1.Experimental animal models in attention deficit hyperactivity disorder

Objective To review the advantages and disadvantages of common attention deficit hyperactivity disorder(ADHD) animal models,published in 2000-2014 at home and abroad,with an attempt to provide new ideas for the future study of the pathogenesis in ADHD.Methods Major online database including CNKI,Wan Fang databases,OVID,Pubmed databases were searched in January 2015,using the key wordsattention deficit hyperactivity disorder, animal modelsand so on,to analysis the advantages and disadvantages of each type of experimental animal models and research value in ADHD.Results Totally 29 studies were enrolled,and the analysis show that the current commonly used experimental animal models of ADHD are spontaneously hypertensive rats,dopamine transporter gene knockout mice,lack of mutant mice model,neonatal 6 hydroxy dopamine damage of juvenile rat model,neonatal hypoxic rats model,X-ray irradiation injury model of rat hippocampus and other brain tissue damage model and isolated feeding model,and each animal model has the advantages,disadvantages and special research value of the pathogenesis of ADHD.Conclusion The specific pathogenesis of ADHD is unclear,but the neuroendocrine changes especially HPA axis attracted much attention in recent years.this paper summarizes the research at home and abroad comparison of several common animal models of ADHD.neuroendocrine changes in SHR and ADHD children have a certain degree of consistency in HPA axis,looking for ADHD children and SHR in neuroendocrine aspects of consistency is likely to open a new path to search the pathogenesis in ADHD.

3.Effect of Acupuncture Therapy of Regulating Yin and Yang on Plasma Levels of Calcitonin Gene-related Peptide and Vascular Endothelin in Stroke Patients

Objective To observe the effect of acupuncture therapy of regulating Yin and Yang for the treatment of stroke,and to investigate the influence on plasma levels of calcitonin gene-related peptide(CGRP) and vascular endothelin(ET).Methods One hundred and twenty stroke patients were equally randomized into 3 groups: group A received acupuncture therapy of regulating Yin and Yang,group B received traditional body acupuncture therapy,and group C received scalp acupuncture therapy.One and a half months constituted one treatment course,and the treatment lasted 2 courses.The therapeutic effect was evaluated and the changes of plasma CGRP and ET levels were observed.Results The total effective rate was 95.0% in group A,85.0% in group B,and 82.5% in group C,the difference being significant between the three groups(P0.05).Conclusion Acupuncture therapy of regulating Yin and Yang exerts a certain effect for the treatment of stroke,and can up-regulate plasma CGRP level and decrease ET level.

5.Tissue distribution of s-oleylpropanolamide in rats detected by liquid chromatography with tandem mass spectrometry.

This paper is to report the development of a rapid and sensitive method for the determination of s-oleylpropanolamide (OPA) in various tissues of rat (brain, heart, lung, liver, spleen, small intestine, kidney, adipose tissue and muscle), and to assess the applicability of the assay to tissue distribution. OPA was extracted by liquid-liquid extraction method with undecylenoylethanolamide as an internal standard. The concentrations of OPA were determined by LC-MS/MS after a single intragastric dose of 50 mg x kg(-1) at 4 time points (5 rats per group). With multiple reactions monitoring mode (MRM) the limit of quantification (LLOQ) was determined at 1 microg x L(-1). The calibration curve was linear from 1 to 2 x 104 microg x L(-1) (r > or = 0.999 0) for tissue homogenates. Validation parameters such as accuracy, precision and recovery were found to be within the acceptance criteria of the assay validation guidelines. The highest concentration was found in small intestine (the highest time point is 15 min) and heart (the highest time point is 90 min). The assay is rapid, sensitive and applicable to studying tissue distribution of OPA in rats.
6.Association of β-3-adrenergic Receptor Gene with Insulin Resistance in the Patients with Polycystic Ovary Syndrome

