1.Quality standard of Yuquan Micropill

AIM:To establish the quality standard of the composition of Yuquan Micropill(Radix Rehmanniae,Fructus Schisandrae Chinensis,Radix Puerariae lobatae,etc.). METHODS: The composition of Yuquan Micropill were identified by TLC and the content was determined by HPLC.Puerarin could be separated through Diamonsil~(TM) C_(18) column(5 ?m,250 mm?4.6 mm) with methanol-water(1.2∶3 by volume) as a mobile phase.The flow rate was 1.0 mL/min,the detection wavelength was set at 250 nm and the column temperature was maintained at 40 ℃.?-Schizandrin could be separated through Kromasil KR100-3.5 C_(18) column(100 mm?4.6 mm) with methanol-water(3.5∶1 by volume) as a mobile phase.The flow rate was 0.6 mL/min and the detection wavelength was set at 254 nm. RESULTS: The spots in the TLC were clear and could be well separated,the blank and adjuvant contrast could not be interfered.HPLC showed a good peak form,good resolution and could not be interfered by the foreign matter.The average recovery of puerarin was 99.24%,RSD=0.84%,RSD of precision was 0.95%,RSD of reproducibility was 0.86%,RSD of stability was 0.44%.The average recovery of ?-schizandrin was 99.74%,RSD=0.78%,RSD of precision was 1.89%,RSD of reproducibility was 1.76%,RSD of stability was 1.82%. CONCLUSION: The method is simple and has good resolution,strong specificity and good reproducibility,It can be used for the quality control of the composition of Yuquan Micropill.
2.Biomechanics of plastic splint on the treatment of Colles fracture

[Objective]To evaluate the clinical effect of the plastic splint on treatment of Colles fracture.[Method]A total of 105 patients with Colles fracture were collected between September 2002 and August 2008.All patients were perfromed using the plastic splint afrer diaplasis.According to Gartland and Werley scoring syste,the function of wrist was evaluated after 3 months.[Result]There are only 2 cases losing during treatment,103 cases were finished.The average excellent to good case was 90.3%,including 80 cases of being excellent,13 of being good,7 of being fair and 3 of being poor.[Conclusion]The plastic splint is exactly effective to treatment Colles fracture,especially,it could effectively relieve swelling and improve the contour of the wrists.Theses all rely on reasonable biomechanics design.

Objective To study the effect of ultrashortwave irradiation and chest-wall vibration therapy on serum eosinophil cationic protein(ECP)and percentage of eosinophil(EOS%)in the sputum of children with mild to moderate asthma. Methods A total of 68 children with asthma were divided into a control group and a treatment group.The control group WaS treated with conventional treatment only,while the treatment group was given ultrashortwave irradiation and chest-wall vibration therapy in addition to the conventional treatment.The serum ECP,EOS% in induced sputum,FEV1.0%,and PEF% were measured before and after treatment.The relationships among ECP,EOS%,FEV1.0% and PEF% were analyzed.Results FEV1.0% and PEF% were negatively correlated with serum ECP and EOS% in children with asthma.Compared with the control group,ECP and EOS% were significantly reduced after treatment,while FEV1.0% and PEF% were significantly elevated. Conclusion Uhrashortwave irradiation and chest-wall vibration therapy can improve ventilation by ameliorating airway inflammation and obstruction.
4.Effect of adrenomedullin on rat renal tubular epithelial cell apoptosis induced by hypoxia-reoxygenation and its mechanism

