1.Study on the molecular mechanisms of sorafenib inhibiting human peripheral blood T cells

Objective To elucidate the molecular mechanisms of sorafenib inhibitting human pe-ripheral blood T cells. Methods CFSE(5, 6-carboxyfluorescein diacetate succinimidyl ester) proliferation assay and MTS [3-(4, 5-diethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl) -2-( 4-sulfophenyl) -2H-etrazoli-um, inner salt] assay were used to examine the proliferation and the viability of T cells; Annexin V-FITC and PI staining was used to detect apoptosis; Flow cytometry was used to detect the expression of CD25, CD69; Western blot was used to detect the expression of cell cycle proteins; ELISA was used to detect the level of IL-2; Picryl chloride-induced delayed-type hypersensitivity model to be used to for the evaluation of in vivo immunocompetency. Results Sorafenib inhibited proliferation of human peripheral blood T cells in-duced by phytohemagglutinin(PHA) in a dose-dependent manner without inducing their apeptosis. Sorafenib caused human blood T cells arrest in the G_0/G_1 phase of the ceLl cycle. Sorafenib decreased CD25 and CD69 expressions and IL-2 production in human T cells. Sorafenib inhibited picryl chloride-induced delayed-type hypersensitivity in mice. Conclusion Sorafenib could inhibit proliferation and activation of peripheral blood T cells. These finding indicated that long term administration of sorafenib might lead to immunosuppressive effects.
2.Effects of simulated weightlessness on the kinase activity of MEK1 induced by bone morphogenetic protein-2 in rat osteosarcoma cells

BACKGROUND: The mRNA expression of α1 chain of type I collagen (COL- Iα1)in rat osteosarcoma (ROS17/2.8) induced by bone morphogenetic protein-2(BMP-2)was reduced under simulated weightlessness. The protein kinase MEK1 in the signal pathway of mitogen-activated protein kinase (MAPK) plays an important role in the expression of COL- I αl mRNA regulated by BMP-2. But there was no report on the kinase activity of MEK1 under simulated weightlessness.OBJECTIVE: To investigate the effects of simulated weightlessness on the activity of MEK1 induced by BMP-2 in ROS17/2.8 cells.DESIGN: A non-randomly control study was conducted.SETTING: Department of Aerospace Biodynamics, Fourth Military Medical University of Chinese PLAMATERIALS: Rat osteosarcoma osteoblast-like cell METHODS: This experiment was conducted at the Laboratory of Aerospace cells and Molecular Biology Laboratory , Institute of Space Medico-Engineering between August 2002 and January 2003. ROS17/2.8 cells were cultured in 1 G control and rotating clinostat simulated weightlessness for 24 hours, 48 hours and 72 hours. The cells were divided into 7 groups as follows: Group 1 was blank control group in which ROS17/2.8 cells were cultured in 1 G condition without BMP2; Group 2, cells were cultured in 1 G for 24 hours;Group 3,in weightlessness for 24 hours; Group 4, in 1 G for 48 hours; Group 5, in weightlessness for 48 hours;Group 6,in 1 G for 72 hours;Group 7,in weightlessness for 72 hours. Cells in Group 2 to Group 7 were all cultured with BMP-2. BMP-2 (500 mg/L) was added into the medium 1 hou before the culture ended. Then the total protein of cells was extracted and the kinase activity of MEK1 was detected by means of Western Blotting.MAIN OUTCOME MEASURES: The content of total ERK1/2 and phosphated-ERK1/2 (p-ERK1/2) protein in the cells RESULTS: ①Total ERK1/2 expression induced by BMP-2 in ROS17/2.8 cells under simulated weightlessness: The total ERK1/2 expression induced by BMP-2 in cells in all the groups showed no significant differences ② Phosphated-ERK1/2 expression induced by BMP-2 in ROS17/2.8 .cells under simulated weightlessness: There was little protein of p-ERK1/2 in ROS17/2.8 cells in Group 1 cultured without BMP-2 in 1 G for 24 hours. The content of p-ERK1/2 in ROS17/2.8 cells in Group 2 cultured with BMP-2 in 1 G for 24 hours was much more than that in Group 1 (P < 0.01). The level of p-ERK1/2 was much lower in simulated weightlessness groups than that in 1 G control groups at the same time point. In other words, the content of p-ERK1/2 in Groups 3, 5 and 7 was respectively lower than that in Groups 2, 4 and 6 (P < 0.01).The expression of p-ERK1/2 showed-a tendency of gradually decreasing in Groups 3, 5 and 7 with the prolongation of time of simulated weightlessness(P < 0.01 ).CONCLUSION: The kinase activity of MEK1 in MAPK signal pathway induced by BMP-2 is reduced under simulated weightlessness.
3.Therapeutic efficacy observation on needling Yangming method for facial palsy

