3.Progress in inflammatory cardiomyopathy
International Journal of Pediatrics 2011;38(2):141-143
Viral myocarditis can cause severe myocardial injury and even acute heart failure, then evolve into dilated cardiomyopathy, known as inflammatory cardiomyopathy. Immune reaction plays a key role in the transformation from viral myocarditis into dilated cardiomyopathy, including virus RNA continuous replication,immune-mediated injury, cellular apoptosis, other cellular immune response , myocardial antibodies and other humoral immune response. In the clinical treatment, besides anti-heart failure and cardiac nutrition, the use of ribavirin, α1-receptor blockers, β-receptor blockers, calcium channel blockers, angiotensin converting enzyme inhibitors and spironolactone should be emphasized.

8.Construction of pEGFP-N1-TGF-β1 recombinant plasmid and transfected it into primary cultured AEC-Ⅱ of neonatal piglet using lipofectamine 2000
Chinese Journal of Emergency Medicine 2016;25(1):50-56
Objective To construct pEGFP-N1-TGF-β1 recombinant plasmid and transfect it into primary cultured neonatal piglet type Ⅱ alveolar epithelium cell (AEC-Ⅱ) by using lipofectamine 2000,in order to provide basis of methodology for producing recombinant plasmids for transplantation of transfected AEC-Ⅱ into ALI/ARDS animal model lungs. Methods PCR primers were designed to amplify the human TGF-β1 cDNA fragment from plasmid.XhoI and EcoRI were used for double digesting the empty plasmid pEGFP-N1 and cDNA fragment of human TGF-β1.Then the products of double enzyme digestion by using T4 DNA ligase were connected and transformed into DH5α and cultured over night for 16 hours. The structure of recombinant plasmid was identified by using PCR and base sequencing to verify the correctness of pEGFP-N1-TGF-β1 recombinant plasmid. It was then transfected into primarily cultured AEC-Ⅱ by lipofectamine2000 mediated transfection and cultured for another 48 hour.Plasmid DNA (pEGFP-N1-TGF-β1 recombinant plasmid) and lipofectamine 2000 were added into serum-free DMEM respectively, then DNA suspension and Lipofectamine 2000 suspension were blended together and added into cells. After 24-48 hours later, the expression level of enhanced green fluorescent protein (EGFP) was evaluated under fluorescence microscope. Results The structure of vector was verified as pEGFP-N1-TGF-β1 recombinant plasmid by using PCR and base sequencing.Green fluorescence found in some cells showed that the pEGFP-N1-TGF-β1 recombinant plasmids had been successfully transfected into primary cultured AEC-Ⅱ,however,the transfection efficiency still need tobe further improved such as repeating the transfection procedure once again or using adenovirus mediated transfection method to improve the efficiceny and to transplant the cells into animal lungs eventually. Conclusions pEGFP-N1-TGF-β1 recombinant plasmid was successfully constructed and,for the first time, transfected into primarily cultured AEC-Ⅱ of newborn piglets. This established method should be useful for investigation of therapeutic effect and outcomes of lung with experimental acute lung injury.
9.The cost-benefit analysis of magnetic resonance cholangiopancreatography before laparoscopic cholecystectomy in cholecystolithiasis patients unsuspected choledocholithiasis
Chinese Journal of Postgraduates of Medicine 2013;36(35):17-19
Objective To study the cost-benefit analysis of magnetic resonance cholangiopancreatography (MRCP) before laparoscopic cholecystectomy (LC) in cholecystolithiasis patients unsuspected choledocholithiasis. Methods The clinical data of 960 cholecystolithiasis patients unsuspected choledocholithiasis were retrospectively analyzed. The patients were divided into MRCP group (665 cases) and non-MRCP group (295 cases) according to the application of MRCP or not before LC.The total hospitalization expense, missing diagnosis rate of postoperative choledocholithiasis, secondary admission rate, hospitalization time and intraoperative bile duct injury between the 2 groups were compared. Results There were no statistical differences in total hospitalization expense and hospitalization time between the 2 groups (P > 0.05).The missing diagnosis rate of postoperative choledocholithiasis and secondary admission rate in MRCP group were significantly lower than those in non-MRCP group [0.15% (1/665) and 0 vs.3.73% (11/295) and 3.73% (11/295)],there were statistical differences (P < 0.05).There was no intraoperative biliary tract injury in the 2 groups. Conclusions In terms of cost-benefit analysis, routine MRCP before LC for cholecystolithiasis patients unsuspected choledocholithiasis is benefiting.Routine MRCP before LC could effectively reduce the missing diagnosis of the postoperative choledocholithiasis and the secondary admission rate without increasing the total hospitalization expense of patients.
10. Observation of synergistic anti-inflammatory effect of combination of sodium ferulate and oxymatrine
Chinese Traditional and Herbal Drugs 2011;42(3):538-541
Objective: To assess the synergistic anti-inflammatory effect of sodium ferulate (SF) and kushenin, i.e. oxymatrine (OMT). Methods: The anti-inflammatory effect of the combination of SF and OMT was evaluated on xylene-induced murine ear edema model, carrageenin-induced rat paw edema model, and rat tampon-induced granuloma model in vivo. In vitro, the effect of SF and OMT on the nitric oxide (NO) production induced by LPS was observed on RAW264.7 cells. The content of NO in the supernatant was detected with Griess reagent. Results: When SF was used in combination with OMT, the edema induced by xylene and carrageenin was markedly inhibited, the weight of granuloma caused by tampon was also reduced in the high dose group, and the inhibition rate was more considerable by combined treatment than those by SF or OMT alone. In vitro, the NO production induced by LPS was depressed by combined treatment, and a synergistic anti-inflammatory effect was observed. Conclusion: The present study demonstrate that the combination of SF and OMT could exert conspicuous synergistic anti-inflammatory effect, and the mechanism may be related to the inhibited effect of NO production in inflammatory factors induced by LPS.
