1.Thyroid Diseases and Cerebrovascular Diseases
International Journal of Cerebrovascular Diseases 2008;16(4):301-305
There are significant correlations between cerebral embolism and thyrotoxic atrial fibrillation in patients with hyperthyroidism. The incidence of thyrotoxic atrial fibrillation increases significantly in patients with subclinical hyperthyroidism with serum thyroid-stimula-ting hormone levels < 0. 1 mU/L. Hyperthyroidism may result in cerebral venous thrombosis, moyamoya disease and giant cell arteritis; while hypothyroidism is closely associated with the risk factors of arteriosclerosis, especially hypertension, hyperlipidemia, and hyperhomocysteine-mia. However, this association remains uncertain in subclinical hyperthyroidism.

Objective To investigate the gene point mutation in the dihydrofolate reductase thymidylate synthase (dhfr) gene of Plasmodium falciparum isolate from Yunnan Province strongly associated with pyrimethamine and cycloguanil resistance. Methods Nested PCR and restriction endonuclease digestion were applied to detect the gene mutation using dried blood filter paper collected from the fields in Yunnan Province. Results Different mutations were found in 4 amino acids at positions 16, 51, 108 and 164 of dhfr gene, particularly, Asn 108 and Ile 51, the mutaiton frequency being 94.1% and 90.1%, respectively. The frequency of the wild type genotype (3D7 type) Ser 108 appeared lower ( 9.1%) , while the frequency of the Ala 16 was high( 61.8%); the mutation type was very high, the ratio of HB3 type, 7G8 type/FCR3 type and Cambodian type was 1∶21∶7.5. Conclusion The investigation first demonstrated that Plasmodium falciparum Yunnan isolate dihydrofolate reductase thymidylate synthase gene(dhfr) at positions 16, 51 ,108 and 164 exhibited different degrees of point mutation. The frequency of mutation of the 7D8 type involved in pyrimethamine resistance was higher, while that of the FCR3 type involved in cycloquanil resistance was lower.

Objective:To investigate the expression of antigens in Plasmodium yoelii sexual stage and transmission blocking effects of monoclonal antibodies(McAbs). Methods:Observing the effects of anti Pys25 McAb4 and anti Pys21 McAb10 on developmental course of parasites in mosquitoes by direct mosquito feeds on passively immunized P.yoelii infected mice and the expression of Pys21 and Pys25 from gametocytes to ookinetes in indicated culture times by IFA and Western blotting. Results:The transmission blocking activity of the anti Pys25 McAb4 was complete and more potent than that of the anti Pys21 McAb10.Both Pys25 and Pys21 were presented in whole developmental course from gametocytes to ookinetes.Furthermore, the expression of Pys25 appeared to be earlier than that of Pys21 on zygote surfact.Conclusion:Pys25 and Pys21 are target antigens of transmission blocking immunity and that anti Pys25 McAb4 has more significantly transmission blocking activity is related with the early stage expression of Pys25 on zygote surface.

Objective To evaluate the effect of free NgR-modified bone marrow stromal cell (BMSC) transplantation on axon regeneration in rats after spinal cord injury. Methods Genes encoding free NgR protein were cloned and transduced into BMSCs at passage 3 using a lentivirus vector.Indirect immunofluorescence was used to detect the expression of free NgR protein.Meanwhile a spinal cord contusion model was established in 36 adult Sprague-Dawley rats at the T10 segment. The rats were then divided randomly into an experimental group and a control group.NgR + BMSCs were transplanted into the injured site 1 week post-trauma in the experimental group.BMSCs were also transplanted at the same time into the control group.Expression of free NgR at the injury site was detected by immunohistochemical staining at 1 week post-transplantation.The functional recovery of both groups was evaluated at 4 and 6 weeks post-transplantation.Longitudinal sections of the spinal cord were studied for axon regeneration using horseradish peroxidase staining. Results Expression of free NgR was found in the cell plasma of BMSCs by indirect immunofluorescence post-transfection.Positive immunohistochemical staining for NgR was found at the transplant site in the experimental group 1 week post-transplantation.Better axon plasticity could be observed in the experimental group. The Basso-Beattie-Bresnahan scoring of the experimental group was significantly higher than that of the controls at both observation times. Conclusions Free NgR-modified BMSCs can prompt injured axons to regenerate and thus to promote the recovery of neurological function. This might provide a new strategy to treat spinal cord injury.

Objective To investigate the genetic diversity of Plasmodium vivax transmission_blocking vaccine candidate antigen (TBV) Pvs25, with P.vivax isolates from Hubei and Zhejiang Provinces, and to compare the genetic polymorphism of Pvs25 with that from Bangladesh. MethodsThe parasite DNA used for the genetic polymorphism assay was obtained from dried filter paper blood spots. The genes were PCR amplified and the products were purified and sequenced directly. Results 45 complete new sequences were analyzed. Only 3 nucleotide changes were found that would result in amino acid substitutions in Pvs25 in comparison with the sequence from P.vivax Sal_I strain. The measurement of nucleotide diversity (?) was remarkably similar for the two populations, indicating that DNA sequences and deduced amino acid sequences were highly homologous among the geographically dispersed isolates or isolates from the same geographical region. Conclusion The results suggest that Pvs25 has limited antigenic polymorphism, especially compared with candidate antigens expressed by hepatic and erythrocytic stage, which may support the development and application of Pvs25_based transmission_blocking vaccine in China.
