1.Relationship between Ala 54 Thr polymorphism of intestinal fatty acid-binding protein 2 gene and plasma lipids in old population

Objective To investigate relationship between Ala 54 Thr polymorphism in the intestinal fatty acid-binding protein (I-FABP) and plasma lipids in old people.Methods We collected fasting blood samples of selected groups,then we extracted DNA from whole blood,and detected lipids from serum.The 54A/T I-FABP genotypes were analyzed in 72 Han Chinese elderly by polymerase chain reaction (PCR) and DNA restriction enzyming.Total cholesterol (TC),triglyceride (TG),high density lipoprotein (HDL-C) and low-density lipoprotein (LDL-C) were detected.Genotype subgroups were Thr54 (-) and Thr54 (+) groups (36 cases for each).Results Compared with Thr54 (-) genotype group,Thr54 (+) genotype group had higher levels of plasma TC[(4.50±0.73) mmol/L vs.(5.48±0.49)mmol/L],TG[(1.08±0.48) mmol/L vs.(2.02±0.53)mmol/L],LDL-C [(3.10±0.44) mmol/L vs.(3.50±0.66)mmol/L],and had lower level of HDL-C [(1.14±0.25) mmol/L vs.(0.96±0.23)mmol/L](t=-6.67,-7.84,-3.03,3.05,all P<0.05).
2.The effect of immunotherapy with purified humulus pollen allergens for asthma patient

Objective To study the effect of specific immunotherapy (SIT) with purified humulus pollen allergens for asthma patients. Methods The humulus pollen allergens were purified partially by gelatin filtration. A double-blinded study was carried out. Eight-six patients with seasonal humulus pollen allergic asthma were divided randomly into two groups: observation group (group A) and control group (group B), which were administered SIT with purified or crude humulus pollen allergens, respectively, before the allergic season for half a year. The clinical effect and side effects were observed. The laboratory observation norms included T lymphocyte subpopulation, the wheals’diameter of IT, the average index of HBDT and the specific IgE before and after SIT . Objective To purify and identif y Platanus acerifoli wild pollen. Methods We carried out intracutaneous test with Platanus pollen extract in 30 patients with allergic as thma who visited our hospital from March to May 2003. Seven subjects who had bee n diagnosed as having Platanus pollen-induced asthma were enrolled. Platanus po llen proteins were separated by gel filtration with Sephadex-G-100. To charact erize allergenic components, Platanus pollen extract was analyzed by means of so dium dodecylsulfate-polyacrylamide gel electrophoresis followed by immunoblotti ng. Results To purity the pollen we separated Platanus poll en extract in a first purification step by using gel filtration with Sephadex G -100. Two elution peaks were observed. Twelve percent SDS-PAGE analysis showed more than 10 protein bands whose molecular mass (Mr) ranged from 16 ku to 71 ku. Six bands abundant with protein at 71, 50, 35, 39, 22 and 16 ku were observed. On SDS-PAGE, the proteins of the first peak whose Mr we re 71, 50, 35, 39, and 22 ku and that of the second peak was 16 ku. SDS-PAGE and IgG-immunoblotting analysis with seven sera showed 4 IgG-binding component s whose Mr was 50, 39, 22 and 16 ku. The protein bands whose Mr was 50 ku and 22 ku had the highest binding capacity. Conclusion The strongest activity exists in the first peak which can be the major sensiti zing components and there is mild allergic activity in the second peak which is the minor sensitizing components.
4.In vitro stability of Endostar

Aim: To compare the stability of Endostar~(TM) and endostatin under different temperatures and pH using polyacrylamide gel electrophoresis( PAGE) and Western blot and to compare the activity of Endostar kept at 4 ℃ and 37 ℃ by inhibition of endothelial cell proliferation. Methods: Endostar and endostatin expressed by Phicha pastoris were kept at 4 ℃, and 37 ℃ for 96 hours, respectively. The electrophoresis of the samples was detected by reduced and non-reduced PAGE. The results were further confirmed by Western blot with rabbit anti-Endostar polyclonal antibody. The inhibitory effect of Endostar stored at different temperatures on HUVEC was also exam-ined by cell-based assay. Results: Single band at 20 kD was detected in all lanes of SDS-PAGE gel loaded with endostatin and Endostar samples under reducing condition. In acidic native PAGE with three different pH values, endostatin showed a smear characteristic, whereas Endostar showed a unique band in acidic non-continuous native PAGE. Although the smear phenomenon was also observed under two conditions of constant native elec-trophoresis, the major band of Endostar could be detected. Similar electrophoretic behavior was found for endosta-tin and Endostar stored at both 37 ℃ and 4 ℃ . Western blot showed similar results to those by PAGE. Furthermore, Endostar stored at these two temperatures also had identical inhibitory effect on proliferation of HUVEC. Conclusion: Endostar and endostatin exhibit similar thermostability at regular conditions, but Endostar was more stable than endostatin expressed in P. pastoris under acidic condition.
5.Polymorphism of human intestinal fatty acid binding protein gene and serum lipid levels in coronary heart disease

