PTEN is a candidate tumor suppressor which has sequence homology with dual-specificity phosphatase. PTEN is a multifunctional protein endowed with a phosphatase activity capable of dephosphorylating both tyrosine phosphate, serine/threonine phosphate residues on proteins and phospholipids of the phosphatidylinositol pathway. PTEN appears to be mutated at considerable frequency in several types of human tumors, including those from brain, breast, endometrium, and prostate. PTEN play an important role in pathogenesis of tumor, tumor cell invasion and metastasis. In this review, we will discuss the chemical structure of PTEN, its phosphatase activity, the ability of affecting signal transduction, and its mutational status in cancer.
2.Changes of aquaporin-4 expressions in experimental rats after intracerebral hemorrhage

Objective To study the relationship between the expression of aquaporin-4 (AQP4) and cerebral edema formation during the pathologic course of hemorrhagic cerebral edema. Methods The intracerebral hemorrhage (ICH) was established by infusing collagenase into rat caudate nucleus. In situ hybridization and immunohistochemistry were respectively used to evaluate AQP4 mRNA and protein expression, and electronic microscopy and microangium perfusion were respectively used to observe the changes of the ultrastructural and local capillaries in perihematomal brain edema. Results As compared with the control group, there appeared a significant increase in AQP4 mRNA and protein expression on the edematous tissue at 6 hours in rats following ICH. Objective To observe the distribution of aquaporin-4(AQP*!4) in brain tissue of intracerebral hemorrhagic rats, and its role in hemorrhagic brain edema.Methods The model of intracerebral hemorrhage(ICH) was made by infusing collagenase into rat caudate nucleuses, the immunohistochemical method was used to observe the expression of AQP*!4 in brain tissue during different time after ICH in rats.Results The expression of AQP*!4 immunoreactive positive cell bodies were observed mainly in the perihematomal brain edema zone and astrocytes of cerebral cortex, periventricular zone, supraoptic nucleus and paraventricular nucleus of ICH rats. AQP*!4 expression level elevated at 6 h and peaked at 72 h after ICH, and it was yet higher than control lever at 1 week after ICH.Conclusion AQP*!4 express increases strongly in the areas of a relevant relationship with the water metabolism. It can be concluded that AQP*!4 plays an important role in the development of brain edema after hemorrhage.
4.Detection of Panton-Valentine Leukocidin Gene and Analysis of Homology in 50 Meticillin-resistant Staphylococcus aureus Isolates

OBJECTIVE To investigate the antibiotic resistance pattern of MRSA,homology and carriage of Panton-valentine leukocidin(PVL) in 50 meticillin-resistant Staphylococcus aureus isolates to guide the clinical treatment.Results of homology provide the proof for molecular epidemiology.METHODS According to CLSI′s guideline,antimicrobial susceptibility tests were performed with disk diffusion method.PVL gene and mecA gene were detected by PCR.The homology was analyzed by repetitive estra-genic palindromic elements(Rep)-PCR method.RESULTS MecA gene was detected in all 50 MRSA isolates.All isolates MRSA were resistant to oxacillin and cefoxitin.But they were all sensitive to vancomycin and teicoplanin.The rates of sensitivity to clindamycin,ciprofloxacin,piperacillin and sulbactam,SMZ-TMP,gentamicin,cefazolin and erythromycin were 7.5%,1.9%,24.5%,92.5%,20.8%,1.9% and 3.8%,respectively.The resistance rates of MRSA were higher than that of MSSA.Twenty PVL genes were detected from 50 MRSA isolates.CONCLUSIONS The resistance rates of MRSA are higher than MSSA.There is no isolate resistant to vancomycin and teicoplanin.40.0% MRSA carry PVL gene and part isolates have high homology.
5.Clinical characteristics and chemotherapy efficacy in patients with newly diagnosed multiple myeloma:observation of 176 cases

