2.Determination of Total Selenium in Salt by HG-AFS

Objective To establish a sensitive ,reliable and convenient method for determination of total selenium(Se) in salt by HG-AFS. Methods The salt samples were dissolved and added hydrochloric acid (HCl),after heating by boiling water ,used HG-AFS to determinate Se. Results The linear range of the method was 0.5-400 ?g/L, The correlation coefficient was larger than 0.999 5, the recovery rates were 95.7%-102.8%. If 1 ml sample was collected, the detection limit was 0.08 ?g/L. Conclusion The method is sensitive, reliable, simple, less sample usage, and is an ideal method for determination of total selenium in salt.
3.Therapeutic Effect of Ursolic Acid on Chronic Nonbacterial Prostatitis in Rats

Objective To investigate the therapeutic effect and mechanism of ursolic acid on carrageenan-induced chronic nonbacterial prostatitis (CNP) of SD rats. Methods Forty-two SD rats were randomly divided into five groups:normal control group, model control group, Pushitai group, ursolic acid high-dose group, and low-dose group.Except for normal control group, chronic nonbacterial prostatitis model was established using carrageenan. The rats of Pushitai group were treated with the solution of Pushitai at the dose of 148 mg??kg-1??d-1 for twenty days.The rats of ursolic acid high-dose and low-dose groups were administered with ursolic acid at the doses of 100 and 50 mg??kg-1??d-1 for twenty days, respectively. The rats of normal and model control groups were treated with isometric 5% CMC-Na for twenty days.The prostate index (PI) was calculated from the ratio of isolated rat prostate weight and body weight of rat.The expressions of IL-1β, TNF-α, and IL-10 were detected by ELISA. The histology and morphology of the prostate was observed with the aid of hematoxylin-eosin staining. Results Compared with the model control group, PI and the levels of IL-1β, TNF-α, and IL-10 in serum were significantly decreased in ursolic acid high-dose,and low dose groups ( P<0. 05 ) . Moreover, the pathological changes of prostate were improved significantly. Conclusion Ursolic acid has certain therapy effects on chronic nonbacterial prostatitis, which may be related to its regulating the expressions of IL-1β, TNF-α, and IL-10.
4.Effects of Gui Curcuma Extract on Hepatic Stellate Cell-LX2

This study was aimed to observe the effects of Gui Curcuma extract on proliferation, collagen production, and matrix metalloproteinase (MMP) expression by hepatic stellate cells (HSC-LX2). HSC-LX2 were cultured in DMEM broth (10% FBS), then treated with Gui Curcuma extract (with the concentration of 0, 22.5, 45, 90 μmol·L-1) for 24 h. The proliferation of HSC-LX2 was measured by MTT assay; injury in HSC-LX2 was indicated with LDH re-lease; collagen I production was detected by ELISA; and MMP-l expression was determined by Western blot. The re-sults showed that Gui Curcuma extract (with the concentration of 22.5, 45, 90 μmol·L-1) dose-dependently inhibited proliferation of HSC-LX2 by 16.2%, 43.9%, 59.4% in MTT assay, respectively. The IC50 was 59.1 μmol·L-1. There was no significant impact on LDH release. The Gui Curcuma extract (with the concentration of 45, 90 μmol·L-1) can significantly reduce the expression of collagen I level (P< 0.01), whereas MMP-1 expression was significantly uplift-ed (P< 0.05) by Gui Curcuma extract (with the concentration of 45, 90 μmol·L-1) in Western blot. It was concluded that Gui Curcuma extract had the potential to prevent hepatic fibrosis, through inhibiting proliferation and collagen I production, and increasing MMP-1 protein expression.
5.Application of nursing quality of bull’s-eye pattern on caring cancer patients with religious beliefs

