1.Neuroprotective effect of acteoside against rotenone-induced damage of SH-SY5Y cells and its possible mechanism

Aim To investigate the neuroprotective effect of acteoside against rotenone-induced cell damage in SH-SY5Y cells and the effect of acteoside on the expression of Parkinson disease (PD)-related proteins Parkin and ?-Synuclein(?-Syn), and to discover the underlying molecular mechanism of neuroprotection by acteoside. Methods The activity of lactate dehydrogenase(LDH) was measured by spectroscopy. Expressions of Parkin and ?-Syn were studied using Western blot analysis, and the distribution of ?-Syn was analyzed using immunofluorescence technique. Results ① Acteoside (10, 20 or 40 mg?L-1) pretreatment for 6 h markedly reduced the release of LDH induced by 0.5 ?mol ?L-1 rotenone; ② Treatment with 0.5 ?mol?L-1 rotenone for 48 h led to significant Parkin cleavage and increased formation of ?-Syn protein dimer; ③ Pretreatment of SH-SY5Y cells with acteoside (10, 20 or 40 mg?L-1) for 6 h markedly reduced the cleavage of Parkin induced by 0.5 ?mol?L-1 rotenone in a concentration-dependent manner, inhibited the aggregation of ?-Syn and decreased ?-Syn-positive SH-SY5Y cells. Conclusion These findings suggest that pretreatment of acteoside has a potent neuroprotective effect against rotenone-induced SH-SY5Y cells damage and its mechanism might involve in reducing the cleavage of Parkin and inhibitng ?-Syn expression induced by rotenone.
3.Atrioventricular nodal ablation for patients with chronic atrial fibrillation

Objective: To evaluate effects of atrioventricular nodal ablation, pacemaker and pharmaco logic agents on ventricular rate control. Methods:Three patients with chronic sustained atrial fibrillation ( AF) were treated by atrioventricular nodal ablation. Fast-slow junctional cardiac rhythm in this course meant the ablation was successful. Unless the ablation was able to get Ⅱ? Ⅱ type or Ⅲ? AVB, R-R intervals ≥3 s, ventricular rate≤40 bpm, it would be canceled. Otherwise implantation of a permanent pacemaker was performed. Results:One patients having complete atrioventricular block and a ventricular rate of 38 -40 bpm had implantation of a permanent pacemaker. Two patients having no atrioventricular block, prolongation of R-R intervals and ventricular rate of 90 - 120 bpm were given antiarrhythmic drugs. Conclusion: Patients with chronic sustained AF can be treaded with atrioventricular nodal ablation, pacemaker and pharmacologic agents effectively.
4.Analysis of mitochondrial DNA gene mutations in hearing loss induced by military noise

Objective To investigate the relationship between mitochondrial DNA gene mutations and the susceptibility to military noise,and to provide a criterion for screening the individuals susceptible to military noise through molecular biology diagnostic method.Methods An investigation of hearing loss was performed among a total of 1032 servicemen including 406 tankers and 626 artillerymen who have endured military noise in their military duty.A total of 122 blood samples were collected,of which 82 samples were collected from susceptible individuals(susceptible group) and 40 from tolerance individuals(tolerance group).DNA was then extracted from the leukocytes of blood samples.The gene fragments of mitochondrial DNA,which might associate with non-syndromal hearing loss,were amplified by polymerase chain reaction(PCR).The PCR products were analyzed by sequencing and contrasting.The gene mutations which were different from the standard sequences of mitochondrial genome were then compared between the susceptible and tolerance groups.Further auditory analysis was performed on those persons who had the gene mutations of hearing loss.Results The gene sequencing revealed there were definite differences in mitochondrial DNA between the susceptible and tolerance groups,and statistically significant difference was found in mitochondrial DNA COII gene T7684C and G7853A mutations between the two groups.Mitochondrial DNA gene mutations were found in the present study,such as A827G,T961insC(heterogeneity),T1005C,T1095C and G7444A,which were reported to be related to deafness in literature.Most of these mutations were detected from the samples of susceptible group,and the others from tolerance group.Conclusion The mitochondrial DNA COII gene T7684C and G7853A mutations might exist in those persons who are susceptible to military noise.
5.Investigation and application of bolus chasing DSA technology

Objective To discuss the application of the bolus chasing DSA technology in angiography in thoracic, abdominal, common iliac and four limbs′ arteries. Methods Bolus chasing DSA with Mask Before program of the system was performed in 40 procedures in 25 patients, using the control handle to control the moving of the bed, and obtained the real time bolus chasing DSA pictures at the blood stream speed. Results In the 40 procedures, photographing in 35 had good qualities, accounting for 87.5%, but not good in 5, accounting for 12 5%, which could still meet the request of diagnosing after non subtraction processing. Conclusion Bolus chasing DSA could display a large scale of anatomical skeleton and blood stream condition from chest to the four limbs exactly. It was a new method that could be taken to observe real time continuous multi sections of blood vessel structure.
6.Effect of improving microcirculation and preventing cell Ca~(2+) overload on halting progress of mild acute pancreatitis to severe acute pancreatitis