Objective To investigate the clinical characteristics and treatment efficacy in newly diagnosed multiple myeloma (MM). Methods The clinical data and treatment methods of 176 patients with newly diagnosed MM were retrospectively analyzed. Results The most common chief complaint in the patients with MM was bone pain. 26.1 % (46/176) patients once experienced misdiagnosis. The overall response rate (ORR) was 65.9 % (116/176), the ORR of bortezomib group (81.0 %, 34/42) was significantly higher than that of M2 regimen group (61.8 %, 34/55) and VAD regimen group (60.8 %, 48/79) (P<0.05). Between normal renal function subgroup and renal insufficiency subgroup in bortezomib group, the efficacy was no significant difference (P>0.05). The most common adverse reactions included hematologic toxicity, infection and peripheral neurotoxicity. Multiple organ failure was the main cause of death (47.1 %, 16/34). Conclusions The clinical manifestations of MM are complex and diverse, which is easy to be misdiagnosed. Molecular genetic abnormalities are closely related to prognosis. Proteasome inhibitor bortezomib can improve the curative effect and not be affected by renal function. To prolong the survival time of the patients needs to prevent and control the infection and renal insufficiency actively.
6.Reduced expression of SIRT2 in serous ovarian carcinoma promotes cell proliferation, migration and invasion

[ ABSTRACT] AIM:To compare the expression of SIRT2 in ovarian surface epithelial ( OSE) cell line and serous ovarian carcinoma ( SOC) cell lines, and to investigate the effects of SIRT2 on the cell proliferation, migration and inva-sion.METHODS:The expression levels of SIRT2 in the OSE cell line and the SOC cell lines were determined by Western blot.The SIRT2 siRNAs and overexpression construct were designed and verified.Transient transfection of SIRT2 siRNAs or overexpression construct was performed, and the effect of SIRT2 on the cell proliferation, migration and invasion was e-valuated.RESULTS:SIRT2 levels in the 5 strains of SOC cell lines were significantly lower than that in the OSE cell line.SIRT2 knockdown in HOSEpiC cells significantly enhanced the ability of cell colony formation and accelerated the cell growth rate.On the contrary, overexpression of SIRT2 in HO8910 cells dramatically repressed the number of cell colonies and cell activity.SIRT2 significantly changed the ability of ovarian cell migration.Knockdown of SIRT2 facilitated the cell invasion.CONCLUSION:The expression of SIRT2 in the SOC cells is significantly down-regulated.In the OSE cells, SIRT2 acts as a tumor suppressor and mediates the inhibition of cell proliferation, migration and invasion.
7.Clin ical significance of Angptl2 expression in tumor tissue and serum in patients with breast canc-er

Objective To explore Angptl2(angiopoietin-like protein 2)expression in tumor tis-sue and serum in patients with breast cancer(BC). Methods A total of 78 BC patients were enrolled in this study. Immunohistochemistry was used to detect Angptl2 expression in tumor and adjacent normal tis-sues. Preoperative and postoperative serum Angptl2 levels were determined via ELISA. Correlation among Angptl2 expression,clinicopathological factors and progression-free survival(PFS)were further evalua-ted. The receiver operating characteristics(ROC)curve was constructed to describe diagnostic specificity and sensitivity. Results Angptl2 was positively expressed in 64(82. 1% )cases. The rates of high and moderate expression of Angpt l2 in tumor and adjacent tissues were 53. 8%(42 / 78)and 7. 7%(6 / 78), respectively(P ﹤ 0. 001). Moreover,the elevated Angptl2 protein was significantly associated with lymph metastasis and adverse PFS. In addition,serum Angptl2 level in BC patients was significantly higher than that in benign controls[(218. 72 ± 88. 02)vs(80. 88 ± 30. 59)ng/ ml,P ﹤ 0. 01]. The area under the curve(AUC)was 0. 909,which indicated a good function for diagnosis. Postoperative serum Angptl2 level was decreased to(142. 43 ± 60. 29)ng/ ml,but still higher than that in controls(P ﹤ 0. 01). Conclusion The expression of Angptl2 may increase in tumor tissue and serum of BC and it may be a potential tumor marker for diagnosis and prognosis.
8.Relationship between the cardiac markers in ST-segment elevation myocardial infarction and quality of life six months later

