to detect the expression of IMP3 and p16 proteins in 67 cases of carcinoma of the uterine cervix,58 cases of cervical intraepithelial neoplasia (CIN) and 15 cases of normal cervical epithelium.Results IMP3 was mainly dedected in the cytoplasm,whose positive expression rate was 85.07％ (57/67) in cervical carcinoma,12.07％ (7/58) in CIN (x2 =66.32,P ＜ 0.05) and 0 (0/15) in cervical epithelium (x2 =44.38,P ＜ 0.05).No significance variations of the expression of IMP3 were observed in cervical adenocarcinoma and squamous cell carcinoma (x2 =0.35,P ＞ 0.05).p16 was found both in nucleus and cytoplasm of cervical carcinoma,whose expression rate was 83.58％ (56/67),higher than 41.37％ (24/58) in CIN (x2 =24.85,P ＜ 0.05) and 0 (0/15) in cervical epithelium (x2 =42.61,P ＜ 0.05).Conclusion Expression of IMP3 and p16 is high in cervical carcinoma,maybe can be used as a potential biomarker of metastasis and progression.

OBJECTIVE:To establish a method for the content determination of isoniazid in Isoniazid tablet. METHODS:Mi-crofluidic chip with contactless conductivity detection was adopted. The effects of electrophoresis parameters,such as the variety, concentration,pH,additive types and dosage,injection time and separation voltage on the separation detection were explored. RE-SULTS:The optimized buffer system was 1.86 mmol/L citrate-0.14 mmol/L sodium citrate buffer solution (pH3.0),no additive , the injection time was 20 s and the separation voltage was 2.5 kV. In the optimal conditions,the linear range of isoniazid was 10-200 μg/mL(r=0.9993);the limits of quantification and detection were 8.0 μg/mL and 2.4 μg/mL;RSDs of precision,stability and reproducibility tests were lower than 2.0%;recovery was 97.8%-102.9%(RSD=1.9%,n=9). CONCLUSIONS:The method is rapid and simple,and suitable for the content determination of isoniazid in Isoniazid tablet.

Objective To investigate the functional protective effect of lycium barbarum polysaccharide (LBP) against light-in-duced retinal degeneration. Design experimental studies. Participants sixty SD rats were divided randomly into 6 groups. Methods Sprague-Dawley rats were exposed for 24 hours to 2000-lux cool white fluorescent light after treatment with LBP. Scotopic flash elec-troretinogram (ERG) were recorded before and on day D1, D6 and D14 after light exposure. The b-wave amplitude was statistic. In addi-tion, rats from each group were killed for retinal morphometric analyses. Main outcome Measure The amplitudes of b-wave of scotopic flash ERG, the histopathological changes. Results In the untreated group, light exposure caused collapse of the b-wave sensitivity curves. Bmax was reduced by 29.97% at D1 without subsequent recovery. Histopathology of the temporal superior central retinas of rats showed outer segment disorganization and shortening, the inner segments swelled and vacuolated, the outer nuclear layer thicknesses decreased markedly with subsequent recovery. In the treated groups, light exposure had a weaker effect on sensitivity curves. The values of b-wave amplitude were significantly increased than those in the exposed-untreated group (P=0.005). Retinal morphometry was pre-served. Conclusion LBP afford functional protection against light-induced retinal damage. (Ophthalmol CHN, 2009, 18: 229-233)