ObjectiveTo study the effect of nursing quality of bull’s eye pattern on caring cancer patients with religious belief. Methods Three hundred and thirty-six cancer patients with religious beliefs were cared according to nursing quality of bull’s-eye pattern.All patients were evaluated by European organization for research and treatment of cancer quality of life questionnaire core 30(EORTC-QLQ-C30)before and after applications of nursing quality of bull’s-eye pattern and the results were compared.Result The scores of the dimensions of overall quality of life and function in EORTC-QLQ C30 questionnaire among those patients with implementation of nursing quality of bull’s eye patterns were higher than those before implementation of the pattern and the symptom score after the use of the pattern was statistically lower than that before its use in respect of dimensions of cancer symptoms(P<0.01). Conclusion The applications of nursing quality of bull’s-eye pattern on caring cancer patients with religious beliefs can improve the patients’ quality of life.
6.Effect of miR-21 on migration and invasion ability in human laryngeal squamous carcinoma cell Hep 2

Objective To explore the effect of microRNA‐21(miR‐21) on the migration and invasion ability in human laryn‐geal squamous carcinoma cell Hep2 .Methods The MTT method was used to detect the viability of Hep2 cells at 48 h after miR‐21 inhibitor and miR‐21 NC transferring into Hep2 cells by LipofectamineTM 2000 .The cell migration ability was detected by using the scratch test .The cell invasion ability was detected by using the Transwell method .The activation of phosphatase and tensin homo‐logue deleted on chromosome 10 (PTEN)/phosphatidylinositol 3 kinase (PI3K) /protein kinase B(Akt) signal pathway and the expression of matrix metalloproteinase 2 (MMP2) ,MMP9 ,reversion inducing cysteine rich protein with kazal motif (RECK) was detected by using the Western blotting .Results Compared with miR‐21 NC ,miR‐21 inhibitor could significantly reduce the Hep2 cellviability[(0.688±0.043)vs.(0.375±0.012)],inhibitedthemigrationability[(6.57±0.02)μm vs.(20.49±2.18)μm]and invasion ability[(100 .7 ± 10 .2) vs .(46 .8 ± 4 .3)] ,and the differences were statistically significant (P<0 .01) ,meanwhile miR‐21 inhibitor could down‐regulate the expression of PI3K ,MMP2 and MMP9(P<0 .01) ,and reduced the phosphorylation level of Akt (P<0 .01) ,up‐regulated the expression of PTEN and RECK (P<0 .01) .Conclusion miR‐21 inhibitor can significantly suppress the migration and invasion ability of Hep2 ,which may be related with the PTEN/PI3K/Akt signal pathway .
7.Effect of stachyose on microbiotic disequilibrium in rhizospheric soil of Rehmannia glutinosa

Objective To testify the screening function of stachyose on soil bacteria by investigating the bacterial culture in ammonium stachyose medium. Methods The turbidimetry was used to determine the absorbance of microbial suspension at 600 nm per 2 h under the same initial concentration of the microbial suspension and to draw their growth curves. Results Most of soil bacteria utilized stachyose ineffectively, while only a few of them grown well in ammonium stachyose medium. Conclusion Since the major soil bacteria can not take stachyose fully as their energy resources, the species and quantity of rhizobacteria may decrease largely and only a few that utilized stachyose better can develop vigorously. Those rhizobateria with better utilization of stachyose may multiply so rapidly as potential ones in the rhizosphere of Rehmannia glutinosa that the disequilibrium of soil microorganism appears.
8.Efficiency of hearing screening combined with gene mutation screening in neonates with high-risk of hearing impairment

