1.The effects of valsartan on the proliferation,migration,p-ERK1/2 and p-P38 MAPK expression of vascular smooth muscle cells

Objective To investigate the effect of valsartan on the proliferation and migration of rat aortic smooth muscle cells and the expression of p-ERK1/2 ,p-P38 promoted by angiotensinⅡ (AngⅡ ) .Methods VSMCs from the rat thoracic aorta were cultured by attachment-block culture .VSMCs from3-5 passages were used .The proliferation of VSMCs was tested by MTT meth-od .Pipettetip wounding injury was used to access the migration of VSMCs .The expression of ERK1/2 ,p-ERK1/2 ,P38 and p-P38 of VSMCs was detected by Western Blot .Results (1) AngⅡ could significantly promote the proliferation of VSMCs .The prolifer-ation of VSMCs promoted by AngⅡ could be inhibited by Valsartan and PD98059 .Valsartan inhibited the proliferation of VSMCs in a concentration-dependence .SB23015 could significantly promote the proliferation of VSMCs .(2 ) The migration activity of VSMCs in the Ang Ⅱ was increased significantly .Valsartan and PD98059 inhibited Ang Ⅱ-induced migration of rat VSMCs and SB23015 was on the contrary .(3) Valsartan and PD98059 could inhibit AngⅡ-induced VSMCs intracellular p-ERK1/2 protein ex-pression ,and this role was enhanced by SB23015 .Valsartan and SB23015 could inhibit Ang Ⅱ-induced VSMCs intracellular p-P38 protein expression ,and PD98059 had no significant effect .(4)VSMCs had no significant effect on cell proliferation ,migration and p-ERK1/2 ,p-P38 protein expression under valsartan alone .Conclusion (1)Valsartan could inhibit Ang Ⅱ-induced proliferation and migration of VSMCs in a concentration-dependence .(2)The effects that valsartan inhibits the proliferation and migration of Ang Ⅱ-induced VSMCs might be related to its inhibiting Ang Ⅱ-induced p-ERK1/2 protein expression .(3)p-P38 might play a negative reg-ulatory role on the AngⅡ-induced p-ERK1/2 activation .
2.A Primary Research in Marriage Quality of Primary Male Infertility Couples

Objective: To investigate the marriage quality of the couples with primary male infertility in order to provide consulting for them.Methods: 37 patients with male infertility and their mates were rated by Olson questionnaire.Results: ①Compared with other married couples,the scores of satisfaction degree of marriage,communication between couples and sexual life of the couples with male infertility are higher and the mates of these patients have higher scores than those of married female in the factors of resolving conflicts,extracurricular activities,relationship with relatives and friends.②The marriage quality between the patients with male infertility and the new wedding males has no difference,and the scores of the extracurricular activities of the wives of the patients with male infertility were higher than those of the new wedding wives,and the scores of the economic arrangement were significantly lower in the patients' wives.Conclusion: Male infertility has no obvious influence on marriage quality.
3.Effect of Cationic Liposome Formulation on Human Carcinoma Hepatic SMMC-7721 Cells' Uptake of Tenofovir and Cytotoxicity

OBJECTIVE:To investigate the effect of liposome formulation in promoting hepatocytes' uptake of tenofovir and cytotoxic effect. METHODS: The tenofovir cationic liposomes were prepared by tert-butyl lyophilization, and its encapsulation efficiency and physico-chemical property were determined. Using human hepatic carcinoma cells SMMC-7721 as a model to investigate the effect of liposome formulation in promoting hepatocytes' uptake of tenofovir. MTT assays were used to examine the toxicity of tenofovir cationic liposomes in different conditions. RESULTS: The prepared tenofovir cationic liposomes had an encapsulation efficiency of (88.3?1.6)%, a size distribution of (278.4?67.6)nm and a Zeta potential of (31?5)mV. The results showed that the liposome formulations containing galactose and PEG modifying lipids resulted in significantly higher and prolonged drug accumulation inside cells as compared with free drugs. The cell survival rates were above 80% when the tenofovir liposome was at a concentration of 7.5?g?mL-1 and lipids at a concentration of 30?g?mL-1, and little toxicity was noted. CONCLUSION: The prepared cationic liposome can greatly enhance hepatocytes' uptake and protection of tenofovir, which is expected to become a highly effective drug delivery system of antiviral drugs such as tenofovir etc.

