1.Clinical study of preoperative immunoguiding therapy of intratumor injection of 131I-3H11 for gastric carcinoma

Objective To investigate the effect of intratumor injection of 131I-3H11 for gastric cancer (GC). Methods 16 patients with GC subjected to endoscopic intratumor injection of 131I-3H11 as a treatment group; 6 GC patients with FAM chemotherapy as a control group. Histological examination of the postoperative specimens of the two group were comparated. Results In treatment group, 75.0%(12/16) of cases were found to have morphological changes with karyopyknosis, karyorrhexis, coagulation of cytoplasm, and invasion of lymphocyte in mesochyma. Most of these changes were medial; but in control group no obvious morphological change was found. Conclusions The results suggest that GC subjected to endoscopic intratumor injection of 131I-3H11 preoperatively has promising application in the clinic.

4.Self-assembly and in vitro and in vivo evaluation of spherical crystallized interferon for sustained delivery.

It is a challenging and important project to prolong the in vivo half life of protein and peptide drugs by physicochemical methods without new molecular entities generation. Protein crystallization provides a new strategy for improving the stability and in vivo delivery of these drugs. We show here that recombinant human interferon-alpha (rhIFN) can form spherical crystals. The physical and chemical features of the crystals were characterized, and drug dissolution was determined in vitro. The pharmacokinetics of crystallized interferon after sc injection in rabbit at 1.5 x 10(7) U x kg(-1) was compared to that of soluble form. The crystals were characterized as mono-dispersed spheres, with yield of > 80%, mean diameter size of about 16 microm and crystallinity of 23.2%. The in vitro dissolution behavior of crystallized rhIFN was featured as low initial burst release (21% within the first 2 h) and prolonged cumulative dissolution time up to 72 h without biological potency lost. After sc administration of soluble and crystallized interferon in rabbits, the peak time (T(max)) and half life (t1/2) were prolonged from (1.80 +/- 0.45) h and (1.35 +/- 0.35) h to (13.20 +/- 2.68) h and (10.68 +/- 1.97) h, respectively. The corresponding peak concentration decreased from (1 411.10 +/- 575.28) U x mL(-1) to (721.37 +/- 206.55) U x mL(-1). PK/PD analysis indicated that (96.87 +/- 20.30) % of relative bioavailability was obtained. The research results of this work will provide important academic value and application prospect for improving clinical therapeutic effect and development of biomacromolecules delivery system for protein and peptide drugs.
5.Effect of Multichannel Functional Electrical Stimulation on Motor and Balance Function of Lower Extremity in Adolescents with Cerebral Palsy

Objective To evaluate multichannel functional electrical stimulation (FES) on motor and balance function of lower limbs in adolescent with cerebral palsy. Methods From January, 2014 to June, 2015, 20 adolescents with cerebral palsy were randomly divided into FES group (n=10) and control group (n=10). Both groups received routine exercise therapy. FES group received active and passive FES treadmill based on normal walking pattern. The Fugl-Meyer Assessment (FMA) was used to evaluate the motor function of lower limbs;the Berg Balance Scale (BBS) was used to assess the balance function, and the Gross Motor Function Measure (GMFM) was used to test the gross motor function. Results The scores of FMA, BBS and GMFM significantly improved 30 days after treatment (F>223.749, P<0.001), and the scores were higher in the FES group than in the control group (t>2.706, P<0.05). Conclusion FES treatment can improve the motor and the balance function of lower limbs in adolescents with cerebral palsy.
6.Analysis of enteral nutrition tolerance in ICU patients and nursing strategy

Objective To explore tolerance-related factors of enteral nutrition(EN) by nasogastric (NG) tube in ICU patients, and to provide several nursing strategies for improving the patients' tolerance. Methods 83 patients using EN with NG tube in ICU were collected.Tolerance-related factors of EN were analyzed by multivariate Logistic regression analysis. Results Among the 83 patients, 36 patients (43.3%) could tolerate very well, 47 patients (56.6%) couldn't tolerate.After treatment 27 patients' tolerance was fair, the other was poor.Multivariate Logistic regression analysis showed that age, APACHE-Ⅱ score, the time of beginning EN, albumin level, drug administration by NG tube or not were the major factors affecting tolerance of NG tube feeding. Conclusions Patients' feeding with NG tube was well-tolerated in ICU.Besides the factors from patients as the severity of disease status and level of serum albumin,the time of beginning EN, drug administration by NG tube were significantly correlated with tolerance.
7.Study of the effect of edaravone on the expression of intercellular adhesion molecule-1 and tumor necrosis factor-α in Rat brain of cerebral ischemia reperfusion

Objective To study the effect the edaravone on the expressions of ICAM-1,TNF-α after focal cerebral ischemia reperfusion in male rats.Methods 45 male SD rats were selected and randomly assigned into 3 groups:sham operation group(SO),ischemia reperfusion group(I/R),and edaravone treatment group(ED),each group had 15 rats.The expression of ICAM-1,TNF-α were checked by immunohistochemistry.All data in the groups were analyzed with t test.Results The expression of ICAM-1 and TNF-α in the I/R group and ED group were higher than the SO group(t=21.919,27.758,26.576,17.395,32.821,43.021,14.195,31.828,21.201,10.195,25.768,17.900,all P<0.05)at reperfusion 3 hours,6 hours,and 12 hours.The expression of ICAM-1 and TNF-α in the I/R group were higher than the ED group(t=9.623,13.845,7.201,11.313,all P<0.05)at reperfusion 3 hours and 6 hours.Conclusion Edaravone may exert the neuroprotective effect against the cerebral ischemia in rats by decreasing the expression of ICAM-1 and inhibiting the activation of TNF-α.This process could inhibit free radical generation.
8.Drinking in male HBV carriers in the outpatient department.