Objective To explore the relationship between FABP2 54 gene polymorphism and serum lipid levels in different ethnic patients with coronary heart disease.Methods We collected fasting blood from participants,then extracted DNA from whole blood,and detected lipids from serum.We performed 54A/T FABP2 genotyping for participants in the case and the control groups by polymerase chain reaction (PCR) and DNA restriction enzyme digestion techniques.Results ( 1 ) In Hans population with coronary heart disease,the allelic frequency of point in 54T gene was 0.542,and 54A gene was 0.458 ; in urban Mongolian with coronary heart disease,the allelic frequency of point in 54T gene was 0.708,and that of 54A gene was 0.292.The allelic frequency of point in 54T and 54A genes were 0.284 and 0.716 respectively in the Han population control group and 0.353 and 0.647 respectively in the normal urban Mongolian group.Compared with the control groups,mutant 54T allele frequency in the case groups of Han and urban Mongolian was significantly higher ( Han:x2 =14.967,P < 0.05 ; Mongolian:x2 =28.083,P < 0.05 ).Compared with the Han Chinese population with coronary heart disease,the mutant 54T allele frequency in urban Mongolian with coronary heart disease was significantly (x2 =7.111,P < 0.05 ).There was no significant difference in mutant 54T allele frequency between the Mongolian control group and the Han control group ( x2 =3.392,P > 0.05 ).( 2 ) Patients with coronary heart disease who carry Thr54 ( + ) had a significantly higher level of fasting plasma TG [ CC Thr54 (-):( 1.89 ± 0.57 ) mmol/L,CC Thr54 ( + ):( 3.92 ± 1.63 ) mmol/L; CM Thr54 (-):(2.23 ± 0.13 ) mmol/L,CM Thr54 ( + ):(4.03 ± 1.14) mmol/L; Hans:P =0.001,Mongolian:P =0.035 ],and LDL-C [ CC Thr54 (-):( 3.09 ± 0.92) mmol/L,CC Thr54 ( + ):(4.05 ± 1.14 ) mmol/L ; CM Thr54 (-):( 4.26 ± 0.08 ) mmol/L,CM Thr54 ( + ):( 5.10 ± 0.56 ) mmol/L; Hans:P =0.025,Mongolian:P =0.045 ] than those with Thr54 (-).Conclusion (1) Threre is FABP2 gene polymorphism in urban Mongolian and Han population.Gene mutation frequency is in accordance with Hardy-Weinberg law of genetic equilibrium,suggesting the sample group representative.(2) FABP2 polymorphism may have a certain contribution to dyslipidemia in patients with coronary heart disease.FABP2 gene polymorphism may be associated with coronary heart disease.Individuals of Mongolian with FABP2 Thr54( + ) has a higher risk of coronary heart disease.
6.Effects of Radix Hedysari Flavonoid on Microvessel Density of Pulmonary Interstitial Fibrosis Model Rats

Objective To discuss the effects of Radix Hedysari flavonoids on microvessel density (MVD) of pulmonary interstitial fibrosis model rats, and provide evidence for its development. Methods SPF Wistar rats were divided into blank group, model group, prednisone group, and Radix Hedysari flavonoids high-, medium- and low-dosage group. Pulmonary interstitial fibrosis model was established by intratracheal instillation of bleomycin. From the second day after model established, each drug treatment group was administered with corresponding drugs intragastrically at different points in time for 14 and 28 days. MVD was detected by immunohistochemistry. Results Neomicrovessel was increased significantly in pulmonary tissue of model rats of 14 days treatment, but slightly decreased in 28 days. MVD in 14, 28 days model group was significantly more than blank group (P<0.01). MVD in 14, 28 days Radix Hedysari flavonoids high dosage group was significantly less than model group (P<0.01). MVD in Radix Hedysari flavonoids medium-dosage group was between high-and low-dosage group, and MVD in medium-dosage group was similar with that in model group. Conclusion Radix Hedysari flavonoids could inhibit the formation of neomicrovessel in a dose dependent manner.
7.Clinicopathologic analysis of 14 cases of atypical polypoid adenomyoma of endometrium with the emphasis on its cancerous transformation

