1.Identification of MicroRNA-like Viral Small RNAs from AaeDV.

Several virus families have been shown to encode microRNAs (miRNAs), which have roles in the infection and replication of viruses in host cells. These virus-encoded miRNAs are identified in double-stranded DNA virus (dsDNA virus) and in several RNA virus families, but not in single-stranded DNA virus (ssDNA virus). We used a bioinformatics approach based on VMir, miRNAFold and MaturePred software to predict virus-encoded miRNA-like small RNAs from the genome of a ssDNA virus: Aedes aegypti densovirus (AaeDV). Northern blotting and stem-loop reverse transcription-polymerase chain reaction (RT-PCR) were used to detect predicted small RNAs. A miRNA-like small RNA termed "AaeDVMD" was identified by stem-loop RT-PCR from predicted candidates. This is the first report demonstrating that a ssDNA virus can encode miRNA-like small RNAs. These data will aid further exploration of the interaction between the AaeDV and its mosquito host.

2.Improved Preparation of Pure Alive Eggs of Schistosoma japonicum

To prepare a large amount of pure alive Schistosoma japonicum eggs, rabbit was infected with 2000 cercariae and its liver was taken aseptically 38-45 days after infection and homogenized. The homogenate was screened through different sieves(60, 120, 200, 300, 360 meshes per inch respectively), and washed with 1.2% NaCl. The eggs and leftover were then digested with 0.25% trypsin for 2 hours, sieved over 360 meshes per inch and washed with RPMI 1640 medium. The collected eggs reached to (95.1?6.4)% of live eggs, with a high efficiency.
3.Effect of extrinsic vascular endothelial growth factor on viability of skin flap treated with radiotherapy in rat at different time after operation

Objective To study the effect of vascular endothelial growth factor(VEGF) on the viability of skin flap treated with radiotherapy at different time after operation and clarify its enhancing effect on angiogenesis.Methods Fifty Wistar rat were randomly divided into 3,5,7,9 d groups and control group.A 3 cm?4 cm full thickness dorsal flap with the pedicle remaining attached at the posterior end in rats and were treated with 60Co radiotherapy at the 2nd,4th,and 6th day postoperatively.The total quantity was 12 Gy.In the experiment groups every rat was given VEGF 120 ng on the 3rd,5th,7th and 9th day.The rats in control group were given the same quality normal saline and radiotherapy.Pathological observation,fluorescence staining and sesuccinic dehyrogenase(SDH) assay were used to measure the blood vessel density,blood vessel diameter,microcirculation and cell vitality of skin flap.Results In 5 d and 7 d VEGF group,the number of average blood vessels and cells of the skin falp was increased compared with control group(P
4.Effect of oxalipatin combined with S-1 on related protein expression in advanced gastric cancer

Objective To analyze the clinical effect of neoadjuvant chemotherapy( oxaliplatin combined with S-1) in the treatment of advanced gastric cancer,and to detect its influence on expressions of vascular endothelial growth factor(VEGF) and matrix metalloproteinase-2(MMP-2).Methods A total of 70 patients with advanced gastric cancer were randomly divided into research group ( n=35 ) and control group ( n=35 ) .The patients in the control group were given surgery treatment and general drug chemotherapy,while the patients in the research group were given the neoadjuvant chemotherapy on the basis of the control group.The incidence of cancer recurrence and the expressions of MMP-2,VEGF of the two groups were observed.Results In the research group,the expressions of MMP-2 or VEGF were significantly different before and after chemotherapy(χ2 =10.03,χ2 =9.11,all P<0.01). The 3-year survival rate in the research group(71.43%) was significantly higher than 45.71% in the control group,there was significant difference(χ2 =11.70,P<0.05).Conclusion Neoadjuvant chemotherapy can reduce the expressions of MMP-2 and VEGF in tumor tissues,indicates that it may play an important role in patients with advanced gastric cancer and can be used in clinic.
5.Construction of the Female Subtractive cDNA Library and Screening of the Specific Expressing Genes