Objective To study the relationship between the cardiac markers in ST-segment elevation myocardial infarction (STEMI) and quality of life (QOL) six months later.Methods N-terminal pro-B-type natriuretic peptide (NT-proBNP),cardiac troponin I (cTnI),creatine kinase isoenzyme-MB (CK-MB)and myoglobin(Myo) was tested in 76 patients with STEMI on admission(0 h) and 24 h after admission.QOL six months later was evaluated by using the SF-36 health survey questionnaire.The relationship between the cardiac markers and QOL was analyzed by multivariate linear regression analysis.Results The total SF-36 scores and the scores of physical function,body pain and general health showed negative linear correlation with NT-proBNP on admission (0 h) and 24 h after admission (P < 0.05).The total SF-36 scores and the scores of physical function and body pain showed negative linear correlation with cTnI on admission (0 h)and 24 h after admission (P < 0.05).The total SF-36 scores and the scores of body pain showed negative linear correlation with CK-MB on 24 h after admission (P < 0.05).The scores of vitality showed negative linear correlation with Myo on admission (0 h) (P < 0.05).Conclusion The higher levels of NT-proBNP,cTnl and CK-MB indicate the lower QOL six months later.
9.The clinical effect of high frequency repetitive transcranial magnetic stimulation combined with cognitive intervention for stroke patients with aphasia

Objective: To discuss the applied effect of high frequency repetitive transcranial magnetic stimulation combined with cognitive intervention for stroke patients with aphasia. Methods: 84 stroke patients with aphasia were divided into observation group (42 cases)and control group (42 cases). Patients of control group were treated by routine intervention of language training, while patients of observation group were treated by high frequency repetitive transcranial magnetic stimulation combined with cognitive intervention. A series of evaluated indicators, such as the language function score of Boston diagnostic aphasia examination(BDAE), the anxiety symptom score of Hamilton anxiety scale(HAMA), the quality of life and degree of satisfaction, were adopted to compare the post-treatment effect for the two groups. Results: After the treatment, there were statistical significance in the difference of understanding, retelling ability, reading ability and writing ability about BDAE scores between the two groups (t=3.572, t=4.482, t=6.238, t=3.013; P<0.05), and the HAMA scores of observation group was significant higher than those of control group (t=3.472, P<0.05). Besides, through different interventions, the series of indicators about quality of life, such as physical function, mental function, social function and material function, of observation group were higher than those of control group (t=4.513, t=3.923, t=4.473, t=4.033; P<0.05). And the satisfaction of observation group was significant higher than that of control group (x2=5.233, P<0.05). Conclusion: The clinical effect of high frequency repetitive transcranial magnetic stimulation combined with cognitive intervention for stroke patients with aphasia is favorable. And this method can increase quality of life and satisfaction for patients, and can extremely reduce medical dispute and improve the therapeutic level of hospital for stroke patients with aphasia.
10.Diagnosis value of minimum exercise test of lactic acid and pyruvic acid for mitochondrial myopathy

17.The abnormal results of METLP were found in 6(100%) patients with other myopathy.Most of them showed a high concentration of lactic acid post-aerobic exercise(L2),and sustainable to after rest,or even more higher(L3).3(21.4%)patients without myopathy showed abnormal results of METLP.Conclusions In all the indexes of METLP,the value of L2/P1 is the reliable index for diagnosis mitochondrial myopathy or no-myopathy;the value of L3/L2 is the most important index in differential diagnosis of mitochondrial myopathy and other myopathy.METLP plays a certain role in the diagnosis of the mitochondrial myopathy,but the clinical and pathological data ought to be combined.
