1.ADR Reports in Our Hospital:Analysis of 270 Cases

OBJECTIVE:To investigate the situation of adverse drug reactions(ADR) in our hospital.METHODS:In this retrospective study,a total of 270 ADR cases reported between 2005 and 2007 in our hospital were analyzed statistically in respect of the age and sex of patients,drug categories and clinical manifestations etc.RESULTS:Of the 270 ADR cases,patients aged ≤18 years or ≥50 years took the highest proportion(51.1% for the two age groups).The ADR were chiefly manifested as lesion of skin and its accessories,followed by the reaction of digestive system.90.74% of the ADR cases were induced by injections,and 191 cases(70.74%) were induced by antibiotics.CONCLUSION:ADR worth the highest alert in the clinic.

4.Changes in deep venous thrombosis after thrombolytic anticoagulant therapies in acute pulmonary thromboembolism

Objective To analyze the treatment effect of deep venous thrombosis (DVT) in acute pulmonary thromboembolism (PTE) with thrombolytic and anticoagulant therapy. Methods Post hoc analysis of data from a prospective multicenter randomized control thrombolytic and anticoagulant trial of 516 patients with acute symptomatic PTE from June 2002 to December 2004. Thrombolytic therapy was performed in patients with massive and sub-massive PTE and anticoagulant therapy was given in patients with non-massive PTE. A total of 362 patients that accepted compression uhrasonography (CUS) before and 14 days after treatment constituted this study. Results The ratio of detected DVT by CUS 14 days after treatment was reduction than that before treatment ( x2 = 22. 667, P < 0. 001 ), but 11.6% patients had new or recurrent DVT. The rates of recanalization in thrombolysis group and anticoagulant group were 56. 5% and 47. 8% respectively (x2 = 1. 435 ,P =0. 231 ). The results after three months follow up showed not recovery in 30. 4% DVT patients and new or recurrent DVT in 10. 4% patients. Conclusions The normalization rate of DVT is low during 14 days treatment, and recurrence rate is high. Thrombolysis has no better rate of recanalization than anticoagulant. The prognosis of DVT hasn't improved significantly during short term treatment.
5.β-arrestin1 mediates the effect of MK-801 on levodopa-induced dyskinesia in Parkinson ', s disease

ObjectiveTo investigate the effect of MK-801 on levodopa-induced dyskinesia (LID)in Parkinson' s disease (PD). MethodsRat models ( n = 25) of Parkinsonism related motor complications were established and were randomly divided into levodopa-induced dyskinesia (LID) group (n = 10), MK801 treatment group (n = 10) and PD group (n =5). Another 5 rats were served as control group. The behaviors of LID rats treated with MK-801 were observed. Immunohistochemistry and Western blot analysis were used to determine the expression of β-arrestin1 in the striate of rats.Results After MK-801 treatment, abnormalinvoluntarymovementscores and peakturning weredecreasedin LIDrats.Immunohistochemistry showed that β-arrestin1-positive cells of the lesioned side ((2. 95 ± 0. 44) × 104) in LID rats were decreased compared to the contralateral side ( ( 3.78 ± 0. 37 ) × 104, t = 5. 415, P < 0. 05 ).Western blot showed that the levels of β-arrestinl in PD group ( presented as lesioned side/contralateral side) were ( 81.02% ± 2. 23% ). The levels of β-arrestin1 (64. 88% ± 3. 10% ) were deceased in LID rats compared to PD rats ( t = 9.47, P < 0. 01 ). However, the levels of β-arrestin1 ( 89. 26% ± 1.90% )were increased in MK-801-treated rats (t = 14. 82, P <0. 01). ConclusionsMK-801 reduces LID in PD rats. The beneficial effect of MK-801 may be mediated through the increased expression of β-arrestinl which in turninhibits the overactivation of glutamate receptors.
6.CT Diagnosis of Gastric Fundus and Cardiac Part Lesions

Objective To study the clinical diagnosis value and CT features of gastric fundus and cardiac part lesions.Methods The CT findings and examining method of gastric fundus and cardiac part lesions were analyzed and summarized in 64 cases.Results (1)Use water as oral contrast can show the gastric mucosa clearly,it was a simple,effective way.(2)CT had important significance for stages and infiltration of cardiac cancer.(3)Varix of gastric fundus had three types:pseudotumor,tuber and thinking-wall.(4)The CT characteristic features of leiomyoma were the continuous linear enhancement in mucosa.Conclusion CT is method for gastrointestinal examining,it has special value to the gastric fundus and cardiac part lesions.
7.Market Research and Analysis of Albendazole

OBJECTIVE: To provide information for clinical rational and safe use of albendazole.METHODS: The medical database and network both at home and abroad were searched and the data regarding the research and development,the market information and indications of albendazole were analyzed.RESULTS & CONCLUSIONS: 5 dosage forms of albendazole are available in the market: tablets,capsules,granules,oral liquid,and syrup.The sales amount of albendazole in recent years dominated the first 3 places in domestic antiparasites market.There are a total of 39 domestic manufactures for albendazole,which can be used to treat 21 kinds of human parasitosis.Its clinical application is widespread.However,severe adverse drug reactions such as the rare encephalitis syndrome and allergic shock etc induced by albendazole pose hidden risks for treatment;therefore,management on its use should be strengthened.
8.A rabbit model of hypospadias induced by finasteride

