1.Role of Fas overexperssion in reversing cisplatin resistance in stomach cancer cells

AIM: To study the effect of Fas on cisplatin resistance in stomach cancer cells and its possible mechanisms.METHODS: The expression of Fas at mRMA and protein levels in SGC-7901 cells and SGC-7901 /DDP cells was determined by RT-qPCR and Western blot.Fas-containing adenovirus vector was transfected into the SGC-7901 /DDP cells to upregulate Fas expression.The cell viability was detected by CCK-8 assay.The cell cycle and cell apoptosis were analyzed by flow cytometry.The protein levels of Fas, P38 /p-P38, JNK/p-JNK, cleaved caspase-8/caspase-8 and cleaved caspase-3 /caspase-3 were detected by Western blot.RESULTS: The expression of Fas at both mRNA and protein levels was significantly downregulated in the SGC-7901 /DDP cells.Fas expression was decreased by cisplatin in a dose-dependent manner in the SGC-7901 cells.Overexpression of Fas suppressed the viability and induced apoptosis in the SGC-7901 /DDP cells, and upregulated the protein levels of p-P38, p-JNK, cleaved caspase-8 and cleaved caspase-3.CONCLUSION:Overexpression of Fas increases the sensitivity of the SGC-7901 /DDP cells to cisplatin, and inhibits the cell growth and promotes cell apoptosis.The mechanism may be related to the activation of JNK and P38 pathway.
2.Design and Realization of Hospital Special Medical Information System

Objective To design and realize hospital special information system and solve the information management questions of special diagnosis, such as emergency observation, radiation therapy, hemodialysis, assisted reproductive of bear and rear better children. Methods The main problems of hospital special medical in China were analyzed and defined the basic concept of hospital special medical. The main flow and system of hospital special medical from 13 aspects were introduced. Results The work mode and management methods about special medical were changed and solved the limitations of traditional management. Conclusion The design and realization of hospital special medical information systems can be improved work efficiency, medical quality effectively and the informationization level of the hospital.
3.Effect of nitric oxide on substance P-induced scratching response in BALB/c mice

Objective To investigate the effect of nitric oxide (NO) on the scratching behavior evoked by substance P (SP) in mice. Methods Forty BALB/c mice were randomly divided into 5 groups to receive intradermal injection of different doses of SP (0, 20, 40, 80, 160 nmol/site) into the rostral part of the back to establish the acute itch model. Another 40 mice were randomly allocated to model, spantide, L-arginine,L-NAME and aminognanidine groups injected intracutaneously with normal saline (NS), spantide, L-arginine,L-NAME and aminoguanidine, respectively, 10 minutes before SP (80 nmol/site) injection. Subsequently,the scratching behavior was observed, iNOS expression and NO level in the injected skin were detected by immunohistochemical staining and nitrate reductase assay, respectively. Results The scratching bouts per hour induced by intradermal NS and SP of 20, 40, 80 and 160 nmol/site were 4.38±4.07, 5.38±3.78,12.75±6.52, 23.50±7.84 and 42.38±15.84, respectively, and only SP at higher doses (40 - 160 nmol/site)elicited a dose-dependent scratching response in mice (P < 0.01 or 0.05) compared with NS. The scratches over 1 hour in SP, L-arginine, spantide, L-NAME and aminognanidine group were 67.13±32.79, 70.75±34.80, 10.75±8.14, 29.00±21.19 and 35.38±22.83, respectively; of them, pretreatment with spantide,L-NAME and aminognanidine significantly inhibited SP-induced scratching (P < 0.01 or 0.05), but L-arginine showed no inhibitory effect (P > 0.05). Compared with SP, the pretreatment with spantide, L-NAME and aminoguanidine significantly downregulated the iNOS expression and NO content (P < 0.01 or 0.05) in the injected skin other than L-arginine (P > 0.05 ). Conclusion Intradermal SP could increase NO synthesis by neurokinin 1 receptor activation, resulting in the scratching behavior in BALB/c mice.
5.Research on performance evaluation model of hospital information engineer based on analytic hierarchy process and entropy method

Objective:To construct the performance evaluation model of hospital information engineer to solve the scientific and objective problems of performance evaluation of hospital information engineer.Methods:AAccording to the requirements of the hospital for the overall performance objectives for the information department,in accordance with the principle of combining qualitative and quantitative analysis,the performance objectives were quantitatively decomposed into five first-level evaluation indicators of core literacy,skill level,work performance,scientific research and teaching,and major tasks,and 23 second-level indicators were refined.The subjective weighting method of the analytic hierarchy process(AHP)and the objective weighting method of entropy method were used to comprehensively calculate and establish the index weight coefficient,and the performance evaluation model of hospital information engineers was constructed.Five information engineers(marked as A,B,C,D and E)from the 900th Hospital of the Joint Logistics Support Force in 2022 were randomly selected by a simple random sampling method for performance evaluation.By collecting relevant data and using model evaluation calculation,the feasibility of the evaluation model was determined by comparing the annual performance level of engineers.Results:After comprehensive calculation,the comprehensive evaluation scores of five information engineers(A,B,C,D and E)were 87.46,80.39,60.38,76.33 and 70.33,respectively.The top engineers,A and B,with the total score were excellent in the annual performance evaluation of the hospital in 2022,and the information engineer C in the lower ranking was evaluated as qualified in the annual performance evaluation of the hospital in 2022,which were consistent with the results of comprehensive calculation and evaluation.Conclusion:The performance evaluation model of hospital information engineers based on AHP and entropy method can objectively and truly evaluate the performance of hospital information engineers,fully mobilize the enthusiasm and creativity of information engineers,promote the scientific and objective evaluation of the performance of hospital information engineers.It can also support the establishment of the evaluation mechanism and the implementation of incentive measures of hospital information department and ensure the construction requirements of information talent team for high-quality development of hospitals.
6.Application of micro-lectures combined with high simulation human in clinical skills teaching for nursing students

