2.Reversal effect study of vitamin D on leukemia multidrug resistance cells

Objective To investigate the reversal effect of vitamin D on drug resistance leukemia cell lines Jurcat vitamin D/ADR,K562/ADR and its mechanism.Methods Non-toxic concentration of vitamin D was chosen by methyl thiazolyl tetrazolium,and on line concentration experiment ;the contents of vitamin D for soft erythromycin in the cell and the cell surface of P-glycoprotein(P-gp) expression level were detected by using flow cytometry instrument and vitamin D for intracellular mdr1 mRNA and mrp1 mRNA expression was detected by Real-time polymerase chain reaction;vitamin D for the change of intra cellular glutathione glycosides peptide (GSH) content was detected by adoping biochemistry method.Results Resistance reversal in experiments showed that vitamin D could decrease soft erythromycin in the accumulation of drug resistant cells,with a dose-dependent relationship; vitamin D treatment Jurcat/ADR,K562/ADR cells after 48 h,chemotherapy drugs could increase the cell concentration,and reduce P-gp and the expression of mdr1 and mrp1 gene ; vitamin D treatment Jurcat/ADR,K562/ADR after 48 h could decrease the intracellular GSH content in K562,K562/ADR,Jurcat and Jurcat/ADR.Conclusions Vitamin D has the drug resistance reversal agents to Jurcat/ADR,K562/ADR,which might be relevant to the inhibition of cell surface of P-gp proteins function and expression and reduction of the mdr1 and mrp1 expressions of resistance genes and reduction of intracellular GSH content.Vitamin D by influencing the above mechanism,which increases the drug concentration in cells,can effectively kill the tumor cells.

4.Formulation of Water-in-oil Microemulsion and Its Quality Evaluation

OBJECTIVE:To select the formula,preparation and quality evaluation of water-in-oil microemulsion. METHODS:Water-in-oil micromulsion has been formulated using Volp-5 as surfactant, several short chain alcohols as co-surfactants, and the different oils, pseudoternary phase diagrams have been constructed to evaluate the phase behavior of the formulations of the water-in-oil microemulsion, and the influence of mixing ratio of various elements on the area of microemulsion was also examined. The morphology, particle size, distribution and stability was assessed. RESULTS:The stable water-in-oil microemulsion was made. The microemulsion delivery could be prepared using Volp-5/ethanol/caprylic-capric-triglyceride/water (3∶2∶2∶4)system and no phase separation was observed in three months. CONCLUSION:The formulation provide a convenient means of the oral delivery taking advantage of microemulsion's merit, which is easy to prepare with consistent quality.
5.The Effect of Pravastatin on Impaired Endothelium-dependent Relaxation of Isolated Rabbit Aortic Rings Induced by Homocysteine

Objective To explore whether pravastatin protects endothelium against the damage induced by homocysteine (Hcy) in isolated rabbit aorta. Methods Endothelium-dependent relaxation responses to acetylcholine (ACh) of thoracic aortic rings were measured by isometric tension recording before and after aortic rings exposed to Hcy in the absence or presence of pravastatin (PT) to estimate the injury effect of Hcy and the protective effect of pravatatin on rabbit aortic endothelium, respectively. Results Incubation of aortic rings with 1~10mmol/L Hcy for 30min significantly inhibited endothelium-dependent relaxation response to ACh of aortic rings in a concentration-dependent manner. Pre-incubation of aortic rings with 0.3~3mmol/L PT for 15min and co-incubation of aortic rings with 3mmol/L Hcy for another 30min markedly attenuated the inhibition of endothelium-dependent relaxation induced by Hcy in a concentration-dependent manner. Conclusion Pravastatin can improve the impairment of endothelium-dependent relaxation induced by Hcy in isolated rabbit aorta.
6.Clinical effect of probiotics combined with saline enema in the treatment of infantile diarrhea and its effect on serum inflammatory factors

