Cancer Research and Clinic 2013;25(7):493-495
Reverse transcription loop-mediated isothermal amplification(RT-LAMP)is a novel nucleic acid amplification technique which has been mainly used in a variety of viruses and other pathogens of gene detection.Compared with reverse transcriptase polymerase chain reaction (RT-PCR),the RT-LAMP has advantages of reaction simplicity,rapidly,cost-effective,detection sensitivity and specificity.Nowadays,RT-LAMP technology is gradually used in various medical fields, but it is still a new method especially in cancer diagnosis of tumor gene. The article reviews the application of the RT-LAMP technology in the cancer
5.Effects of proliferation of rat hepatic stellate cells and collagen-Ⅰ production by antisense oligodeoxynucleotides against platelet-derived growth factor B-chain
Chinese Journal of Pathophysiology 1986;0(03):-
AIM: To investigate the effect of antisense oligodeoxynucleotides against platelet-derived growth factor B-chain(PDGF-B) in the proliferation of cultured rat hepatic stellate cells and collagen-Ⅰ production.METHODS: The phosphorothioate antisense oligodeoxynucleotides and missense oligodeoxynucleotides were synthesized. The rat hepatic stellate cells(HSCs) were treated with these oligodeoxynucleotides at different concentrations, respectively. The inhibition of the proliferating of cultured rat hepatic stellate cells was assayed by MTT method. The expressions of PDGF-B and collagenⅠ mRNA were detected by means of reverse transcription-polymerase chain reaction(RT-PCR).The expression of endogenous PDGF-B was determined by flow cytometry(FCM).Collagen-Ⅰin the supernatant was determined by ELISA.RESULTS: The results showed that the ASODN against the PDGF-B at a final concentration of 10 ?mol/L inhibited the HSCs proliferation, the expressions of PDGF-B and collagen-ⅠmRNA.FCM and ELISA demonstrated that the expression of endogenous PDGF-B and the production of collagen-Ⅰin HSCs were significantly lower than those in controls, whereas the controls(missense oligodeoxynucleotides) had no effect at the same concentration.CONCLUSION:PDGF-B ASODN inhibits the proliferation of cultured rat hepatic stellate cells, the endogenous expression of PDGF-B and the production of collagen.PDGF-B ASON might be useful for gene therapy in liver fibrosis.

OBJECTIVE: To put forward proposals for improving essential medicines system in China.METHODS: The problems existing in the essential medicine system in China and the successful experience of some other countries were analyzed.RESULTS & CONCLUSION: Considering the current medical economy level and the capacity on research & development(R&D) of drugs,China should draw inspiration from WHO and other developed countries about their successful experiences on essential drugs to improve the screening methods for essential medicines list, assign legal status for essential medicines and build up system of production, procurement and distribution of essential medicines according to the basic national situation of our country and finally establish the essential medicine system suitable to our country's condition.
7.Diagnosis strategy of Guillain-Barré syndrome spectrum disorders
Chinese Journal of Applied Clinical Pediatrics 2016;31(12):884-886
Guillain-Barré syndrome(GBS) has clinical characteristics:flaccid, symmetrical, ascending paralysis.Cranial nerves and respiratory muscle related, albuminocytologic dissociation in cerebrospinal fluid, and electrophysiological changes.GBS was believed to be an autoimmune perineuropathy.Recently, there were more and more reports about GBS spectrum disorders or GBS variant correlated with anti-GQ1b antibody or anti-GM1 IgG antibody et al.The GBS Classification Group presented the new clinical criteria in 2014,to enable neurologists and non-neurologists to diagnose GBS and all its variants using a simple yet all-inclusive classification system.
8.Effects of L-tetrahydropalmatine on hemeoxygenase-1 in cerebral tissue of rats and endogenous carbon monoxide during global cerebral ischemia/reperfusion
Chinese Journal of Integrated Traditional and Western Medicine in Intensive and Critical Care 2006;0(02):-
Objective: To observe the changes of hemeoxygenase-1 (HO-1) activity in cerebral tissue, carboxyhemoglobin (COHb) level in blood of rats during global cerebral ischemia/reperfusion (I/R) and the influence of L-tetrahydropalmatine (L-THP) on them. Methods: Seventy-seven Sprague-Dawley (SD) rats were randomly divided into sham-operated group (S group, n=7), I/R group (I group, n=35) and L-2THP treatment group (T group, n=35). Cerebral I/R model was reproduced in SD rats of I group and T group. HO-1 activity, cyclic GMP (cGMP) in the brain and COHb in blood were evaluated respectively at 1, 3, 12, 24, and 48 hours after global cerebral I/R and compared to those of the S group. Results: ① HOCD*21 activity, COHb content and cGMP level in I group increased as compared with those in S group after global cerebral I/R (all P

10.Expressions of SOCS3 and IL-6 in Triple-negative Breast Cancer and Their Chinical Significance
Acta Medicinae Universitatis Scientiae et Technologiae Huazhong 2015;(1):82-86
Objective To investigate the expressions of SOCS3 and IL‐6 in triple‐negative breast cancer(TNBC)and their clinical significance. Methods The expressions of SOCS3 and IL‐6 in TNBC and normal breast cells were detected by Western blot and fluorescence quantitative PCR.Immunohistochemical staining was used to detect their expressions in 84 TNBCs ,33 breast fibromas and 11 normal breast tissues. The correlation of their expressions in TNBC and the relationship between their expressions and clinical pathological parameters were analyzed as well. Results SOCS3 was not expressed or down‐expressed in TNBC cell lines while IL‐6 was over‐expressed. The positive rate of SOCS3 protein was 32.1% in TNBCs ,63.6% in breast fi‐bromas(P<0.01)and 72.7% in normal breast tissues (P< 0.01);and that of IL‐6 protein was 72.6% in TNBCs ,30.3% in breast fibromas(P< 0.01)and 27.3% in normal breast tissues (P< 0.01).Moreover ,a significant negative correlation was found between the expression of SOCS3 and that of IL‐6 in TNBC(rs = -0.590 ,P<0.01). The expressions of SOCS3 and IL‐6 were markedly associated with tumor size ,lymph node metastasis ,clinical stages and expressions of Ki‐67 and p53(P<0.05) , but not with age ,histological grades or menopausal status(P>0.05).Conclusion The expressions of SOCS3 and IL‐6 are asso‐ciated with the development of TNBC ,suggesting that SOCS3 and IL‐6 can serve as biological markers for evaluating the prog‐nosis ,and therapeutic targets of T NBC.
