1.Study on the role of local immunity in the female genital tract with nongonococcal urethritis

Objective To study the role of local immunity in the female genital tract with non- gonococcal urethritis(NGU).Methods Forty-four cervical secretions from patients with NGU were collected.After treatment with Azithromycin,22 cases were followed up.One month later,pathogen was eliminated in 15 patients.Another 27 un-infected people were included in our study.The level of secretory immunoglobulin A(SIgA)was detected using the automatic special protein analysor,and interferon-?(IFN-?)was quantified by enzyme-linked immunosorbent assay(ELISA).All data were analyzed with ANOVA.Results The levels of SIgA and IFN-?in NGU infected group were(34.29?15.92)mg/L and(97.13?24.00)ng/L respectively,while those of uninfected group were(47.57?19.61)mg/L and(120.71?33.78)ng/L,respectively(all P0.05).Conclusions SIgA and IFN-?secreted by genital tract mucosa may help pre- vent and resist the local NGU infection.However,the effect is limited,and is insufficient to eliminate infection completely and prevent reinfection.
3.THE RESEARCH ON THE RECOMBINANT ADENO-ASSOCIATED VIRUS AS A VECTOR FOR THE GENE THERAPY OF LIVER CANCER

Objective To explore the feasibility of recombinant adeno-associated virus (rAAV) as a vector for the gene therapy of liver cancer. Methods The rAAV/enhance green fluorescein protein (EGFP) recombinant was prepared by the routine method of two plasmids cotransfection. Results The experiment showed that one 10cm plate could produce 107-108 infection unit recombinant by the method of two plasmids cotransfection, and the transduction of HepG2 cell was increased with the increase of infection dosage of rAAV. About 100 multiplicity of infection (MOI) AAV vector could make all the tumor cell light. Conclusion Liver cancer cell can be efficiently transduced by rAAV, and AAV vector may be a valuable vector for the gene therapy of liver cancer.
4.Effect of ritodrine hydrochloride combined with radix salviae miltiorrhizae on fetal growth restriction

Objective Ritodrine hydrochloride(RH) combined with radix salviae mihiorrhisa(RSM) is ap- plied to treat fetal growth ristriction(FGR). Curative effect was recorded during observation. Methods 62 pregnant women with slowly growing fetuses were divided into two groups randomly. Group A(test group) :including 31 preg- nant women who received RH combined with RSM treatment. Group B(control group) :including the other 31 preg- nant women who only received RSM treatment. Group B received 250ml of fat emulsion injection. One time every three days within one week. At the same time,gronp B also received injection of 500ml 10 % dextrose, coenzyme A 100U ,ATP 40mg,vitamin C 2g,dextran-40 and compound salvia injection 10ml one time a day,ten days in all. Be- sides the above-mentioned medicament for group B, group A received RH 100ml injection every day. Ten days was a period of treatment. After the treatment, uterine height, abdominal circumference, weight were measured and bipari- etal diameter, head circumference, abdominal circumference were checked by B-type ultrasonic inspection. After birth,weight of the baby and weight of placenta were measured too. Results Measurement indexes from group A, weight of the new-born baby and weight of placenta are obviously higher than those from group B. Conclusion RH combined with RSM can obviously extend blood vessel, loose placenta, improve blood circulation, accelerate cell metabolism, make microeirculation better, reduce permeability of capillary vessel, keep placenta's function and help foetus' growth.
5.Cloning and sequence analysis of 4 amastin genes from Leishmania parasites

Objective To clone amastin coding genes from different strains of Leishmania parasites. Methods Using amastin cDNA sequence as the reference, dbEST data base established by National Center Biotechnology Information (NCBI), USA, was searched by BLAST tool. A 309 bp DNA fragment of Leishmania major was found and used as the probe for the screening of a DNA library. The amastin gene of Leishmania major Abdou was cloned and sequenced. Specific primers were designed and amastin genes for Leishmania mexicana WR972, Leishmania brizeliensis and Leishmania amazonensis joseph were amplified by polymerase chain reaction. Results The amastin genes from 4 strains of Leishmania parasites were cloned and sequenced. It was found that all 4 amastin genes contained unique open reading frame of 552 bp and encoded amastin protein of 183 amino acid residues. Conclusion The amastin genes of 4 strains of Leishmania parasites were successfully cloned.
6.Study on the distribution of auto-antibodies against platelet in adults and children with primary immune thrombocytopenia in Nanning

Objective Study on the distribution of auto‐antibodies against platelet in adults and children with primary immune thrombocytopenia(ITP) .Methods Platelet auto‐antibodies were detected in 83 ITP patients and 58 non‐ITP patients by enzyme linked immunosorbent assay (PAKAUTO kit) .Anti‐GP Ⅱ b/Ⅲ a ,GP Ⅰ b/Ⅸ and GP Ⅰ a/Ⅱ a auto‐antibodies were analyzed for 46 a‐dults and 37 children with ITP .Results The positive rate of autoantibody was 66 .27% in ITP patients ,which was much higher than that of non‐ITP patients ,the difference was significant(P < 0 .05) .As for ITP patients ,female ITP patients in adult group were 28 ,while in children group were 24 ,the difference was not significant(P > 0 .05) ,but the morbidity of ITP in females was higher than that of males in both of the two groups ,the difference was significant(P< 0 .05) .There was no significant difference on the distribution of platelet auto‐antibodies in adult and children group(P> 0 .05) ,the main of them was resistance against GP Ⅱ b/Ⅲ a and GP Ⅰ b/Ⅸ antibodies .Conclusion The detection of platelet auto‐antibodies could distinguish ITP and non‐ITP patients ,and GP Ⅰ a/Ⅱ a has an important reference value about therapy .
7.Relationship between Complications and Neurologic Subtype or Gross Motor Function in Preterm Infants with Cerebral Palsy

