1.Application of tissue-cultured method on preservation of articular cartilage

[Objective]To study the effects of different storage method on the viability of cheodroeytes so as to find out the idem method for preservation of articular cartilage.[Method]Under the aseptic condition,acquiring 240 pieces of osteoehondral plugs from both condyles of femur and tibial plateau.The control group was not treatment using any preserved means.The tissue-cultured group was to used F12-DMEM medium long-term preserved osteochondral plugs under the conditions of 37 ℃.At preserve 8 weeks,takes the osteochondral section to carry on examination observation ohondrocytes Viability activeness change separately.[Result]The control group chnadrocyte survival rate was 94.4%.At preserves 8 weeks,the cell survival rate of the tissue-cultured group was 76%,the grading-cooling cryopreservation group was 66%,the constant-cooling cryopreservation group only was 41.80%.[Conclusion]Under the condition of 37 ℃,tissue culture method can long term preservation of vital articular cartilage in vitro.The chondrocyte survival rate of the preservation of vital articular cartilage in vitro.The chondrocyte survival rate of the tissue-cultured method obvious higher than constant-cooling cryopreservation method.
2.The clinicopathological significance of transforming growth factor-β1, transforming growth factor-β1 receptor Ⅱ ,Smad4 and Smad7

Objective To investigate the clinicopathological significance of transforming growth factor-β1 (TGF-β1), transforming growth factor-β1 receptor Ⅱ (TGF-βRⅡ ), Smad4 and Smad7 protein expressions in gastric carcinomas. Methods 109 cases of gastric carcinomas,28 of high grade dysplasia,20 of low grade dysplasia,30 of chronic atrophic gastritis,29 of intestinal metaplasia, and 21 of normal gastric mucosa were selected for the detection of the expression level of TGF-β1,TGF-βRⅡ , Smad4 and Smad7 proteins by immunohistochemistry. Results The expression of TGF-β1,TGF-βRⅡ and Smad7 proteins in gastric carcinomas and dysplasia (high and low grade) were significantly higher than those in chronic atrophic gastritis, intestinal metaplasia and normal gastric mucosa (all P <0. 001 ) ,whereas the expression of Smad4 was significantly increased in high and low grade dysplasia (cytoplas-mic IS :4.89±2.38,5.80±1.54, respectively; nuclear IS: 3.89± 1.52,3.80±1.33, respectively), and decreased in gastric carcinomas (cytoplasmic IS:2.41±2.27,nuclear IS:2.02±2. 14) (P <0.001 ,P <0.001 ,respective-ly). Meanwhile,the expressions of TGF-β1,TGF-βRⅡ and Smad7 were higher in advanced gastric cancer (4.36± 2.66,3.05±1.93,4.84±3.06, respectively) than those in early gastric cancer ( 2.93±1.85,2. 17±1.87, 4. 14±2.46, respectively) ( P < 0.01, P < 0.05, P < 0. 05, respectively ) ; however the expression of Smad4 protein was much higher in early gastric cancer (P < 0. 001 ). The expression of Smad4 protein was significantly correlated with tumor size( P < 0.05 ), lymph node metastasis ( P < 0. 001 ), T stage ( P < 0. 001 ) and chnical stage ( P < 0.001) ,and the expressions of TGF-βRⅡ and Smad4 proteins were correlated with patients' 5-year survival rate (P<0.05 ,P < 0.01 ). TGF-β1 showed the positive correlation with TGF-βRⅡ and Smad7 (r1= 0.45 ,P <0. 05; r2 = 0.49, P <0. 05), but negative correlation with Smad4 ( r = - 0.21,P < 0.05 ). Conclusion TGF-β1, TGF-βR Ⅱ, Smad4 and Smad7 protein may play an important role in the carcinogenesis of gastric carcinoma,and smad4 is the key in TGF-β pathway. Also, the detection of these proteins expression can be a useful marker for predicting the gas-tile cancer invasion,metastasis and patients' prognosis.
3.Expression and clinical pathological significance of CD138 in cervical lesion tissues

