Objective To investigate the effect of berberine on the radiosensitivity of cervical cancer cells.Methods 5,10,15,20 μmol/L of berberine were used to treat cervical cancer cell lines of Siha,HeLa,Caski.DMSO was applied as control of drug treatment.Cell proliferation was detected by the CCK-8 method,and then the half inhibitory concentration of berberine was calculated.Cell apoptosis and cell cycle distribution were detected by flow cytometry.Protein expressions of Cleaved Caspase-3,Cyclin B1,CDK1,STAT3 and p-STAT3 were detected by Western blot.Cervical cancer cells of Siha were treated by berberine with a half inhibitory concentration for 24 h and then irradiated with 0,2,4,6,8 Gy of X-rays.Cell clone assay was used to detect cell survival.Results Berberine could inhibit the growth of cervical cancer cells with a half inhibition concentration of(16.84 ± 3.52),(23.54 ± 8.67),(21.86 ± 6.35)μmol/L for Siha,Caski,and HeLa cells,respectively.The berberine at 17 μmol/L could induce apoptosis (t =56.847,P ＜ 0.01) and G2/M phase arrest (t =47.251,P ＜ 0.01) in Siha cells,which also inhibited the expressions of Cyclin B1,CDK1 and p-STAT3 and promoted the expression of cleaved Caspase-3,but did not influence the expression of STAT3 in cervical cancer cells.Treatment of cells with 17 μmol/L berberine increased the radiosensitivity of cervical cancer cells with a sensitivity enhancement ratio of 1.55.Conclusions Berberine can inhibit cell proliferation,promote apoptosis,block cell cycle,and increase radiosensitivity of cervical cancer cells.

Objective To study the influences of hepatolobectomy on space learning capability and the expressions of Drebrin and PSD95 in the hippocampus in APP/PS1 transgenic mouse model of Alzheimer's disease.Methods A total of fifty-four 3-month-old APP/PS1 transgenic mouse were randomly divided into 3 groups:3-month-old control group,sham surgery group,hepatolobectomy group,3-month-old littermates control group and 6-month-old control group.Morris water maze test was used to observe space learning capability on the 1st,3rd,7th,14th day after surgery,meanwhile the expressions of Drebrin and PSD95 in hippocampus were measured by Western blotting.Results Compared with the sham surgery group,the results of the Morris water maze test were decreased in hepatolobectomy group at day 3,7,14 after surgery [(62.9±6.9) s vs.(35.7±12.2) s,(66.3± 9.5) s vs.(39.3±8.3) s,(67.1±7.5) s vs.(32.6±14.1) s],and 6-month-old control group [(75.9±12.1) s] (all P＜0.05).The escape latency were (62.9±6.9)s,(66.3±9.5)s,(67.1± 7.5)s and (75.9±12.1)s,the probe trials were (2.1±0.7) times,(1.83±1.5) times,(2.5±1.9) times and (1.8±0.8) times respectively in hepatolobectomy group at day 3,7,14 after surgery and 6-month old control group.No significant differences in the results of the Morris water maze test were found among 3-month-old control group,3-month-old littermates control group,and sham surgery group.Compared with 3-month-old control group and sham surgery group atday 1,3,7,14 after surgery the expressions of Drebrin were decreased in 6-month-old control group and the hepatolobectomy group at the same time points.Meanwhile,the expression of Drebrin in hepatolobectomy group was increasedat day 14 versus day 7 after surgery.Compared with 3 month-old control group,the expression of Drebrin was increased at day 7 after sham surgery.Compared with 3-month-old control group and sham surgery group at day 3,7 and 14,the expressions of PSD95 were decreased in hepatolobectomy group at the same time points.Compared with 3-month-old control group,the expression of PSD95 was increased in sham surgery group at 7th day (P＜0.05).Between 3 month-old control group and 3-month-old littermates control group,the expressions of Drebrin and PSD95 had no significant differences.Concltsions Hepatolobectomy can impair the capabilities of space learning and memory in 3-month-old APP/PS1 transgenic mice,which may be associated with the decreased expressions of Drebrin and PSD95 in hippocampus.

