1.Challenge in prenatal diagnosis of monogenic and microdeletion/microduplication disorders

Along with the development of screening, diagnostic and therapeutic technologies, the spectrum of fetal abnormalities has been constantly expanded. This brings increasing challenges to the clinical diagnosis and treatment, including but not limited to optimizing multidisciplinary cooperation, options of prenatal genetic testing methods, the uncertainty in the transition period of new technology implementation, and the comprehensiveness of genetic counseling before and after testing. We discuss the above issues aiming to meet the dilemma and achieve the leap of fetal medicine to the advanced level through multidisciplinary collaboration resulting in the improvement of diagnosis and treatment efficiency.
2.Expression of placental glucose transporters in pregnant women with abnormal glycometabolism

Objective To investigate the expression of glucose transporters(GLUTs)in placentas of pregnant women with abnormal glyeometabolism,and to explore its effect on glucose transport between mother and fetus and its relation with the birth weight.Methods Placentas of 41 pregnant women with abnormal glycometabolism(7 cases of DM,10 GDM A1,10 GIGT and 14 GDMA2)and 15 normal pregnant women as control were selected.The expression of GLUT1 and GLUT3was detected by immunohistochemistry.The birth weight was measured at delivery.Results GLUT1 was expressed in the syncytiotrophoblasts and cytotrophoblasts,whereas GLUT3 in some endothelial cells.The expressions of GLUT1 and GLUT3 were significantly different among the five groups(P<0.01).Positive correlation was shown between the GLUT1 expression and the birth weight(rs=0.532,P<0.01),but not in GLUT3 expression.Conclusions The expression of GLUT1 and GLUT3 in placentas of pregnancy with abnormal glycometabolism is enhanced,and GLUT1 may play a predominant role in the fetal glucose uptake.
3.HISTOCHEMICAL STUDIES ON BASIC SUBSTANCES AND ENZYMES CONCERNED WITH ENERGY METABOLISM IN THE ENTERIC PLEXUS OF GUINEA PIG

The present study deals with the histochemistry of intramural plexus in stomach, duodenum, ileum, caecum, proximal colon, distal colon and rectum of normal adult guinea pig, including the reaction of Mg~(++)-ATPase, Ca~(++)-ATPase, CCO, SDH, G6PDH, carbohydrate, protein, nucleic acid and lipid in the enteric neurons. The reactions of all the enzymes mentioned above are observed under the light microscope and estimated semiquantitatively, among which, Mg~(++)-ATPase and Ca~(++)-ATPase are measured quantitatively with microphotometer (Leitz MPV II). Both results of light microscope, and microphotometer were analysed statistically. It was found that the above mentioned enzyme activities in the enteric neurons, suggesting that the neurons carry out active metabolism with these concerned enzymes. There were significant differences in some enzyme activities between myenteric plexus and submucous plexus, the former showed higher activity in SDH, while the latter in LDH, G6PDH, and two kinds of ATPase. It is suggested that there are some differences in. the major metabolic manner of carbohydrate, energy metabolism and functional activity between, two plexus. Besides, there were marked difference in enzyme activities among myenteric plexus of different part of digestive tract, the highest activity was showed in duodenum and proximal colon, the lowest was in caecum. These results indicate that enteric neurons differ greatly in functional state, but whether or not the variation of enzyme activities is related with the different types of neurons is unknown.
4.Action of Fufang Danpu Decoction on experimental gastric ulcer

AIM: To study the action of Fufang Danpu Decoction(FFDPD) on experimental gastric ulcer in mice. METHODS: The anti-gastric ulcer action of FFDPD was observed on the gastric ulcer induced by water immersion stress,absolute ethyl alcohol,reserpine and pyloric ligation.The effect on gastric secretion in rats was studied by pyloric ligation.The effect on gastrointestinal motility was observed by gastric emptying in mice. The analgesic effect was tested by the hot-plate test. RESULTS: FFDPD markedly inhibited gastric ulcer induced by water immersion stress,absolute ethyl alcohol,reserpine and pyloric ligation.FFDPD significantly inhibited the secretion of gastric acid and pipsin.FFDPD markedly delayed gastric emptying and enhanced hot pain threshold in mice. CONCLUSION: FFDPD may have anti-gastric ulcer effect,in relation to its inhibition of the secretion of gastric juice,of activity of gastric smooth muscle and analgetic effect.
5.Determination of Emodin in Niuhuang Anti-inflammatory Tablets by HPLC

Objective To establish a HPLC method for the determination of emodin in Niuhuang anti-inflammatory tablets. Method Thermo-Hypersil C18 (250 mm?4.6 mm, 5 ?m) was used, mobile phase was methanol-0.1% phosphoric acid (70∶30), with flow rate of 1.0 mL/min, detection wavelength of 254 nm, and column temperature was 40 ℃. Results There was a good linear relationship of emodin at the range of 0.020 11~0.402 2 ?g. Conclusion The method is simple, accurate, with better separation efficiency, wide linear range and high sensitivity. It can be used for quality control of the preparation.
6.Comparison of the Content of Tanshinone Ⅱ_A in Different Dosage Form of Guanxin Danshen Preparations

