1.Effect of comprehensive rehabilitation treatment for osteoporotic bulging disc combine diabetes patients

Objective To analysis the effect of using comprehensive rehabilitation treatment for osteoporotic bulging disc combine diabetes otients.Methods Ninety cases osteoporotic bulging disc combine diabetes patients, who treated in Guanganmen Hospital of Chinese Academy of Traditional Chinese Medicine from January 2011 to January 2011, were randomly divided them into observation group and control group, 45 cases in each group, control group was given conventional treatment, the observation group was given comprehensive rehabilitation treatment, compare visual analogue scale (VAS) score and Oswetry functional disability index (ODI) changes in two groups after treatment, and compared the efficient of treatment.Results VAS scores after treatment for one course and 2 courses in observation group were respectively (2.05± 1.31) and (1.26± 1.03) points,ODI score was respectively (26.37±5.10) and (21.82±4.57) points, VAS score in the control group was (5.85±1.38) and (3.29±1.94) points,ODI score was respectively (37.29±6.32) and (28.33±5.30), all obviously improving than before treatment, but compared with control group, the observation group improved more apparent,the difference between groups was statistically significant(P<0.05).Treatment effective rate in observation group was 95.56% (43/45), the control group was 80.00% (36/45), the difference between groups was statistically significant(x2=5.075 ,P<0.05).Conclusion Comprehensive rehabilitation therapy applied in treatment for osteoporotic bulging disc combine diabetes patients can obviously improve the patient's pain symptomsand and the lumbar spine, obtain better treatment effect, worthy of clinical popularization and applications.
2.Community-based intensive health management for type Ⅱ diabetics

Objective To discuss the effectiveness of community-based intensified health management on type Ⅱ diabetes patients.Metbods From June 2010 to December 2011,1 024 type Ⅱ diabetes patients in Mishi Lane community were enrolled and assigned to the intervention group (n=501,obtaining intensive health management) and the control group (n=523,obtaining routine community health management).Fastingbloodglucose (FBG),2-hour postprandial blood glucose (PBG2h),HbA1c,blood pressure (BP),triglyceride (TG) and body mass index (BMI) were tested and recorded before and after the intervention.Student's t test or Chi-square test were used for data analysis.Results Differences of age (t=0.124,P>0.05) and gender (x2=2.0,P>0.05) were not statistically significant (both P>0.05).Following the intensive health management,the control rates of HbA1c,BP,TG and BMI were increased to 75.4%,30.4%,85.6% and 49.0%,respectively (x2 values were 4.54,4.71,6.54 and 3.67; all P<0.05).TG,FBG and PBG2h of the intervention group were (1.5 ± 0.9),(6.2 ± 1.3) and (8.3 ± 2.4) mmol/L,respectively,which were significantly lower than those of the control group (all P<0.05).Conclusion Community-based intensive health management could be effective for diabetes patients.
3.Subtype classification of ceftriaxone sodium and its influence on the quality of product.

Powder X-ray diffraction (PXRD) technology combined with cluster analysis method was used to classify 75 batches of crystalline ceftriaxone sodium into subtypes, the crystalline characteristics of each subtype were measured with scanning electron microscope (SEM). By comparing some parameters of these subtypes correlated to crystallization process of ceftriaxone sodium, such as salification rate, water content in different subtypes, as well as by studying different lattice stabilities, different compatibilities with rubber closures during accelerated stability tests, the key point to improve the quality of domestic ceftriaxone sodium was disclosed. The results of this paper indicated that the fine structure of the products could be controlled well by improving the salification and crystallization process. As a result, the subtype II of ceftriaxone sodium with high stability can be produced.
4.Observational Study of Calcitonin and 1, 25 Vitamin D Preventing Periodontitis-Induced Alveolar Bone Loss in Rats

Objective To evaluate the effects of calcitonin and 1, 25 vitamin D on the bone loss in experimental periodontitis in rats. Methods A total of 125 male Wistar rats were randomly allocated to five groups according to the treatments during 8 weeks: normal control (group A, n=25), periodontitis(group B, n=25), calcitonin (group C, prepared in sterile saline at 2 mg/L, and 2μg/kg was administered daily, s.c. , n=25), 1,25 vitamin D (group D, prepared in corn oil daily at a concentration of 2 mg/L, and 2μg/kg was administered daily, p.o. , n=25), 1,25 vitamin D plus calcitonin (group E, n=25). The experimental model of periodontitis was induced by ligating floss around mandibular first molars with orthodontic wires in B,C,D and E groups. Five rats from each group were sacrificed, and the specimens were prepared at 2, 4, 6 and 8-week. The probing depth (PD) and alveolar bone level were observed. The serum bone-specific alkaline phosphatase (BALP) and osteocalcin (OC) were measured by enzyme-linked immunosorbent assay (ELISA) at 8-week in each group. Results The values of PD were significantly lower after 4, 6 and 8 weeks in E group than those of B, C and D groups (P<0.05). The alveolar bone loss was significantly lower after 6 and 8 weeks in group E compared with that of B, C and D groups (P<0.05). The serum levels of BALP and OC were significantly higher after 8 weeks in E group than those of B, C and D groups (P<0.05). Conclusion The present study suggests that 1, 25 vitamin D and calcitonin can partially inhibit the alveolar bone loss induced by periodontitis. Especially, the application of both is more effective than either drug treatment alone.
5.Knowledge-attitude-practice(KAP)-based health management of type 2 diabetes among community residents

