Tumor vaccine is current hotspot of medicine. The article explicates the status quo, merit, side effects of tumor vaccine and brings up the ethic principle we should follow in clinic application.

Objective To explore the relationship between Mycoplasma pneumoniae infection and bronchial asthma, and provide a theoretical basis for the diagnosis and treatment of bronchial asthma .Methods 54 cases of children with bronchial asthma were selected as the observation group , treatment of children with herpes infection of the upper respiratory tract of 54 cases as the control group .Two groups of children were assayed by eosinophil count , interleukin-13 (IL-13), immunoglobulin IgE (MP-IgM) and Mycoplasma pneumoniae antibody detection .The positive rate of MP-IgM test in two groups were compared .According to the observation group , MP-IgM detection results were divided into MP -IgM positive group and MP -IgM negative group .Positive contrast group , MP-IgM MP-IgM negative group and a control group of children with eosinophil count , IL-13 and IgE were detected .Re-sults The positive rate of MP-IgM in children with observation group detection was 75.93%;The positive rate of MP-IgM in children with control group of detection was 27.78%;The positive rate of MP-IgM detection in chil-dren with the observation group was significantly higher than the control group ( p<0.05); The MP-IgM positive group, MP-IgM negative groups eosinophil count , the levels of IL-13 and IgE levels were significantly higher than that of control group (all p<0.05); In MP-IgM positive group, Eosinophils count (538.18 ±41.63) ×106/L, IL-13 level (253.27 ±40.36) ng/L and IgE level (327.13 ±56.91) kU/L were significantly higher than in MP -IgM negative group (all p<0.05).Conclusion The incidence of asthma is associated with mycoplasma pneumonia infection , and we should strengthen the detection and anti -infection treatment of Mycoplasma pneumoniae in children with asthma .

Objective To investigate the prognosis and its influencing factors of post-encephalitic epilepsy ( PEE) .Methods Fifty viral encephalitis patients with PEE were followed up, which prognosis was observed by whether their no seizures for at least 1 year or not.The clinical data of patients was collected, and the relate risk factors on the poor prognosis of PEE were analyzed.Results The disturbance of consciousness and the seizure presence during acute encephalitis, more than ten seizures in poor prognosis group were much higner than the good one (all P<0.01).The age, sex, seizure type, dysphrenia and neurological defect in acute encephalitis, status epileptisus in acute encephalitis, abnormal EEG and abnormal head CT/MRI had no correlation with poor prognosis ( all P>0.05 ) .Multiple logistic regression analysis demonstrated that disturbance of consciousness during acute encephalitis、seizures presence in acute encephalitis were independent risk factors of poor outcome in PEE ( OR=7.269,95%CI:1.22 -43.35,P=0.029;OR =22.893,95%CI:4.02 -130.43,P=0.000).Conclusion Disturbance of consciousness during acute encephalitis, seizure presence in acute encephalitis both are the single risk factors of poor prognosis in PEE.

AIM　Five Isoflavonoid glycosides were isolated from the pericarps of Sophora japonica L. and their structures were elucidated by chemical and spectral method as genistein 7,4′-di-O-β-D-glucoside(Ⅰ), sophorabio-side(Ⅱ), prunetin 4′-O-β-D-glucoside(Ⅲ), sophororicoside(Ⅳ) and genistin(Ⅴ). Among them, compounds Ⅰ, Ⅲ and Ⅴ were isolated from in S. japonica for the first time.

Analyzing and discriminating incorrect cases is a good method in medical statistics teaching,which can not only help students review the knowledge they have learned but also guide their self-learning.Teachers can start teaching based on the reverse thinking,propose incorrect cases with moderate difficulty and in accordance with the reality and arouse students' creativity through introducing the incorrect cases,reviewing the incorrect cases and conducting group discussion.

Objective To investigate the differences of the correct memorization of a sentence with auditory between the hearing-impaired children after rehabilitation training and normal children.Methods Twenty-four hearing-impaired children and 14 normal hearing children at age 3~4 were selected.The hearing-impaired chil-dren were divided into two groups according to the length of the hearing device.The closed-set sentence test of e-valuation of auditory response to speech (EARS)was used.Results 1 .In the first test,the hearing-impaired chil-dren with hearing device 2 to 3 years whose correct rate was 57.14%were close to the normal children whose correct rate was 58.93%.The hearing-impaired children with hearing devices 1 to 2 years whose correct rate was 32.5%were worse than the children of other two groups 2 .In the second test,the hearing-impaired children with hearing devices 2 to 3 years whose correct rate was 44.64% were better than the hearing-impaired children with hearing devices 1 to 2 years whose the correct rate was 22.5%.The normal children whose the correct rate was 55.36%were better than the children of other two groups.3.There was no significant difference to correctly memorize a sen-tence between three groups in the first test,but there was significant difference to correctly memorize a sentence between the hearing-impaired with hearing device 1 to 2 years and the hearing-impaired children with hearing de-vice 2 to 3 years (P<0.05),and significant differences also existed between the hearing-impaired with hearing de-vice 1 to 2 years and normal children (P<0.05),but there was no significant difference between the hearing-im-paired children with hearing device 2 to 3 years and normal children (P>0.05).Conclusion The length with hear-ing device strengthens the ability of hearing-impaired children in correctly memorizing a sentence after rehabilitation training.And the content of test affects the result ,it provides reference to the hearing-impaired children's auditory training .

