1.Effects of ligustrazine on the secretion of hydrogen sulfide in the ventricular tissues of rats under physiological condition

Objective: To explore the effects of ligustrazine on the secretion of hydrogen sulfide in the ventricular tissues of rats under physiological condition.Methods:32 rats were divided into control group,high dose group of ligustrazine(150?g/ml),medium dose group of ligustrazine(15?g/ml) and low dose group of ligustrazine(1.5?g/ ml) at random.The left and right ventricular tissues were incubated in 37℃ thermostatic water bath for 4 hours with Krebs solution as supernatant.After incubation of the ventricular tissues,the sensitive electrode-based method was used to detect the content of H2S in the supernatant.Results: In the left ventricular tissues,there were on dif-ference of the H2S level betweenthe 1.5?g/ml ligustrazine group(0.17?0.03),15?g/ml ligustrazine group(0.17?0.03)and control group.However,compared with control group, the H2S level in hige dose group of ligustrazine decreased obviously(P0.05),but the H2S level in the 150?g/ml ligustrazine group(0.46?0.09)was significantly lower than that in the control group(0.64?0.13)(P
2.The effect of hydrogen sulfide donor on the pathogenesis of atherosclerosis in apolipoprotein E knock out mice

Aim To explore the effect of hydrogen sulfide (H2S) donor on the pathogenesis of atherosclerosis in apolipoprotein E knock out mice (Apo E-/-).Methods 6-week old C57BL/6J and Apo E-/-mice were divided into normal group, Apo E-/-group and Apo E-/-+ sodium hydrosulfide(NaHS)group,with 8 mice in each group. They were fed chow diet until 16-week old. The serum levels of total cholesterol, low density lipoprotein and high density lipoprotein were determined and the changes of atherosclerosis plaque size were observed. Sulfide electrode-based method was used to measure the serum level of H2S.Results The serum levels of total cholesterol[(12.59?3.11 vs 2.32?0.40) ?mol?L-1]and low density lipoprotein[(1.33?0.43 vs 0.13?0.03) ?mol?L-1] in Apo E-/-mice were significantly higher than those in normal group, while the serum levels of high density lipoprotein[(0.45?0.13 vs 1.49?0.21) ?mol?L-1]and H2S in Apo E-/-mice[(44.64?4.52 vs 57.69?7.03) ?mol?L-1]were significantly lower than those in normal group. Meanwhile, the atherosclerosis plaque[(139316.6?30362.93 vs 0) ?m2] was obvious in Apo E-/-mice. After NaHS was administrated, there were no obvious changes in the serum levels of total cholesterol, low density lipoprotein and high density lipoprotein in Apo E-/-mice, while the serum level of H2S[(52.21?7.24 vs 44.64?4.52) ?mol?L-1]increased significantly and the size of atherosclerosis plaque[(85927.84?1922.73 vs 139316.6?30362.93) ?m2]diminished significantly.Conclusions H2S postponed the proceeding of atherosclerosis and diminished the size of atherosclerosis plaque.

4.Characterization of one novel mutation of the low density lipoprotein receptor gene in patients with familial hypercholesterolemia-one case report

Objective To analyze the genetic variation of low density lipoprotein receptor(LDLR)gene in patients with familial hypercholesterolemia(FH).Methods Polymerase chain reaction(PCR)was used to amplify the promoter region and the 18 exons of LDLR gene family with the patients' genomic DNA as templates.DNA sequencing of the PCR production was used to find mutations in patients with FH.Results A novel missense mutation(G→T)in exon 4 was identified by DNA sequencing in the proband and his mother,and the mutation was characterized at nucleotide site of 385 in exons of coding region,with tyrosine substitute for aspartate at amino acid site of 108.Conclusion Characterization of the novel mutation provides an example of the genetic basis of LDLR causing FH.
5.Improvement of acetic acid tolerance and fermentation performance of industrial Saccharomyces cerevisiae by overexpression of flocculent gene FLO1 and FLO1c.

Flocculent gene FLO1 and its truncated form FLO1c with complete deletion of repeat unit C were expressed in a non-flocculent industrial strain Saccharomyces cerevisiae CE6 to generate recombinant flocculent strains 6-AF1 and 6-AF1c respectively. Both strains of 6-AF1 and 6-AF1c displayed strong flocculation and better cell growth than the control strain CE6-V carrying the empty vector under acetic acid stress. Moreover, the flocculent strains converted glucose to ethanol at much higher rates than the control strain CE6-V under acetic acid stress. In the presence of 0.6% (V/V) acetic acid, the average ethanol production rates of 6-AF1 and 6-AF1c were 1.56 and 1.62 times of that of strain CE6-V, while the ethanol production rates of 6-AF1 and 6-AF1c were 1.21 and 1.78 times of that of strain CE6-V under 1.0% acetic acid stress. Results in this study indicate that acetic acid tolerance and fermentation performance of industrial S. cerevisiae under acetic acid stress can be improved largely by flocculation endowed by expression of flocculent genes, especially FLO1c.

