OBJECTIVE:To observe clinical efficacy of clomiphene combined with metformin in the treatment of polycys-tic ovary syndrome. METHODS:112 patients with polycystic ovary syndrome admitted in gynaecology and obstetrics depart-ment of our hospital were selected,and randomly divided into control group and observation group,with 56 cases in each group. Control group was treated with clomiphene orally,since fifth day of menstruation,once a day,50 mg/time;observa-tion group was additionally treated with metformin orally,30 min before meal,3 times a day,500 mg/d. Treatment course last-ed for 2 months. Biochemical indicators,control time of disease,actual treatment time,HOMA-IR,HOMA-β and clinical effi-cacy of patients were observed in two groups. RESULTS:After treatment,LH,FSH,T and FINS of patients in observation group were lower than before treatment and control group,with statistical significance(P<0.05);control time of disease and actual treatment time of observation group were significantly shorter control group (P<0.05),with statistical significance;HOMA-IR and HOMA-β of observation group were significantly lower than those of control group,with statistical significance (P<0.05);clinical effective rate of patients in observation group was significantly higher than control group,with statistical significance (P<0.05). CONCLUSIONS:Clomiphene combined with metformin in the treatment of polycystic ovarian syn-drome can improve biochemical indicators,shorten control time of disease and actual treatment time and enhance clinical effi-cacy.

Objective To observe the neurologicaI functional recovery after intravenous transplantation of human umbilical cord blood(HUCB)CD34~+ cells transfected with glial cell-derived neurotrophic factor (GDNF) and green fluorescent protein (GFP) in SD rats with middle cerebral artery occlusion (MCAO).and tO investigate the survival,migration and neural differentiation of the graft cells.Methods (1)CD34~+ cells were isolated from HUCB using centrifuge combined with immune beads and then identified by flow cytometry,and transfected by the recombinant plasmid of GDNF.GFP or GFP plasmid by liposome method.(2) Sixty aduh male SD rats with MCAO were randomly divided into three groups (n=20 in each group):GDNF-GFP-CD34~+ cells group,in which the GDNF-GFP-CD34~+ cells were transplanted intravenously at the 24th h after the establishment ofmodels of MCAO;GFP-CD34~+ cells group:in which the GFP-CD34~+ cells were transplanted intravenously at the Same time;Normal saline group,in which normal saline was injected at the same time.Fifteen SD rats served as sham-operated group.(3) Neurological functional measurements were performed using the modified neurolc'gical severity score.Quantitative histological determinations of infarct volume were performed using standard TTC staining and quantitative image analysis. The GDNF level in the cell culture or the cerebral tissue was measured by ELISA. Meanwhile, the survival and migration of GFP-labeled CD34~+ cells and the expression of astrocytie marker-GFAP and the neuron marker-neuronal nuclei (NeuN) were detected by immunohistochemical and fluorescent staining. Results (1) The GDNF level in the cell culture was significantly higher in GDNF-GFP-CD34~+ cells than in GFP-CD34~+ cells (P<0. 05). (2) No significant difference was found in the modified neurological severity score at the day 7 after transplantation among the three groups with MACO, but at day the 28 after transplantation, the neurological function in GFP-GDNF-CD34~+ group was improved significantly (5.0±1. O) as compared with the GFP-CD34~+ cells (5. 9 + 1.4) or saline groups (7. 0±1.7) (P<0. 05). The cerebral infarct volume in GFP-GDNF-CD34~+ group (142±44mm~3) was significantly decreased as compared with the GFP-CD34~+ group (196±58 mm~3) (P<0. 05) or saline group (233<50 mm~3 ) (P<0. 01 ). The GDNF level in cerebral tissue in GFP-GDNF-CD34~+ group was significantly increased as compared with the GFP-CD34~+ group (P<0. 05) at the 28th day after treatment. At the 28th day after treatment, the NeuN positive cells (6.7±2.0), and GFAP positive ceils (14.1±3.3) in GFP-GDNF-CD34~+ group were significantly increased as compared with the GFP-CD34~+ group (P<0. 05), but there were no positive cells in sham-operationgroup. Conclusion Intravenous transplantation of GDNF gene-modified CD34~+ cells from human umbilical cord blood could improve the neurological function in rats with MCAO. The increased GDNF level in cerebral tissue was one of possible mechanisms responsible for the different improvements.

This article was aimed to observe the intracardiac pressure under different states by electric acupuncture (EA) on yuan-source point of PC7-Daling and to explore the acupuncture effect features. Catheterization was applied through the right common carotid artery to the left ventricle in order to record the left intraventricular pressure. The intracardiac pressure was used as an indicator. The effects of EA on PC7-Daling to the left intraventricular pressure of rats were observed under the states of normal physiology, isoproterenol and propranolol, respectively. The results showed that for the immediate effects of acupuncture, EA on PC7-Daling can obviously excited the intracardiac pressure of normal rats. Under the state of intravenous application of propranolol and isoproterenol, no obvious reac-tions during EA on PC7-Daling. From the effects of acupuncture, under normal conditions, EA on PC7-Daling had excitatory effect; under propranolol state, it had inhibitory effects; and the effect of isoproterenol state was variable. It was concluded that according to the specificity of acupoint effect, PC7-Daling can excite intracardiac pressure.