Objective: To study the expression of CD138 in normal cervical mucosal tissue,cervical intraepithelial neoplasia ( CINⅠ/Ⅱ and CINⅢ) and cervical squamous cell carcinoma.And to investigate the clinical pathologic significance of CD 138 expression with lymph node metastasis , microvascular density ( MVD ) , CD68 expression and clinical stage in cervical squamous cell carcinoma.Methods:To detected the expression of CD 138 in 120 cases of cervical squamous cell carcinoma ,106 cases of CINⅢ,14 cases of CINⅠ/Ⅱand 54 cases of normal cervical mucosa tissues.Results: The expression of CD138 was lowest in cervical cancer tissue,followed by normal cervical mucosal tissue ,CIN I/II and CINIII,that there had significant difference (P<0.001).The expression level of CD138 was higher in without lymph node metastasis group than in lymph node metastasis group ( P<0.05 ).In cervical cancer , the expression level of CD 138 was higher in early stage ( stage 0 andⅠ) than in advanced stage (Ⅱ) ( P<0.001 );and higher in CD68 expression negative group than in positive group (P<0.05).Furthermore,MVD was higher in CD138 expression negative group than in positive group ( P<0.05 ).Conclusion: CD138 play an important role in the process of development of cervical squamous cell carcinoma,especially for the role of lymph node metastasis is more obvious.CD138 could be used as a indicators for determinate the process of progress in cervical squamous cell carcinoma.
4.Survey on Toxoplasma gondii infection among key populations in Wuxi City

Objective To investigate the prevalence and risk factors of Toxoplasma gondii infection among key populations in Wuxi City,Jiangsu Province,so as to provide evidences for developing preventive and control interventions for T. gondii in-fection. Methods A questionnaire was designed to conduct face-to-face interviews to HIV/AIDS carriers/patients,tumor pa-tients,pregnant women and pet breeders. The blood samples were collected to test anti-Toxoplasma IgM and IgG antibodies by using ELISA method. Results Totally 404 participants were investigated,and 38 were positive in testing T. gondii antibodies (9.4%),and among these,37(97.4%)were IgG positive and 1(2.6%)was IgM positive. The multivariate logistic regression indicated that age,regularly contacting cats/dogs and being HIV positive were associated with the infection of T. gondii(OR=2.1,2.6 and 2.3,all P < 0.05). Breeding pets,being HIV positive and being tumor patients were more likely to infect T. gondii than being pregnant(OR = 1.5,8.1 and 2.8,all P < 0.05). Conclusion The high infection rate of T. gondii is observed among key populations in Wuxi City,so that the effective measurements should be taken to enhance the intervention among key popula-tions including immunocompromised people,pet breeders or people having bad dietary habits of eating raw food.
5.Resveratrol ameliorates cerebral ischemia/reperfusion injury by inhibiting endo-plasmic reticulum stress in rats

Objective:To investigate the effect of resveratrol ( Res ) on endoplasmic reticulum stress induced by cerebral ischemia/reperfusion(I/R)injury in rats.Methods:The seventy-two male SD rats were divided randomly into three groups(n=20):sham operation group ( group S ) , I/R group and Res-treatment group ( group R ) .Focal cerebral I/R model was induced by electrocoagulation of left middle cerebral artery and occlusion of bilateral common carotid arteries followed by reperfusion after 30 min.The rats in Res group were treated with Res(50 mg/kg)i.p.7 d before the operation,once a day for 7 d.Neurological deficits were assessed at 24 h post-injury,followed by collecting the brain tissues.Cerebral infarct size was detected by TTC staining,and the water content of brain tissue were measured by wet-dry weight method.The expression of GRP78,p-PERK and CHOP proteins were deter-mined by immunohistochemistry and Western blot analysis.Results:Compared with sham group,the neurological deficit score and the brain water content were significantly increased(P<0.05),cerebral infarct size was enlarged(P<0.05),and the expression of GRP78, p-PERK and CHOP were up-regulated in I/R group(P<0.05).At the corresponding time,compared with I/R group,the neurological deficit score and the brain water content were markedly decreased(P<0.05),cerebral infarct size was smaller(P<0.05),the level of GRP78 was notablely increased(P<0.05),while the expression of p-PERK and CHOP were down-regulated in Res group(P<0.05). Conclusion:Resveratrol plays a protection role in ischemia-reperfusion injury, through inhibiting the endoplasmic reticulum stress in rats.
6.Detection of chromosome aberration and lymphocytes subsets of the population around a nuclear power plant