Objective To evaluate the fiber altering and clinical relationship in patients with occipital and temporal lobe cerebral infarction with DTI.Methods Fifty patients suffering from cerebral stroke were analyzed with conventional MRI,DWI and DTI. The fibers of visual pathway were depicted ,and their fractional anisotropy (FA),apparent diffusion coefficient (ADC),isotropic im-age (Iso)and T2-weighted trace were measured and analyzed.Results Regions of interest (ROIs)were placed in infarction and nor-mal contralateral tissue for tracking the visual pathways.The different parameters were measured.FA is useful to differentiate hy-per-acute stroke from acute and sub-acute stroke.FA values increased slightly in hyper-acute and decreased in acute,sub-acute and chronic phase.The values of ADC in infarction regions in hyper-acute,acute and sub-acute phases were lower than those of the con-tralateral tissue.Iso and T2-weighted trace increased in acute phase and decreased in chronic phase.DTT was used to show the way and chang of visual pathway.Conclusion DTI could track the fiber of visual pathway,and depict the fiber of infarction zone.DTT could be used in evaluating the prognosis of patients.
7.Anti-tumor effects of p53N15-based fusion peptide in the transfected H1299 lung cancer cells

ObjectiveLoss-of-function mutation of p53,a tumor suppressor gene,is an important mechanism for the development of human cancers.In this study we tried to transfect p53N15-based fusion peptide into H1299,a lung cancer cell line,and evaluate the anti-tumor effects of the fusion peptide.MethodsAdeno-associated virus ( AAV) vectors were used for transfecting p53N15 fusion peptide into p53-null lung adenocarcinoma H1299 cells.The anti-replication effects of p53N15 fusion peptide were evaluated with inverted microscopy,MTT test for cell viability and flow cytometry.ResultsFusion peptides in H1299 cells was highly expressed and had detectable suppressive effects on cell proliferation.A large amount of dead cells were seen under microscope after the transfection of recombinant viruses for 72 hours.Cells activity was reduced significantly in the virus-transfecting groups as demonstrated by MTT test.The flow cytometry showed that a large number of dead cells were present in the virus-transfecting groups.ConclusionThe growth of H1299 lung adenocarcinoma cells could be inhibited in vitro by being transfected with p53N15 fusion peptide,which may be a potential gene therapy alone or as an adjuvant option in the treatment of lung cancer.
8.Reconsideration of indications for gallbladder-preserving cholecystolithotomy and preventive measures for postoperative recurrence of gallstones

Choledochoscopic minimally invasive gallbladder-preserving cholecystolithotomy is a surgical method for preserving the gallbladder in the treatment of gallstones, and recurrence of gallstones is a hot topic for gallbladder-preserving cholecystolithotomy. The postoperative recurrence rate of gallstones can be reduced by selecting appropriate surgical indications and adopting effective preventive measures after surgery. This article reviews the indications for minimally invasive gallbladder-preserving cholecystolithotomy and postoperative preventive measures for the recurrence of gallstones, in order to better understand the influencing factors for recurrence and provide guidance for the treatment of gallstones by minimally invasive gallbladder-preserving cholecystolithotomy.

Objective To explore the diagnostic value of tumor necrosis factor-alpha (TNF-α) , interleukin-6 (IL-6) and neuron enolase (NSE) in the cerebrospinal fluid of children with central nervous sys-tem infection (CNSI).Methods 54 cases of CNSI hospitalized children ,admitted in the hospital from October 2015 to January 2017 ,were enrolled in the study and divided into viral meningitis group (30 cases) and suppu-rative meningitis group (24 cases).Another 20 cases who underwent cerebrospinal fluid examination and other related examinations were enrolled in the study as the control group.The levels of three biomarkers TNF-α , IL-6 and NSE in cerebrospinal fluid of three groups were detected by enzyme linked immunosorbent assay (ELISA).Results The levels of TNF-α ,IL-6 and NSE in purulent meningitis group were the hightest ,and the levels of these three factors in viral meningitis group were highter than the control group ,and the differ-ence was statistically significant in three groups (P<0.05).But there were a lot of data overlaps.There was no significant difference in the levels of TNF-α ,IL-6 and NSE between the brain group and the control group (P>0.05).Conclusion The detection of TNF-α ,IL-6 and NSE in cerebrospinal fluid has a certain clinical val-ue for the diagnosis of CNSI ,but it needs to be further verified by a large sample clinical trial.
10.Social skill training for children with autistic spectrum disorder on the uncertainty of the disease from mothers

Objective:
To improve the social skills of children with ASD by using Program for the Education and Enrichment of Relational Skills(PEERS ), and to reduce the uncertainty towards ASD and negative emotions for mothers of ASD children.
Methods:
From September to October 2017, 30 dyads of autistic mother and child were recruited and divided into intervention group and control group (15 mother child dyads each). Based on the content of PEERS social skill, cognitive behavior therapy was delivered in group format, through demonstration, role play and group exercise. At the same time, mother child dyads were trained using parallel social technology. Mothers and children with ASD were investigated using Parents Perception of Uncertainty Scale (PPUS), Patient Health Questionnaire 9 (PHQ-9), Chinese Version of the Beck Depression Inventory II(BDI-Ⅱ-C), Beck Anxiety Inventory (BAI), State Trait Anxiety Inventory(STAI-Form Y), and Autism Behavior Checklist (ABC), Cildhood Autism Rating Scale (CARS), and Social Communication Questionnaire (SCQ).
Results:
Changes in ASD symptom score in children and emotional score of mothers in the intervention group were less than 0. The total score of mother disease uncertainty(74.93±13.58, 90.40± 9.21 ), ambiguity(31.13±7.07, 38.93±4.73), lack of clarity information(11.93±2.09, 13.80±2.54), unpredictability(9.60±1.99, 12.07±2.89), significantly changed after intervention( t =-3.65, -3.55, -2.20, -2.72, P <0.05).
Conclusion
Social PEERS group intervention can enhance the social skills of children with ASD, reduce uncertainty of illness among mother of ASD children. Timely disease related information, guidance for mothers to actively participate in child care and training, might help to reduce cognitive bias, depressive and anxiety symptoms among mothers.