OBJECTIVE:
To understand the relation among drinking, characteristics and emotion in outpatient male carriers with hepatitis B virus (HBV), and to provide reference for alcohol intervention.
METHODS:
We used alcohol use disorders identification test (AUDIT), self-rating anxiety scale (SAS), and Beck depression inventory (BDI) to investigate 980 male HBV carriers in the outpatient department.
RESULTS:
The questionnaires were responded by 544 people with drinking experience for nearly a year (drinking rate 58.18%). The prevalence of moderate drinking was 37.8% (354 patients), hazardous and harmful drinking 17.97% (168 patients) and alcohol dependence 2.35% (22 patients). In groups with different ages, education levels, occupations, income and symptoms, the constituent ratio of the hazardous harmful drinking and alcohol dependence (AUDIT 7-26 scores) was different (P<0.05). The total score in SAS in the alcohol dependence group was much higher than that in the normal group (35.95±11.55 vs 29.78±0.46, P =0.020); the total score in SAS and in BDI was significantly higher in the alcohol dependence group than that in the moderate drinking group and the hazardous harmful drinking group (35.95±11.55 and 10.45±8.95 vs 29.65±7.97 and 6.35±5.65 vs 29.68±7.06 and 6.44±5.27, respectively).
CONCLUSION
Male HBV carriers of 30-49 years old, with education level lower than elementary school or higher than bachelor degree, cadres/professionals with high income and major symptoms show higher harmful/alcohol dependence (AUDIT 7-26 points). Anxiety and depression in the alcohol-dependent male hepatitis B virus carriers are obviously higher than in the moderate drinking group and the hazardous harmful drinking group.

9.Impact of Ticagrelor on Myocardial Microcirculation in Patients of Non-ST Elevation Myocardial Infarction After Percutaneous Coronary Intervention

Objective: To explore the impact of ticagrelor on myocardial microcirculation in patients of non-ST elevation myocardial infarction (NSTEMI) after percutaneous coronary intervention (PCI). Methods: A total of 80 NSTEMI patients treated in our hospital from 2015-03 to 2015-12 were enrolled. All patients received coronary angiography (CAG) and PCI, they were randomly divided into 2 groups: ticagrelor (TA) group and clopidogrel (CA) group, n=40 in each group. The baseline condition, PCI related parameters, plasma levels of SOD, MDA at pre- and 24h, 72h post-PCI were compared, the incidence of major adverse cardiac events (MACE) within 30 days after PCI was also compared between 2 groups. Results: The baseline condition, proportion of infarction-related vessels, average length and diameter of implanted stents were similar between 2 groups, all P>0.05. Compared with CA group, TA group had the less post-operative CTFC in LAD and RCA, P<0.05; while CTFC in LCX was similar between 2 groups, P>0.05. The ratios of acute pre-operative, post-operative TIMI grade 3 and slow flow incidence were similar between 2 groups, P>0.05. The peak values of CK-MB and cTnI in TA group were lower than CA group, P<0.05. Compared with baseline condition, MDA levels at 24h and 72h post-operation were increased in both groups, all P<0.001; compared with CA group, TA group had the lower MDA level at 24h post-operation, P=0.023, MAD was further reduced at 72h post-operation in TA group, P=0.043. Compared with baseline condition, SOD levels at 24h and 72h post-operation were decreased in both groups, all P<0.001; compared with CA group, TA group had the higher SOD level at 24h post-operation, P=0.013, SOD was elevated at 72h post-operation in both groups, the elevation was more obvious in TA group, P=0.049. The incidence of MACE was similar between 2 groups within 30 days of PCI,. Conclusion: Ticagrelor could improve myocardial microcirculation in NSTEMI patients after PCI; it was safe and without obvious adverse effects.
10.Changes of the level of phosphorelated ERK1/2 in the spinal trigeminal nucleus of migraine model rats

Objective To explore the role of ERK1/2 in the central pathogenesis of migraine.Methods Healthy adult male SD rats were randomly divided into five groups:normal group (group C),sham operation group(group C),migraine model group(group M),DMSO group (group D)and PD-98059group (PD group),with 12 rats in each group.The extracellular discharge frequency in the spinal trigeminal nucleus was recorded and ERK1/2 phosphorylation was tested.Results (1) The percentage of extracellular discharge frequency change:Two hours after treatment,the percentage of discharge frequency change was (325.9±47.32)%.The percentage of extracellula discharge frequency change in group M (325.9±47.3)% was higher than that in group N (100.0± 0.0) % and group C(107.3± 16.4)%.There was no significant difference in the percentage of discharge frequency change between group D(319.3±42.5) % and group M (325.9±47.3) %.The percentage of discharge frequency change in group PD(218.5±31.7)% was lower than that in group M(325.9±47.3)% and group D(319.3± 42.5)%.(2) ERK1/2 phosphorylation:the ERK1/2 phosphorylation in group M and group D was higher than that in group N and group C.There was no significant difference in ERK1/2 phosphorylation between group D and group M.The ERK1/2 phosphorylation in group PD was lower than the other four groups.Conclusion During the process of central sensitization to migraine,neuronal excitability and ERK1/2 phosphorylation were increased.ERK1/2 inhibitor (PD98059) reduced ERK1/2 phosphorylation and neuronal excitability.These indicated that ERK1/2 may play a role in central sensitization of migraine in rats.