4 cm in diameter(Z=1.966,P=0.048).A positive relation was seen between HSP70 expression and CD8~+ T cell number at the border zone of RFA sphere(r=0.489,P=0.046).The HSP70 expression and the CD8~+ T cell number at the border zone of RFA sphere were higher in recurrence-free patients than in recurrent patients,respectively(Z=2.009,P=0.045;Z=2.007,P=0.045).Conclusions Following the RFA,the HSP70 expression and the CD8~+ T cell number at the border zone of RFA sphere will be elevated,indicating that RFA induces an immune response against tumor antigens and a tumor-specific T cell reaction.
8.Clinical study of trimetazidine,telmisrtan and low molecular weight heparin in treatment of chronic pulmonary heart disease in acute exacerbation

Objective To explore the effect of trimetazidine, telmisartan and low molecular weight heparin (LMWH) on improving cardiopulmonary function, hemorheology and clinical symptom of chronic pulmonary heart disease in acute exacerbation. Methods A total of 96 patients with chronic pulmonary heart disease in acute exacerbation were randomly divided into group A (40 cases) and group B (56 cases ).The group A was treated with general method and the group B was treated with trimetazidine, telmisartan and LMWH besides general method. After 3 weeks' treatment, ca_rdiopulmonary functional parameters,hemorbeology parameters and clinical symptoms were observed. Results After treatment, the left ventricular ejection fraction (LVEF), and the ratio of forced expiratory volume in one second and forced vital capacity (FEV_1/FVC ) improved in group B (P<0.05 ), and there were significant difference in LVEF, FEV_1/FVC after treatment between two groups (P<0.05). The clinical total effective rate in group B (92.9%, 52/56 ) was significantly higher than that in group A (67.5%, 27/40 ) ( P<0.05 ). Conclusion Trimetazidine, telmisartan and LMWH is effective to improve patients' cardiopulmonary function, hemorheology parameters and clinical symptoms in the treatment of chronic pulmonary heart disease in acute exacerbation.
9.Effects of VEGF on proliferation and morphology of hBMSCs

Objective To observe the influences of VEGF transfection on the proliferation and ultrastructure of hBMSCs. Methods Three groups were enrolled in the experiment: group of Ad-VEGF transfection, group of empty Ad transfection, and the control group. Cells were observed continually by inverted phase contrast microscope, and stained by HE. Proliferation of cells was tested by MTT and by flow cytometry analysis. Results Histological observation and observation through inverted phase contrast microscope showed that the cells were of the similar morphology in the 3 groups. As time elapsed, the amount of cells increased, but still no obvious differences were found in optical density (OD) value of hBMSCs.Groups B, C, A had a similar percentage of DNA G1 period and a similar proliferation index (PrI) ( P >0. 05). Conclusion Transfeetion of VEGF has no obvious influence on the prohferation and morphologyof hBMSCs in vitro.

Objective To evaluate the safety and efficacy and explore the clinical application value of the enhancement controlla-ble output channel narrowed ileal bladder suspension surgery and traditional ileal neobladder after total cystectomy.Methods From January 2001 to August 2009,42 patients with bladder cancer received enhancement controllable output channel narrowed ileal blad-der suspension surgery after total cystectomy;and 46 patients received ileal neobladder after total cystectomy.Their clinical data, perioperative situation,postoperative complications and tumor progress were analyzed.Results In regard to the blood loss,postop-erative hospital stay,and postoperative recent or far complications,both surgical methods had no significant statistical difference (P >0.05).The group of Enhanced controllable output channel narrowing ileal bladder suspension surgery was good control of uri-nation after operation(P <0.05).Conclusion Enhancement controllable output channel narrowed ileal bladder suspension surgery has good clinical effect and safety.It is especially suitable for the patients who need control inurine and don′t accepted orthotopic neobladder.