OBJECTIVE: To determine and compare the contents of tanshinone Ⅱ A in difference dosage form of Guanxin danshen preparations(tablets,capsules,granules and dripping pills).METHODS: The tanshinone Ⅱ A in different Guanxin danshen preparations was extracted by ultrasonic extraction with methanol as solvent,and the content of tanshinone Ⅱ A was determined by HPLC on Agilent Zorbax C8(150 mm?4.6 mm,5 ?m) column with mobile phase consisted of methanol-water(72∶28)under a detection wavelength of 270 nm.RESULTS: The linear ranges for tanshinone Ⅱ A stood at 2.76~138 ?g?mL-1(r=0.999 9).The average recoveries of Guanxin danshen tablets,Guanxin danshen capsules,Guanxin danshen granules and Guanxin danshen dripping pills were 99.29%,100.37%,100.18% and 98.71%,respectively with RSD at 1.78%,2.10%,1.55%,and 1.09%(all n=6),respectively.CONCLUSION:The content of tanshinone Ⅱ A in differed greatly in different Guanxin danshen preparations;therefore it is urgent to establish the quality standard for Guanxin danshen preparations as soon as possible.
7.Brief Introduction on the Disclosure of FDA Information in US and Its Implication

OBJECTIVE:To provide reference for food & drug regulatory authorities in China to disclose government information.METHODS:In the light of the development and status quo of the disclosure of information in US FDA,the access,content and procedure for the disclosure of information in FDA were discussed.RESULTS & CONCLUSIONS:There are many similarities between US and China in the disclosure of information.In view of the practical situation of governmental information disclosure in food and drug regulatory authorities in China,Chinese government should be able to find out such key issues as the human and financial support is much needed in FDA information disclosure,the disclosure level should be in line with our national condition,a three-dimensional publicity system of government information at different level should be established and emphasis should be laid to the key disclosed information etc based on the research on the development,access,content and procedure of information disclosure in FDA.
8.The differential diagnosis of pulmonary inflammatory mass and peripheral lung cancer by computed tomography

Objective To analyze the CT characteristic of pulmonary inflammatory mass and peripheral lung cancer, and to look for the method of diagnosis solitary pulmonary nodule. Methods 28 patients of pulmonary inflammatory masses and 33 patients of peripheral lung cancers were included in the study,all patients conformed by pathology. The lesions were divided into typical and atypical masses according to morphology. Observing the signs of lesions and using the results ameliorate the morphological classification. Tested the method by ROC. Results According to the standard of morphology, the diagnosis sensitivity, specificity and accuracy of inflammatory mass are 64.3%, 72.7% and 68.9% respectively. The ameliorated sensitivity, specificity and accuracy are 75%, 81.8% and 78 7% respectively. Conclusion The method of making morphology as basement and sign as supplement can improve the sensitivity, specificity and accuracy of diagnosis.
9.Mechanism for baclofen inhibition on quantal glutamate release in spinal dorsal horn neurons

Aim To investigate the inhibition mechanisms of baclofen, a specific GABA B receptor agonist, on quantal glutamate release in the rat spinal dorsal horn neurons.Methods Whole-cell voltage-clamp technique was performed on dorsal horn neurons in rat spinal cord slice to record glutamatergic spontaneous miniature excitatory postsynaptic currents (mEPSCs). Baclofen action on quantal glutamate release was assessed by analyzing the change of mEPESC to baclofen perfusion.Results Baclofen(10 ?mol?L -1,50 s) depressed the frequency, but not amplitude distribution of glutamatergic mEPSCs, indicating baclofen presynaptic depression on glutamate release. The depression on frequency of mEPSCs persisted in Ca 2+-free solution, or in the presence of K + conductance blocker, 4-AP. On the other hand, the depression was occluded by forskolin, an activator of adenylate cyclase, but not protein kinase C (PKC) activator phorbol 12,13-dibutyrate (PDBu). N-ethylmaleimide (NEM), a sulphydryl alkylating agent, which destroys G protein, abolished baclofen depression.Conclusion Not presynaptic K +, Ca 2+ conductance or PKC, but G protein and/or cAMP pathway are involved in the baclofen depression on glutamate release in rat spinal dorsal horn;this depression might contribute to the analgesic action of baclofen at spinal level.
10.Role of PI3K/Akt in endothelial progenitor cells differentiation in vitro

BACKGROUND:The molecule mechanism underlying endothelial progenitor cells(EPCs) differentiation into mature endothelial cells remains poorly understood.It requires for the regulation of the expression of various genes inside cells by various signal pathways,PI3K/Akt maybe included.OBJECTIVE:To investigate the role of PI3K/Akt in EPCs differentiation.DESIGN,TIME AND SETTING:A group control in vitro experiment was performed at the Pathology Laboratory in Shengjing Hospital of China Medical University from September 2007 to March 2008.MATERIALS:Wistar rats aged 4-6 weeks,of either gender.METHODS:EPCs were isolated from rat bone marrow with density gradient centrifugation.Then difference-speed adherence screening method was used to obtain the second time adherent cells,which then were inoculated in culture flask.After 5 days of culture,cells were collected to be indentified with laser confocal microscopy.The differentiating EPCs were those that have positive results of both AC133 and vWF stainings.MAIN OUTCOME MEASURES:The mRNA and the protein expression of AC133,vWF,PI3K and Akt in EPCs were detected with Western blot and reverse transcription-polymerase chain reaction(RT-PCR) after 0,3,7,10 and 14 days of inoculation respectively.RESULTS:According to the RT-PCR and Western blot detection,AC133 showed the strongest expression at day 0,the weaker at day 3 and none at day 7,10 and 14(P
