1.Preliminary clinical study of regional cerebral blood flow and hemodynamics in patients with chronic cerebrovascular insufficiency

Objective To explore the changes in regional cerebral blood flow(rCBF)and cerebral hemodynamics in patients with CCVI.Methods Twenty patients diagnosed as CCVI were enrolled to undergo single photon emission computed tomography(SPECT)rCBF imaging or transcranial Doppler uhrasonography(TCD)examination.All the patients were free from cerebral structural abnormalities as demonstrated by X-CT or magnetic resonance imaging(MRI).Cranial arteries blood flow velocity,pulsating index(PI)were measured by TCD and compared with the mean of rCBF as measured by SPECT.Results On SPECT images, decreased rCBF lesions were found in 95 percent of patients(19/20).The areas of hypoperfusion were mainly located in frontal,temporal,parietal lobes and fondues nodus.Increased cranial arteries blood flow velocity was found in the anterior cerebral artery(ACA)and middle cerebral artery(MCA)in 80 percent of patients (16/20).There were no correlations among rCBF,the decreased percentage of rCBF,average velocity,and PI of cranial arteries.Conclusions By performing SPECT rCBF and TCD on CCVI patients,the degree of changes in rCBF and hemodynamics Can be evaluated easily,which is valuable for early clinical diagnosis and treatment of CCVI.
2.Determination on adenosin of Safflower Injection by RP-HPLC

Objective: To establish a RP HPLC method to determine the content of adenosin in safflower Injection. Methods: The sample was determined by RP HPLC with a Diamonsil TM C 18 column, UV detection wavelength was at 270nm. adenosin used as an external standard and CH 3OH 1% HAC as a mobile phase. Results: The average recovery was 101.86%, RSD was 1.34%. Conclusion: The method is simple and accurate. It can be used to control the quality of Honghua Injection.
3.Analysis of Saussurea involucrata Kar. et Kir.from different sources by HPLC-FPS

Objective:To study comparatively HPLC FPS of seven kinds of snowlotus, Saussurea involucrata Kar. et Kir. from different sources of Xinjian area. Methods: HPLC fingerprint analytical method of Snowlotus, from Urumqi was developed, and the HPLC FPS of seven samples were established. Results: The methodological evaluation showed that this method had a good repeatability, and the ratio of common peaks area of different samples were different. Conclusion: The method can be used to distinguish Snowlotus from different sources conveniently.
4.Evaluation of the prenatal screening mode for fetal congenital heart diseases by ultrasound

Objective To assess prenatal heart disease screening program by ultrasound. Methods A total of 11 544 second-trimester screening scans were performed before 24 weeks′ gestation on 11 410 women between February 2004 and May 2007 in Obstetrics and Gynecology Hospital of Fudan University. Fetal heart screening was based on four-chamber and outflow tract views( left ventricular outflow + three vessel view). The sensitivity and specificity of different views were evaluated. Follow-up data of newborns was obstained. Results (1) Among 11 544 cases, 48 cases of congenital heart disease (CHD) were diagnosed in utero. Six cases were false negative, and 2 cases were false positive. The incidence of CHD was 0.47% (54/11 544). (2) Thirty-three CHDs were detected based on the four-chamber view, including 18 ventricular septal defect (9 with conotroncal anomalies),6 anomalous atrioventricle valve, 9 disproportion of left/right ventricle. The sensitivity of the four-chamber view alone was 61.11% (33/54), and the specificity was 99. 98% ( 11 488/11 490). Fifteen CHDs were detected based on the left ventricular outflow and three vessel views, including 1 pulmonary atresia, 3 pulmonary valve stenosis, 2 transposition of the great arteries ( TGA ), 1 pulmonary stanosis with TGA, 6 tetralogy of Fallot, and 2 pulmonary stenosis. The sensitivity of the combination of the four-chamber and outflow tract views was 88. 89% (48/54), and the specificity was 99. 98% (11 488/11 490). (3)Of 48 CHDs,11 cases were eceompanied by other malformations. Eleven cases were performed amniocentesis, amony whom 5 cases were trisemy 21. Conclusion The screening program based on four-chamber and outflow tract views shows good sensitivity and excellent specificity. Our prenatal heart screening program is clinically feasible.
5.MR R2*map for tracing superparamgnetic ironoxides labeled endothelial progenitor cells in vitro.