Objective To study the effect of the long-term operation of a nuclear power plant (NPP) on population health,and to collect the health related data of the population around (NPP).Methods Four research areas,named 0-10 km area,10-20 km area,20-30 km area and control area,were set up interms of the distance from the nuclear power plant.Human blood samples were collected to detect chromosome aberration and lymphocytes subsets.Results The rates of cells with chromosome aberration at 0-10 km area,10-20 km area,20-30 km area and control area were 0.157%,0.167%,0.070% and 0.111%,respectively.The differences of rates in the four areas were not statistically significant.The percentages of T lymphocytes,helper T lymphocytes,suppressor T lymphocytes,B lymphocytes and natural killer lymphocytes were all normal.The difference of percentages among 0-10 km area,10-20 km area,20-30 km area were not statistically significant.Conclusions In the operational condition,less significant impact is caused to the population around nuclear power plant.
7.Effect of matrix metalloproteinase-9 on aquaporin-4 expression in astrocytes after hypoxia and glucose deprivation and its regulatory mechanism

Objective To investigate the effects and regulating mechanisms of matrix metalloproteinase 9 (MMP-9) gene silencing on aquaporin 4(AQP4) expression in astrocytes induced by oxygen-glucose deprivation.Methods Cerebral cortical astrocytes from 2 days newborn SD rats were undergone the primary culture.The ischemic cell model was established by oxygen-glucose deprivation.This experiment were divided into control group,negative control group and MMP-9 gene silencing group.The leakage rate of lactated dehydrogenase (LDH)was detected by chromatoptometry.The MMP-9 gene silencing was carried out by Lentivirus transfection.Real-time fluorescence quantitative PCR and Western blot were used to detect the expression levels of AQP4 and MMP-9.The expressions of PKA,PKC,PKG and CaMK Ⅱ were determined by Western blot.Results Compared with control group,LDH leakage rate was significantly higher in astrocytes induced by oxygen-glucose deprivation(t=13.35,P<0.01).The expression of MMP-9 mRNA in astrocytes in MMP-9 gene silencing group(0.412±0.297) decreased significantly compared with that in negative control group(1.118 ± 0.240) (t =-4.964,P< 0.05).The expression of AQP4 mRNA in astrocytes in MMP-9 gene silencing group(1.002±0.082) decreased significantly compared with that in negative control group(1.442±0.066) (t=-9.886,P<0.01).The expression of AQP4 protein in astrocytes in MMP-9 gene silencing group(0.643±0.036)decreased significantly compared with that in negative control group(1.000± 0.069)(t=-11.073,P<0.01).The expression of PKC protein in astrocytes in MMP-9 gene silencing group (0.198±0.110)decreased significantly compared with that in negative control group (0.980± 0.232) (t =-7.218,P<0.01).The expressions of PKA(t=0.875),PKG(t=0.818) and CaMK Ⅱ (t=0.933) protein had no statistically significant difference between negative control group and MMP-9 gene silencing group(all P>0.05).Conclusion The permeability of astrocytes is increased by oxygen-glucose deprivation.Gene silencing MMP-9 could induce expression of AQP4 mRNA and protein decreased,and MMP-9 may regulate AQP4 expression by regulating PKC activity.
8.Chinese Medicine Regulates Hepatocellular Carcinoma-related Signaling Pathways: A Review

Hepatocellular carcinoma (HCC) is a common tumor in the digestive tract, the formation mechanism of which remains to be fully elucidated. Although surgery, radiation, chemotherapy, targeted therapy, and immunotherapy have achieved significant results in the treatment of HCC, these methods are accompanied by a considerable number of adverse reactions and complications. In recent years, Chinese medicine has shown remarkable efficacy in the treatment of HCC, and both basic experiments and clinical studies have confirmed the effectiveness of Chinese medicine, which exerts therapeutic effects via multiple components and multiple targets. However, the pathogenesis of HCC is exceptionally complex and not fully understood, which means that studies remain to be carried out regarding the specific mechanism of Chinese medicine in preventing and treating HCC. Network pharmacology and molecular biology can be employed to decipher the mechanism of Chinese medicine in the treatment of diseases. Studies have shown that Chinese medicine can regulate various pathways such as the mitogen-activated protein kinase (MAPK), phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt), Hedgehog, Wnt/β-catenin, nuclear factor-κB (NF-κB), Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3), and transforming growth factor-β (TGF-β)/Smad signaling pathways. Chinese medicine can exhibit its anti-HCC effects by inducing cell apoptosis, inhibiting cell proliferation and migration, and blocking the cell cycle via the above pathways. However, the specific mechanisms remain to be systematically studied. This study comprehensively reviews the regulatory effects of Chinese medicine on HCC-related signaling pathways to reveal the molecular mechanisms of Chinese medicine in the treatment of HCC. This view holds the promise of providing new targets, new perspectives, and new therapies for HCC treatment and advancing the modernization and development of Chinese medicine.
