Objective To explore the influence of IL-2 pretreatment on splenic lymphocyte following mobilization with G-CSF, which may provide a new approach to attenuate acute graft-versus-host disease (GVHD). Methods Splenic cells and na(i)ve CD4+T cells from C57BL/6N mice receiving G-CSF-mobilized were pretreated with IL-2(50 U/ml) , and cocultured with the allogeneic antigens from BALB/c mice. The proliferation responses and the polarization of T cells were determined. C57BL/6N mice were randomly divided into 4 groups: G-CSF + IL-2 group, G-CSF group, IL-2 group , control group. Results Compared with the control group, IL-4 increased obviously while IFN-γ decreased significantly in the group of G-CSF + IL-2. The proliferation responses were also suppressed in vitro. Conclusion The proliferation responses of splenic cells and naive CD4 + T cells from C57BI/6N mice receiving G-CSF-mobilized to the allogeneic antigens were significantly abrogated by the pretreatment with IL-2, T cells were polarized toward the production of type-2 cytokines. The combination of G-CSF and IL-2 is potentially synergetic in the induction of T lymphocyte immune tolerance.

International Journal of Cerebrovascular Diseases 2009;17(5):373-377
Although there are many drugs for the treatment'of cerebral infarction, no satisfactory efficacy has achieved. Mesenehymal stromal cells (MSCs) have multilineage differentiation potential and self-renewing capability, and can differentiate into neurons. The transplanted MSCs can improve neurological deficit after cerebral infarction, promote the recovery of neurological function, and thereby reduce the disability of cerebral infarction through the mechanisms such as releasing nutritional factors and growth nutritional factors, promoting angiogenesis. The use of MSCs for cell transplantation is likely to be an ideal means in the treatment of cerebral infarction.

AIM: To investigate the molecular mechanism of Epstein-Barr virus encoded latent membrane protein 1 regulated cellular proliferation in nasopharyngeal epithelial cells. METHODS: The nasopharyngeal epithelial cells NP69 were infected with RV-pLNSX (the empty vector) and RV-LMP1 retroviruses, respectively. Therefore, the NP69-pLNSX and NP69-LMP1 cell lines were established. Sequentially, cellular proliferation of NP69-pLNSX and NP69-LMP1 cells was compared to draw the cellular growth curve. The experiments of plate clone formation and forming of soft agar colony were conducted. Meanwhile, the differential expression of proteins were identified between NP69-pLNSX and NP69-LMP1 cell lines by proteomic methods, and the expression levels of partial identified proteins were verified. RESULTS: (1) LMP1 was able to accelerate cellular proliferation of nasopharyngeal epithelial cell NP69 (n=3, P<0.05). (2) Twenty two proteins (9 up-and 13 down-regulated) of LMP1 mediated regulation were identified from infected NP69 cell lines, and the differential expression of partial identified proteins was confirmed by Western blotting and fluorescent real-time quantitative RT-PCR. CONCLUSION: LMP1 probably mediates the regulation of vimentin protein and keratin 19 protein expression to promote cellular proliferation in NP69 cells.

BACKGROUND: As unspecific antagonist of opiate receptor, naloxone is widely used for multiple diseases which are related with abnormal release of endogenous opium. At present, researches suggest that large dosage of naloxone is used at early period can decrease death rate of patients with acute craniocerebral injury and promote neural functional recovery.OBJECTIVE: To investigate the effect of naloxone on improving the nervous function of rats with acute craniocerebral injury and to analyze effectively.DESIGN: Randomized grouping design based on the experimental animal.SETTING: Beijing Neurosurgical Institute.MATERIALS: Totally 250 SD rats were divided randomly into 0.3, 1.0,3.0, 9.0 mg/kg naloxone group, positive control group and negative control group.METHODS: Craniocerebral injured model was established with Feenly free fall struck, and the medicine was given 30 minutes after injury. The rats of the first four experimental group were injected transpeniponeally with naloxone hydrochloride by 0.3 mg/kg, 1.0 mg/kg, 3.0 mg/kg and 9.0 mg/kg respectively once a day; meanwhile, the control groups were given 2 mg citicoline sodium for injection and 0.5 mL normal saline per rat respectively. The longest time was 14 days.MAIN OUTCOME MEASURES: MNSS neural functional score was used every day. The brain edemas of 8 rats in each group were measured with wet-dry weight methods on the second and the fourth day after head trauma.RESULTS: Among 250 rats, 172 entered the final analysis. The nervous function of rats in naloxone groups was better than the two control groups (P < 0.01), and that in 1, 3 and 9 mg/kg naloxone group were better than 0.3 mg/kg group (P < 0.05), but there were no significant differences a mong the three naloxone groups (P > 0.05). The brain edemas of rats in naloxone groups were lighter than that in the control groups (P < 0.05), and that of 1, 3 and 9 mg/kg groups were lighter than 0.3 mg/kg (P < 0.05), but there were no significant differences among these three groups (P > 0.05).CONCLUSION: Naloxone can decrease the brain edemas of rats with traumatic brain injury, promote the nervous function recovery, and the treatment effect changes with the dosage during some range. Therefore, the experiment illustrates that naloxone can decrease the brain edemas of experimental brain injury in SD rats and improve the nervous function, but the effect of naloxone is associated with the dosages in some range.