Objective To evaluate the effect of improving microcirculation and preventing cell Ca 2+ overload in preventing the development of acute pancreatitis (AP).Methods 278 AP patients admitted from 1990 to 1994 and from 1995 to 1999 were retrospectively analyzed. Routine conservative managements were performed in the first stage,improving microcirculation and preventing cell Ca 2+ overload were performed in the second stage.Results During the first period,there were 124 patients with AP.The conversion of mild AP to severe AP developed in 28 of 120 patients with mild AP.18 of 32 patients with severe AP developed systemic complications,24 patients developed local complications,there were 9 deaths.During the second period,154 cases were treated ,the conversion of mild AP to severe AP presented in 16 of 149 patients with mild AP. 4 of 21 patients with severe AP presented systemic complications,19 patients presented local complications, 3 patients died.Conclusion Improving pancreatic microcirculation and preventing pancreatic cell Ca 2+ overload are helpful not only to prevent the conversion of mild pancreatitis to severe pancreatitis,but also to prevent progressive pancreatic necrosis, and improve the prognosis of acute pancreatitis.
7.The correlation analysis of interleukin-1 family genotypes with coronary heart disease in elderly patients

Objective To investigate the correlation of interleukin-1 family genotypes,including interleukin-1 (IL-1α,IL-1β) and interleukin-1 receptor antagonist (IL-1Ra),with coronary heart disease (CHD) and serum lipoprotein level in the elderly.Methods Interleukin-1 family genotypes were detected in 318 elderly controls and 329 elderly CHD patients by polymerase chain reaction and restriction fragment length polymorphisms method.Serum levels of lipoproteins were inspected simultaneously.Results The TT and Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype frequency of IL-1Ra was 90.3% in elderly CHD patients,but 82.4% in controls.Carriers with TT,Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype of IL-1Ra were at an increased risk with an odds ratio of 1.98 in elderly CHD patients as compared with controls (x2=8.55,95% CI:1.25-3.16).The TT and Ⅰ/Ⅰ or Ⅰ/Ⅳ genotype frequency of IL-1Ra was 96.2% in elderly CHD patients with acute coronary syndrome,but 84.8% in elderly CHD patients with stable angina.Carriers with TT,Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype of IL-1Ra were at an increased risk with an odds ratio of 4.54 in acute coronary syndrome group as compared with stable angina group (x2=12.17,95%CI:1.81-11.36).The CT or TT genotype frequency of IL-1α-889 was 22.8% in acute coronary syndrome group,but 7.6 % in stable angina group.Carriers with CT or TT genotype of IL-1α-889 were at an increased risk with an odds ratio of 3.59 as compared with stable angina group (x2 =14.93,95%CI:1.82-7.03).There were no significant differences in levels of serum lipoproteins among the different genotypes (P>0.05).Conclusions In elderly patients with coronary heart disease,IL-1α(-889) CT or TT genotype carriers are at high risk for acute coronary syndrome,but IL-1Ra CC,TC,Ⅰ / Ⅱ or Ⅱ / Ⅱ genotype carriers are at a low risk for CHD or severe CHD.

9.Efficacy of Hydrochloride Phenazopyridine on Postoperative Pain of Cystoscopy

Objective To explore the clinical effect of hydrochloride phenazopyridine on relieving the pain after cystoscopy. Methods 60 male patients undergoing cystoscopy were divided into observation group and control group according to the block randomization method with 30 patients in each group. 1 h after cystoscopy, the observation group was given orally hydrochloride phenazopyridine 0.2 g, 3 times a day for 0.6 g totally. The control group was not treated with oral medication for pain. The pain degree was evaluated with Numerical Rating Scale (NRS), and the side effects were recorded. Results There was no significant difference in NRS score immediately after cystoscopy between 2 groups (P=0.725). The NRS score was lower in the observation group than in the control group 24 h after cystoscopy (P=0.002).Conclusion Hydrochloride phenazopyridine can effectively relieve the postoperative pain of cystoscopy.
10.Effect of electroacupuncture combined with acupoint injection of deer melon polypeptide on bone morphogenetic protein and rheumatoid factor in rheumatoid arthritis

ObjectiveTo detect the effect of electroacupuncture combined with acupoint injection of deer melon polypeptide on the bone morpho-genetic protein synovial, BMPs and serum rheumatoid factor IgA-RF, IgM-RF.MethodsA total of 246 patients with Rheumatoid arthritis were divided randomly into control group and combined treatment group, 123 patients in each group. The control group received the treatment of electro-acupuncture, drug and Ultrashort wave therapy, and the combined treatment group continue to receive acupoint injection of deer melon polypeptide after 30 minutes when the treatment of control group was completed. Both groups were treated for 2 weeks. The expression of BMPs, MMP-1 and the ESR, serum rheumatoid factor IgA-RF, IgM-RF were detected. The morning stiffness time, Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and visual analogue scale (VAS) were recorded for outcome assessment.ResultsCompared with control group after treatment, the positive expression rates of ESR (8.79 ±1.61 mm/hvs.12.41 ± 2.57 mm/h,t=3.654), IgM-RF (22.38% ± 5.45%vs.39.57% ± 6.18%,t=2.365), IgA RF (32.84% ± 6.57%vs.49.84% ± 4.20%,t=2.574) in the combined treatment group significantly decreased. Compared with the control group, the expression of BMPs (15.43 ± 2.94vs.7.22 ± 1.52,t=3.914) in the combined treatment group significantly increased, while MMP-1 (8.01% ± 1.54%vs. 15.34% ± 2.47%,t=2.015) in the combined treatment group significantly decreased (P<0.05). Compared with control group, the outcomes VAS (3.37 ± 0.35 vs.5.75 ± 0.39, t=3.251), WOMAC (22.36 ± 3.49vs.30.50 ± 4.11,t=4.021), and morning stiffness (34.32 ± 5.57 minvs. 49.84 ± 4.20 min,t=3.914) in the combined treatment group significantly decreased (P<0.05). The total effect rate (82.11%vs. 71.55%,χ2=12.764) of combined treatment group was significantly higher than control group (P<0.05).Conclusion Electroacupuncture combined with acupoint injection of deer and melon peptide can improve effect rate.