Objective To establish a rabbit model of hypospadias finasteride to further stadying molecular mechanisma of hypospadias etiology. Methods forty pregnant new zealand rabbits were randomly divided into five groups. Finasteride was fed with 0days(group A), 4 days (group B), 5 days (group C), 6 days (group D), 7 days (group E) which began at GD(gestation days) 19, 10 mg?kg-1?d-1. The appearance of penis and the position of urethras orifice were exmamined to observe hypospadias on postnatal day 35; the second observation was performed to observe retained testicle and the Severity of hypospadias on postnatal day 70, all of the male rabbits were anatomized to observe the development of didymus. Results Hypospadias was seen in group A(0)、group B(22.2%)、group C(95.5%)、group D(85.2%)、group E(100%),and Severity of hypospadias can be divided into 3 degrees. Retained testicle was seen in group A(0)、group B(0)、group C(36.4%)、group D(40.7%)、group E(73.3%). Conclusions The experimental model of hypospadias induced by finasteride is stable.
9.Identification of serum protein of prostate cancer with osseous metastasis

Objective To identify the biomarkers which can be used of estimating the biological behaviors of prostate cancer with osseous metastasis by SELDI-TOF-MS. Methods Screening for potential tumor biomarkers of serum samples from 19 prostate cancer patients and 35 prostate cancer patients with osseous metastasis by using the technology of SELDI-TOF-MS and CM 10 protein chips (Ciphergen Inc. USA).The PBS Ⅱ protein chip reader was used to analyze the CM10 protein chips and transform the protein information into the form of spectra. The protein contents of two groups in the same mass-to-charge ratio (M/Z value) were analyzed and preceded the analysis of variance by Biomarker Wizard software. The proteins whose contents in serum were significantly different, which was distinguished the correctly groups by Biomarker Pattern software. Results The contents of 4 proteins in the two groups were significantly different and the M/Z values of these 6 proteins were from 2000 to 20 000. The relative protein content of prostate cancer patients group was higher than that of Prostate Cancer patients with osseous metastasis group at the M/Z value of 2089,4281, 3507 and 4178 [(4.63±8.03) vs (9.88±10.77), (19.78±21.46) vs (26.73±19.41), (5.46±10.14) vs (8.10±8.74), (38.01 ±26.27) vs (45.25±20.40), (P<0.05)]. The relative protein content of prostate cancer patients group was lower than that of prostate cancer patients with osseous metastasis group at the M/Z value of 15 900 and 16 081 [(11.52±16.80) vs (4.84±5.83), (8.55±12.64) vs (3.56±3.90), (P<0.05)]. Conclusion The associated serum protein in prostate cancer with osseous metastasis can be quickly and exactly diagnosed by SELDI-TOF-MS with high sensitivity and specificity. This new method will be widely used in clinical application.
10.Relationship between G protein-coupled receptor kinase 6 and N-methyl-D-aspartate receptor in the mechanism study underlying motor complications in Parkinson's disease

Objective To investigate the relationship between G protein-coupled receptor kinase 6 (GRK6) and N-methyl-D-aspartate (NMDA) receptor in the mechanism study underlying motor complications in Parkinson's disease (PD).Methods The rat models (n= 25) of Parkinsonism related motor complications were established and were randomly divided into levodopa-induced dyskinesia (LID) group (n= 10,intraperitoneal injection of levodopa for 23 d),MK-801 treatment group (n= 10,intraperitoneal injection of MK-801 at day23 after intraperitoneal injection with levodopa for 22 days) and PD group (n= 5,intraperitoneal injection of vitamin C).Another 5 rats were served as controls (sham-operation group).The behavior changes of rats in MK-801 treatment group were observed,and the expression of GRK6 in the striate of rats was detected by immunohistochemistry and Western blot.Results After the chronic treatment with levodopa methyl ester,PD rats displayed abnormal involuntary movements,which was similar to levodopainduced dyskinesia in PD patients.Immunohistochemistry showed that GRK6-positive cells of lesion side were decreased in LID rats as compared with PD rats [(3.23±0.41 ) × 103 vs.(4.81 ± 1.31 ) ×103,P<0.01].Rats in MK-801 treatment group displayed the decreased AIM scores and increased peak rotation,and the increased GRK6-positive cells of lesion side as compared with LID rats (P<0.05).Western blot showed that the levels of GRK6 was 83.7% ±2.1% in PD group (presented as lesion side/unlesion side),76.8% ± 2.2% in LID group and 91.1% ± 2.7% in MK-801 treatment group (intergroups comparison:all P<0.05).These results were in accordance with the results of immunohistochemistry.Conclusions Antagonist of NMDA receptor can be used to reduce the motor complications in rats.It may be due to increased GRK6 which inhibits the overactivation of glutamic acid receptors.