Objective To study the apllication value of cerebraspinal fluid minimal residual leukemia cells(MRLCs) combined with lactate dehydrogenase(LDH) detection in early diagnosis of central nervous system leukemia(CNSL) to provide a laboratory basis for clinical diagnosis and evaluation of disease progression and therapeutic effect.Methods Thirty inpatiernts without blood disease and central nervous system(CNS) organic diseases,and normal CSF routine and detected biochemical indexes were selected as the control group and 96 cases of acute leukemia (AL) diagnosed by bone marrow puncture FAB morphology and flow cytometry typing served as the disease group.Flow cytometry was used to detect the MRLCs in CSF,the activity of LDH in CSF was detected by the rate method,and the detection results were performed the comparative analysis.Results Among 96 cases of AL,30 patients with CNSL had 13 cases of positive MRLCs in CSF,the positive rate was 43.33%,among 66 cases of non-MRLCs,MRLCs was not detected detected.Among 96 cases of AL,CSF LDH level in 30 cases of CNSL was (30.54±10.29)U/L,which was significantly higher (16.16±7.12)U/L in the patients without CNSL,the difference between the two groups was statistically significant (P<0.05).Conclusion The detection of MRLCs and LDH in CSF of AL can be used as a laboratory diagnostic index for early diagnosis of CNSL,and MRLCs combined with LDH detection can improve the positive diagnosis rate of CNSL.

Objective To detect the mRNA expression of KPNA2 in hepatocellular carcinoma(HCC) tissue and to investigate its relationship with the clinicopathological features of HCC so as to provide the basis for predicting HCC infiltration ,metastasis and early treatment .Methods The HCC tissue and paracarcinoma normal tissue from 30 operative cases of HCC in the hepatobiliary surgery department of this hospital from January 2011 to November 2013 were collected and detected KPNA2 expression at the transcriptional level by RT-PCR .The correlation between the mRNA expression of KPNA2 with the clinicopathological factors of HCC was analyzed by consulting the medical record data .Results Among 30 cases of HCC ,the ratio of KPNA2 mRNA in the HCC tissue and the paracarcinoma normal tissue in 25 cases(83 .33% ) was up-regulated (grey value >2 times) .Therefore ,the KP-NA2 gene exhibited the high expression in the HCC tissue and low expression in the paracarcinoma normal tissue .Conclusion KP-NA2 is highly expressed in the HCC tissue and lowly expressed in the paracarcinoma normal tissue ;the high expression of KPNA2 is positively correlated with the stage and pathological grading of HCC ;KPNA2 may be used as an important indicator for early di-agnosing HCC and judging the malignant degree and potential metastais ability of HCC .

Obgective To explore the risk factors of preterm infants with bronchopulmonary dysplasia(BPD).Methods A prospective cohort study was conducted to analyze the risk factors of preterm infants with BPD.Preterm infants (gestational age ＜ 32 weeks,and admission within 24 h since birth,and survival time more than 28 d since birth) who were sent to the Ward of Extremely Preterm Infants in Bayi Children's Hospital Affiliated to Clinical Medical College in Beijing Military General Hospital of Southern Medical University were enrolled from November 2013 to May 2014.According to the diagnostic criteria of BPD,the subjects were divided into 2 groups(BPD group and non-BPD group).Factors such as maternal information,neonatal basic information,neonatal diseases and treatments were compared between the 2 groups.Risk factors of preterm infants with BPD were analyzed by using t test,Chi-square test,Fisher's exact probability method and Logistic regression analysis.Results (1) A total of 298 cases were enrolled in this study.Among these infants,180 cases were male and 118 cases were female.The gestational age ranged from 25.6 to 31.9 weeks with the average age of (29.9 ± 1.4) weeks and the birth weights ranged from 740 to 2 300 g with the average weight of (1 428.3 ± 289.0) g.There were 19 cases of extremely low birth weight and 175 cases of very low birth weight.Sixty-nine cases of these infants were diagnosed as BPD (43 cases were mild,10 cases were moderate,16 cases were severe) with incidence of 23.2％.(2)The incidence of BPD was negatively related to gestational age and birth weight:the incidence of BPD in preterm infants with gestational age ＜ 28 weeks,28-30 weeks and ≥ 30-32 weeks were 70.4％,41.9％ and 6.2％;the incidence of BPD in preterm infants with birth weight ＜ 1 000 g,1 000-1 500 g and ≥ 1 500-1 800 g were 78.9％,29.5％ and 8.8％.(3) Multivariate Logistic regression found gestational age (OR =4.52),birth weight (OR =3.38),gender (OR =3.04),cytomegalovirus infection (OR =55.27),duration of invasive ventilation ≥ 7 d (OR =3.22),the highest concentration of inspired oxygen ≥400 mL/L (OR =4.14),patent ductus arteriosus(PDA) in need of surgical ligation (OR =7.30),and transfusion of packed red blood cells within 14 d since birth (OR =3.51) were the independent risk factors of BPD (all P ＜ 0.05).(4) Factors such as birth weight (P =0.015),duration of invasive ventilation (P =0.003),duration of inspired oxygen (P =0.000),and PDA in need of surgical ligation or not(P =0.017) were related to the severity of BPD.Conclusions BPD is a multifactorial disease.Taking effective measures to control risk factors is the key for preventing BPD.

