1.Comparison of clinical characteristics of acute and chronic central serous chorioretinopathy

Objective:To compare clinical characteristics of acute and chronic central serous chorioretinopathy (CSC).Methods:41 patients with CSC in the First Affiliated Hospital of Soochow University were studied, according to the course, they were divided into acute central serous chorioretinopathy group (A group, 29 eyes of 29 cases) and chronic central serous chorioretinopathy group (B group, 12 eyes of 12 cases). All patients underwent the best corrected visual acuity (BCVA), subretinal fluid thickness (SRF) and subfoveal choroidal thickness (SFCT) examination. The imaging characteristics of fundus fluorescein angiography (FFA) and optical coherence tomography angiography (OCTA) were compared between the two groups.Results:The BCVA, SRF and SFCT in A group were higher than those in B group ( P<0.05). FFA in group A showed that the leakage of ink diffusion or smoke type fluorescein leakage. FFA in group B showed that the fluorescein leakage was single or multiple in macular area, and atrophy of retinal pigment epithelium (RPE) cells was observed. OCTA of group A showed that the choroidal capillary layer showed high blood flow signal, low blood flow signal around the outside, or coarse granular high signal, and no choroidal neovascularization (CNV) was found. OCTA in group B showed that the image of choroidal capillary layer was similar to that of group A, but in group B, 2 eyes (2/12) had CNV with obvious morphology. Conclusions:Clinical characteristics of acute and chronic central serous chorioretinopathy are different. OCTA has definite application value in follow-up and detection of CNV.

Objective: Studying the affect of chronic stress to ultrastructure and the expression of vascular cell adherence molecules-1 in thoracic aorta in rats. Methods: 24 male Wistar rats are divided into two groups randomly, including the test group and control group. The number of each group is 12. The test group are compelled to receive a series of stimulation after they are fed 1 week in quiet surrounding, but the control group are not. All of rats are executed at the same time. After have drawn the materials from the thoracic aorta, we observe the ultrastructure by electronic microscope and the expression of vascular cell adherence molecules-1 by the method immunohistochemistry in thoracic aorta. Results: ①The ultrastructures in test group are changed, but the control group are normal; the modification is concentrated on the obvious swelling, vacuolicitical transformation in mitochondria, but the control are not. ②The VCAM-1 is highly expressed in thoracic aorta of the test group, there is not in control group(P
3.Diagnosis of congenital anomoly of pediatric kidneys by high frequency ultrasound

Objective To discuss the significance of high frequency ultrasound for diagnosis of congenital anomoly of pediatric kidneys. Methods Totally 118 patients with congenital anomoly of pediatric kidneys were examined with high frequency ultrasound,the characteristics of the images were analysed.Results Congenital anomoly kidneys had great diversity of variety in number,size,position,axial direction,abnormal vein and renal pelvis. In the 118 cases,pelvi ureteric stenosis was in 72 cases,renal duplication in 23,renal hypoplasia in 10,kidney deficit in 7,rotatory anomaly in 1,fused kidney in 1,and double renal pelvis in 4. Conclusions High frequency ultrasound is beneficial for diagnosing and differential diagnosing congenital anomoly of pediatric kidneys.
4.The efficacy of cognitive-behavioral intervention on anxiety and oral health-related quality of life in adult orthodontic patients

Objective To explore the influence of cognitive-behavioral intervention on the anxiety and oral health-related quality of life in adult orthodontic patients.Methods 152 adult orthodontic patients whose MDAS scores were greater than 11 were divided into the intervention group and the control group according Multistage Grouping Process.The former were treated with MBT appliance combined with cognitive-behavioral intervention,and the latter were treated with MBT appliance only.The State-Trait Anxiety Inventory(STAI) was adopted to evaluate anxiety in all patients during orthodontic treatment,and the UK oral health-related quality of life questionnaire(OHRQoL-UK) was adopted to evaluate the oral health-related quality of life in all patients before and after orthodontic treatment,respectively.Results 1.After 3 and 6 months cognitive-behavioral intervention,the scores of SAI in the intervention group(50.37±9.18,35.65±8.92) were significantly reduced compared to the control group(56.62± 10.52,54.13± 10.03) (P<0.01).After 6 months cognitive-behavioral intervention,the scores of TAI in the intervention group(47.92±6.82) were significantly reduced compared to the control group(51.52±7.41) (P <0.05).2.After the orthodontic treatment,the scores of OHRQoL-UK in the intervention group(58.27±13.43) were significantly improved compared to the control group (51.99± 11.69) (P< 0.01).3.The oral health-related quality of life after the orthodontic treatment were significantly related to post-treatment anxiety of adult orthodontic patients(r=-0.192 ~-0.459,P< 0.05).Conclusion Cognitive-behavioral intervention can effectively reduce anxiety and improve the oral health-related life quality in the adult orthodontic patients.
5.Effects of ascites derived exosomes on the stemness and invasion ability of ovarian cancer stem-like cell