Objective:To observe the clinical efficacy of Prof. Gao Yu-chun’s needling Yangming method for facial palsy, as well as to inherit and further develop the academic idea.
Methods:A total of 70 cases were randomly allocated into a treatment group (n=35) and a control group (n=35). Cases in the treatment group were treated with needling Yangming method, whereas cases in the control group were treated with traditional acupuncture method. The treatment was done once a day in both groups (6 times a week) for a total of 6 weeks. Then the therapeutic efficacies were evaluated before and after treatment using the House-Brackmann (H-B) scale and scores of signs and symptoms.
Results:After 2 weeks of treatment, patients in the treatment group obtained a better H-B score and recovery rate than those in the control group (P<0.05). After 4 weeks of treatment, the H-B score, scores of signs and symptoms and recovery rate in the treatment group were better than those in the control group (P<0.05,P<0.01,P<0.05). After 6 weeks of treatment, there were no between-group statistical differences in the above aspects (P>0.05).
Conclusion:Needling Yangming method can obtain a faster effect, a higher early-stage recovery rate and shorter course of treatment than traditional acupuncture method.
4.Changes of Hippocampus Somatostatin and Learning Ability in Rats after+Gz Exposure

Objective To investigate changes of learning ability and somatostatin (SS) changes after positive acceleration (+Gz) exposures. Mehtod Eighty male SD rats were randomly divided into 3groups: control group(Con), +6 Gz/3 min group ( +6 Gz), and +10 Gz/3 min group ( +10 Gz),8 rats in each group. Changes of learning ability in rats were observed at 0 d, 2 d, 4 d and 6 d after + Gz exposure. SS in hippocampus was measured by RIA at 0 d, 2 d and 4 d after + Gz exposures ( there were 8 rats every time, in each group). Result In Y-maze test,number of correct response decreased significantly (P <0.01 ), and total reaction time increased significantly(P <0.01 ) in +6Gz and +10 Gz groups as compared with control group; number of correct response and total reaction time in +10 Gz group changed significantly at 0 d(P <0.01 or P <0.05) as compared with +6Gz group. RIA showed that, content of SS in hippocampus declined at 0 d and 2 d(P <0.05 or P <0.01) in +6 Gz and + 10 Gz groups as compared with control group. Conclusion + Gz exposure could impair learning ability of rats, and inhibit expression of SS in hippocampus.
5.Evaluation microscopic observation drug susceptibility assay in detecting mycobacterium tuberculosis in sputum

Objective To study the effect of microscopic observation drug susceptibility assay (MODS)for testing Mycobacteri-um tuberculosis in sputum ,and to evaluate its clinical value .Methods A total of 150 sputum samples were collected from patients with pulmonary tuberculosis ,and Mycobacterium tuberculosis were detected by Lowenstein-Jensen (L-J) method and MODS ,and compared the results of the two methods .Results The concordance rates of the test results of sputum sample between MODS and L-J was 77 .8% .The sensitivity and specificity of MODS were 93 .3% and 70 .0% ,positive predictive value (PPV ) was 60 .8% , negative predictive value (NPV) was 95 .4% ,as well as accuracy by MODS was 77 .8% .Conclusion MODS assay could be used as a rapid tuberculosis detection method ,with rapid ,sensitive ,simple ,and other advantages .
6.Clinical efficacy of 810 nm diode laser in treating keratosis follicularis

Objective To observe the efficacy of 810 nm diode laser therapy for keratosis follicularis.Methods A total of forty-eight patients with keratosis follicularis were treated by 810 nm diode laser,energy range from 9 to 10 J/cm2,frequency 10 Hz,pulse width 400 ms.Treatments were carried out in three times at 8-week intervals,and clinical efficacy was evaluated after third treatment (6 months).Results The total effective rates of keratosis follicularis were 91.7%.With the increase of the number the curative effect were obviously improved.The treatment effective rate was 52.1% (25/48) for the first time.The treatment effective rate was 75.0% (36/48) for the second time.And the third time was 91.7% (44/48).the patients skin texture was obviously improved in the six-month of follow-up except adverse reaction appeared in five patients in the short term.Conclusions 810 nm diode laser is safe and effective for keratosis follicularis.
7.The regulatory effect of 5-aza-2,-deoxycytidine in cervical cancer cells

