1.Study on plasminogen and BDNF expression in the hippocampus of depression rat mode and its relationship

Objective To study on the relationship between plasminogen and brain-derived neurotrophic factor(BDNF),and explain the molecular mechanism of depression ,then provide new clew for diagnosis and treatment of depression .Methods The chronic unpredictable mild depression rat mode was established ,then depression symptoms including absence of delight ,the decline of actions and activities ,and weight reduction of rat were tested .The levels of individual plasminogen and BDNF in hippocampus were determined by Western blot .Results The expression of BDNF and plasminogen in depression rat mode and control group was significantly different(P<0 .01) ,and there was a positive correlation between BDNF and plasminogen(r=0 .65 ,P<0 .01) .Accord-ing to the linear-regression analysis ,there was a dependence relationship between them(r2 =0 .423) .The equation of regression was YBDNF=0 .750XPlasminogen +0 .201 .Conclusion Stress could affect the growth and survival of nerve cell ,which lead to the depression behavior of rats ,meanwhile ,the decline of plasminogen and BDNF levels ,the positive correlation between them illustrate that plas-minogen and BDNF take part in the mechanism of depression .

3.Effects of lead exposure on bone lead mobilization and bone microstructure in pregnant rats

Objective:
To investigate the effect of lead exposure before pregnancy on bone lead mobilization and bone microstructure in pregnant rats, so as to provide the evidence for illustrating the potential mechanisms of bone lead mobilization during pregnancy.
Methods:
Twenty-six weaning female specific pathogen-free (SPF) rats of the Wistar strain were randomly divided into the exposure group and the control group. Rats in the exposure group were given 0.05% lead acetate solution for weeks, while animals in the control group were given 0.05% sodium acetate solution. Then, rats in both groups were given distilled water. Following removal of lead exposure for 4 weeks, female rats were co-caged with healthy males at the same age until pregnancy. The blood, femur and tibia specimens were collected from female rats on days 3 (GD3), 10 (GD10) and 17 (GD17) at pregnancy, and the blood and bone lead levels were measured using inductively coupled plasma mass spectrometry (ICP-MS). The unilateral rat femur was scanned using micro-computed tomography (micro CT), and the microstructure changes of cortical and trabecular bones were investigated. The structural and morphological changes of rat femur were observed using hematoxylin-eosin (HE) staining.
Results:
During the study period, satisfactory mental status and activity and good coat glossiness were observed in female rats in both groups, and there was no significant difference in the increase of rat body weight between groups. The blood lead level at GD17 and bone lead levels at GD3, GD10 and GD17 were significantly higher in rats in the exposure group than in the control group (P<0.05), and the trabecular bone lead level was significantly lower in rats in the exposure group at GD17 than at GD10 (P=0.015). The trabecular bone lead level correlated negatively with blood lead level (r=-0.578, P=0.049), and bone lead contributed 26.8% to blood lead. The bone mass, trabecular number, thickness and density of female rat trabecular bones all reduced in the exposure group at GD17, with an increase in trabecular space, and the proportion of trabecular areas reduced by 27.34% in the exposure group relative to the control group (t=2.851, P=0.046).
Conclusions
Lead exposure before pregnancy promotes the release of lead from trabecular bones into blood and affects bone microstructure in rats. There is bone lead mobilization during late pregnancy.
4.Efficacy of 308-nm excimer light in the treatment of stable vitiligo

Objective To evaluate the efficacy and safety of 308-nm excimer light in the treatment of stable vitiligo. Methods Thirty patients with stable vitiligo were enrolled in this clinical trial. All the subjects received the treatment with 308-nm excimer light on a twice-weekly schedule for 3 months. Results The repigmentation rate was 95.0%, 75.0% and 66.7% for lesions in the face and neck, trunk and limbs, with the treatment sessions averaging 10.22 ± 1.60, 19.10 ± 2.38, 37.74 ± 3.06, respectively, and accumulative irradiation dose averaging 7.50 ± 3.45, 10.60 ± 1.01, 18.56 ± 3.05 J/cm2 respectively. Significant differences were observed in the repigmentation rate and treatment sessions between the lesions in the face and neck, trunk and limbs (all P < 0.05). No severe side effects were seen during the treatment. Conclusion 308-nm excimer light is effective and safe for the treatment of vitiligo.

To investigate the biological role of snake venom cystatin(sv-cystatin) in tumor progression, the cDNA of sv-cystatin amplified by PCR from pUC18-cystatin plasmid was cloned into methanol-inducible expression vector pPICZ?A. The linearized recombinant plasmid pPICZ?A-cystatin was transfered into Pichia pastoris, strain GS115 by electrophoration. Transfermants with phenotype Mut+ selected were identified by PCR analysis and induced in 1.0% methanol. The reombinant sv-cystatin protein was examined by SDS-PAGE, Western blot analysis. The molecular mass of expression product was about 14 kDa and approximately 16 mg/L of recombinant sv-cystatin was produced from one of GS115-cystatin transformants. Objective To explore the values of CT virtual gastroscopy (CTVG), three dimensional (3D) and axial imaging by multi slice spiral CT (MSCT) in the preoperative assessment of gastric carcinoma. Methods Fifty two patients with gastric carcinoma proven by means of biopsy underwent CTVG, 3D imaging using the surface shaded display (SSD) and Raysum, and axial imaging. MSCT was first performed after the ingestion of gas, and a second tri phase MSCT scan was performed after the ingestion of water. The results were compared with those of fibrous gastroscopy (FG) and surgical pathological findings. Results The detection rates of gastric lesions with CTVG, 3D and axial imaging by MSCT were 98.1%, 96.2%, and 96.2%, respectively. 8.Effects ofYinqiao Powder on Mouse Models with Upper Respiratory Trace Mucosal Immunity Dysfunction Infected with Influenza Virus A

