1.Comparative analysis of radiotherapy alone,thermoradiotherapy and radiotherapy and chemotherapy of esophageal cancer effect

Objective To comparative analysis the effect of radiotherapy,thermoradiotherapy and chemora-diotherapy on esophageal cancer.Methods 120 patients with esophageal carcinoma were selected and randomly divided into the radiotherapy group,thermoradiotherapy groupand chemoradiotherapy group by digital table method, with 40 cases in each group,which were treated with radiotherapy alone,thermoradiotherapy alone and radiotherapy combine with chemotherapy respectively.The effects of treatment of the three groups were compared.Results The main toxic reaction in the three groups during the treatment for adverse reaction were digestive system reaction,hema-tologic toxicities and radioactive esophagitis.The incidence of toxic reaction of the three groups were not statistically significant(P >0.05).Total response rate of simple radiotherapy group,the thermal radiation group,chemoradiothera-py group were 52.5%,75.0% and 85.0%.,The total remission rate of chemotherapy group was higher than those of the radiotherapy group and the thermal radiation group(χ2 =5.218,3.857,all P <0.05).Conclusion The different treatment methods of esophageal cancer can obtain certain curative effect,but there are some adverse reactions.Among different treatment methods,radiotherapy and chemotherapy in the treatment of esophageal carcinoma can get better effect than the radiotherapy and chemoradiotherapy.
2.Imaging Features of Hypertrophic Spinal Pachymeningitis: a Report of 2 Cases

Objective To investigate the imaging features of hypertrophic spinal pachymeningitis(HSP).Methods 2 patients with HSPproved pathologically received both CT and MRI scan. The literatures were reviewed, the clinical data and imaging features of HSP wereretrospectively analyzed.Results The characteristic HSP manifestation of HSP on imaging was the crescent-shaped thickening of spinaldura maters , the thickness of the dura was 5.2 mm and 13 mm respectively in this two cases . Conclusion HSP is of typical imaging characteristic. The diagnosis can be made correctly in combination with clinical data.
3.Effects of Melatonin on Oxidative Stress and Neuronal Apoptosis in Hippocampus of Rats with Epilepsy

Objective To investigate the effects of melatonin on oxidative stress and neuronal apoptosis in hippocampus of epileptic rats and the mechanism. Methods Seventy-two adult male Sprague-Dawley rats were equally divided into control group, model group, low dose group and high dose group. The model group was injected coriamyrtin 50μg/kg in the lateral ventricle, while the low dose group and high dose group were injected melatonin 20 mg/kg and 60 mg/kg, respectively, 30 minutes before injection of coriamyrtin. The contents of malo-ndialdehyde (MDA) and superoxide dismutase (SOD) were detected with ultraviolet spectrophotometer, the apoptosis was detected with TU-NEL, and the ultrastructural changes of neurons and mitochondria in hippocampal CA3 region were observed with electron microscopy, af-ter 60 minutes of epilepsy. Results The neurons and mitochondria in hippocampus were damaged, the number of apoptotic cells significant-ly increased (P<0.001), the content of SOD decreased (P<0.001), and the content of MDA increased (P<0.001) in the model group, com-pared with the control group. In the low dose group, the ultrastructural damage relieved, the number of apoptotic cells decreased (P<0.01), the content of SOD increased (P<0.05), and the content of MDA decreased (P<0.05);and for the high dose group, the ultrastructural damage relieved very much, the number of apoptotic cells decreased (P<0.001) and was not significantly different from the control group (P>0.05), SOD increased (P<0.001), and MDA decreased (P<0.001), compared with the model group. Conclusion Exogenous melatonin may signifi-cantly reduce neuronal apoptosis in rat hippocampal after epilepsy, and high dose is more effective, which may relate with resistance of oxi-dative stress, alleviate neuronal mitochondrial damage.
4.Research progress in radiation-induced organ fibrosis

Radiation-induced organ fibrosis is a common complication of tumor radiotherapy and radiation accident, which poses a serious threat to patients′life.TGF-β, CTGF, PDGF-BB and NF-κB all play important roles in this pathological process, and drugs targeting these key molecules are also being developed, some of which have shown positive therapeutic effects.This review focuses on the molecular mechanisms and therapeutic progress involved in radiation-induced organ fibrosis, intended to provide a new clue to the complex process of radiation-induced organ fibrosis, and to provide a new strategy for treatment.
5.Application of sorting peripheral blood cells by flow cytometry in the detection of peripheral Mood cell sorting

