The paper expounds the importance and role of circulation service in college and university libraries, how to do circulation work better is discussed from three aspects: scientific management of collections, creating a better environment and fully promoting the quality of staffs, thus to make library play a greater role in campus culture construction.

Objective:To prepare recombinant pCDNA3.1+/Ag85A DNA vaccine encoding tuberculosis Ag85A gene mediated with LipofectamineTM 2000 and to study the effect on T cell subpopulation via oral vaccination.Methods:Recombinant plasmid containing Ag85A using liposome as a vector was constructed and administered to C57BL/6 mice via oral route.Determination of the contents of IFN-r,IL-4 in serum of C57BL/6 mice was performed by double antibody sandwich ELISA.Furthermore,the ability of splenocytes to secret IFN-? and IL-4 was tested by ELISPOT method.When the mice were vaccinated with recombinant eukaryotic expressing vector 5 weeks later,titers of serum antibody against Ag85A were detected by ELISA.The percentage of the CD4+ and CD8+ T cell subsets in the splenocytes was determined by flow cytometry.Results:Lymphocytes obtained from the spleen of liposomal pcDNA3.1+/Ag85A vaccine-immunized mice exhibited lower IFN-? production and higher IL-4 production than those of pcDNA3.1+ vector immunized mice.The number of spleen MNC secreting IFN-? stimulated by Ag85A protein in vitro was significantly lower than that of plasmid vector group.Liposomal pcDNA3.1+/Ag85A vaccine immunized mice elicited higher Ag85A-specific antibodcy titres.The percentage of the CD4+ and CD8+ T cell subsets in the splenocytes was decreased.The subset both were shown the profile of Th2 responses.Conclusion:The oral recombinant plasmid pCDNA3.1+/Ag85A mediated with LipofectamineTM 2000 It shows down-regulation effect on the subsets of CD4+ T cells and CD8+ T cells.The regimen has good immunogenecity and could induce Th2 type humoral response in C57BL/6 mice.The immunization suppresses secretion of IFN-?.It can greatly enhance the titres of Ag85A-specific antibodies.LipofectamineTM 2000 can act as an adjuvant through comparation with negative control group.

And the optimum incubation time was 24h, the optimum temperature was 30℃, the optimum broth initial pH was 6.5. The optimum media component was: lactose 30g, trypone 15g, soybean peptone 20g, meat extract 30g, peptonethe 20g, Tween 80 1mL, K_(2)HPO_(4 )2g, NaAc 5g, Tri-Ammonium citrate 2g, (MgSO_(4)) 0.58g, MnSO_(4) 0.25g, H_(2)O 1000mL. Production of bactericoin under the optimum broth was 640AU/mL, 8 times than MRS media.

Saliva and samples containing saliva were regarded as a kind of potential biological trace evidencefor individual identification in forensic science practice. In this paper, the authors have studied theDNA analysis of saliva and saliva containing samples. The results demonstrated that DNA can be extracted from saliva and saliva-containing samples. RFLPs analysis by DNA fingerprinting and PCRbased typing can be performed according to the amount of DNA extracted from these samples.

Objectives To compare the effects of different early enteral nutrition formula on improving metabolic status of burned patients. Methods Thirty cases of burned patients with total burn surface area (TBSA) more than 30% were randomly divided into two groups:⑴Group F, enteral feeding with Fresubin 750MCT containing MCT/LCT as fat source, ⑵Group N, enteral feeding with Nutrison containing only LCT as fat source. All of the thirty patients received isocaloric, isonitrogenous and isolipidic enteral feeding with intragstric tube and enteral feeding pump, within 24h of postburn. Plasma insulin, glucagons, tumor necrosis factor (TNF) and 3-methylhistidine in urine were measured on day 1,4,7and 10 days of postburn. Results The results showed that the patients of group F had an increased blood concentration of insulin and the ratio of insulin/ glucagons, a decreased plasma concentration of TNF, and a reduced excretion of 3-methylhistidine in urine as compared with the patients of group N. Conclusions An enteral compound diet containing MCT/LCT in the early enteral feeding in burned patients can improve the metabolic status of burned patients more effectively.

