1.Effect of scraping therapy on Th1/Th2 balance in rats with lumbar disc herniation

Objective: To observe the effect of scraping therapy (ST) on the immune balance of serum helper T (Th) 1/Th2 cells in autologous nucleus pulposus transplantation non-compressive lumbar disc herniation (LDH) model rats, and explore the immune mechanism of ST in reducing LDH pain. Methods: Seventy-two male Sprague-Dawley (SD) rats were randomly divided into a model group, a sham operation group and a ST group, with 24 rats in each group. Autologous nucleus pulposus transplantation non-compressive LDH models were established in the model group and ST group, while the rats in the sham operation group underwent sham operation without model establishment. On the fifth day after the model was successfully prepared, rats in the ST group received ST, once every other day, 3 times as a course for a total of 3 courses. Six rats in each group were randomly selected to observe their pain thresholds, and peripheral blood of the rats was collected before the first scraping treatment and at the end of the first, second, and third courses of treatment. The serum was isolated and enzyme-linked immunosorbent assay (ELISA) was applied for the detection of rat serum interferon (IFN)-γ, interleukin (IL)-4 and IL-10. Results: Compared with the model group, the pain threshold in LDH rats in the ST group increased (P<0.05), the serum IFN-γ level was significantly reduced (P<0.05), but the levels of IL-4 and IL-10 did not change significantly (both P>0.05). At the end of the second and third courses of treatment, the IFN-γ/IL-4 ratio was negatively correlated with the pain threshold in the rats, and the IFN-γ/IL-4 ratio was significantly reduced in the ST group (P<0.01). Conclusion: ST can help suppress the Th1 immunity in LDH rats triggered by the autologous nucleus pulposus, restore the immune balance of Th1/Th2, and reduce the pain of LDH.
2.Effect of GPS On Stimulating Murine Lymphocyte Proliferation

Glycyrrhiza uralensis Fish Polysacchoride(GPS)was observed to have mitogenic activity on murine spleen cell proliferation in vitro,The results showed that this mitogenic activity of GPS may be directed to B Iymphocytes,i.e.GPS is a new mitogen of murine B lymphocytes.Furthermore,it was found that B lymphocytes stimulated by GPS was M? depedent and T cell-indepedent.On the contrary,antibodies and IL-2 production was inhibited by GPS in vivo.
3.The effect of amygdaloid nuci ei on immunogical function induced by stress

Rat with amputation operation stress possessed a decline of NK activity and splenocyte pro-liferation to mitogen ConA.Amygdaloid nuclei lesion could antagonize the decrease of those ofcell-mediated immunity.Further results showed that serum corticosterone and methionine-enkephahn was high in stress' rats.Amugdaloid Nuclei lesion could antagonize the increase ofserum corticosterone level,but seem no dffeet on serum methionine-enkephalin level,thereforesuggesting that it is related to ratio of corticosterone to enkephalin changed by decrease of corti-costerone that amygdaloid neuclei lesion antagonized the decline of immunogical function inducedby stress.
4.Healing effect analysis of rhegmatogenous retinal detachment in 288 myopic cases

AlM: To understand the prognostic factors of myopic rhegmatogenous retinal detachment ( RRD) surgery, and provide guidance for clinical treatment.
METHODS: This retrospective study included 288 myopic RRD eyes ( excluding trauma, aphakia, congenital diseases, tractional and exudative retinal detachment ) from April 2007 to April 2014 in our department. The surgical anatomic reduction situation was statistically recorded, the effects of hole's size, retinal detachment duration on sight restoration and surgery success rate were analyzed
RESULTS:The statistical results showed that 245 eyes (85. 1%) healed after their first operation, about 95. 1%cases finally healed after their second or third operation. The degree of myopia and aging factor influenced the RRD operation:the higher degree and the older age were, the lower success rates were (P<0. 05). Through surgery, postoperative visual acuity was significantly better than the preoperative ones ( P<0. 01 ). A higher degree of myopia RRD led to poor visual acuity ( P < 0. 01 ). Postoperative visual acuity of 152 eyes ( 52. 9%) were improved, of which 106 eyes' visual acuity was 0. 05 or more.
CONCLUSlON:Surgery is an effective means of treating myopic RRD. The older patients have lower success rates. A higher degree of myopia led lower reset rate after retinal detachment surgery and poor postoperative visual acuity. When treat myopic RRD, we should pay attention to the recovery of visual function.
5.Changes of Nogo-A Expression in Spinal Cord of Rats with Experimental Allergic Encephalomyelitis