OBJECTIVESTo investigate the value of R2*map for quantitatively tracing superparamgnetic ironoxides (SPIO) labeled endothelial progenitor cells (EPCs).
METHODSThe EPCs were isolated from Balb/c mice bone marrow and cultured in vitro. After 7 days, expression of acetylated low-density lipoprotein (acLDL) and Ulex europaeus agglutinin-1 (UEA-1), two markers of EPCs, was observed by double staining using fluorescence microscope, the expression of stem cell antigen-1 and vascular endothelial growth factor receptor-2 (VEGFR-2) was confirmed by flow cytometry. EPCs were labeled by incubating with 50 microg/ml SPIO and 6 microl/ml lipofectamine2000, SPIO labeled EPCs were observed under transmission electron microscope (TEM). Labeled and unlabeled EPCs were mixed with 10 g/L agarose and scanned using a 3.0T MR scanner, R2* map and R2 map images were obtained on workstation.
RESULTAfter 7 days of in vitro culture, most of the cells showed characteristics of EPCs. There was no morphological difference between SPIO labeled EPCs and unlabeled EPCs. R2* and R2 values exhibited a linear correlation with the number of labeled cells in the agarose gel. Compared to R2, R2* was a better indicator of the number of labeled cells.
CONCLUSIONMR R2* map can be used to trace SPIO labeled EPCs quantitatively.

6.Expressions of EGF and bFGF in human prostate

Purpose:To study the expression and functional way of EGF and bFGF in human prostatic tissue.Method:To detect the expression of EGF and bFGF in 6 cases of human normal prostate (NP) and 27 cases of benign prostatic hyperplasia (BPH) specimens using mRNA dot blot, in situ hybridization and immunohistochemistry.Results:There was no expression of EGF mRNA in any of the prostate exmined,weakly immunohistochemical staining of EGF was observed in 2 cases of 6 NP and 5 of 27 BPH sepcimens,there was no statistic difference between NP and BPH (P>0.05).The total amount of bFGF mRNA in BPH tissue is much than that in NP tissue,aboundant bFGF mRNA was revealed in epithelial cells of 6 cases of NP tissues,but no bFGF;low amount of bFGF was expressed,according with the level of its mRNA both in basal and stromal cells of NP tissues;No signal for bFGF mRNA was found in epithelial cells of BPH tissues but bFGF protein was found on surface of local proliferating epithelial cells.Markedly increased expression of bFGF mRNA and its protein were present in basal and stromal cells of BPH tissues,especially in the region of local proliferating stromal cells.Conclusion:There are no EGF secretory cells in human normal or hyperplastic prostate; overexpression of bFGF in basal and stromal cells of human prostate caused irregular hyperplasia both of stromal elements and glandular epithelium via autocrine and paracrine pathways.
7.Study on the association of single nucleotide polymorphisms and haplotypes of GPR154 gene with allergic asthma in Han nationality in Hubei Chinese population.

OBJECTIVETo investigate whether two polymorphism sites of the G protein-coupled receptor 154 gene (GPR154) are associated with asthma in Han nationality of Hubei province in China.
METHODSThe polymorphisms were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) in 145 cases of allergic asthma and 120 healthy controls.
RESULTS(1)The genotype frequencies of SNP563704 were 0.324 for CC, 0.524 for CT, 0.152 for TT in allergic asthma patients in a Chinese population. The genotype frequencies of SNP522363 were 0.289 for CC, 0.521 for CG, 0.190 for GG in allergic asthma patients in a Chinese population. (2)No significant differences in the distributions of SNP563704 and SNP522363 polymorphisms were found between allergic asthma and healthy control subjects (chi square is 1.880, P> 0.05; chi square is 0.700, P> 0.05, respectively). (3)No significant difference in serum total IgE was found between patients with allergic asthma with different genotypes of SNP563704 and SNP522363 (F is 0.714, P> 0.05; F is 0.083, P> 0.05, respectively). (4) The distribution of frequencies of 4 haplotypes showed significant difference(chi square is 16.50,P< 0.01). The haplotype frequencies of CT and GT were remarkably higher in asthma subjects than those in controls.
CONCLUSIONThe polymorphisms of SNP563704 and SNP522363 are not associated with allergic asthma in Han nationality in Hubei Chinese population. However, the haplotypes are associated with allergic asthma.