OBJECTIVE:To establish a method for identification of Cornus officinalis and its Preparations. METHODS: LC-MS method was adopted. The separation was performed on Hypersil C18 column (150 mm?2.1 mm,5 ?m) with a mobile phase of methanol-acetic acid(84 ∶ 16) at a flow rate of 0.2 mL?min-1. The column temperature was set at 30℃. Mass spectra were equipped with an electrospray ionization (ESI) source with negative ion detection model as ion scanning. The conditions of determination were sheath gas flow rate of 1.5 L?min-1,sweep gas flow rate of 0.45 L?min-1,spraying voltage of 4.5 kV, high temperature capillary temperature of 350℃ and scan range of 110～600 amu. RESULTS: The three-level mass spectrometry of the ursolic acid and oleanolic acid was well separated. The results were stable. Three-level mass spectrometry of ursolic acid and oleanolic acid were established and identification of C. officinalis preparations was carried out.CONCLUSION:Three-level fragment peaks of fat-soluble constituents of C. officinalis can be used for indentification of C. officinalis and its preparations.

Objective To investigate the correlation between the D-dimer level and the acute exacerbation of chronic obstructive pulmonary disease (AECOPD).Methods The changes of D-dimer levels in 156 patients (AECOPD group) before and after treatment were measured and compared with the D-dimer levels of 156 healthy subjects(control group).Results The concentration of plasma D-dimer in the AECOPD group was (1.35 ± 1.46) mg/L,which was significantly higher than (0.25 ± 0.16)mg/L in the control group,and the difference was statistically significant(t =9.354,P ＜0.05).The level of plasma D-dimer in the AECOPD group was (0.69 ± 0.87)mg/L,and compared with (1.35 ± 1.46)mg/L before treatment,the difference was statistically significant(t =4.850,P ＜ 0.05).Conclusion D-dimer level is correlated to the condition of patients with chronic obstructive pulmonary disease,it can be used as one of the indicators to diagnose whether a patient is in hypercoagnlable state or suffers thrombotic diseases.

Objective To study the association of peptidylarginine deiminase 4 (PADI4) (padi4_94 and padi4_104) genetic single nucleotide polymorphism (SNP) and rheumatoid arthritis (RA) susceptibility in Han population of Hebei province. Methods This hospital-based case-control study included 115 RA patients and 106 healthy controls. All the individuals were recruited from Han Hebei residents and were randomly selected. The genotype and allele frequencies of PADI4 gene polymorphisms (padi4_94 and padi4_104) were analyzed by PCR-DNA sequencing method. Results The distribution of padi4_94 and padi4_104 genotypes between the two groups was not significantly deviated from that expected by Hardy-Weinberg equilibrium (P>0.05). The combined effect of padi4_94 and padi4_104 SNPs was analyzed and three haplotypos (AA, AG and GG) were found but without GA haplotype. The genotype and allele distribution of padi4_94 and padi4_104 in the patients with RA was not significantly different from that in healthy controls,and the analysis of haplotypes revealed that no haplotypes were risk factors for RA. Conclusion In Han population of Hebei province, the SNPs of PADI4 (padi4_94 or/and padi4_104) is no associated with RA susceptibility. Therefore, it should not be regarded as a genetic risk factor for RA.