Objective To investigate the effect of adrenomedullin on rat renal tubular epithelial cell line (NRK-52E) apoptosis induced by hypoxia-reoxygenation (HR) injury and its mechanism.Methods NRK-52E cells were cultured and randomly allotted to the following 4 groups:control group,HR group,empty plasmid + HR group,ADM + HR group.NRK-52E cells were transfected with pcDNA3.1-myc-his B empty vector or pcDNA3.1-ADM by transfection complex comprising optimal proportion of plasmid and Fugene HD reagents.Cells were counted by trypanblau and cell survival rate was computed.The concentration of lactate dehydrogenase (LDH) was detected by spectrophotometric method to evaluate cell vitality.The apoptotic rate of NRK-52E cells was measured by flow cytometry.The transfer efficiency was detected by immunocytochemistry.The mRNA expressions of ADM,Bax,Bcl-2 and Fas were determined by Semi-quantitative RT-PCR,and active caspase-3,8,9 protein expression were examined by Western blotting.Results Compared with control group,the expression of ADM significantly increased in HR group (P < 0.05).The expression of ADM significantly increased in ADM + HR group than that in empty plasmid + HR group.Compared with control group,in HR group,the living cell counts and cell survival rate significantly decreased; the LDH concentration in media,apoptotic rate and the levels of Bax,Bcl-2,Bax/Bcl-2,Fas,active Caspase -3,8,9 significantly increased (all P < 0.05).Compared with HR group,in ADM + HR group,the living cell counts and cell survival rate significantly increased; the LDH concentration in media,the cell apoptotic rate and the levels of Bax,Bax/Bcl-2,Fas,active Caspase-3,8,9 significantly decreased,while Bcl-2 was promoted (all P < 0.05).The above indexes had no differences between empty plasmid + HR group and HR group (all P > 0.05).Conclusion The increased expression of ADM can inhibit NRK-52E apoptosis induced by HR,and the mechanism might be achieved by inhibiting mitochondrial and death receptor-mediated apoptotic pathways.

We present that the modern basic theory of traditional Chinese medicine is a new science in this article. To demonstrate the problem new discipline faced and needed to be understood, firstly we want to clear the related concepts of discipline and traditional Chinese medicine from an international perspective achieving the original inten-tion of deepening the research field, forming a discipline branch and towards modern teaching. Secondly, we explore the discipline classification system and in which the location of traditional Chinese medicine and theory of traditional Chinese medicine discipline, and then explore the discipline overall regularity of occurrence, development and demise focusing on regularity and evaluation criteria reveal of medicine and traditional Chinese medicine disciplines. On this basis, we demonstrate the discipline property and location of new scientific disciplines of modern basic theo-ry of traditional Chinese medicine, subjects and fields, direction and meaning and then draw a credible conclusion in accordance with deductive reasoning.
6.Application of fMRI Technology in Exploration of Central Brain Localization among Premenstrual Syndrome Patients with Liver-qi Depression in Resting-state

This study was aimed to explore brain regions which were closely related to the disease onset of premenstrual syndrome (PMS) with liver-qi depression. The BOLD-functional magnetic resonance imaging (fMRI) was used in the study. The processing of imaging data was based on the SPM 8 software and the REST software of the matlab platform. Each cluster was more than 389 continuous voxel. The brain region with single voxel of P < 0.05 (corrected) was defined as region with statistical significance. The 2 Sample T-Test was applied in the case group and the control group. The results showed that compared with the normal control group, the frontal lobe, occipital lobe, insula, limbic lobe, basal nuclei, and cingulate gyrus were activated in the PMS with liver-qi depression cases. It was concluded that the disease onset of PMS with liver-qi depression cases was related to brain regions such as frontal lobe, occipital lobe, insula, limbic lobe, basal nuclei, and cingulate gyrus.

8.The expression of protection of telomeres 1 in acute myeloid leukemia and its significance

Objective To analyze the relationship between the expression of protection of telomeres 1 (POT1) and the pathogenesis of acute myeloid leukemia (AML). Methods 62 patients with de novo AML (case group) and 10 patients with iron deficiency anemia (control group) were enrolled in this study. The quantitative real-time polymerase chain reaction (PCR) and Western blot were used to detect the expression of POT1 in AML patients. Results There were 62 de novo AML patients, including 2 cases M1, 14 cases M2, 12 cases M3, 14 cases M4, 17 cases M5, 2 cases M6 and 1 case AML without classification. According to the risk stratification, high risk group (24 cases), medium risk group (22 cases) and low risk group (16 cases) were divided. Compared with that in the controls, POT1 expression levels in patients with AML were significantly decreased both in mRNA and protein level (P< 0.05). The relative expression levels of POT1 mRNA and protein in patients with M2, M4 and M5 were significantly lower than those in the controls (P< 0.05). The expression levels of POT1 in high risk group, medium risk group and low risk group were significantly decreased than those in the controls (P<0.05). Compared with that in the controls, The relative POT1 mRNA expression was significantly decreased in M3 patients (P< 0.05), but not in protein level. POT1 protein expression was showed both in the cytoplasm and nucleus. There was no significant difference of the expression of POT1 protein between cytoplasm and nucleus (P> 0.05). Conclusions POT1 may be involved in the pathogenesis of AML. POT1 protein expresses in both cytoplasm and nucleus, and the regulatory mechanism may be related to the telomere length.
9.Research of the effect in intermedin on cell proliferation repair of renal tubular cell hypoxia-reoxygenation injury