Objective To Investigate the expression of metallothionein (MT) and p53 in non-small cell lung carcinoma(NSCLC). Methods The expression of MT and p53 was detected by immunohistochemical method in the paraffin-embedded specimens of 73 NSCLCs and 15 normal bronchial mucosal tissues(NBMT). Results [WTBZ]The positive rates of MT and p53 expression in NSCLC were 54.8%(40/73)and 65.8%(48/73), respectively, and in NBMT were 20.0%(3/15)and 33.3%(5/15), respectively. The positive rates of both MT and p53 in NSCLC were significantly higher than those in NBMT(P0.05). Objective To study the technical method of immunohistochemical staining in pathological tissue.Methods 80 cases of hospitalized patients in our hospital from May 2015 to May 2016 were selected, they were randomly divided into immunohistochemical staining group(immunohistochemistry group, n=40)and biopsy group(n=40)two groups, then the AFP(alpha-Fetoprotein), Glypican-3 positive situation of the the two groups and diagnostic Resultsof patients with different types of disease in the immunohistochemistry group were statistically analyzed.ResultsThe positive rate of AFP(a-Fetoprotein), Glypican-3 of the immunohistochemistry group 92.5%(37/40)was significantly higher than the biopsy group 50.0%(20/40)(P<0.05), Among the 37 cases of positive patients, metastatic tumors 19 cases, hepatocellular carcinoma 9 cases, intrahepatic bile duct carcinoma 7 cases, hemangioma 3 cases, the positive rates were 100.0%, 75.0%, 100.0%, 100.0%, 26 cases died, the mortality rate was 70.3%, of which metastatic tumors 13 cases, hepatocellular carcinoma 6 cases, intrahepatic bile duct carcinoma 5 cases, hemangioma 2 cases, the mortality rates were 72.2%, 66.7%, 71.4%, 66.7%.Conclusion Immunohistochemical staining technic in pathological tissue can promote positive rate, and standard operation procedures can reduce errors, which is worthy of clinical attention.

Objectives To analyze the mechanisms and implications involved in the generation of platelet associated antibodies in cancer patients.Methods HLA IgG antibody from 116 cancer patients who had received repeated platelet transfusion (more than 3 times) was screened by ELISA and the GTI QUICKSCREEN kit. The positive samples from the initial screening were confirmed for HLA and HPA antibodies using the PAKPLUS reagent. Patients with antibodies were observed for symptoms and clinical effectiveness of platelet transfusions. Results A total of 44 cases of HLA IgG antibody were screened out of 116 samples, with an antibody rate of 37.93%. Among those 44 samples, 18 were confirmed to contain anti-HLA-Ⅰantibody, 8 with anti-HPA antibody, 10 with both anti-HPA and anti-HLA antibodies, and 8 were not identified. Objective:To prepare a colloid gold kit for the qualitative detection of the dopes met-amphetamine and morphine by one-step chromatography immunoassay in human samples based on the principle of the highly specific immunochemical reactions between the antigens and their antibodies which were used for the analysis of specific compounds in human urine samples at ideal cut-off concentrations.Methods:The monoclonal antibodies(McAb) of either mouse anti-morphine or mouse met-amphetamine were prepared by hybridomas and the limiting dilution respectively. The titers of McAb were tested by ELISA.The kit was assembled and the specificity,sensitivity,and stability were investigated.In addition,these characters were compared with those of issued Pan Probe single test strip.Results:①The titer of the first McAb against morphine was 1∶3.2?103, as for another, 1∶1.6?103. ②The colloid gold kit was found with following cut-off for the dope concentrations,morphine at 300 ng/ml and met-amphetamine at 1 000 ng/ml. ③The colloid gold kit showed fine specificity without cross-reaction with routine medicines for cold treatment or with the solf-drinks of cokocola or with tobacco for the individuals who did not take the dopes the 7 days.④The colloid gold kit was even more sensitive than the issued strip,because urine sample of the junkies were still positive 4-5 days post-intake.Conclusion:The colloid gold kit for indentifying the drug-users is a rapid,specific, sensitive immunoassay suitable for the simultaneous, qualitative detection of drug abuse.
