1.Effect of ginsenoside-Rg1 on the proliferation of paraurethral fascia fibroblasts derived from women suffering from stress urinary incontinence

Objective To investigate the effect of ginsenoside-Rgl on paraurethral fascia fibroblastsmultiplication and the expression of proliferation cell nuclear antigen (PCNA) of stress urinary incontinence(SUI) women in vitro. Methods Specimens of human paraurethral fascia were obtained from 4 SUI womenduring tension-free vaginal tape (TVT) or tension-free vaginal tape-obturator (TVT-O) procedure.Fibroblasts were isolated and cultured by outgrowth technique. After reaching confluency fibroblasts weresubcultured every 5 days and cells after passage number 3 to 5 were used for assessment. The paraurethralfascia fibroblasts were treated with ginsenoside-Rgl at different concentrations (5, 10, 20 μmol/L) andfibroblnsts without Rgl were used as controL The multiplication conditions of paraurethral fascia fibroblastswere respectively detected by methyl thiazolyl tetrazolium (MTr) assay and the expression of PCNA byhistochemistry. Results ( 1 ) Compared with the control group, the growth rate of cells treated with differentconcentrations of Rgl after 72h [ (29±5 )%, (40±5 )%, (26±4)% respectively ] was significantly higher(P<0.01). (2)Compared with the control group, the stimulatory effect of Rgl on fibroblast growth wassignificant at 24 h (P<0.01), and peaked at 72 hi (29±5)% ,(40±5)%, (26±4)% respectively, P<0.01]. (3)Compared with the control group(28.77% ), there was a significant increase of PCNA-positivecells (P<0.01) after 48 h treatment with different concentrations of Rgl (49.24%, 83.48%, 54.50%respectively). Objective To further develop SDE for structured documentation of clinical data based on medical ontology. Methods OpenSDE was customized for the broad domain of medical ontology: medical concepts and its descriptors from history taking and physical examination were modeled into a tree structure. Results An expandable EMR system with structured data entry was developed. Conclusion Structured data entry is significant to clinical decision based on EMR.
3.Effect of arsenic trioxide in inducing apoptosis and expression of Fas/FasL of gastric tumor cells in nude mice

Objective To study the effect of arsenic trioxide (As_2O_3) on the expression of Fas/FasL and in inducing apoptosis of gastric tumor cells in nude mice. Methods The xenograft model was established in 30 male BALB/C-nu/nu mice. The mice were randomly divided into 3 groups and injected intraperitoneally with As_2O_3 (2.5mg/kg or 5mg/kg) or with saline solution of the same volume respectively. Tumor growth was detected by measuring tumor volume. Apoptosis of the tumor cells was measured with transmission electron microscope (TEM) and TdT-mediated dUTP nick end labeling (TUNEL). The expression of Fas/FasL in the tumor cells was detected with immunohistochemistry. Results Tumor volume in As_2O_3-treated groups was smaller than that in control group (P<0.05). Apoptotic cells in the As_2O_3-treated groups significantly outnumbered than those in control group (P<0.01). The expression of Fas in tumor cells was higher in the As_2O_3-treated groups than in control group (P<0.05), but FasL expression of tumor cells did not differ between the treated groups and control group (P>0.05). Conclusion As_2O_3 can obviously inhibit the growth of human gastric tumor. One of the mechanisms is up-regulation of Fas gene that can induce the apoptosis of gastric cells.

5.Effects of Melatonin on Oxidative Stress and Neuronal Apoptosis in Hippocampus of Rats with Epilepsy

Objective To investigate the effects of melatonin on oxidative stress and neuronal apoptosis in hippocampus of epileptic rats and the mechanism. Methods Seventy-two adult male Sprague-Dawley rats were equally divided into control group, model group, low dose group and high dose group. The model group was injected coriamyrtin 50μg/kg in the lateral ventricle, while the low dose group and high dose group were injected melatonin 20 mg/kg and 60 mg/kg, respectively, 30 minutes before injection of coriamyrtin. The contents of malo-ndialdehyde (MDA) and superoxide dismutase (SOD) were detected with ultraviolet spectrophotometer, the apoptosis was detected with TU-NEL, and the ultrastructural changes of neurons and mitochondria in hippocampal CA3 region were observed with electron microscopy, af-ter 60 minutes of epilepsy. Results The neurons and mitochondria in hippocampus were damaged, the number of apoptotic cells significant-ly increased (P<0.001), the content of SOD decreased (P<0.001), and the content of MDA increased (P<0.001) in the model group, com-pared with the control group. In the low dose group, the ultrastructural damage relieved, the number of apoptotic cells decreased (P<0.01), the content of SOD increased (P<0.05), and the content of MDA decreased (P<0.05);and for the high dose group, the ultrastructural damage relieved very much, the number of apoptotic cells decreased (P<0.001) and was not significantly different from the control group (P>0.05), SOD increased (P<0.001), and MDA decreased (P<0.001), compared with the model group. Conclusion Exogenous melatonin may signifi-cantly reduce neuronal apoptosis in rat hippocampal after epilepsy, and high dose is more effective, which may relate with resistance of oxi-dative stress, alleviate neuronal mitochondrial damage.
6.The impact of burden, positive feelings, coping style and social support on caregivers' fatigue of dementia patients