Aim To evaluate the effect of intermedin (IMD)on cell proliferation and regeneration in rat tu-bular epithelial cell line (NRK-52E)that was subjec-ted to hypoxia-reoxygenation (H/R)injury.Methods The NRK-52E cells were divided into control group and three model groups (H/R,H/R +primitive vec-tor,H/R +IMD vector).The content of LDH was de-tected to observe the influence of IMD on H/R injury. The cell proliferation was detected by MTT.The cell cycle was detected by flow cytometry.Real-time PCR and western blotting were used to determine mRNA and protein levels.Results ① In comparison to the con-trol,H/R treatment decreased the cell viability and in-creased LDH activity (P <0.01 );in contrast,com-pared to H/R,IMD treatment ameliorated cell viability (79.1 5 ±1 .421 % vs 61 .22 ±1 .63%,P <0.05)and decreased LDH activities by 33.85% (P <0.01 ).②The proliferation of NRK-52E cells was significantly in-hibited by H/R treatment.In comparison to the con-trol,H/R treatment of NRK-52E cells increased the proportion of cells in the G0 /G1 phase but decreased the proportion of cells in the S and G2 /M phases. Moreover,the over-expression of IMD resulted in S and G2 /Mphase redistribution and the accumulation of G2 /M-phase cells.The real-time PCR and western blotting results indicated that the mRNA and protein expression levels of cyclin D1 ,CDK4 and p57 were increased in H/R-treated cells.IMD further stimulated this up-reg-ulated expression of cyclin D1 ,CDK4 and decreased the expression of p57 in NRK-52E cells.④Cyclin D1 had a predominantly nuclear localization in NRK-52Ecells,although cytoplasmic localization was also ob-served.Conclusion The study shows that the over-expression of IMD may promote renal cell proliferation and regeneration after renal tubular cell H/R injury via the up-regulation of cyclin D1 ,CDK and the down-reg-ulation of p57.
10.Thermotherapy plus chemotherapy in treatment of retroperitoneal lymph node metastasis after radical gastrectomy and sequential radiochemotherapy

Objective To observe the efficacy and toxicity of thermotherapy plus chemotherapy in treatment of retroperitoneal lymph node metastasis after radical gastrectomy and sequential radiochemotherapy. Methods Sixty patients with retroperitoneal lymph node metastasis after radical gastrectomy and sequential radiochemotherapy were randomly divided into of microwave hyperthermia combined with chemotherapy group (experimental group, 30 cases) and chemotherapy group (control group, 30 cases) by using random number table method. The control group: oxaliplatin 130 mg/m2, intravenous drip (2 h), d1; S-1: 80 mg·m-2·d-1, P.O 2 times/d (after breakfast and after dinner), d1-14. 3 weeks was 1 cycle, a total of 4 cycles. The experimental group: chemotherapy on the basis of control group combined with microwave hyperthermia, d1, 8. the efficacy and toxicity of two groups were evaluated. Results The efficacy rate of the experimental group was higher than that of the control group, and the difference was statistically significant [66.7 % (20/30) vs. 33.3 %(10/30), P< 0.05]. The improvement of Karnofsky score in the experimental group was better than that in the control group, and the difference was statistically significant [73.3%(22/30) vs. 23.3%(7/30), P<0.05]. The improvement of pain score in the experimental group was better than that in the control group, and the difference was statistically significant [75.0 % (15/20) vs. 17.6 % (3/17), P< 0.05]. There was no significant difference in gastrointestinal reactions of two groups [30.0%(9/30) vs. 26.7%(8/30), P>0.05]. There was no significant difference in bone marrow suppression of two groups [33.3%(10/30) vs. 30.0 % (9/30), P> 0.05]. Conclusion Microwave thermotherapy plus chemotherapy has a good efficacy for retroperitoneal lymph node metastasis after radical gastrectomy and sequential radiochemotherapy, and the patients can well tolerated, it is worthy of clinical promotion.