Objective To investigate the effects of partial hepatectomy on spatial learning and memory abilities and the expression of spinophilin,which was used as a marker to quantify changes in dendritic spine density in the hippocampus of APP/PS1 transgenic mice.Methods 3-month-old and 6-month-old male APP/PSI transgenic mice and 3-month-old wild-type male littermates were divided into five groups based on their genetic background:3-month-old control (group A,n =8),anesthesia (group B,n =32),surgery (group C,n =32),littermates control(group D,n =8),6-month-old control (group E,n =8).According to the different time points at day 1,3,7,14 after anesthesia or surgery,group B and C were divided randomly into 4 subgroups:B1,B3,B7,B14 and C1,C3,C7,C14.The spatial learning and memory abilities were detected by Morris water maze test in each group at the corresponding time points.Meanwhile,spinophilin expression was detected by Western blot and Immunocytochemistry in the hippocampus.Results Compared with the same time point subgroup B,the latency to find a hidden platform was longer and the frequency of passing through the platform was decreased in Group C at day 3,7,14(P ＜ 0.05).No significant differences were found among group A,group D,and group B (P ＞ 0.05).The grey value and Western blot relative integrated optical density of spinophilin in group A(1 609 453 ±92 801/0.471±0.015),approached the Group D(1 329 398.4±77 783/0.434±0.008)(P＞0.05),were significantly increased compared with that(801 678 ± 10 505/0.053 ± 0.003) in group E (P ＜ 0.01).Compared with the Group A,the same time point subgroup B and subgroup C14,the grey value and Western blot relative integrated optical density of spinophilin were decreased in Group C at day 1,3,7 (P ＜ 0.05).Conclusion Partial hepatectomy can impair spatial learning and memory abilities of the APP/PS1 transgenic mice,which is closely related to the decrease expression of spinophilin in hippocampus.

Objective To explore the effect of meta-mood ability training for improving job satisfaction of nurses.Methods A total of 120 nurses were randomized into the observation group and the control group (60 cases in each group).The observation group received meta-mood ability training,while the control group was given routine training.The meta-mood ability and job satisfaction degree were investigated.Results After the intervention,job satisfaction degree and the meta-mood ability level of the observation group nurses were improved than that of the control group.Conclusions The meta-mood ability training is effective in improvement of job satisfaction degree of nurses,as well as increase clinical service quality of nurses.

Objective To compare the contrast-enhanced ultrasonography (CEUS) manifestations of pNENs with the pathological changes and to explore the clinical value of CEUS in diagnosis of pNENs. Methods The CEUS of sixteen pathologically diagnosed pNENsfrom April,2012 to February,2014 were retrospectively observed.The CEUS of thirty pathologically diagnosed pancreatic carcinomas during the same period were taken as the control group.The enhancement extent and enhancement patterns of CEUS in pancreatic tumors were analyzed. The specimens of pNENs were stained with HE and CD34 immunochemistry.The grading of interstitial content and Ki-67 index were obtained on HE-stained slices and microvascular density(MVD) were obtained on CD34 immunochemistry stained slices.Spearman rank test analysis was employed to analyze the correlation.Results Among the 16 cases of pNENs,12 cases (75.0%) demonstrated hyper-or iso-enhancement,which included 6 cases (37.5%) showing homogeneous enhancement and 6 cases (37.5%) showing inhomogeneousen hancement,and the other 4 cases demonstrated hypo-enhancement. Among 30 cases of pancreatic carcinomas,25 cases (83.3%) demonstrated hypo-enhancement and 5 cases (16.7%) hyper- or iso-enhancement,and 21 cases (70.0%) demonstrated homogenous enhancement,and 9 cases(30.0%)inhomogeneous enhancement.Taking hyper-or iso-enhancement on CEUS as the diagnostic criteria,its diagnostic sensitivity,specificity,positive predictive value,negative predictive value,and accuracy were 75.0%,83.3%,70.6%,86.2%,80.4%respectively.Negative and positive correlation of the enhancement extent of pNENs with the interstitial content(r =-0.880,P < 0.001) and MVD (r =0.658,P =0.003) were observed,respectively.Six tumors with inhomogeneous enhancement contained cystic degradation or necrosis under microscope. Conclusions CEUS is an effective and non-invasive diagnostic approach for pNENs and can reflect the pathological changes.Hyper-or iso-enhancement of CEUS is typical appearance of pNENs.