Objective:To investigate the effects of ovarian cancer ascites-derived exosomes on the stem cell properties and invasion ability of ovarian cancer stem-like cell (OCS-LC).Methods:(1) A2780 cells were induced into OCS-LC in serum-free medium, and authenticating their stem-like properties by sphere-forming test, differentiation test and CD 133 marker detection. (2) Exosomes from ascites and A2780 cell were extracted by ultracentrifugation, then authenticating them. The morphology of exosomes was observed by the transmission electron microscope, exosome particle size was measured by nanoparticle tracking analysis (NTA). The expressions of heat shock protein 70 (HSP-70), CD 63 and CD 9 were detected by western blot. (3) The exosomes from ovarian cancer ascites and tumor cell supernate were co-cultured with OCS-LC. The groups were divided into control group, ascites-derived exosomes (ADE) group (ADE+OCS-LC group), and cells-derived exosomes (CDE) group (CDE+OCS-LC group). The sphere-forming ability was evaluated by sphere-forming cycle, maximum sphere diameter and sphere-forming rate of each group; the expression of CD 133 was detected by immunofluorescence staining under microscope; quantitative real-time (qRT)-PCR was used to detected the expression levels of octamer-4 (Oct-4), Nanog mRNA of the signature genes in the stem cells of each group; the metastasis ability of each group was measured by transwell assay. Results:(1) Identification of OCS-LC: sphere-forming experiment showed that the suspension of OCSC single cells was grown in serum-free medium in secondary sphere-forming. Differentiation function experiment showed that OCS-LC were differentiated into adherent A2780 cells by changing the growth mode in serum containing medium. Flow cytometry showed that the proportion of CD 133 positive ( CD133+) cells in OCS-LC group was (18.9±0.9)%, significantly higher than that of control group (0.6±0.5)% ( t=38.570, P<0.01). (2) Under transmission electron microscope, clear lipid bilayer structure was observed in ADE and CDE, and one side presented a concave hemispheric or cup like structure. NTA showed that the diameter of exosomes mainly ranged from 30 to 100 nm, with an average diameter of 67.2 nm. Western blot analysis showed that the specific protein molecules HSP-70, CD 63 and CD 9 were positive. (3) Three groups′ OCS-LC could continue to grow into spheres, and the group of ADE+OCS-LC showed two growth modes, most of the cells continued to grow into spheres, while a small part of cells grew in adherent differentiation. The sphere-forming rate of OCS-LC in the control group, ADE+OCS-LC group, and CDE+OCS-LC group were (1.05±0.20)%, (4.15±0.10)%, and (10.45±0.25)%, the sphere-forming cycle of OCS-LC in the three groups were (15.3±1.5), (10.3±0.6), and (6.7±0.6) days, and the maximum diameters of OCS-LC were (100.3±3.2), (145.2±5.1) and (170.0±2.1) μm, respectively. And the differences were statistically significant (all P<0.05). After co-culture of exosomes with OCS-LC, the sphere-forming ability of cells in the group of CDE+OCS-LC was prior to ADE+OCS-LC group (all P<0.05). Immunofluorescence staining showed that the number of CD 133 green fluorescent chromophore cells in OCS-LC groups [(46.2±2.1)%, (58.4±2.2)%] was significantly higher than that in the control group [(26.6±1.5)%] after the addition of exosomes in co-culture, the positive rate of CD 133 was higher than that in the control group( F=187.588, P<0.05). The qRT-PCR results showed that the expression levels of Oct-4 mRNA in ADE+OCS-LC and CDE+OCS-LC groups were 3.46±0.24, 4.03±0.31, compared with that in control group (1.04±0.12), the differences were statistically significant ( F=134.932, P<0.05). The mRNA expression levels of Nanog were 1.57±0.32, 2.66±0.15, which were significantly higher than that in the control group (1.00±0.07), and the differences were statistically significant ( F=49.329, P<0.05). And the expression of both in CDE+OCS-LC group increased more significantly than ADE+OCS-LC group (all P<0.05). The number of invasive cells in the three groups of OCS-LC were: control group 30±5, ADE+OCS-LC group 102±4, CDE+OCS-LC group 210±7, and there were statistically significant differences among three groups ( F=820.800, P<0.05). Compared with the control group, the number of invaded cells in the co-culture group were significantly increased ( P<0.05), and the CDE+OCS-LC group had the higher cell invasion ability then the ADE+OCS-LC group ( t=23.202, P<0.05). Conclusions:Exosomes derived from ovarian cancer ascites could enhance and maintain the stemness of OSC-LC, and promote the invasion of tumor cells. Moreover, CDE is superior to ADE.
6.Sodium alginate film promotes the regeneration of injured sciatic nerve in rats