Objective To explore the clinical effect of probiotics combined with saline enema in the treatment of infantile diarrhea and its effect on serum inflammatory factors.Methods 186 cases of infantile diarrhea were selected,they were randomly divided into three groups according to the random number table method:probiotics group,montmorillonite group and control group.Patients in probiotics group were administered with clostridium butyricum combined with saline enema,and patients in the montmorillonite group were treated by montmorillonite combined with saline enema,and the control group was given routine treatment.Then,the clinical symptom relief time,total effective rate,serum inflammatory factor concentration and adverse reaction were observed and compared.Results The total effective rate of the probiotics group was 98.39%,and that of the montmorillonite group was 96.77%,both of which were significantly better than 80.65% of the control group,and the differences were statistically significant(χ2 =11.408,P =0.000;χ2 =8.432,P =0.018).After treatment,the serum levels of inflammatory factor tumor necrosis factor -α(TNF -α)in the probiotics group,montmorillonite group,control group were (8.30 ±2.33)pg/mL,(8.54 ± 2.47)pg/mL,(12.49 ±3.44)pg/mL,and the difference was statistically significant(F =5.120,P =0.014),also, the serum levels of interleukin -6(IL -6)in the three groups were (10.08 ±5.46)pg/mL,(11.24 ±5.84)pg/mL, (14.02 ±5.97)pg/mL,the difference was statistically significant(F =34.015,P =0.000).There were 2 cases of constipation in the montmorillonite group,the incidence rate was 3.23%,and there was no adverse reaction in the other two groups.Conclusion The effect of probiotics combined with saline enema in the treatment of infantile diarrhea is similar to that of montmorillonite,which can promote the children to alleviate the symptoms,shorten the length of hospital stay,reduce inflammation,and has no adverse reactions such as constipation,it is worth to be recom-mended widely for clinical application.
8.Efficacy on borderline hypertension treated with acupuncture combined with anti-hypertensive plaster intervention.

OBJECTIVETo compare the efficacy difference in treatment of borderline hypertension between acupuncture combined with anti-hypertensive plaster and simple acupuncture.
METHODSSixty patients of borderline hypertension were randomized into an observation group and a control group, 30 cases in each one. In the observation group, acupuncture was applied to Fengchi (GB 20), Taichong (LR 3) and the others, once a day, 5 treatments made one session. There were 2 days at interval among the sessions. Totally, 4 sessions were required. Additionally, in combination, the anti-hypertensive plaster was prepared with the fine powder of Chuanxiong (ligusticum wallichii) and Wuzhuyu (fructus evodiae) at 1.1 and mixed with vinegar. The plaster was applied to the umbilicus, once every two days, totally 15 times were required. In the control group, acupuncture was given simply. The selected acupoints and operation were the same as the observation group. Before and after treatment, blood pressure and syndrome score were observed and the short-term and long-term efficacies were assessed in the two groups.
RESULTSAfter treatment, the systolic pressure and diastolic pressure were reduced as compared with those before treatment in the two groups (all P<0.001), and the differences were significant in comparison between the observation group and the control group (P<0.01, P<0.05). The syndrome score after treatment was reduced as compared with that before treatment in the two groups (both P<0.001) and the difference was not significant in comparison between the two groups (P>0.05). In follow-up of the effective cases and remarkably effective cases in 3 months, the total effective rate in the observation group was 89.3% (25/28) and was 60.0% (15/25) in the control group, indicating the significant difference in comparison (P<0.05).
CONCLUSIONAcupuncture combined with anti-hypertensive plaster is the effective method in prevention and treatment of borderline hypertension. Regarding the long-term efficacy, this combined therapy achieves the superior improvement in the symptoms and physical signs of the patients of borderline hypertension as compared with simple acupuncture.

10.Evaluation the performance of the body fluid mode on the automatic analysis system-XT-4000i

Objective To evaluate the performance of the body fluid mode on the automatic analysis system-XT-4000i .Methods XT-4000i analyzer was used to detect the precision ,accuracy ,carryover rate ,meanwhile to validate the detection limit of the pa-rameters(WBC and RBC) .Results The range of the precision for WBC-BF was 0 .90% -5 .54% ,for RBC-BF was 1 .13% -5 .13% .The carry over rates were 0 .09% for WBC and 0 .14% for RBC ,respectively .As for as the analyzer range ,the results of the linearity and the detection limit verification were in line with the requirements of the analyzer ;the comparison between the man-ual microscopy results and the analyzer results meet the requirements of XT-4000i analyzer .Conclusion The performance of XT-4000i analyzer is in line with the requirements of the instrument range ,which can be used for clinical body fluid analysis and detection .