Objective To observe the effect ofYinqiao Powder on the mouse models with upper respiratory trace mucosal immunity dysfunction infected with influenza virus A, and explore mechanism of action.Methods The mouse models of upper respiratory trace mucosal immunity dysfunction induced by cold stimulation with the influenza A/PR/8/34 (H1N1) virus through the nasal cavity were established. The mice were randomly divided into normal group, model group, positive medicine (Ribavirin) group, andYinqiao Powder group. All administration groups received gavage with relevant medicine, and then mortality, the life prolonging rate, average survival time and the lung index of each group were observed.Results Compared with the model group, the mortalities in positive medicine group andYinqiao Powder group decreased significantly (P<0.05,P<0.01), with longer survival time. The lung indexes in positive medicine group andYinqiao Powder group decreased significantly (P<0.05,P<0.01), and the inhibition ratios of lung index were 35.5% and 24.6%, respectively.ConclusionYinqiao Powder can realize the protective effects on upper respiratory infection through upregulating the mucosal immunity of the upper respiratory tract of mouse models.
9.Serum anti-phospolipase A2 receptor antibodies and glomerular IgG4 in the diagnosis of membranous nephropathy

Objective To discuss the relationship between serum anti-Phosphalipase A2 receptor (PLA2R) antibodies and glomerular IgG4 subclass in patients with membranous nephropathy and evaluate the diagnostic value of the two markers.Methods Patients diagnosed as membranous nephropathy from October 2011 to April 2014 in Peking Union Medical College Hospital were included and divided into IMN and SMN groups accoding to their clinical diagnosis.Serum anti-PLA2R antibodies and glomerular IgG subclasses were both detected by indirect immunofluorescence assay.Receiver operator characteristic curves were used to evaluate the diagnostic efficiency of anti-PLA2R antibodies and glomerular IgG4.Results Prevalence of serum anti-PLA2R antibodies of IMN patients was 69.5% (41/59); prevalence of MLN patients was 4.8% (1/21).Within the IMN group,thirty-five patients showed positive results of both serum anti-PLA2R antibodies and glomerular IgG4; Six patients were positive for serum anti-PLA2R antibodies but negative for glomerular IgG4; Seventeen patients were positive for glomerular IgG4 but negative for serum anti-PLA2R antibodies; one patient was negative for both tests.The sensitivity of serum anti-PLA2R antibody was 69.5% and the specificity was 95.2%; the sensitivity of glomerular IgG4 was 89.8% and the specificity was 52.3%.The sensitivity of the combined marker consisting of serum anti-PLA2R antibody and glomerular IgG4 was 59.3% and the specificity was 100%.Four out of the six patients secondary to HBV infection,one out of the three patients secondary to Sj(o)gren syndrome,one out of the three patients secondary to malignant tumor showed positive results of serum anti-PLA2R antibodies.Conclusions Serum antiPLA2R antibodies were of high prevalence among IMN patients; the prevalence among SMN patients varied with etiologies.Results of serum anti-PLA2R antibodies and glomerular IgG4 were helpful to rule out secondary etiologies in the diagnosis of membrnous nephropathy.
10.Comparison of the effects of Baimai ointment and baclofen in stroke patients with spasticity

Objective To compare the effects of Baimai ointment and baclofen in stroke patients with spas-ticity.Methods 84 cases accompanied by limb spasticity in stroke patients by digital table were randomly divided into Baimai ointment group and baclofen group,42 cases in each group.The Baimai ointment group were treated with Baimai ointment on the spastic limbs,the baclofen group received oral baclofen tablets 30 -75mg/days for 2 weeks, 4 weeks,8 weeks.The curative effects of the two groups were compared before and after treatment.Results Before and after treatment in the two groups,the levels of spasticity,pain and activities of daily living (ADL)differences were statistically significant and Baimai ointment in the treatment of spasm.After 4 weeks and 8 weeks,the Ashworth score of the Baimai ointment group were (1.59 ±0.46)points,(0.89 ±0.56)points,and those of baclofen group were (1.75 ±0.64)points,(1.45 ±0.48)points,the differences were statistically significant(t values were 2.916, 3.367,all P <0.05).After 2 weeks,4 weeks and 8 weeks,the VAS score of the Baimai ointment group were (2.72 ± 0.54)points,(2.02 ±0.24)points,(1.24 ±0.12)points,and baclofen group were (3.56 ±0.44)points,(3.15 ± 0.48)points,(2.58 ±0.26)points,the differences were statistically significant(t values were 2.975,3.359,5.416, all P <0.05),activities of daily living (ADL)was higher than that of the baclofen group.After 8 weeks,the MBI score of the Baimai ointment group was (64.46 ±10.78)points,and baclofen group was (50.74 ±9.18)points,the difference was statistically significant between the two groups (t values was 3.562,P <0.05).Conclusion Baimai ointment has the better antispasmodic effect than baclofen in patients with stroke.