BACKGROUND:The sodium alginate film, jointly developed by our research team and the Institute of Polymer Material of Qingdao University, has good biocompatibility and is often used for the preparation of a variety of composite materials. OBJECTIVE: To observe the effect of sodium alginate film wrapping and covering nerve anastomotic stoma on the regeneration of injured sciatic nerve in rats. METHODS: A total of 36 male Wistar rats were randomly divided into two groups after the right sciatic nerve was cut. Control group: The nerve stump was sutured through epineurial end-to-end anastomosis; Experimental group: The nerve stump was sutured through epineurial end-to-end anastomosis, and wrapped with sodium alginate film, forming a regenerative environment for sciatic nerve regeneration. The degradation and absorption of sodium alginate film, as wel as the adhesion at suture site were observed post-operation. Interleukin-2 and interleukin-4 positive expression was detected with immunohistochemistry method. The histological sections were also detected with hematoxylin-eosin staining and osmium tetroxide staining. RESULTS AND CONCLUSION:At 4-6 weeks post-operation, sodium alginate film was mostly degraded and absorbed in the experimental group. The experimental group rats had less local adhesion, slighter infiltration of inflammatory cels, and fewer fiber tissue hyperplasia than control group rats. The contents of interleukin-2 and interleukin-4 were almost the same between the control and experimental groups at 1, 7, 14 days post-operation. At 6 weeks, the regenerated nerve fibers were distributed evenly with uniform shape, the number of nerve fibers, axons and myelin sheath were significantly better than the control group (P < 0.05). Sodium alginate film has good biological degradability and histocompatibility and plays an important role in promoting sciatic nerve regeneration.
7.Effects of the Ligustrazine Injection on Retina Injury Rat Models Induced by Retina Ischemia-reperfusion

Objective To investigate the therapeutic effect of the ligustrazine injection on rats models of experimental high intraocular pressure induced by retina ischemia-reperfusion.Methods Thirty two SD rats were divided into four groups randomly:the ligustrazine group,the model group,the normal group and the vitamine E group.The ischemia-reperfusion model is established by increasing intraocular pressure to 110mmHg in rat eyes.The ligustrazine group was treated with the ligustrazine injection,the vitamine E group with gastric gavage of vitamine E,and the model and normal groups with distilled water.The period of treatment was 7 days.The histopathologic changes were observed under the optical microscope and the electron microscope,meanwhile the SOD and MDA were measured.Results After ischemia for 60 min and reperfusion for 48 h,the cells in retinal ganglion cell layer and inner nuclear layer were thinned under optical microscope,perinuclear karyopyknosis and apoptotic bodies appeared in retinal ganglion cell layer and inner nuclear layer under electron microscope,SOD activity decreased and MDA level increased.After treatment,the morphology of retinal ganglion cell layer and inner nuclear layer was improved obviously,SOD activity increased and MDA level decreased.It is suggested that ligustrazine injection exerts anti-ischemic and anti-hypoxic and cytomembrane stabilizing effects to a certain extent.Conclusion The ligustrazine injection is an effective traditional Chinese medicine in treating retinal ischemia.
8.Role of Cdk5-CRMP pathway in sevoflurane-induced dendritic develop-mental disorder of neurons in prefrontal cortex of neonatal rats

[ABSTRACT]AIM:Toinvestigatetheeffectofsevoflurane(Sevo)onthedendriticdevelopmentinprefrontal cortex ( PFC) of neonatal rats and the role of cyclin dependent kinase 5 ( Cdk5 )-collapsin response mediator protein ( CRMP) pathway in it.METHODS:Eighty-eight postnatal day 7 Sprague Dawley ( SD) rats were randomly divided into 4 groups (n=22):Air+NS group, Air+roscovitine (Ros) group, Sevo+NS group and Sevo+Ros group.The rats in Air+NS group and Air+Ros group were exposed to the air for 4 h, while the rats in the other 2 groups were exposed to 2.8%sevoflurane for 4 h.The rats received intraperitoneal injection of 150μL normal saline 15 min before exposure in the Air+NS group and Sevo+NS group, while the rats in the Air+Ros group and Sevo+Ros group received intraperitoneal injection of 150μL roscovitine ( in DMSO solution, 10 mg/kg) 15 min before exposure.At the end of exposure, the corti-ces of the rat brain were collected and the protein levels of P35, P25, Cdk5, CRMP1, CRMP2, CRMP4 and p-CRMP2 Ser522 in PFC were detected by Western blot.On the postnatal day 30, the rat brains were sectioned for Golgi-Cox staining and morphological analysis of dendrites in the PFC neurons.Open-field test and contextual fear conditioning test were per-formed on postnatal days 25~27 and 31~32, respectively.RESULTS:Compared with Air+NS group, the expression of P35 in the Sevo+NS group was significantly decreased, and the expression of P25 was dramatically increased (P<0.05), whereas roscovitine partly reversed the changes above induced by sevoflurane (P<0.05).The expression of Cdk5 was not significantly different among all groups.Compared with the Air+NS group, the expression of CRMP1, 2, and 4 in the Se-vo+NS group were decreased, and the protein level of p-CRMP2 Ser522/CRMP2 was increased ( P<0.05 ) , whereas roscovitine partly reversed the changes above induced by sevoflurane (P<0.05), except for the expression of CRMP2. Compared with Air+NS group, the total dendrite length, secondary dendritic length and interactions on 60 and 80 μm shells in the Sevo+NS group were decreased (P<0.01), whereas roscovitine partly reversed the changes above induced by sevoflurane (P<0.05).Compared with Air+NS group, the percentage of freezing time in the Sevo+NS group was de-creased (P<0.01), whereas roscovitine partly reversed the changes induced by sevoflurane (P<0.05).No significant difference among groups in the open-field test was observed.CONCLUSION:Sevoflurane exposure disturbed dendritic de-velopment of neurons in PFC, learning and memory ability of neonatal rats, which may be mediated by Cdk5-CRMP path-way.
9.High Frequency Convex Ultrasound Probe in the Diagnosis of Ovarian Tumor in Infants and Young Children