Objective To analyze the clinicopathologic characteristics of atypical polypoid adenomyoma (APA) of endometrium,and investigate the special characteristics of cancerous transformation from APA.Methods Fourteen cases of APA were collected in General Hospital of People' s Liberation Army from January 2007 to March 2013.The clinical data,morphologic features,immunohistochemistry and the related literature were reviewed.Results The median age of the 14 patients was 38 years (ranged from 23 to 72 years),only 1 patient was postmenopausal.The most common symptom was irregular vaginal bleeding (4/14),and 4 patients were identified during routine physical examination for infertility.Among 14 cases,4 cases were diagnosed as well differentiated endometrioid adenocarcinoma originating from APA,and their median age was 35 years (ranged from 28 to 41 years); color Doppler flow imaging (CDFI) of ultrasound showed rich blood flow signal.The tumors with cancerous components were obviously larger than the usual APA (mean diameter:4.7 versus 1.8 cm).Histologically,irregular and branched glands were embedded in fibromuscular stroma and the glandular epithelium were atypical hyperplasia in varying degrees.While carcinoma developed in the APA,the sieve,solid and papillary structures were noticeable,and necrosis were common.Conclusions APA is a rare lesion of the uterus.Although the clinical behavior is benign in most cases,there may be possible for some cases developing carcinomas.If the APA mass is more than 4 cm in diameter,and microscopically demonstrates prominent sieve,solid,papillary structures and necrosis,the diagnosis of carcinoma developed from APA can be made.Thorough analysis should be done before the most proper therapeutic regimen is drawn up.
8.Morphological characteristics of eyes of female Uygur youth: an anthropometric study

Objective To study the morphological characteristics of eyes of female Uygur youth for providing the relevant data for eye aesthetic plastic surgery.Methods Compared with 150 cases of Han youth in the same school,the somatological survey was carried out in 150 female Uighur college students with the morphological characteristics of the eyes and good facial harmony.SPSS 17.0 was used for statistical analysis of the measured data in groups divided according to different race.Results 59.3% (89/150) of the Uighur young women had prominent brow ridges tall nose and superior orbit cave,18.7% (28/150) of them were similar to the Han type of eye,and 22.0% (33/150) in between.The rate of epicanthus in Uighur women was 28.0% (42/150),which was significantly lower than the Han nationality women 62.7% (94/150).Double eyelid incidence was 96.7% (145/150),which was significantly higher than that of Chinese Han women 67.3% (101/150).There was a no statistically significant difference between them in palpebral fissure length inner canthic diameter and outer canthic diameter (P>0.05),but a statistically significant difference was found in the height of palpebral fissure,the width of double eyelid and the distance between the eyebrow eyelid (P<0.05).Conclusions The Uighur women's eye morphology has dual characteristics of Caucasoid and Mongoloid.The data have great significance and value in guiding the eye's cosmetic plastic surgery.

Objective:To explore the association of the genes GSTM1 and GSTT1 with oligozoospermia infertility.Methods:PCR technique was used to analyze the GSTM1 and GSTT1 genes polymorphisms in 75 with oligospermia infertility and 36 healthy individuals of Zhuang population from Guangxi Baise area,and then the possibility function of GSTM1 and GSTT1 genes in human oligozoospermia were studied.Results:Analyses of the polymorphisms in GSTM1 and GSTT1 genes showed that GSTM1 defect or combined defects of both GSTM1 and GSTT1 was found more frequent in patients with infertile oligospermia than in the healthy control.Conclusion:GSTM1 and GSTT1 genes may have modulating effects on human spermatogenesis,whose mechanism needs further studies.
10.Establishment of a mouse model of Humulus pollen allergen-induced allergic asthma

Objective To develop a mouse model of allergic airway inflammation by using crude Humulus pollen extract.Methods A total of 24 Balb/c mice were divided randomly into two groups: control group and asthma model group with 12 in each.After sensitization by intraperitoneal injection,the mice were challenged by intranasal instillation of crude Humulus pollen extract.The total cell number and eosinophile percentage of bronchoalveolar lavage fluid(BALF) samples were determined.The lungs of mice were stained with hematoxylin and eosin to evaluate the degree of allergic inflammation.The cytokines in BALF and spleen homogenate were assayed by enzyme-linked immunoassay(ELISA).The total IgE in the serum was also measured by ELISA.Results Compared with the mice in normal control group,those in asthma group developed obvious allergic inflammation in the airways.Histological scoring for pulmonary inflammation significantly differed between normal group and asthma group(P