Objective To screen the Schistosoma japonicum female specific expressing genes. Methods{S.japonicum} adult worms were collected from the rabbits’ vein after six-week infection by affusing method. The adult worms were stabilized by RNA-later liquid, the male and female worms were carefully separated with nipper. The high quality total RNA was extracted and mRNA was obtained after purification. Double stranded cDNAs were synthesized after reverse transcription. Female subtractive (female as tester, male as driver) and male subtractive (male as tester, female as driver) cDNA libraries were constructed. The differentially expressed genes were further screened by dot-blot hybridization. The clones were selected and sequenced, which showed apparently higher signals when hybridizing with the female subtracting male probes, than those signals when hybridizing with the male subtracting female probes. The homology of these sequences was searched with BLAST program. The semi-quantitative PCR was applied to test the differential gene expression in female and male adult worms. Result Female subtracting male and male subtracting female cDNA libraries were constructed with SSH technique. After dot-blot hybridization, 50 clones were tested to be the potential female differentially expressed genes and were sequenced. 42 expressing sequence tags (ESTs) were received. After bioinformatics analysis, 17 fragments (about 40^5%) showed high identity with the S.japonicum egg-shell protein genes, 17 sequences(about 40^5%) were highly homologous to unknown S.japoniucm genes and partly homologous to female specific 800 protein. 8 fragments (about 19^0%) showed high identity with other S.japonicum unknown genes. The fragments in clones of 577, 579, 668, 695, 720, and 708 were tested by RT-PCR to be the differentially expressed genes in female adult worms using S.japonicum actin gene as the internal standard. These fragments were highly homologous to S.japonicum egg shell protein gene AY222885, AY222895, AB017097, AF519182, M32281, and S.japonicum unknown gene AY813556 respectively. Conclusion SSH is essential to screen the differentially expressed genes of {S.japonicum} female worms. A number of female specific genes have been found by this method.
6.Value of procalcitonin and high sensitivity C-reactive protein in early diagnosis of neonatal septicemia

Objective Too investigate the application value of procalcitonin(PCT)and high sensitivity C-reactive protein(hs-CRP)in early diagnosis of neonatal septicemia and disease condition assessment.Methods 48 patients with neonatal septicemia treated in the hospital from November 2011 to May 2013 were collected.The data of PCT,hs-CRP and blood culture were recorded and performed the comparative analysis with the serum PCT,hs-CRP detection results in contemporaneous 48 neonates without septicemia.Results The serum PCT and hs-CRP was 93.75% and 10.42% in the neonates with septicemia,which were signifi-cantly higher than 79.17% and 50% in the neonates without septicemia(P <0.05),the positive rate had statistical difference be-tween the two groups.Conclusion PCT and hs-CRP have remarkable change in the early stage of neonatal sepsis,the combination detection of serum PCT and hs-CRP can be used as the indicators for early diagnosis of neonatal sepsis,moreover the sensitivity and specificity of PCT for diagnosing neonatal septicemia are higher the those of hs-CRP,their combined detection can provide fast and accurate diagnostic basis for clinic.
7.Effects of Percutaneous Kyphoplasty on Osteoporosis Vertebral Compression Fractures in Elderly

Objective To observe the clinical effect and safety of percutaneous kyphoplasty (PKP) on osteoporosis vertebral compres-sion fractures (OVCF) in elderly. Methods From January, 2014 to May, 2015, 88 patients with OVCF treated with PKP were retrospective studied. Visual Analogue Scale (VAS) and Oswestry Dysfunction Index (ODI) were used to evaluate the pain and quality of life, and the ky-photic angle (Cobb's angle) and anterior vertebral body height were recorded before operation, one week after operation and the last fol-low-up. The cement leakage and its effect on nerve function were recorded. Results Bone cement leakage was found in eleven patients, in which two in pedicle, three in venous leakage, three in intervertebral disc and three in vertebral posterior. Nerve root symptoms appeared in one patient, who was treated with decompression. The scores of VAS and ODI, and the Cobb's angel were significantly lower (t>29.518, P<0.001), and the vertebral body height was significantly higher (t>35.173, P<0.001) one week after operation and the last follow-up than be-fore operation. Conclusion PKP is safe and effective for OVCF in elderly. It can efficiently reduce the pain, correct kyphotic deformity and improve the quality of life.
8.The multiple mediation effect of adult attachment on the relationship between borderline personality disorder and childhood trauma in recruits

