Chinese Journal of Pathophysiology 1999;0(09):-
Neuronal growth associated protein (GAP-43) is a membrane bound phosphoprotein found in the axonal growth cones of sprouting central nervous system axons. It is neuron specific and is proposed to play a critical role in growth cone function during development of the nervous system or the regenerative recovery from axon injury. The injury leads to the transfection of fibroblasts with the GAP-43 gene, which result in increasing GAP-43 expression. GAP-43 is expressed at high levels during the course of recovery of brain injury. GAP-43 acts with calmodulin and G protein to regulate metabolic responses that initates axonal growth. It is important to reestablish neuronal structure and axonal sprouting. The level of GAP-43 returns to basal level once sprouting has finished. So more and more researchers attach importance to the study of GAP-43, which will improve the level of treatment with brain injury. [

Background and purpose:Over-expression of Hypoxia-inducible factor-1? in tumors was known to be associated with resistance to radiation, chemotherapy and with the more malignant tumor phenotypes relative to increased invasiveness, metastatic potential.Furthermore, research has shown that HIF-1? over-expression is associated with aberrant P53 accumulation in human tumors. So we investigated the significance of HIF-1? expression and the relationships among expression of HIF-1?,P53 and P-gp in gastric carcinoma. Methods:The expressions of HIF-1?,P53 and P-gp were investigated by immunohistochemistry in 74 specimens of gastric carcinoma. Results:The positive percentage of P53 protein was higher in the group with HIF-1? positive than the one with HIF-1? negative(70.45% vs 30.0%).The positive rate of P-gp protein was also higher in HIF-1? positive group than in HIF-1? negative group(61.36% vs 36.67%),The positive expression of HIF-1? was significantly related to expression of P-53 and P-gp(r_(s)=0.372,0.256).The positive rate of P-gp protein was higher in P53 positive group than in P53 negative group.Spearman rank correlation test showed a positive correlation between P53 expression and P-gp(r_(s)=0.0283,P

Objective:To explore the relationship between serum cortisol and attention deficit hyperactivity disorder (ADHD), and to investigate the application of cortisol in the diagnosis of ADHD, so as to provide clues and theoretical basis for the comprehensive prevention and treatment of ADHD in the future. Methods:Serum cortisol levels were detected in 159 ADHD children [ADHD group, 58 cases of predominately inattentive presentation (ADHD-I), 32 cases of predominately hyperactive/impulsive presentation (ADHD-HI), 69 cases of combined presentation (ADHD-C)], and 58 healthy control children (healthy control group) from July 2018 to June 2019, at Fujian Medical University Union Hospital. The receiver operating characteristic (ROC) was used to evaluate the diagnostic value of serum cortisol levels in ADHD groups.Results:(1) The serum cortisol levels of ADHD-I group[(216.58±70.55) nmol/L], ADHD-HI group[(182.26±51.34) nmol/L]and ADHD-C group[(222.81±75.70) nmol/L]were significantly lower than that of the healthy control group[(344.83±98.17) nmol/L](all P<0.001). The level of cortisol in ADHD-HI group was lower than that in ADHD-I group and ADHD-C group ( P<0.05). (2)According to the ROC analysis of serum cortisol, the area under the ROC curve for the diagnosis of ADHD group was 0.866 (95% CI: 0.814-0.917), the maximum Youden index was 0.583, the corresponding sensitivity was 89.3%, the specificity was 69.0%, and the cut-off was 302.88 nmol/L.When the specificity was 85.0%, 246.13 nmol/L was the diagnostic threshold and its corresponding sensitivity was 71.1%.Under the ROC curve for the diagnosis of ADHD-I group, the area was 0.857 (95% CI: 0.792-0.922), the maximum Youden index was 0.552, the corresponding sensitivity was 69.0%, the specificity was 86.2%, and the cut-off was 243.39 nmol/L.Under the ROC curve for the diagnosis of ADHD-HI group, the area was 0.934 (95% CI: 0.887-0.980), the maximum Youden index was 0.745, the corresponding sensitivity was 96.9%, the specificity was 77.6%, and the cut-off was 261.55 nmol/L.Under the ROC curve for the diagnosis of ADHD-C group, the area was 0.841 (95% CI: 0.774-0.908), the maximum Youden index was 0.559, the corresponding sensitivity was 87.0%, the specificity was 69.0%, and the cut-off was 302.82 nmol/L.In view of parents′ lack of understanding of ADHD behavior, it is suggested that the diagnostic threshold of cortisol level of 246.13 nmol/L should be based on 85.0% specificity principle, combined with behavior verification in clinical practice, so as to improve the diagnostic accuracy. Conclusions:Cortisol levels in ADHD-I, ADHD-HI and ADHD-C groups are lower than that in control group. There is functional impairment of hypothalamic-pituitary-adrenal axis in ADHD children. The level of cortisol has certain accuracy in the diagnosis of simple ADHD and can be used in clinical diagnosis.

Chinese Journal of Clinical and Experimental Pathology 2001;(2):136-138
Purpose To investigate relationship between androgen receptor and apoptotic index and to further understand the tumor biology of prostate cancer. Methods Fifty-six patients with histologically proven prostate cancer and 20 cases with benign prostatic hyperplasia(BPH) were collected. Androgen receptor(AR) were stained by 2H12 monoclonal antibody using immunohistochemical method. The apoptotic index (AI) was determined by the terminal deoxynucleotidyl transterase-mediated dUTP biotin nick end labeling (TUNEL) technique on serial sections of formalin fixed, paraffin embedded tissues. Results AR was not found significantly difference (P>0.05) between prostate cancer and BPH. The association of AR with AI according to Gleason score were not observed in prostate cancer. AI were significantly higher in prostate cancer compared to BPH (P<0.05).AI were also significantly higher in AR-positive prostate cancer than in AR-negative prostate cancer(P<0.05). Conclusion AR expression can induce prostate cancer cells to become apoptosis, and may be a useful predictor in functional classification and endocrine response of prostate cancer.

Objective To explore the association of the Arg64 polymorphism β-3-adrenergic receptor(β3-AR) gene with polycystic ovary syndrome(PCOS)and its characterization with obesity, insulin resistance(IR),blood pressure (BP), blood lipid spectrum. Methods The polymorphism of the β-3-AR gene was determined in 163 patients with PCOS and 100 controls. Body mass index(BMI), waist to hip circumference ratio(WHR), BP were examined. Blood glucose, blood lipid spectrum, insulin were also measured. Insulin resistance index was calculated. Results The frequency of Arg64 allele in PCOS women was significantly higher than that of controls(0.33 vs 0.13 respectively, P<0.05). Fasting insulin, insulin resistance index, WHR in Arg64 group were significantly higher than those of Trp64 group. The frequency of Arg64 allele was positively correlated with FINS, insulin resistance index, WHR. Conclusion The Arg64 mutation in the β-3-adrenergic receptor(β-3-AR) gene might contribute to development of PCOS and IR.