Objective To apply fluo-rescencc-activated cell sorting(FACS) in sorting T lymphocyte (CD_3~+) and granulocyte (CD_(15)~+), which establish the separating method of series of cells from human peripheral blood, so that the scientific research and clinical research could be carried out were specifically. Methods 10 cases of normal peripheral blood were collected and T lymphocyte (CD_3~+) and granulocyte (CD_(15)~+) were stained with florescence conjugated antibodies. The positive cells were sorted by FACS. Results Before sorting the peripheral blood, the proportion of the T lymphocyte(CD_3~+) and granulocyte (CD_(15)~+) in the leukocyte is 48.8 % and 30.8 %; after sorting by FACS, the purity of T lymphocytes (CD_3~+) is up to 98 % and the recovery is about 95 %; the purity of granulocyte (CD_(15)~+) is up to 97 % and the recovery is about 96 %. Conclusion The FACS could Mlow us to quickly sort T lymphocyte (CD_3~+) and granulocyte (CD_(15)~+) with higher recovery and higher purity from the peripheral blood.
6.Comparison of methods of natural killer cell sorting and purification from human peripheral blood

Objective To apply fluorescence-activated cell sorting (FACS) and magnetic activated cell sorting (MACS) in sorting natural killer (NK) cells from human peripheral blood. The efficiency of these two methods were compared so that the basic conditions for the further study on the lower proportion of peripheral blood cells could be created. Methods Twenty cases of normal peripheral blood and 3 cases of post-hematopoietic stem cell transplantation (post-HSCT) were collected and NK cell were sorted by FACS and MACS. Results Before sorting the normal peripheral blood, the proportion of the NK cell in the leukocyte was (12.86±3.62) %, the purity of NK cell was up to (96.15±2.03) % and the recovery was (95.08±2.16) %after sorting by FACS, the purity was up to (93.35±3.61) % and the recovery was (94.11±3.01) % by MACS.And before sorting the post-HSCT peripheral blood, the proportion of the NK cell in the leukocyte was (11.01±2.08) %, the purity of NK cell was up to (96.22±2.16) % and the recovery was (95.27±1.18) % after sorting by FACS. The purity was up to (90.98±1.94) % and the recovery was (94.54±3.52) % by MACS. Thereis difference in purity of NK cell (t = 5.925, P <0.05), while no difference in rate of recovery between 2methods (t = 0.789, P >0.05). Conclusion FACS is more efficient, rapidly and easily than MACS in sorting NK cell, especially for post-HSCT cases.
7.Clinical significance of detecting the NKT cell content and its surface receptors NKG2A and NKG2D in the peripheral blood of patients with esophageal carcinoma

Objective NKT cells are very important as a kind of non-specific immune cells. Much attention in antitumor significance has been received in the study of its effect on malignant diseases. The aim of this study was to detect the expression of NKT cells and its CD+8 NKT subsets in peripheral blood of esophageal patients and normal person, and to analyze the changes in the expression of NKG2D and NKG2A receptorsand its clinical pathological factors. Methods By flow cytometric analysis, 53 patients with esophageal carcinoma and 39 normal controls were analysed for peripheral blood of NKT cells and CD+8 NKT subsets, and the expression of NKT cells NKG2A and expression of NKG2D receptor. The clinical pathological factors were collected for the comparative analysis. Results Compared with the normal control group, the expression of NKT cells in peripheral blood of esophageal patients increased [(4.32±0.73) %, (5.97±1.29) %] (t =3.562, P <0.01), and the expression level of its surface receptor NKG2D reduced [(17.56±5.92) %, (15.12±1.56) %] (t =3.892, P <0.05), but the express levels of NKG2A [(4.02±1.41) %, (5.99±4.59) %] in creased (r = 4.015, P <0.05), those expression change with the development of the esophageal cancer. Conclusion The increased expression of NKT cells and CD+8 NKT subsets in the peripheral blood of patients with esophageal carcinoma refletcs that the immune feedback of patients' antineoplastic effect is strengthened. The decresed expression of the active receptor NKG2D and the increased expression of the inhibitory receptor NKG2A on NKT cells might be one of mechanisms leading to the reduction of NKT cell activity and immune escape of patients with esophageal carcinoma. The changes of surface receptors of NKT cells may be associated with the development of the esophageal cancer.
8.Correlation and its clinical significance of regulatory T cells and NKG2D-expressing NK cells in lung cancer patients