Objective To explore the mediation effect of adult attachment on the relationship between childhood trauma and borderline personality disorder(BPD) in recruits.Methods Childhood trauma questionnaire-28 short form (CTQ-SF),adult attachment scale (AAS) and personality diagnostic questionnaire-4+ (PDQ-4+) were administered to 1 750 recruits in the autumn of 2013.Results The scores of physical neglect,emotional neglect,emotional abuse,sexual abuse,physical abuse,attachment anxiety,attachment close and dependence and BPD were 11.05±3.04,10.00±3.54,6.73±2.32,5.92±2.06,5.83± 1.97,3.02 ± ±0.53,3.19±0.44,2.45± 2.04,respectively.BPD had significantly positive correlation with childhood trauma and attachment anxiety (r=0.353,0.284,P< 0.01),and significantly negative correlation with attachment close and dependence(r=-0.198,P<0.01).Childhood trauma,attachment close and dependence and attachment anxiety could positively predict BPD(F=142.172,P<0.01),which could explain 19.6% of the total variation.Structure equation model and Bootstrap test showed that childhood trauma not only directly predict BPD in recruits,but also indirectly predict BPD in recruit through attachment close and dependence and attachment anxiety (x2/df=11.472,RMSEA =0.077,NFI =0.943,RFI =0.901,IFI =0.948,TLI =0.902,CFI =0.948),and the mediational path through attachment close and dependence and attachment anxiety with the effect size were 10.8% and 8.1%,and the total mediational effect size was 18.9%.Conclusion Adult attachment exerts a multiple mediation effect on the relationship between childhood abuse and BPD in recruits.
9.Imaging features of Benign fibrous histiocytoma of bone

Objective To evaluate the imaging features of benign fibrous histiocytoma (BFH).Methods Imaging data were retrospectively collected and reviewed in 11 patients with pathologically proved BFH.Of the 11 patients,X-ray was performed in all patients,MR scans in 6 patients,and CT scans in 4 patients.Results ALL lesions detected were a solitary lesion.The distribution of BFH was in the tibia (n =5 ),femur ( n =3),fibula ( n =1 ),sacrum ( n =1 ),and thoracic vertebrae ( n =1 ).X-ray features included eccentric osteolytic lesions in 7 patients and centric in 2 patients,with clear boundary and thinning of the cortex,and 7 patients with varying degrees of ossified border were found. CT scan shows bone destruction with density similar to soft tissue.The majority of lesions ( n =3 ) were observed in the expanding shell of bone,2 patients in the tibia and 1 patient in the thoracic lesions with cortical bone perforation.The thoracic lesion as soft tissue mass was detected. All of the lesions detected in CT showed no periosteal reaction.In patients with MR images,hypo to isointense signal intensity on T1WI and hyperintense signal intensity on T2WI was found. All lesions on post-contrast T1WI were detected with homogeneous or heterogeneous lesion with moderate or significant enhancement.Conclusion Imaging features were typical for MFH which is useful tool helping correct diagnosis of MFH.
10.Multi-slice spiral CT in the diagnosis of congenital heart diseases associated with tracheobronchial stenosis in children

Objective To explore the diagnostic value of post processing techniques of MSCT for diagnosing congenital heart disease associated with tracheobronchial stenosis in children. Methods Thirty four patients with congenital heart disease complicated by tracheobronchial stenosis were evaluated with MSCT. MPR, CPR, MinIP and VR were performed to show the tracheobronchial morphology. Findings in 43 segments of 32 cases were compared with the findings of surgical operation. Mann-Whitney test was employed to assess the significance of measurement between the post processing techniques and the operation. Results The lenghth of stenotic segments were variable seen at operation, with values between 4-39 mm in trachea, 4-33 mm in main bronchi and 3-12 mm in lobe bronchi, respectively. The biggest difference between the measurement of operation and MSCT was 4 mm (3 segments). Other differences ofstenotic segments were within 3 mm. Six segments exhibitee mild, 16 moderate and 21 severe stenosis at surgery. By contrast, five segments exhibited mild, 17 moderate and 21 severe stenosis by MPR or CPR,2 mild,9 moderate and 32 severe stenosis by MinIP and 4 mild, 11 moderate and 28 sever stenosis by VR,respectively. There was no significant difference in measuring the degree of tracheobronchial stenosis between MPR or CPR, VR and the surgery (Z =-0. 105,- 1. 479;P >0.05), while MinIP frequently overrated the degree of stenosis compared with the measurement at surgery (Z=-2.484,P =0. 013). Conclusion The integrated three dimensional reformations of MSCT scan can accurately evaluate the degree and extent of congenital heart disease associated with tracheobronchial stenosis in children and provide valuable information for clinical management.
