1.Investigation of correlation and mechanism between moderate hypoxia and tumor malignant transformation
Chongqing Medicine 2015;(16):2174-2176
Objective To investigate the correlation between moderate hypoxia and tumor invasiveness in colon carcinoma cell line HT‐29 and detect the expression of NF‐κB protein in nucleus and activities of MMP‐2/9 ,so as to explore its latent mecha‐nism of malignant transformation .Methods The moderate hypoxia model was established according to the pressure of O2 of tumor in vivo .After different period in moderate hypoxia ,activities of MMP‐2/9 secreted by HT‐29 cells were measured by gelatin zymog‐raphy ,NF‐κB levels in nucleus were assessed by Western blot after extraction of nucleic protein .Results MMP‐2/9 activities under moderate hypoxia gradually up‐regulated in 12 h and reached the peak at 24 h .The expression of NF‐κB protein in nucleus was simi‐lar to that of MMP‐2/9 .Conclusion Moderate hypoxia could induce malignant phenotype in HT‐29 cells through up‐regulating MMP‐2/9 activities ,in which NF‐κB may involve in this progression .

Objective To evaluate the safety of biopsy of the prostate guided by transrectal ultrasound. Methods Three hundred and seventeen patients suspected to have prostate cancer were examined and 208 patients were followed up.Results One hundred and eighty-one patients followed up did not feel severe pain during the biopsy of the prostate. Clinical manifestations induded 12 cases of hematuria,4 dases of hematospermia,1 case of fever, 2 cases of pollakisuria and urodymia,2 cases of uroschesis, which disappeared after corresponding treatment. Conclusions Biopsies of the prostate guided by transrectal ultrasound is advantageous in diagnosis of prostatic carcinoma.

Chinese Journal of Immunology 2016;32(7):992-995
Objective:To analyze Bufei Huo Xue Tang combined with Western medicine for chronic obstructive pulmonary disease( COPD) and its effect on airway inflammation reduction and lung function improvement. Methods: 100 patients with stable COPD in our department from February 2013 to March 2015 were randomly divided into combination group and control group. The control group received conventional oxygen, salmeterol and fluticasone propionate inhalation powder(1 absorption /times,2 times /day) and expectorant treatment. Combination group were based on the addition of lungs Bufei Huo Xue Tang with a course of 12 weeks. The clinical efficacy and α1-AT and IL-8 levels in two groups of patients were compared. Results:The cough, sputum, difficulty breathing, wheezing, wheezing symptom score in the combined group after treatment was significantly lower than the control group ( P<0. 05 ) . FVC,FEV1 and FEV1 /FVC in the combined group after treatment was significantly lower than those in the control group(P<0. 05).α1-AT and NO in the combination group was significantly higher after treatment. IL-8 and TNF-α was significantly lower than the control group(P<0. 05). BODE scores in the combined group after treatment were significantly lower than the control group(P<0. 05). Conclusion:Bufei Huoxue soup combined with Western medicine for COPD can inhibit inflammation, improve clinical symptoms, lung function and quality of life.
4.Protection Mechanism of Neferine in Learning and Memory Function of Rats with Chronic Cerebral Ische-mia
China Pharmacist 2016;19(7):1267-1272
Objective: To investigate the effects of neferine ( Nef ) on the learning and memory function and the expression of Notch1 and SYN in hippocampus in the rats with chronic cerebral ischemia .Methods:Male SD rats (250-300g) were randomly divid-ed into the sham operation group(Sham), the model group(Mod), nimodipine (Nim) positive control group and Nef treatment group with 5 ones in each .The chronic cerebral ischemia ( CCI ) model was established using bilateral common carotid artery ligation ( 2-VO).The rats were orally administered with NS(5 mg· kg-1),Nim(1 mg· kg -1) and Nef (20 mg· kg-1),respectively for 21 days after the first day of operation .The body weight was recorded .The number of hippocampal neurons in the rats was observed by Nissel staining. The learning and memory function was evaluated by Morris water maze test .Notch1 and SYN protein expressions in hippo-campus were detected by IHC staining and Western blot .Results:Compared with the Mod group , Nef group could reverse the reduc-tion of body weight and the number of hippocampal neurons in hippocampus CA 1 region induced by 2-VO, increase the route and the time of platform finding, prolong the escape latency and decrease the number of platform cross on the 21st day, and the effects were bet-ter than those of Nim (P<0.05).Moreover, the down-regulated SYN and Notch1 protein expressions in CCI group were both increase after the Nef treatment on the 21st day. Conclusion:Nef has a protective effect on the function of learning and memory in CCI rats in-duced by 2-VO,which may be related with the strengthened Notch 1 pathway in CA1 region of hippocampus and synaptic plasticity .
5.The effect of quercetin in the rats of LPS-induced acute Lung Injury
The Journal of Practical Medicine 2016;32(4):531-534
Objective To investigate the effect and mechanism of quercetin in LPS-induced acute lung injury. Methods The PaO2, PaCO2, RR and pH, the expression of IL-6、 IL-1β、 TNF-α、 JAK2、 STAT3、 p-JAK2、 p-STAT3, the pathological change and Dry/Wet of Lung tissues were evaluated in rats. Results Quercetin can significantly decrease the pathological change of Lung tissues, the PaCO2, RR and the expression of IL-6、IL-1β、 TNF-α、 p-JAK2 and p-STAT3 with increasing the PaO2, pH and D/W. Therefore, with on influence on the expression of JAK2. Conclusion Quercetin pretreatment decreased LPS-induced acute lung injury by suppressing JAK2/STATA3 signallingto improve exchange ability though reducing inflammation.
