1.Role of P38 mitogen-activated protein kinase in expressing of cyclooxygenase-2 in human umbilical vein endothelial cells

Objective: To investigate the role of P38 mitogen-activated protein kinase (P38MAPK) in expressing cyclooxygenase-2 (COX-2) in human umbilical vein endothelial cells (HUVEC), in order to study the role of P38MAPK on diabetic atherosclerosis. Methods: HUVEC are separately stimulated with high glucose, advanced glycosylation end products (AGE), high insulin and H_(2)O_(2), the expression of phospho-P38MAPK and COX-2 is detected; Simultaneously, HUVEC pre-treated with SB203580 (P38MAPK special inhibitor) are stimulated with above stimulating factors, the expression of COX-2 is detected. Results: High glucose, AGE, high insulin and H_(2)O_(2 ) can activate P38MAPK and increase the expression both of phospho-P38MAPK and COX-2 significantly; But the expression of COX-2 is inhibited by SB203580. Conclusion: P38MAPK regulates the expression of COX-2; P38MAPK is a upstream signal molecule of COX-2 and P38MAPK may be one of initial signals on atherosclerosis occurrence.
2.Efficacy of modified urethral dilatation in the treatment of female bladder neck obstruction

Objective:To investigate the efficacy of modified urethral dilatation in the treatment of female bladder neck obstruction.Methods:The clinical data of 33 female patients with bladder neck obstruction who underwent modified urethral dilatation in the Third People's Hospital of Qingdao from March 2015 to March 2020 were retrospectively analyzed. Before treatment, physical examination, routine urine examination, International Prostate Symptom Score, ultrasound examination, urodynamic examination and cystourethroscopy were performed to confirm the diagnosis. All patients were treated with α-blocker for more than 3 months, but obvious effect was not obtained. Under local anesthesia, they underwent modified urethral dilatation. After 3 months of treatment, International Prostate Symptom Score and urodynamic examination were performed to determine residual urine volume, the maximum urinary flow rate, and detrusor pressure at the maximum urinary flow rate. The curative effects of modified urethral dilatation were evaluated.Results:After modified urethral dilatation, dysuria was obviously alleviated in 25 patients. Eight patients who had no obvious improvement in dysuria were scheduled to undergo transurethral bladder neck incision. International Prostate Symptom Score after treatment was significantly lower than that before treatment [(15.18 ± 6.19) vs. (24.86 ± 7.26), t = 3.782, P < 0.001). Residual urine volume after treatment was significantly smaller than that before treatment [(53.69 ± 48.35) mL vs. (181.45 ± 92.15) mL, t = 15.328, P < 0.001]. The maximum urinary flow rate after treatment was significantly higher than that before treatment [(16.21 ± 4.22) mL/s vs. (7.91 ± 1.69) mL/s], t = 6.358, P < 0.001]. Detrusor pressure at the maximum urinary flow rate after treatment was significantly lower than that before treatment [(27.38 ± 5.13) cmH 2O vs. (57.15 ± 8.43) cmH 2O, t = 9.584, P < 0.001]. Conclusion:Modified urethral dilatation is an effective treatment method of female bladder neck obstruction. It can be used as a supplement for surgical treatment.
3.Comparison clinical curative effect of Sanxuemingmu tablets and compound Xueshuantong capsulein treatment of RVO patients with qi stagnation and blood stasis syndrome

Objective To investigate the efficacy and mechanism of Sanxuemingmu tablets and compound Xueshuantong capsule in treatment of RVO patients with qi stagnation and blood stasis syndrome.Methods 1200 cases of RVO patients with qi stagnation and blood stasis syndrome from April 2014 to April 2016 months in our hospital were selected,and randomly divided into two groups,600 cases in each group.The observation group was treated with Sanxuemingmu tablets,the control group were treated with compound Xueshuantong capsule,the curative effect and relative score of the two groups were compared.Results After treatment,the visual acuity of the two groups was significantly higher than that before treatment,the visual acuity of the observation group was higher than control group (P<0.05);The absorption of fundus hemorrhage in the observation group was significantly better than that in the control group(P<0.05);The total effective rate of FFA in observation group was significantly higher than that in control group (P<0.05);Compared with before treatment,the whole blood viscosity and plasma viscosity of the two groups were decreased,and the whole blood viscosity and plasma viscosity of the observation group were lower than those of the control group(P<0.05);The TCM syndrome scores of both groups were lower than those before treatment,and the observation group was significantly lower than the control group( P <0.05);The total effective rate and the total effective rate of the treatment group were significantly higher in the observation group than in the control group(P<0.05).Conclusion Sanxuemingmu tablets has significant clinical effect in treatment of RVO patients with qi stagnation and blood stasis syndrome,can accelerate the absorption of retinal hemorrhage,treatment of retinal vein obstruction qi stagnation of blood stasis,promote vision recovery and improve treatment efficiency.
4.Effect of isokinetic training on the function recovery of knee meniscus injuries following arthroscope