Objective To explore the association of the Arg64 polymorphism β-3-adrenergic receptor(β3-AR) gene with polycystic ovary syndrome(PCOS)and its characterization with obesity,insulin resistance(IR),blood pressure (BP), blood lipid spectrum. Methods The polymorphism of the β-3-AR gene was determined in 163 patients with PCOS and 100 controls. Body mass index(BMI), waist to hip circumference ratio(WHR), BP were examined. Blood glucose,blood lipid spectrum,insulin were also measured. Insulin resistance index was calculated. Results The frequency of Arg64 allele in PCOS women was significantly higher than that of controls(0.33 vs 0.13 respectively, P<0.05). Fasting insulin, insulin resistance index, WHR in Arg64 group were significantly higher than those of Trp64 group. The frequency of Arg64 allele was positively correlated with FINS, insulin resistance index, WHR. Conclusion The Arg64 mutation in the β-3-adrenergic receptor(β-3-AR) gene might contribute to development of PCOS and IR.
7.Ketogenic diet therapy in the epilepsy of infancy with migrating focal seizures associated with TBC1D24 gene mutation: two case reports and literature review

Objective:To investigate the efficacy and safety of ketogenic diet (KD) therapy in the epilepsy of infancy with migrating focal seizures (EIMFS) associated with TBC1D24 gene mutation.Methods:Clinical data of two children with TBC1D24 gene-related EIMFS were collected retrospectively, who were admitted to Department of Pediatrics, Fujian Medical University Union Hospital from 2019 to 2020. Their clinical characteristics and the efficacy and safety of KD therapy were analyzed, and literature review was conducted.Results:Seizures were onset before six months old in the two children with TBC1D24 gene-related EIMFS. Multifocal myoclonic seizures were manifested and happened frequently, lasting for more than 30 minutes sometimes. Developmental retardation was obvious in the two children. A small amount of focal sharp, spike, sharp-slow complex, and spike-slow complex waves were showed in the interictal electroencephalography (EEG). TBC1D24 gene mutations were found in the two children, one with a compound heterozygous mutation (c.1025C>T, p.S342L; c.229_c.240delATCGTGGGCAAG,p.I77_K80del), and the other with a homozygous mutation [c.119G>A,p.R40H(Arg40His)]. Both of those were potentially pathogenic. A variety of anti-epileptic drugs showed poor outcome for the two children. The epilepsy was drug-refractory one. After four to 17 months of KD therapy, the epilepsy in the two children was controlled effectively. There was not obvious adverse reactions. Among six children with TBC1D24 gene-related EIMFS in the literature review, four cases were effective or partially effective for KD therapy, one was discontinued due to insufficient ketogenic ratio, and one was discontinued without effect. There were no obvious adverse reactions in the six children.Conclusions:TBC1D24 gene-related EIMFS is mostly drug-refractory epilepsy. Early KD therapy may help to control seizures.

9.Nurses′experience and attitudes toward truthful communication with dying cancer patients about the prognosis:a qualitative study

Objective To elucidate the experience and attitude of bad news delivery from nurses to advanced cancer patients, in order to offer better nursing care to the dying cancer patients as well as to provide evidence for end-of-life cancer care. Methods A phenomenological research method of qualitative study was employed in this study. Fifteen nurses caring for dying cancer patients were in-depth interviewed with semi-structured interview guide. Colaizzi analytical method was adopted to collect and analyze the data, which was then organized into themes and subthemes. Results Three themes wereconflict between will and behavior barriers of bad news delivery delivery with comprehensive assessment of dying cancer patients. Conclusions Though the oncology nurses thought the dying cancer patients should be told they were dying, few nurses did that. Communication and hope maintenance related to end-of-life cancer care training should be offered to oncology nurses, as well bad news delivery to ensure cancer patients′safety.
10.Clinical value of computer-aided algorithm on radiologist's diagnosis of masses using breast ultrasound imaging

Objective To propose a novel computer-aided segment algorithm, and retrospectively investigate its effect on radiologist's sensitivity and specificity for discriminating malignant masses from benign masses basing on ultrasound images. Methods Four hundred and sixty-six images of 100 masses obtained by conventional ultrasound were processed by a novel segment algorithm. Radiologists who were blind to the histology results were invited to analyse the original and computerized images respectively. The sensitivity and specificity were calculated by means of a binary outcome using receiver operating characteristic(ROC) analysis. Results By using the segment algorithm, the quantity of the image was obviously improved, especially the margin and calcification of the masses. The diagnostic performance of the radiologists was also improved. The sensitivity rose from 70.32% to 90.52% while the specificity was 74.31%. The area under the ROC curve increased significantly (P<0.01) from 80.8% to 90.5%. Conclusions The proposed segmentation algorithm can improve the diagnosis performance of the radiologists by meliorating the quality of the ultrasound image.