Objective To discuss the effectiveness of knowledge-attitude-practice(KAP)-based health management on type 2 diabetes in community residents.Methods From June 2010 to December 2011,930 type 2 diabetes patients living in Mishi Lane Community were enrolled and assigned to the intervention group(n=494) and the control group(n=436).The KAP mode was used for the intervention group,and the routine management mode was applied to the control group.Fasting blood glucose(FBG),PBG2 h,HbAlc,blood pressure(BP),triglyceride(TG) and body mass index(BMI) were tested and recorded.Student's t test or Chi-square test was used for data analysis.Results Age(t=0.124,P>0.05),gender(x2=2.0,P>0.05) and other demographics showed no statistically significant difference between the two groups(all P>0.05).After one-year KAP-based health management,HbA 1 c of the intervention group reduced to(6.1 ± 1.2)%,FBG and PBG2 h reduced by 1.7 and 3.23 mmol/L,respectively(t=4.926,P<0.05; t=4.306,P<0.05; t=4.523,P< 0.05).Diet,physical exercises,medication,regular inspection,unhealthy habits and questionare test awaveness of the intervention group were improved(86.03%,82.19%,85.63%,70.45% 76.11% and 88.46%,respectively.Drinking,dietary structure,overweight and physical exercises of the intervention group were also improved(14.17%,15.79%,70.65% and 68.83%,respectively; all P<0.001).Conclusion The KAPbased health management is proved to be effective in the control of community diabetic patients.
6.Screening of proteins interacting with the cytoplasmic tail of NECL1 by yeast dual-hybrid system

Objective To screen the proteins interact with cytoplasmic tail of NECL1.Methods The cytoplasmic tail sequence of human NECL1 gene was inserted into the "bait" vector pGBKT7 in frame.The recombinant pGBKT7-NECL1C was then transformed into yeast cells and followed by screening of human fetal brain cDNA library.Then the GST pull down assay was used to confirm the interactions between the proteins.Results Nine different sequences by sequencing the positive clones and five alternative proteins were obtained by the amino acid sequence blast.Using the GST pull down analysis,we confirmed that two of them can interact with the cytoplasmic tail of NECL1 in vitro.Conclusion The alternative proteins obtained with the yeast dual-system may interact with the cytoplasmic tail of NECL1.

8.Optimization of PCR Components and Selection of Its Primers for Detection of Trichomonas Vaginalis

Objective To screen primers used in polymerase chain reaction (PCR) for detecting Trichomonas vaginalis. Methods Three pairs of PCR primer reported in the literatures (TVA5-TVA6, TV1-TV2 and TVK3-TVK7) were screened. For each PCR, four components, including primers, Mg2+, dNTPs and Taq polymerase, were optimized using Taguchi methods to determine the optimal PCR conditions. With the optimal conditions, the sensitivities of three PCR were compared. Vaginal swabs were collected to detect Trichomonas vaginalis by culture and PCR, and the PCR with highest sensitivity was evaluated. Results All three PCR were of high specificity, and the PCR with primers of TVK3-TVK7 had the highest sensitivity. Of 25 clinical vaginal swabs, T. vaginalis was detected in 7 samples by the culture, however, it was detected in 8 samples by the PCR. All culture-positive samples were also positive by PCR. Conclusions The PCR with the primers of TVK3-TVK7 is highly sensitive and specific, which could be useful to detect T. vaginalis in vaginal swab samples.
9.Isolation,cultivation and identification of Long-Evans rat bone mesenchymal stem cells

Objective To isolate and cultivate Long-Evans rat bone mesenchymal stem cells (BMSCs) and identify their biological characteristics. Methods BMSCs were obtained from femurs of Long-Evan rats and dealt with 0.85 NH4Cl. The cell growth curve and cell cycle were measured and surface antigens were detected by flow cytometry. Osteoblast differentiation was studied by Alizarin red staining. Results BMSCs had active proliferative ability. The growth velocity of passage 5 cells was slower than that of passage 3 cells. The cell cycle analysis showed that 93.79 of BMSCs was at G0/G1 phase. BMSCs were positive for CD29 and CD44, but negative for CD45. Alizarin red staining of BMSCs after osteoblast induction was positive. Conclusion Highly purified BMSCs obtained from Long-Evan rats by 0.85 NH4Cl show stable biological properties.
10.Preparation and characterization of polyclonal antibodies against VLDL receptor.

The polyclonal antibodies against VLDL receptor were prepared and identified. Rabbits were immunized with polypeptide fragment of VLDL receptor as antigen. The collected blood serum of the immunized rabbits was analyzed and identified by using ELISA and Western Blot. The results showed that the rabbit against mouse and human VLDL receptor antibodies were obtained with high titer and could recognize the natural VLDL receptors through Western blot. The prepared polyclonal antibodies against VLDL receptor provide a new tool to study the protein of VLDL receptor.