Objective To detect the levels of CD+4 CDHi25 CDLo127 regulatory T (Treg) cells and NKG2D-expressing NK cells in peripheral blood of lung cancer patients and investigate their relationship,and explore their roles in the immune escape of lung cancer and clinical significance.Methods The levels of CD+4 CDHi25 CDLo127 Treg cells,NK cells and NKG2D-expressing NK cells were measured in peripheral blood of 70 lung cancer patients and 50 healthy controls.Results Compared with control group,the levels of CD+4 CDHi25 CDLo127 Treg cells enormously increased [(8.4±4.1) %,(6.7±1.7) %] (t =3.09,P < 0.05),while the value of NKG2D obviously decreased [(83.3±4.9) %,(87.4±2.9) %] (t =3.16,P < 0.05); And the group of NK cells is not different [(15.6±8.3) %,(17.2±4.2) %] (t =-1.33,P > 0.05); NKG2D-expressing NK cells had a perfect negative correlation with CD+4 CDLo25 CDLo127 Treg cells (r =-0.302,P < 0.05).Conclusion Accumulation of Treg cells in serum may lead to the down-modulation of NKG2D-expressing NK which participates the tumor immune escape.They may be potential indicators evaluating immune functions and prognosis of lung cancers.
9.Expression and clinical significance of CD+8 CD-28 and CD+4CDhigh25 CDlow127 regulatory T cells in peripheral blood of lung cancer patients

Objective To observe the changes of CD+8CD+28,CD+8CD-28 and CD+4CDhigh25CDlow127 regulate T (Treg)cellsin peripheral blood of lung cancer patients,and to analyze the correlation between CD+8CD-28 and CD+4CDhigh25CDlow127 Treg cells to reveal the role and clinical significance of them in lung cancer patients.Methods Flow cytometry was applied to evaluate the level of CD+8CD+28,CD+8CD-28 and CD+4CDhigh25CDlow127 Treg cells in peripheral blood of 60 untreated lung cancer patients and 60 healthy controls group.The association of each term with clinical features was analyzed.Results The percentage of CD+8CD-28 and CD+4CDhigh25CDlow127 Treg cells in lung cancer group[(58.430:15.749) %,(7.365±2.025) %]was significantly higher than those in healthy group [(41.057±15.436)%,(6.648±1.669)%,(t=6.102,P<0.05;t=2.115,P<0.05)],while the percentage of CD+8CD+28cells is lower(41.570±15.739)% than that in healthy group[(58.700±15.298)%,(t=-6.043,P<0.05)].No close associations were found between three index and gender,age,and biological characteristics.With the increase of TNM stage,The percentage of CD+4CDhigh25 CDlow127 Treg cells increased gradually,which was remarkably higher in patients rith stage Ⅳ than that with stage ⅢA(t=-3.898,P<0.05).The percentage of CD+4CDhigh25 CDlow127 Tregcells was uncorrelated with CD+8CD-28 cells(r=-0.169,P>0.05).Conclusion The higher percentage of CD+8CD-28 and CD+4CDhigh25 CDlow127 Treg cells,the lower percentage of CD+8CD+28 cells may be the important reasons of immune suppression in lung cancer patients.Though there is no correlation between CD+8CD-28 and CD+4CDhigh25 CDlow127 Treg cells,it is may be helpful to understand immunologic function and it may look for more specific therapy and provide a new reference in the prognosis of lung cancer.
10.Detection and its clinical significance of CD+4 CDHi25 CDLo127 regulatory T cells in peripheral blood of patients with thyroid neoplasm

Objective To analyze CD+4 CDHi25 CDLo127 regulatory T cells (Treg) in peripheral blood of patients with thyroid nodules and differentiated thyroid cancer and their change regularity, and to investigate the immunosuppression mechanism. Methods The peripheral blood was collected from 175 patients with thyroid nodules, including 43 patients with differentiated thyroid cancer and 132 patients with nodular goiter.By using monoclonal antibodies, the blood samples were evaluated with the flow cytomertry for lymphocyte subsets and Treg cells. Results The results showed the prevalence of the CD+4 CDHi25 CDLo127 Treg in differentiated thyroid cancer group [(6.48±1.49) %] and nodular goiter group [(6.23+1.67) %] was higher than those in the healthy group [(5.62±1.48) %], and the difference was significant(P < 0.005), but there was no significant difference between the nodular goiter group and differentiated thyroid cancer group (P >0.005).Conclusion It is concluded that the relative increase of CD+4 CDHi25 CDLo127 Treg in peripheral blood of patients with nodular goiter and differentiated thyroid cancer may be related to immunosuppression and tumor progression.