Objective To observe the time interval changes of the expression of Nogo-A in the intumescentia lumbalis of rats with experimental allergic encephalomyelitis(EAE),and explore the function of Nogo-A in the happening and development of EAE.Methods One hundred and six Wistar rats(female) were randomly divided into 2 groups:EAE group(70 rats)and control group(36 rats).Female Wistar rats were used to make EAE animal models,immunized by fresh cavia cobaya spinal cord homogenate and complete Freund adjuvant(CFA),at the same time,their blood brain barrier permeability was lifted by using pertussis stock solution.The rats those were coincident with the standard of being picked up were divided into 6 groups randomly according to the time of falling ill.In control group,fresh guinea pig spinal cord homogenate was replaced by sodium chloride,and rats were also divided into 6 groups randomly.Immunohistochemistry SP was used to detect expression of Nogo-A.Amyelination was detected by improved acid fuchsin and azulin myelin staining.Results In the control group,differences of Nogo-A expression in the intumescentia lumbalis were not obvious in each time group.In the EAE group,expression of Nogo-A could be observed in each time group,but their quantities were very different in contrast with those in the control group.One day after the rats had clinic symptoms,the expression of Nogo-A decreased to some extent;the third day,it reached the lowest point,and was significantly lower than that in control group(P

The systemic anaphylaxis murine model was induced by the injection of oval albumin (OVA)in aluminum hydroxide gel adjuvant. The death rate varied with the mouse species: 40-90% in Swiss and 69% in average,no death in C57BL/6 and 615 but some symptoms. developed. The levels of c-fos mRNA in U937 cell line were determined with in situ hybridization.Theresults showed that TPA induced the expression of c-fos gene with a relationship of dosage-ef-fect.It was also proved that GM-CSF,TNF,IL-9,IL-3 and IL-6 increased the expression of c-fos gene.
9.Study on the changes in intestinal phylum firmicutes in elderly patients with type 2 diabetes

Objectives To compare the differences in intestinal phylum firmicutes between elderly patients with type 2 diabetes (T2DM)and the healthy elder people.Methods 37 elderly patients with T2DM and 69 healthy controls in Shaoxin city were recruited.DNA of phylum firmicutes from fecal samples was extracted.The real-time quantitative PCR was used with special primers for bacterial genus including Faecalibacterium prausnitzii,Eubacterium rectale,Clostridium leptum,and Peptostrepyococc.The differences in content of different bacteria between two groups were analyzed and compared.Results In healthy elderly group versus the elder patient with T2DM,the contents of intestinal phylum firmicutes were[(6.22±1.41) × 107 versus(5.41± 1.40) × 107,t=2.83,P=0.006] in Eubacterium rectale,[(7.46 ± 0.98) × 107 versus (6.96 ± 1.40) × 107),t =2.13,P =0.036] in Faecali bacterium prausnitzii,[(7.89±0.89) × 107 versus(7.46±1.11) × 107,t=2.15,P=0.034]in Clostridium leptum,and[(4.86 ± 1.33) × 107 versus (4.21 ± 1.24) × 107,t=2.45,P =0.016] in Peptostrepyococc,which showed that the contents of intestinal phylum firmicutes were less in T2DM group than in healthy elderly group.Conclusions There are some differences in intestinal flora between the elderly patients with T2DM and healthy people.These intestinal flora may play an important role in the development of T2DM.This study may provide new evidences for probiotic treatment of T2DM.
10.Comparison of Two-incision Microinvasive and Modified Harding Approaches for Total Hip Arthroplasty in Aged Patients

Objective To compare the efficacy of two-incision microinvasive and modified Harding approaches for total hip arthroplasty in aged patients.Methods From May 2003 to December 2006,21 old patients(aged 82 on average) received two-incision microinvasive total hip arthroplasty in our hospital.The outcomes of the surgery was compared with those of 39 cases(mean age,83),who underwent the operation through the modified Harding approach.Results No significant difference was observed in intraoperative blood loss between the two groups [(270?94) ml vs(280?107) ml,t=-0.360,P=0.720].On the 2nd day after the surgery,the patients in the two-incision group could raise their legs straight upward without needing help.At day 10,they could raise the legs straight up to a mean of(53?12)?,which was significantly higher than that in the Harding group [(32?16)?,t=5.262,P=0.000].However,the Harris scores were similar in both the groups in 6 months after the operation(80.6?12.6 vs 79.5?13.2,t=0.313,P=0.756).The incidence rates of orthopedics and systemic complications in the two-incision group was not significantly different from those in the Harding group [9.5%(2/21) vs 8.1%(3/37),?2=0.000,P=1.000;19.0%(4/21) vs 37.8%(14/37),?2=2.210,P=0.137].Conclusions The short-term outcomes of microinvasive two-incision total hip arthroplasty is better than that of modified Harding approach in aged patients.However,the long-term results of the two procedures are similar.
