1.Effects of aerobic exercise time on cardiovascular and renal functions of 2K1C renal vascular hypertensive rats

AIM: To explore the impact of aerobic exercise (AE) time on mean arterial pressure (MAP), sinus bradycardia, heart and kidney histological changes and oxidative damage in the two-kidney and one-clip (2K1C) hypertensive rats, and to assess the specific effects of AE time on alleviation of hypertension.METHODS: Healthy male Wistar rats (n=60) were randomly divided into sham operation (sham) group and 2K1C group.After operation, the rats in these 2 groups were divided into sham sedentary (sham+SED) group, sham exercise (sham+AE) group, 2K1C se-dentary (2K1C+SED) group and 2K1C exercise (2K1C+AE) group.The rats in sham+AE group and 2K1C+AE group were subjected to AE for 10 weeks: swimming for 1 h/d.The MAP, sinus bradycardia and the histological changes of heart and kidney in these 4 groups were tested regularly.RESULTS: Compared with 2K1C+SED group, the rats in 2K1C+AE group had significantly improved MAP and sinus bradycardia at the 4th week, with reduced collagen deposition in the myocardium and kidney, decreased level of thiobarbituric acid reactive substances (TBARS) in the left ventricle and increased catalase (CAT) activity in the left ventricle and kidney.The results of the 6th to 10th weeks showed that the differences of MAP and sinus bradycardia between the rats in 2K1C+AE group and 2K1C+SED group had gradually narrowed.The TBARS levels in the left ventricle and kidney and CAT activity in both kidneys of the rats in 2K1C+AE group were partly restored at the 8th week.The collagen deposition in the heart and kidney of the rats in AE group was reduced to the lowest level at the 10th week.CONCLUSION: Short-time (about 4 weeks) AE effectively improves the MAP and sinus bradycardia of 2K1C renal vascular hypertensive rats and attenuates the damages of the heart and kidney by regulating oxidative stress, thus improving the cardiovascular and renal functions of 2K1C renal hypertensive rats.
2.Content Determination of L(-)-Anabasine in Different Medicinal Parts of Alangium chinense by HPLC

OBJECTIVE:To establish a method for the content determination of L(-)-Anabasine in different medicinal parts (stem,rhizome,fibrous root,rootlet) of Alangium chinense,and provide reference for its development and utilization. METH-ODS:HPLC was performed on the column of Thermo C18 with mobile phase of methanol-phosphate buffer solution(22∶78,V/V)at a flow rate of 1.0 ml/min,the detection wavelength was 259 nm,column temperature was 30 ℃;the injection volume was 10 μl. RESULTS:The linear range of L(-)-Anabasine was 0.020 62-0.257 80μg(r=0. 999 9);the limit of quantitation was 1.7 ng,limit of detection was 0.5 ng;RSDs of precision,stability and reproducibility tests were lower than 2%;recovery was 97.38%-98.86%(RSD=0.6%,n=6). The content of L(-)-Anabasine in different medicinal parts was the fibrous root>the rootlet>the rhizome>the stem. CONCLUSIONS:The method is simple,accurate,reproducible and specific,and suitable for the content determination of L(-)-Anabasine in different medicinal parts(stem,rhizome,fibrous root,rootlet)of A. Chinense.
3.Comparison of propofol dosage in morning and at afternoon undergoing gynecological clinic short surgery

Objective To explore propofol dosage difference between in morning and at after-noon undergoing gynecological clinic short operation.Methods One thousands and nine hundred fifty eight female patients,aged 1 6-50 yr,of ASA physical status Ior II,undergoing gynecology clinic short operation,according to the operation time,were divided into two groups,Group morning (8:30-1 1:30,n =1047)and group afternoon (14:00-1 7:00,n =91 1).Each patient was intravenously injected fentanyl 0.01 μg/kg,and then propofol 2.5 mg/kg.When the observer’s assessment of alertness/se-dation (OAA/S)scores reached 0 score,the operation began.If the body moved,the surgery was stopped and 25 mg propofol was intravenously injected quickly.After 10 seconds observation,if the body movement existed,25 mg more propofol was performed,until the movement disappeared.Re-cord patients’year,height,weight,body mass index,and the heart beat,blood pressure of pre-op-eration,the number of times of cervix uteri expansion per vagina,the history of labor per vagina,the total dosage of propofol and the time length of sedation.Results The dosage of group afternoon (1 52.1 6±65.90)mg was higher than group morning’s (135.69±37.67)mg (P <0.05).There was no significant difference of time lengths of sedation between the two groups.Additional use of propofol in the afternoon group 387 (42.5%)was higher than group morning’s 1 99 (23.5%)(P <0.05).Conclusion In gynecological clinic short operation,the propofol dosage in afternoon was higher than that in the morning group.But the sedation time length in afternoon was not longer than that in the morning.