Objective To explore the effect of micro-lectures withadvanced simulation man in improving the practical skills teaching of nursing students, so as to promote the students' post competency. Methods Totally the 186 nursing internswere divided into control group and observation group by random number method with 93 people in each group. The control group used the traditional teaching modein the teaching of practical skills.The observation group used the micro-lectures with high simulation teaching.Comparing the two groups of nursing students comprehensive assessment test simulation results in the theory, skills, scenarios, and nursing students the evaluation of the curriculum. Results The scores of the two groups were all above the qualification line, but the scores of the observation group were significantly higher than those of the control group (P<0.05). The theoretical examinations and the situation simulation comprehensive testswas(77.89 ± 7.79), (75.60 ± 7.92)points in control group, and (93.87 ± 3.90),(92.87 ± 4.08)points in observation group, there was significant difference between two groups (t=17.67, 18.70,all P=0.000). The curriculum evaluation results of improving learning initiative, active curriculum atmosphere, clear operation demonstration, exercise clinical thinking, improve the clinical interest were 81.72%(76/93), 72.04%(67/93), 93.55%(87/93), 60.22%(56/93), 67.74%(63/93)in control group, and 96.77%(90/93), 95.70%(89/93), 100.00%(93/93), 92.47%(86/93), 98.92%(92/93)in observation group, there was significant difference between two groups(χ2=20.39, P=0.016).Conclusions The effect is significant of micro-lectures combined with high simulation of human using in clinical skills teaching. Thismold can conducive to the cultivation of clinical thinking of nursing students, and improve clinical comprehensive ability, and promote the promotion of nursing students post competency.
7. Clinical significance of next-generation sequencing technology for monitoring molecular minimal residual disease in acute myeloid leukemia

Objective:
To investigate the value of next-generation sequencing (NGS) technology in the prognosis monitoring and treatment guidance for molecular minimal residual disease (MRD) in acute myeloid leukemia (AML) patients with complete remission (CR).
Methods:
The clinical data of 68 AML (non-acute promyelocytic leukemia) patients who received gene mutation spectrum by using NGS technology at initial diagnosis and in CR phase in Tangdu Hospital of Air Force Military Medical University from January 2016 to July 2018 were retrospectively analyzed. The recurrence and survival of both molecular MRD positive group and negative group were analyzed and compared, and the value of NGS technology and multiparameter flow cytometry (MFC) were also analyzed in MRD monitoring.
Results:
There were 39 males (57.4%) and 29 females (42.6%) in 68 patients, and the median age was 52 years old (8-82 years old). Molecular MRD positive group included 38 patients, while negative group included 30 patients. Residual mutation gene type in CR phase was most frequently detected in epigenetic regulator gene mutations, such as ASXL1, TET2, DNMT3A and IDH1/IDH2. Statistical analysis showed that the 2-year cumulative recurrence rate (CIR) in the molecular MRD positive group was higher than that in the molecular MRD negative group (86.8% vs. 51.3%;
8.The inhibitory effect of GSDME on tumor progression and its underlying mechanisms in mice

This study aims to explore the role of mouse GSDME in tumor progression and its mechanisms.Wide type and GSDME knockout mice were used to establish tumor models and the tumor growth was monitored;CRISPR-Cas9 was employed to knockout GSDME in MC38 cells.Bone marrow-derived macrophage and dendritic cells were culture in vitro and adoptively transferred into tumor-bearing mice,respectively.Western blotting was conducted to detect protein expression in cells;flow cytometry was used to analysis the immune cell number,CD8+T cell function,and Cleaved Caspase-3 expression in TAM and TADC cells.According to the above experiments,we found that GSDME deficiency promoted tumor progression in mice.GSDME knockout reduced the number of CD8+T cells and decreased IFN-γ and Perforin in CD8+T cells in tumors.GSDME was mainly expressed in macrophages and dendritic cells,and adoptive transferring GSDME deficient macrophages or dendritic cells did not affect tumor progression.However,GSDME deficient mice reduced the ratio of Cleaved Caspase-3 positive macrophages and dendritic cells in tumors,and Cleaved Caspase-3 positive macrophages expressed more iNOS and MHCII,while Cleaved Caspase-3 positive dendritic cells expressed more CD80 and CD86.In general,GSDME increases Cleaved Caspase-3 positive macrophages and dendritic cells that promote CD8+T cell function,thereby inhibiting tumor progression.