Purpose To explore the value of high frequency convex ultrasound probe in the diagnosis of ovarian tumor in infants and young children. Materials and Methods Sixty-nine infants and young children with ovarian tumor were examined with high frequency convex ultrasound probe, and the ultrasound findings were analyzed. Results Out of 69 cases, 45 had tumors on the right side of ovary and the rest 24 on the left side;27 cases of simple cyst, 24 cases of benign teratoma, 9 cases of borderline teratoma, 3 cases of serous cystadenoma, 3 cases of mucous cystadenoma, 3 cases of yolk sac tumor;24 accompanied with torsion or necrosis. The ultrasound of ovarian tumor presented the following features:polycystic structures were common;the strong echo posterior acoustic shadow was not obvious; no specific manifestation was shown to distinguish benign from malignant tumors. Doppler ultrasound of tissues around the tumor was significant in detecting ovarian tumor torsion and necrosis. Conclusion High frequency convex ultrasound probe can provide clear images and distinct dissection information, which is beneficial in diagnosis and differential diagnosis of ovarian tumor in infants and young children.
10.Effect of combination of bone marrow stromal cells and bcl-2 gene therapy on expression of basic fibroblast growth factor in focal cerebra ischemic rat brain

Objective To investigate the efficacy of the combination of bone marrow stromal cells (BMSCs) and bcl-2 gene in the treatment cerebral ischemia taxi its effect on the expression of basic fibroblast growth factor (bFGF) in rats.Methods Forty Wistar rats were used to establish middle cerebral artery occlusion model. They were randomly divided into 4 groups: saline control, bcl-2, BMSCs and BMSCs + bcl-2 groups (n = 10 in each group). Every group was redivided into 3- and 14-day after reperfusion subgroups (n = 5 in each subgroup). Neurological scores of the experimental rats were assessed. BMSCs were labeled with bromodeoxyuridine (BrdU). The distribution and numbers of BMSCs, the expressions of Bcl-2 and bFGF were detected by immunohistochemistry, and apoptotic cells were detected with TUNEL staining in rat brain. Results The neurological score at day 3 after reperfusion in the BMSCs + bcl-2 group was significantly lower than that in the saline control group (P < 0. 05), and at day 14, it was significantly lower than that in the other 3 groups (all P <0. 05). A large number of BrdU-positive BMSCs were observed in the infarcted hemisphere in the BMSCs + bcl-2 and BMSCs groups. The numbers of BrdU-positive BMSCs at day 3 and 14 after reperfusion in the BMSCs + bcl-2 group were significantly higher than those in the BMSCs group (all P <0. 05). The expressions of Bcl-2 in the infarcted hemisphere at day 3 and 14 after reperfusion in BMSCs +hel-2 group wre significantly higher than those in the other 3 groups (all P <0. 05). The expressions of Bcl-2 at all time points were increased more significantly than those in the other 3 groups (all P <0. 05). The numbers of apoptosis in brain at all time points in the BMSCs + bcl-2 group were decreased more significantly than those in the other 3 groups (all P <0. 05). Conclusions Both BMSCs and bcl-2 genes have the therapeutic effect on cerebral ischemia. The efficacy of combination of both ,of them is significantly superior to monotherapy. They may significantly improve the neurological function and increase the expression of bFGF in rats. Its mechanism may be that bcl-2 genes have inhibited BMSCs apoptosis at the same time of anti-apoptosis in brain.