8.Effects of benzo(a)pyrene exposure on oxidative stress and ATPase in the hippocampus of rats.

OBJECTIVETo investigate the effects of benzo[a]pyrene (B[a]P) exposure on the behaviors and hippocampal oxidative stress and ATPase in rats and the molecular mechanism of neurobehavioral toxicity of B[a]P.
METHODSA total of 120 male SD rats (21 days old) were randomly and equally assigned to five groups: blank control group, vegetable oil (solvent control) group, and 2.5, 5, and 10 mg/kg B[a]P exposure groups. The rats in B[a]P exposure groups were injected intraperitoneally with B[a]P once a day for 4 consecutive weeks. Then, Morris water maze and shuttle box were used to evaluate the learning and memory abilities of rats; colorimetric assay was used to measure the activities of superoxide dismutase (SOD), Na(+)/K(+)-ATPase, and Ca(2+)/Mg(2+)-ATPase and the content of malonaldehyde (MDA) in the hippocampus; the concentration of Ca(2+) in the hippocampus was measured by fluorescent labeling.
RESULTSCompared with the blank control group and solvent control group, the B[a]P exposure groups exhibited significant increases in escape latency, active avoidance response latency, and passive avoidance response latency and significant decreases in number of platform crossings and active avoidance response frequency in the last test (P < 0.05 for all comparisons), with a dose-effect relationship. In addition, the B[a]P exposure groups had significantly lower activities of SOD, Na(+)/K(+)-AT-Pase, and Ca(2+)/Mg(2+)-ATPase and significantly higher MDA level and Ca(2+) concentration than the blank control group and solvent control group (P < 0.05 for all comparisons), with a dose-effect relationship.
CONCLUSIONThe neurobehavioral toxicity of B[a]P may be related to increased oxidative stress and decreased activities of Na(+)/K(+)-ATPase and Ca(2+)/Mg(2+)-ATPase in the hippocampus of rats.

9.Development of a predicting model of survival rates for patients with salivary adenoid cystic carcinoma.

OBJECTIVETo establish a predicting model of survival rates and to evaluate the weighted contributions of each key prognostic factor of the patients with salivary adenoid cystic carcinoma (SACC).
METHODSOne hundred and eighteen follow-up cases with SACC were analyzed for the survival study with retrospective cohort method. Ten possible clinical and pathologic factors were selected. A multivariate analysis was performed by Cox proportional hazard model and prognostic index (PI) was calculated. According to the PI, all cases were divided into three risk subgroups respectively: lower, intermediate and higher risk subgroups. Ten-year survival rate and median survival time were calculated and the predicting models of survival rates were established.
RESULTSThe significant prognostic factors influencing the survival rate were age at diagnosis, clinical presentation, TNM clinical stage, treatment, surgical margins (P < 0.05). The predicting formula was PI = 0.031X(2) + 0.665X(5) + 0.420X(6)-0.576X(7) + 0.999X(10). According to the value of PI, the prognosis of the patients was significantly different among the three subgroups (P < 0.05). In the three risk subgroups, 10-year survival rates were 83.56%, 31.45% and 11.20% respectively, the median survival time was 18 years, 7 years and 4 years respectively.
CONCLUSIONSThe established predicting model of survival rates can predict the prognosis of the patients with SACC.

10.Study of using shattering extraction with solvent to extract ingredient of three kinds of traditional Chinese medicine.

OBJECTIVETo prove if it is possible for using the shattering extraction with solvent to extract ingredients of traditional Chinese medicine.
METHODThe shattering extraction with solvent, the refluxing extraction and the ultrasonic extraction were used to extract paeoniflorin from Radix Paeoniae rubra, and to extract baicalein from Radix Scutellariae, and to extract chlorogenic acid from Flos lonicerae japonicae respectively, using ingredient content and extract yield as the measuring indexes.
RESULTThe content of each every ingredient obviously higher by using shattering extraction with solvent than using refluxing extraction or the ultrasonic extraction.
CONCLUSIONThe shattering extraction with solvent is a high efficiency, simple and quick extraction. It may be used to extract the ingredient of three kinds of traditional Chinese medicine.