BACKGROUND: Rehabilitation training after sports injury is of so great importance in the remaining and relieving of exercise ability that the researches in thisfield need to develop.OBJECTIVE: To evaluate the influence of isokinetic training on the functional recovery of knee flexors and extensors and the muscle force around the joint after knee meniscus injuries receiving arthroscopy.DESIGN: Case-control observation SETTING: Department of Police Training, Liaoning Advanced Police Officer School PARTICIPANTS: A total of 20 patients with acute meniscus injury of lateral knee joint were selected from the Department of Surgery, Affiliated Hospital of Dalian Medical University between September 2004 and January 2005. They were randomly divided into experimental group and control group,including 11 cases respectively. METHODS: All the patients were treated with arthroscope operation, additionally the control patients received routine blocking, physiotherapy and massage, etc. To recover the function. From the 2nd to 4th days postoperative, the patients of experimental group began to carry out the functional rehabilitation, and received isokinetic exercise in both knees flexors and extensors with the Cybex-6000 isokinetic dynamometer 3 weeks later.MAIN OUTCOME MEASURES: Peak torque values, total work, torque accelerating energy and average power in both knee flexors and extensors at different angular velocities [60, 120 and 180(°)/s].RESULTS: A total of 22 patients were involved in the result analysis.①After arthroscope operation and isokinetic training, the range of joint movement were extended, and the maximum flexion angle changed from (132±25)° to (158±21)°. There were significant differences before and after training by t test (P < 0.01).②The experimental group had statistical significance compare with control group in the test index at 60, 120 and 180(°)/s(P < 0.05).CONCLUSION: The arthroscope combining with isokinetic training can speed up the rehabilitation after knee meniscus injury, enhance the muscle force around knee joint and maintain the stability of knee joint and motor ability.
5.Cell death induced by nanomaterials:research progress

With the wide use of nanomaterials in biomedicine,materials,chemicals and other fields,their environmental exposure and cellular toxicity have come to the attention of researchers as a new research area in the field of nanotoxicology and nanomedicine. Researches on the toxic effects of nanomaterials have not only provided of a theoretical basis for safety evaluation of nanomaterials ,but also promoted the have applications of nanotechnology. Numerous studies have revealed cell death is closely associated with the toxicity of nanomaterials. In this paper,we elaborated on the roles of three key cell death modes in nanotoxicity,including autophagy,apoptosis,and necrosis. Furthermore,the pos?sible mechanisms involved in these three modes were explored. All this will provide reference for safety evaluation of nanomaterials.
6.Comparison of the effect of myomectomy and uterine artery embolization in the treatment of uterine fibroids

Objective To compare the effect of myomectomy and uterine artery embolization in the treatment of uterine fibroids .Methods 158 patients who were pathologically diagnosed uterine fibroids were randomly divided into the uterine artery embolization group and myomectomy group ,79 cases in each group .The blood loss 3 days after operation,pelvic pain and pressure symptoms ,hospitalization time,follow-up and time of normal activity were recorded and compared.Results The pelvic pain,pressure symptoms,hospitalization time,cure rate,incidence rate of compli-cations and recurrence rate of the two groups had no statistically significant differences ( t=1.21,0.97,1.06,χ2 =0.98,1.21,1.32,all P>0.05).The postoperative bleeding score ,time of return to normal activity in the uterine artery embolization group were (5.8 ±0.5) points,(9.8 ±1.8) d,which were better than those of the myomectomy group[(4.9 ±0.4)points,(25.9 ±5.1)d](t=3.76,5.98,all P<0.05).Conclusion The effect of the myomec-tomy and uterine artery embolization treatment for uterine leiomyoma is similar ,and both have high efficiency and safe-ty,but the uterine artery embolization treatment has less impact on the body of patients .
7.Construction and expression of inset expression vector pIZT/V5-His harboring mIL-4

Objective To establish stable insect cell line expressing mIL4 on the basis of construction of inset expression vector pIZT/V5-His.Methods Amplified mIL-4 and pIZT/V5-His were treated with EcoRI and XbaI,and mIL-4 were ligated into pIZT/V5-His vector using T4 DNA ligase.Sf9 cells were transfected with recombinant DNA and ELISA was employed to detect soluble mIL-4 production by transfected Sf9 cells.Results Transfected Sf9 cells could significantly produce mIL4(1 mg?L~(-1)) compared!with control group(0.003 mg?L~(-1)).Conclusion Inset expression vector pIZT/V5-His is an ideal expression vector for mIL-4 production in transfected cells.
8.Construction of prokaryotic expression vector harboring different domains of STAT6

Objective To explore the influence of different domains of STAT6 expressed by the vector pGEX-6p-2 on signal transduction.Methods Different domains of STAT6 were amplified by PCR technique and ligated respectively into the prokaryotic expression vector.Recombinant DNA was transformed into HB101 cells.Transformants were lysed by supersonic method and lysates run on SDS-PAGE.Results Western blotting analysis showed a high level expression(60 000,56 000 and 50 000) of different domains of STAT6 SHST,DST and NST,and the empty vector only had GST expression.Conclusion Expression vector pGEX-6p-2 can be used for high level expression of different domains of STAT6.

10.Laparoscopic sacral colpopexy with prolene mesh in treatment of pelvic organ prolapse

The clinical data of 42 patients with pelvic organ prolapse, who underwent laparoscopic sacral colpopexy using prolene meshes were retrospectively reviewed. During the operation prolene meshes were placed in the rectovaginal septum and vesicovaginal septum ,and the uterus was preserved in all cases.The mean operation time was (92 ± 12)min and the mean blood loss during the operation was (98 ± 11 )ml.Postoperatively, both prolapse and symptoms were significantly improved ( P <0. 01 ) according to the pelvic organ prolapse quantification (POP-Q) system. The operation is an effective and minimally invasive new technique which offers a chance for patients with pelvic organ prolaps who desire to preserve uterus.