ObjectiveTo observe the clinical effection of acupuncture using Xingnao Kaiqiao needling method and rehabilitation training on patients with stroke.Methods70 patients with stroke were randomly divided into the observation group(n=35)and the control group(n=35), both groups receiving routine treatment and early rehabilitation training, while acupuncture using Xingnao Kaiqiao needling method was given additionally to the observation group, 4 weeks for a course. The Fugl-Meyer Assessment, assessment of neurological deficit, the Barthel Index and the clinical effect were compared among two groups.ResultsThe neurological deficit decreased obviously, and the Fugl-Meyer Assessment and Barthel Index increased significantly in two groups after the treatment(P<0.05). The theraputic effect was significantly better in the observation group than in the control group(P<0.05).ConclusionAcupuncture using Xingnao Kaiqiao needling method combined with rehabilitation training can more effectively promote the recovery of nerve function, enhance motor function, and significantly improve the quality of life and the prognosis of patients with stroke.

Objective To improve the in vivo analgesic activity of acacetin and find leads for the development of new drugs, novel acacetin derivatives containing alkyl amide groups with different length of carbon chain were designed and synthesized according to the molecular structure of the active hit compound found in our previous work. Methods Using apigenin as the initial chemical ma-terial,the acacetin was synthesized through 3 steps,then the target compounds were prepared by conjugating hydroxy group of acace-tin at position 7 with bromoalkyl amides. The analgesic activity of the target compounds was evaluated by acetic acid writhing model of mice. Results and Conclusion Novel acacetin alkyl amide derivatives showed more potent analgesic activities than that of clinical medicine diclofenac,which could be used as leads for further development of new drugs.

Objective To explore the manifestation of pancreatic metastases (PM)using contrast‐enhanced ultrasound (CEUS). Methods Eleven patients with PM confirmed by pathology or CT/MRI combined with clinical data were analyzed retrospectively. The manifestation on conventional ultrasound and the enhancement appearance on CEUS were analyzed. Results The primary malignant tumors included lung cancer(n=6), breast cancer(n=1), renal clear cell carcinoma(n=1), gastric cancer(n=1), rectal cancer(n=1)and leiomyosarcoma of uterus(n=1). Eight patients had one lesion and three were multiple. The maximum lesion diameter was 2. 0-5. 0cm. Eight of the 11 patients showed hypoechoic on conventional US. Three patients were diagnosed as probably PM, one probably malignant and 7 undetermined by US. In the early stage of CEUS, two patients showed hyper‐enhancement, six showed iso‐enhancement and 3 showed hypo‐enhancement I.n the late stage of CEUS, nine patients (9/11) showed hypo‐enhancement. Seven patients showed homogeneous enhancement S.ix patients were diagnosed as PM, three malignant, one neuroendocrine tumor and one undetermined by CEUS. Conclusions PM showed certain characteristics on CEUS. The hyper‐or iso‐enhancement at the early stage of CEUS and history of primary cancer is helpful for diagnosis of PM.

Objective To investigate the prevalence and risk factors of needle-sharing behavior among intravenous drug users (IDUs).Methods Data regarding socio-demographics,drug abuse characteristics and HIV related health literacy of 474 heroin IDUs were collected.Antisocial personality disorder of IDUs was diagnosed through Mini international neuropsychiatric interview.Results The prevalence of needle sharing among IDUs was 53.8％.Needle-sharing behavior of IDUs was significantly associated with male gender,marital status of being single and separated/divorced,local dwelling,an early age of first drug abuse and antisocial personality disorder (OR =1.11 ～ 6.69,P＜0.05).Conclusion Heroin IDUs have high prevalence of needle sharing.A comprehensive social,behavior and psychology based intervention is needed to effectively prevent HIV infection in IDUs.

Object To establish the quality standard for Wuhu Powder. Methods Radix Angelicae Sinensis (RAS), Radix Angelicae Dahuricae (RAD), and Flos Carthami (FC) were determined qualitatively by TLC. The content of cimicifugoside and 5-O-methylvisammioside were determined by HPLC. Results The linear ranges of cimicifugoside and 5-O-methylvisammioside were 0.370 0—1.850 0 ?g and 0.191 0—0.955 0 ?g. The recoveries were 100.74% and 98.33%, RSD were 0.67% and 1.91% (n=5), respectively. Conclusion The method is simple, accurate, credibility, which can be used as the quality control standard of Wuhu Powder.