ObjectiveTo investigate the significance of hearing screening combined with gene screening for neonates with high-risk of hearing impairment.MethodsNeonates admitted to the Neonatal Department of Guangdong Women and Children Hospital between July 2013 and June 2014 were enrolled in this study. They were divided into high-risk group (with high-risk for hearing impairment) (n=3 129), and control group (n=5 106). Neonate hearing screening was carried out using otoacoustic emission and automated auditory brainstem response. Blood samples were collected using a standard protocol for detecting the mutations of four common deafness genes, includingGJB2,GJB3,SLC26A4 and mitochondrial 12s rRNA.Chi-square test was used to compare the differences of the pass rate of hearing screening and positive rate of gene mutations between the two groups.ResultsThe rates of failure on otoacoustic emission, automated auditory brainstem response or both in the high-risk group were 11.92% (373/3 129), 10.32% (323/3 129) and 4.83% (151/3 129), respectively, higher than those in the control group [5.03%(257/5 106), 6.56%(335/5 106) and 2.02% (103/5 106)] (χ2=130.265, 37.354 and 51.196, allP=0.000). In the high-risk group, the overall positive rate of gene mutations was 5.63% (176/3 129), and theGJB2 andSLC26A4 gene mutation rates were 3.04% (95/3 129) and 2.40% (75/3 129)], all higher than the control group [3.15% (161/5 106), 2.04% (104/5 106) and 1.06% (54/5106)] (χ2=30.301, 8.216 and 22.517, allP<0.01). But the mitochondria 12S rRNA gene andGJB3 gene mutation rates were the same in high-risk group and control group [0.19% (6/3 129) vs 0.06% (3/5 106); 0.03% (1/3 129) vs 0.00%(0/5 106), bothP>0.05]. The rates of failure on otoacoustic emission and automated auditory brainstem response of the neonates with deafness gene mutations were 9.50% (32/337) and 10.39% (35/337), respectively, higher than the neonates without [1.14% (90/7 898) and 1.29% (102/7 898)] (χ2=154.621 and 163.399, both P=0.000).ConclusionCombined hearing screening is of clinical significance for neonates with high-risk of hearing impairment.
9.Application of intelligent hydrogel in the tissue engineering

BACKGROUND:Compared with traditional hydrogels, intel igent hydrogels can appear to exhibit different responses to different stimuli such as temperature, pH value, light, and magnetic field, produce two-stage structure and alter chemical structure to generate the ordered supramolecular structure by self-assembling, and ultimately cause the formation of the gel with three-dimensional structure.
OBJECTIVE:To review the research status of intel igent hydrogels and its application in tissue engineering.
METHODS:A computer-based search of CNKI and PubMed databases was performed to retrieve articles addressing intel igent hydrogels in tissue engineering published before 2014. The keywords were“hydrogel, tissue engineering”in Chinese and English.
RESULTS AND CONCLUSION:Intel igent hydrogels are classified into temperature sensitivity, pH sensitivity, photosensitivity, magnetic susceptibility and temperature/pH dual responsive hydrogels. The change of the external environment can be automatical y detected and responded. Intel igent hydrogels exhibit a series of outstanding performances in drug delivery systems, drug delivery, repair and improvement of defected tissues, which are not possessed by traditional materials. In particular, the intel igent hydrogels show considerable superiorities in tissue engineering, including low immunogenicity that reducing inflammation and rejection, biodegradability, realizing the three-dimensional scale simulation of cel microenvironment that is conducive to cel adhesion, growth, migration and differentiation.
10.Expression of 4-hydroxynonenal in the kidney of diabetic rats and the effect of probucol

Objective To investigate the expression of 4-hydroxynonenal (4-HNE) in the kidney of diabetic rats and the effect of probucol.Methods The rats were being intraperitoneal injected with STZ (60 mg/kg) to establish diabetic models.Then diabetic rats were randomly divided into diabetic group (group D,n =24),probucol treated group (group P,n =24).Normal rats were taken as control group (group C,n =24).Rats in group P were treated by probucol (110 mg·kg-1·d-1); rats in group D and group C were given equal volume water instead.Scr,BUN,triglyceride (TG),total cholesterol (TC) and 24-hour urinary proteinin were measured at the 4th,8th and 12th week.PAS staining and HE staining were used to evaluate the pathological changes of the kidney.The immunohistochemistry and Western blotting were used to detect the expression of 4-HNE in renal tissue.Results Levels of Scr,BUN,TG,TC and 24-hour urinary protein in group D were higher than those in group C at the 4th,8th and 12th week(all P < 0.05); Levels of Scr,BUN,TG,TC and 24-huor urinary protein in group P were lower than those in group D at 4th,8th and 12th week (all P < 0.05).The pathological changes of the kidney in group D were more serious than that in group P.The expression of 4-HNE in group D were higher than group C at the 4th,8th and 12th week (all P < 0.05);The expression of 4-HNE in the kidneys of group P decreased significantly compared to that of group D at the same time (P < 0.05).Conclusions As an indicator of lipid peroxidation,the expression of 4-HNE significantly increases in the kidney of diabetic rat.Probucol may protect the diabetic kidney through decreasing the expression of 4-HNE and the level of lipid peroxidation.