Objective To investigate the distribution of complications and the relationship with neurologic subtype and gross motor function in preterm infants with cerebral palsy (CP). Methods The type, grade of Gross Motor Function Classification System (GMFCS), intelligence,speech, ophthalmologic consultation, brainstem auditory evoked potential and electroencephalogram of 135 preterm infants with CP were reviewed. Results There were 284 complications in total, (2.10±1.33) per child, and was significantly different among various types of CP (F=5.50, P<0.001). The incidence of mental retardation and speech disorder was significant different among various types (P<0.05). The incidence of mental retardation, speech disorder, visual impairment and epilepsy increased significantly (P<0.05) in spastic quadriplegia infants,compared with those with diplegia and hemiplegia. The incidence of mental retardation, speech disorder, visual impairment and musculoskeletal disorder was significantly different (P<0.05) among various grades of GMFCS. The frequency of complications was more in children unable to walk (GMFCS Ⅳ~Ⅴ) than able to walk (GMFCS Ⅰ~Ⅲ) for children over 2 years old (t=70.05, P<0.001). Conclusion The incidence of mental retardation, speech disorder, visual and hearing impairment, secondary musculoskeletal disorder and the multiple disorders are related with neurologic subtype and/or the grade of GMFCS.
8.Clinical efficacy of Shuxuening injection combined with epalrestat for diabetic nephropathy

Objective To explore the clinical efficacy of Shuxuening combined with epalrestat in the treatment of diabetic nephropathy. Methods Eighty patients from March 2014 to June 2015 in Metabolic Disease Department of the Second Hospital of Tianjin were randomly divided into observation group and control group with 40 patients in each group.The control group received epalrestat, and the observation group received Shuxuening injection on the baisis of control group. The related indicators were compared between two groups.Results After treatment, the 24 h-urine microalbumin, plasma low density lipoprotein and serum creatinine in observation group [(239.31 ±106.54)mg/L,(2.45 ±0.55)mmol/L,(95.54 ± 22.13)mol/L]were lower than those in control group [(349.90 ±148.40),(3.41 ±0.52),(108.76 ±34.30)](P<0.05).The β2-microglobulin (β2-MG) and plasma high-sensitive C-reactive protein (hs-CRP) in observation group [(0.39 ±0.06),(6.31 ±1.58)mg/L]were lower than those in control group [(0.49 ±0.12),(7.89 ±1.35)](P<0.05).The total efficacy in observation group was higher than that in control group (70.0%vs.45.0%,P<0.05).Conclusion Shuxuening injection joint epalrestat has the exact efficacy in treatment of patients with diabetic nephropathy.
9.The relationship between eosinophil and in-stent restenosis in ACS patients

Objective To investigate the relationship between eosinophil and in-stent restenosis in acute coronary syndrome patients. Methods One hundred and fifity-one ACS cases werenenrolled in this study. According to the results of coronary angiography (the stented segment lumen loss over 50% was judged to be ISR), patients were divided into the restenosis group and the non-restenosis group. Results Based on the logistic analysis, no significant association was found between eosinophil and ISR, and even after adjustment of related risk factors (P > 0.05). The stratification analysis showed that the high level of eosinophil might increase the risk of ISR in ACS patients with history of hypertension (P = 0.038) and myocardial infarction (P = 0.032). Conclusion Eosinophil may be associated with the risk of ISR in ACS patients with history of hypertension and myocardial infarction. The underlying mechanisms need to be elucidated in further study.
10.Anti-tumor activities of CIK cells derived from lymph nodes of lung cancer patients

Background and purpose:Cytokine-induced killer (CIK) has both the advantages of T lympho-cytes’ powerful anti-tumor activity and NK cells’ tumor killing capacity without MHC restriction. It could directly kill tumor cells, regulate and enhance immune function, without damaging the structure and functions of the immune sys-tem. Its effects on the treatment of malignant solid tumors has been widely recognized. This study aimed to evaluate the anti-proliferation effects of CIK cells obtained and cultivated from lung cancer patients’ lymph nodes. Meanwhile, the safety of clinical transfusion was observed.Methods:The peripheral blood and lymph nodes of 6 surgery patients with lung cancer from Shanghai Chest Hospital were used to cultivate CIK cells for 14 days. The phenotypes of CIK cells were detected by lfow cytometry. The anti-proliferation activities of CIK cells on A549 lung cancer cells were detected by CCK8 assay. The morphological changes of CIK cells were observed by invert microscope. The expression of CEA level and adverse events were evaluated after CIK transfusion.Results:The proportion of CD3+CD56+T lymphocyte in two groups were both more than 30%. The CCK8 assay showed that the suppression rate of lymph nodes group was higher than that of peripheral blood group at each effect/target ratio(P<0.05). The adverse effect of CIK transfusion was mild and tolerable. The expression of CEA level decreased in patients.Conclusion:Lymph nodes of surgery patients with lung cancer can be used for cultivation of CIK cells. The anti-tumor activity of CIK isolated from lymph nodes is better than that of CIK cells cultivated from peripheral blood. Preclinical experiments showed high safety.