Objective:To establish a trace chromogenic substrate method for the determination of anti-FIIαactivity in compound heparin sodium cream .Methods: The anti-FIIαactivity in compound heparin sodium cream was determined by a trace chromogenic substrate method according to the completely random design of experiment based on the amount reaction principle of 4*4 parallel lines in the biological test statistics method .Results:The calibration curve was linear within the range of 0.005 04 IU· ml-1-0.021 IU· ml-1(r=0.992).The average recovery was 101.6% with RSD of 2.76% (n=9).Conclusion: The method is accurate, reliable and reproducible , and can be used for evaluating the quality of compound heparin sodium cream .

Aim To obtain genetically engineered cytokine with potentialities of clinical application. Methods Anti-human hepatocellular carcinoma (HCC) single chain gene fragment(scFv25) was linked with human TNFα gene to form a fusion gene of anti--HCC cytokine， then it was subcloned into prokaryotic GST fusion expression vector pGEX 4T-1 and expressed in the host E.coli. Indirect immunofluorescent staining was performed on HCC cell smearing slides in order to evaluate the activity of purified aim protein, then initial tumor regression trial was carried out in nude mice bearing HCC to evaluate the targeting therapeutic value of scFv25-TNFα . Results scFv25-TNFα had similar specificity as parental antibody HAb25 for SMMC-7721 antigen. The tumor regression trial to the 2 mm HCC xenografts in nude mice showed that scFv25-TNFα had certain targeting cytotoxicity capacity and the efficiency was 3/3 with complete remission. The cytotoxicity of scFv25-TNFα was better than that of TNFα , whose efficiency was only 2/3 and no complete remission was seen. Conclusion scFv25-TNFα is the anti-HCC cytokine, which have certain potentialities of clinical application.

Objective To obtain the Hiwi gene encoding full-length and construct human Hiwi adenoviral vectors carrying green luorescence protein (GFP), and to establish foundation for a further study on Hiwi function and mechanism of inducing leukemia stem cell differentiation and apoptosis.Methods All coding areas of human Hiwi gene full length were amplified using method of overlapping extension PCR technology, and the full length coding aeras were inserted into the vector of Flag-IRES-hrGFP carrying GFP with Gateway technology to construct pDown-Hiwi-3 × flag-IRES-hrGFP. The cloning vector pDown-Hiwi-3 × flag-IRES-hrGFP and expression vector pAV. Des1d were used for homologous recombination reaction to obtain recombinant adenovirus vector pAV.Ex1d-Hiwi-3× flag-IRES-hrGFP.The positive clones were selected by PCR to extract the recombinant adenovirus plasmid and to pack into recombinant Ad-Hiwi-3 × flag-IRES-hrGFP adenovirus. Results The human recombinant Hiwi was successfully cloned and the recombinant adenovirus vector pAV.Ex1d-Hiwi-3×flag-IRES-hrGFP was found to be successfully constructed via restriction enzyme digestion and sequencing methods. The adenovirus vector pAV.Ex1d-Hiwi-3×flag-IRES-hrGFP was transfected into the HEK293A cells using lipofectamine mediated gene transfection method. Under fluorescence microscope, the transfected cells with green fluorescence could be observed.Conclusion The expression plasmid of adenovirus vector pAV.Ex1d-Hiwi-3 × flag-IRES-hrGFP is successfully constructed and it can express in HEK293A cells.

Objective To explore the incidence ofpost-encephalitic epilepsy (PEE) and its influencing factors in patients with viral encephalitis at acute symptomatic seizures (AESS).Methods Seventy-six patients with AESS were collected in our hospital from June 2006 to June 2012.All patients were followed up for at least two years.PEE was defined as patients appearing at least one unprovoked seizure after acute periods of encephalitis and confirming by EEG.The PEE incidence of patients with AESS was observed.With Logistic regression,the factors affected patients with AESS to be PEE were explored.Results Twenty-nine patients developed into PEE among the 76 patients with AESS,with PEE incidence of 38.2％.Multiple Logistic regression analysis indicated that the status epilepticus during acute encephalitis was the independent predictor of PEE (OR=8.000,95％CI:1.890-33.950,P=0.005),and antiepileptic drugs after acute encephalitis was the protective factor of PEE (OR=0.330,95％CI:0.120-0.940,P=0.038).Conclusion PEE incidence of patients with AESS is high;status epilepticus during acute encephalitis is an independent risk factor for patients with AESS to be PEE,and these patients should continue take antiepileptic drugs after acute encephalitis.