Objective To discuss the regulatory effect of 5-aza-2 ,-deoxycytidine on P16 and MGMT in cervical cancer cells. Methods After four kinds of cervical cancer cells (HeLa, SiHa, C33A and CaSki) were treated with 5-Aza-dC , MSP was used to detect the methylation variation of P16 and MGMT , and fluorogenic quantitative PCR and Western blot were employed for determination of P16 and MGMT expression. MTT and Annexin V-FITC/PI double staining were adopted for detection of cell proliferation and apoptosis. Results Both P16 and MGMT exhibited methylation in four kinds of cervical cancer cells , and after treatment with 5-Aza-dC ,their methylation levels were reversed. 5-Aza-dC was able to inhibit p16 and MGMT expression in the cervical cancer cells, and can also suppress cell proliferation and promote apoptosis. Conclusions Although methylation of P16 and MGMT are present in cervical cancer cells, their expression level was still high. Therefore, regulation of P16 and MGMT expression may be affected by other factors. 5-Aza-dC can suppress the growth of cervical cancer cells. Although 5-Aza-dC reverse the methylation levels of P16 and MGMT, it inhibits their gene expression. More experiments are needed to verify the hidden reasons and mechanisms.
8.Assessment of atherosclerosis in ApoE knockout mice feeding with western diet using ultrasound biomicroscopy

Objective To evaluate the value of ultrasound biomicroscopy (UBM) in assessment of atherosclerosis in apolipoprotein E (ApoE) knockout mice feeding with western diet.Methods Sixteen ApoE knockout mice in 8 weeks age were selected,then divided into two groups.One group was fed with west diet as high-fat group,and another group was fed with normal diet as control group.Intima-media thickness (IMT) and plaque area in the aortic root were assessed by UBM in two groups after 8 weeks and 16 weeks.And the measurements of UBM were compared with results of histopathology and blood-fat.ResultsThicken wall and plaque could be find in aortic root in control group and high-fat diet group byUBM.IMT and plaque area in high-fat diet group was significantly higher than those of control group ( P < 0.05).The IMT and plaque area in UBM were good correlation with histopathology ( rwas 0.81 and 0.70 respectively).The triglyceride(TC) and total cholesterol in high-fat diet group was significantly higher than those of control group ( P <0.05),and IMT in UBM were increased with the elevated level of TC,there was a positive correlation between IMT and TC( r =0.528).ConclusionsWestern diet can accelerate the process in formation of atherosclerotic plaque in ApoE knockout mice.UBM can be used to observe this prograss noninvasively in vivo mice.
9.Research progress of DNA methylation in idiopathic male infertility

Male infertility is a multifactorial disease, of which the cause of male infertility cannot be determined, which is called idiopathic male infertility, and the incidence rate is gradually rising. Because its cause is unknown, it has become a major problem in the department of reproductive endocrinology. With the in-depth study of epigenetics, the diagnosis and treatment of idiopathic male infertility also has a new direction, especially the important role of DNA methylation in spermatogenesis and embryonic development. More and more gene fragments and loci have been found by scholars, which makes it possible to achieve accurate identification and targeted treatment. This article reviews and summarizes the research progress of DNA methylation related to idiopathic male infertility in recent years, aiming to further elaborate the pathogenesis of idiopathic male infertility and provide new ideas for clinical diagnosis and treatment.
10.Qualitative and Quantitative Analysis of Chest CT Features of Anti-MDA5 Antibody Positive Dermatomyositis

Purpose To observe the CT imaging features of dermatomyosistis patients with anti-melanoma differentiation related genes 5(MDA-5),and to analyze the relationship between CT imaging indicators and short-term prognosis of patients.Materials and Methods Sixty-seven MDA-5-positive dermatomyosistis patients admitted to China-Japan Friendship Hospital from January 2017 to December 2018 were retrospectively included.Prognosis of included patients in one year were followed-up.Analysis of the relationship between chest CT imaging and short-term poor prognosis.Results Among 67 patients,nine patients died in one year.There was a statistically significant difference between the two groups(the death group and the survivor group)in the interstitial lung disease(ILD)imaging types(χ2=0.198,P=0.025)and pulmonary/aortic diameter ratios(U=103.0,P=0.004).CT imaging of dermatomyosistis-ILD with anti-MDA5 antibody positive patients showed organizing pneumonia.Mortality of diffuse alveolar damage type was significantly higher than that of other types.Logistic regression analysis showed that pulmonary/aortic diameter ratio(OR:4.208,P=0.002)was a strong independent risk factor for the death of patients with anti-MDA5 antibody.Conclusion Most patients with anti-MDA5 antibody positive dermatomyositis show ILD,with the main feature of organic pneumonia.Patients with poor prognosis within 1 year have different types of ILD,while the pulmonary/aortic diameter ratio is a strong independent risk factor for the death of dermatomyosistis-ILD with anti-MDA5 antibody positive patients.