5.The research on MRI for differential diagnosis of benign and malignant lesions of SPN

Objective:To investigate the clinical value of magnetic resonance imaging (MRI) in differential diagnosis for benign and malignant lesions of solitary pulmonary nodules (SPN).Methods: 120 patients with SPN were enrolled in the retrospective analysis, and all of them underwent scan of MR including MRI, DCE-MR and diffusion weighted imaging (DWI) before their surgery. All of patients received detection of pathology or histopathological examination after their surgery, and the ADC values under different b values of dispersion sensitivity factor (300 s/mm2, 500 s/mm2 and 800 s/mm2) were compared.Results:Under the conditions both of 300 s/mm2 and 800 s/mm2, the differences of ADC value between benign nodule and malignant nodule were not statistically significant (t=1.195,P>0.05). And under the condition of 500 s/mm2, the ADC value of benign nodule was significantly higher than that of malignant nodule (t=4.313,P<0.05), and at this time, the sensitivity, specificity, rate of missed diagnosis and misdiagnosis rate of the differential diagnosis for benign and malignant lesions of SPN by using MR were 85.71%, 74%, 14.9% and 26%, respectively.Conclusion: MR examination has higher sensitivity in differential diagnosis for benign and malignant lesions of SPN, and it has a certain clinical value when it is used as early detecting method.
6.Clinical efficacy and safety of ravidavir-based antiretroviral regimen in treatment of patients with chronic hepatitis C

China has largest number of chronic hepatitis C (CHC) patients, with complex distribution of genotypes, high proportion of refractory types and low diagnosis and treatment rate. It is necessary to explore accessible pan-genotype antiviral treatment protocols with high cure rate. Currently, the direct acting antiviral agents (DAAs) are recommended as the main treatment regimes by international guidelines for hepatitis C. Ravidasvir (RDV) is a new generation of NS5a inhibitor, which can be widely used in treatment of gene type 1, 2, 3, 4 and 6 CHC with strong antiviral activity and high resistance barrier. A number of clinical studies demonstrate that the combination of ravidavir with DAA S, such as second-generation protease inhibitor danoprevir (DNV) and nucleoside NS5b inhibitor sofosbuvir (SOF) have gained high cure rate and good safety for CHC patients. In this article, the results of related studies on ravidavir-based antiretroviral regimens in treatment of CHC are reviewed.

AIM:To clarify if interferon-?(IFN-?), tumor necrosis factor-?(TNF-?)and interleukin-1?(IL-1?)can induce apoptosis of human airway smooth muscle cells (ASMCs) in vitro. METHODS: Human ASMCs were isolated and cultured in DMEM containing 10% fetal bovine serum. Passage 4-6 cell was used in the experiment. IFN-?,TNF-? and IL-1?, were used separately or together in the treatment of human ASMCs. The effects of IFN-?,TNF-? and IL-1? on the growth of the cells was detected by MTT method at the hour 0,24,48 and 72. Light microscopy and electron microscopy were used to examine the morphological change. DNA fragmentation was analyzed by agarose gel electrophoresis. SP immunohistological staing method was performed to detect the change of expressions of p53 , bcl-2 and bax gene. The apoptosis cell percentage were detected by in situ end labeling technique (TUNEL)of fragmental DNA. RESULTS: (1)IFN-? or IFN-? together with TNF-? and IL-1? decreased the number of viable cells in a time dependent manner. (2) Light and electron microscopic examination showed cell shrinkage, membrane blebbing, nuclear contraction, chromatin condensation and nuclear fragmentation in human ASMCs. (3) Agarose gel electrophoresis showed a characteristic"ladder"of DNA bands representing integer multiples of the internucleosomal fragments (about 180-200 bp) in cytokine cotreated human ASMCs. (4)The expression of p53 and bax gene in cytokine cotreated group was significantly higher than in control group, but the expression of bcl-2 gene was lower than in control group. (5)Stimultaneous treatment with IFN-?(4?10 5 U/L),TNF-?(4?10 5 U/L)and /or IL-1? (10?10 4 U/L) induced apoptosis of human ASMCs. AIM: To study the effects of Rhodiola(Rho), nitric oxid e (NO), hem oglobin (HB) on the multiple organ dysfunction syndrome(MODS) in early stage aft er burn in rabbits. METHODS:The rabbits were divided into the sh am burn group (SB), burn group (B), orally taken Rho group (R), burn and Rho therapy group (BR ). The changes of hemodynamics were monitored. The index of pulmonary permeabili ty was calculated. These data reflected separately the functions of heart, live r, lung, kidney and blood coagulation system were also determined. NO contents i n ser um and bronchoalveolar lavage fluid (BALF) were measured by Griess method. The l e vels of serum HB were measured. RESULTS:① The dysfunctions of hear t, liver and kidney achieved the criterion of MODS in group B at 48 h postburns (B 48 h ). The N O content of group B significantly increased in serum and BALF at B 48 h . ② The cardiac index (CI) and creatine phosphokinase (CK), urea nitrogen (BUN) markedly raised or decreased in group BR at 48 h postburns (BR 48 h ) than B 48 h . The NO cont ents in serum and BALF markedly raised. ③ HB contents in serum markedly raised in group B and BR at 0 h postburns (B 0 h , BR 0 h ) than group SB, B 48 h , R, BR 48 h , but NO was reverse. CONCLUTIONS: ① HB contents in serum markedly raise d at 0 h post burns, but NO was reverse. ② Rho promoted the increases of NO synthesis and the blood perfusion of organs, which might be one of mechanisms to prevent the devel opment of MODS.