Objective To explore the impact of burden,positive feelings,coping style and social support on caregivers' fatigue of dementia patients.Methods 190 caregivers of dementia patients were investigated by using the questionnaires of Caregivers Burden Inventory(CBI),Positive Aspects of Caregiving(PAC),Simplified Coping Style Questionnaire (SCSQ),Social Support Rating Scale (SSRS) and Fatigue Scale-14 (FS-14).Results The caregivers' fatigue of dementia patients was positively correlated with burden(r=0.56,P<0.01),negative coping style(r=0.31,P <0.01),and negatively associated with positive feelings(r=-0.33,P <0.01),social support (r =-0.36,P < 0.01) and positive coping style (r---0.55,P < 0.01),respectively.Path analysis showed coping style and social support had direct impacts on fatigue; burden had a direct impact on fatigue,and also had an indirect influence on it through coping style or social support; fatigue might be decreased by positive feelings and increased by negative coping style.Conclusion To prevent and relieve the caregivers'fatigue of dementia patients,caregivers should be encouraged to take positive coping style and increase social support so as to reduce burden and increase positive feelings.
7.Correlation between miR-320d and prognosis of diffuse large B-cell lymphoma

Objective To study the expression of miR-320d in diffuse large B cell lymphoma (DLBCL),and its correlation with prognosis of DLBCL.Methods Sixty cases of DLBCL with the follow-up data were collected from Shanxi Cancer Hospital,and were examined by immunohistochemical EnVision method for CD3,CD10,CD20,bcl-6 and Mum-1.The DLBCL were classified into germinal center B cell-like (GCB) and non-germinal center B cell-like (non-GCB) subtypes according to Hans' algorithm.Agilent Human miRNA Microarray 16.0 was used to select the miRNAs on 24 cases paraffin-embedded tissue of DLBCL.The expression levels of miR-320d in 62 cases were examined by TaqMan real-time polymerase chain reaction.Eleven cases of reactive hyperplasia of lymph node were selected as control.Results In 62 cases of DLBCL,22 cases (35.5 %) were GCB and 40 cases (64.5 %) were non-GCB subtypes.The expression of miR-320d in GCB was 3.43 times as much as non-GCB subtypes (P =0.034),and in reactive hyperplasia of lymph node it was 5.65 times as much as in DLBCL (P < 0.001).The low expression groups of miR-320d was significantly correlated with shorter overall survival (P =0.021).Multivariate Cox proportional hazard regression analysis (including the IPI scores) revealed that the down-regulated miR-320d was the independent predictor in DLBCL (RR =2.434,95 % CI 1.148-5.159,P =0.020).Conclusions The down-regulated expression of miR-320d might be considered as a distinct subgroup with poor prognosis.
8.Research on Open Structure Data Entry for Electronic Medical Records and Clinical Knowledge Expressive Strategy

Objective To focus on the expressiveness and flexibility of OpenSDE: an application that supports recording of structured narrative data. Methods OpenSDE enabled data entry with (customizable) forms based on trees of medical concepts. The relevant scope for data entry could be tailored per medical domain by construction of a domain-specific tree. OpenSDE was intended for structuring narrative data to make these available for both care and research. Results The OpenSDE application was used at two departments in aspects of radiology, neurology, pediatrics, and child psychiatry. Conclusion OpenSDE is superior to traditional text entry method in aspects of correct and full expression and clinical decision support.
9.Teratogenecity of Ethylene Oxide Sterilized Medical Devices on Cells Used for Biotherapy

OBJECTIVE To observe the teratogenecity of ethylene oxide sterilized materials to cells which are used for biotherapy.METHODS Three kinds of cells were cultivated in ethylene oxide sterilized culture flasks which were conserved for different time and high temperature/high pressure sterilized vitric culture flasks.NK/CIK cells induced from cord blood,human mesenchymal stem cell and CD34+ cells were cultivated in ethylene oxide sterilized culture flasks which were conserved for one day(group A),for three months(group B),for more than six months(group C) and cultivated in high temperature/high pressure sterilized vitric culture flasks(group D).After ten days,not only the growth and morphology of cells but also the breakages fragmentations and gaps in chromosomes and replication of chromosome in nucleus were observed.According to these data,the malformation rate was calculated.RESULTS Compared with groups C and D,the inductivity of groups A and B obviously degraded(P0.05).CONCLUSIONS Compared with the control group,the materials which were sterilized by ethylene oxide and conserved for less than one day and three months can bring damage to cells.However,the damage is not discovered in the materials which were conserved for more than six months.Ethylene oxide can induce cell aberrant and mutation.Therefore,it is suggests that materials be conserved for more than six months to ensure safe and effective enforcement of biotherapy.
10.Development of Hospital EMR with IHE XDS Technical Framework in Clinical Practice

Objective To explore the application ways of IHE XDS technology in electronic medical record (EMR) system and develop application system of clinical EMR integration. Methods The clinical EMR integration plan was worked out based on IHE XDS technical framework. Results A EMR solution based on IHE XDS was presented. It was consisted of the components of the clinical Data Entry, XDS Registry, clinical document Repository, EMR application server and the i-node accessing server. The system was tested and the result indicated that the system was effective and scalable. Conclusion IHE XDS profile can be not only used to realize the sharing of EMR documents across multiple hospital, but also support the collection clinical data within a hospital clinical system integration.