Objective To investigate the changes and roles of the long non-coding RNA (IncRNA)during the reprogramming of human induced pluripotent stem cells. Methods Agilent Human lncRNA (4 × 180K) chip was used to check the expression of lncRNA in somatic cells, induced pluripotent stem cells and embryonic stem cells. Compared with differentially expressed lncRNA in somatic cells and induced pluripotent stem cells, lncRNA was selected that may play an important role during the reprogramming of human pluripotent stem cells. Results The lncRNA expression profiles in induced pluripotent stem cells were similar to embryonic stem cells, but were different from the somatic cells. A total of 3 156 differentially expressed lncRNAs were found between stem cells and somatic cells by cluster analysis, and 222 differentially expressed lncRNAs were found during the reprogramming process of human pluripotent stem cells by biological analysis. Conclusion lncRNA may play an important role in reprogramming process of human pluripotent stem.

Objective To observe the effect of Tongguanluo capsule in treating oviduct obstructive infertility and analyze the potential mechanism. Methods 106 patients with oviduct obstructive infertility were randomly divided into the observing group and the control group, 53 cases of each group. The observing group was treated by Tongguanluo capsule and the control group was treated by Huoxue Huayu Capsule. The effects were compared after 4 weeks. OBJECTIVE:To develop an HPLC method for the determination of L-Carnosine in Polaprezine granules.METHODS:The Chromatographic column was Agilent ODS with detection wavelength of 215 nm and flow rate of 1 mL?min-1.The mobile phase consisted of sodium subsulfite buffer solution(pH=3)-acetonetrile(77:23).The column temperature was set at 30 ℃ and injection size was 10 ?L.RESULTS:The good linearity was obtained in the range of 0.1559～3.1584 ?g for L-Carnosine(r=1.0000).The average recovery was 99.65%(RSD=0.078%).CONCLUSION:The method is simple,accurate and sensitive,and it is suitable for content determination of L-Carnosine in Polaprezine granules.

Objective:To screen the potential characteristic gene spectrums and signal pathways of osteoarthritis based on gene chips.Methods:We analyzed 2 microarrays of human joint synovial tissue (GSE82107 and GSE55235) derived from the Gene Expression Omnibus (GEO) database and included for this study 20 osteoarthritis (OA) samples and 17 healthy control samples. The differentially expressed genes (DEGs) between OA and HC were screened by GEO2R tool. Analyses of Gene Ontology function and Kyoto Encyclopedia of Genes and Genomes pathway enrichment were performed using the Database for Annotation, Visualization and Integrated Discovery to identify the pathways and functional annotations of DEGs (https://david.ncifcrf.gov/). Protein-protein interaction of these DEGs was analyzed based on the Search Tool for the Retrieval of Interacting Genes database and visualized by Cytoscape software (http://www.string-db.org/).Results:191 up-regulated DEGs and 49 down-regulated DEGs were screened out from the 2 microarray databases. Enrichment of DEGs was mainly found in regulation of such biological functions as "inflammation" , "bone cell differentiation" and "positive apoptotic cell regulation" , HTLV-I infection, silk crack on the original amp-activated protein kinase (MAPK) signaling pathway, swine flu, tumor necrosis factor (TNF) signaling pathway, the nf-kappa B signaling pathway, PI3 kinase/Akt pathway, toll-like receptor pathway, legionella, salmonella and other 14 signaling pathways. In 2 modes of MNC and Degree, the top 10 core genes were screened, of which interleukin-6 (IL6), JUN, chemokine 8 (CXCL8), early reaction growth factor (EGR1) and cyclin (CCND1) were identified as valuable biomarkers of OA.Conclusions:Based on GEO chips, 10 characteristic gene profiles such as IL6, JUN, CXCL8, EGR1, CCND and 14 signal pathways such as tumor necrosis factor (TNF) signal pathway, NF-κB signal pathway, PI3 kinase/Akt pathway and Toll-like receptor pathway were screened, which may provide new clues for understanding of the pathogenesis of osteoarthritis.

BACKGROUND: How to make growth factor plays a role persistently and efficiently is a key in constructing bone tissue engineered bone. Currently, varied microspheres or scaffolds were used as release carriers, however, the delayed release effects needs elevating.OBJECTIVE: To prepare chitosan microosPheres/nano-hydroxyapatite/poly (lactic-co-glycolic acid) (CMs/nHA/PLGA) scaffolds, and to measure its characteristics of delayed release of bovine serum albumin (BSA).METHODS: CMs were prepared by an emulsifying cross linking method with BSA as a model protein. Using ice particulates as porogen, composite CMs/nHA/PLGA scaffolds were prepared by freeze-drying. The characteristic and morphology of the composite were observed by scanning electron microscope, later particle size analyzer, mercury porosimeter and universal testing machine, and the release behavior of BSA was investigated in vitro.RESULTS AND CONCLUSION: The CMs were spherical shape with a regular surface, with diameters of 20-40 μm. The encapsulation efficiency of the CMs was 86.5%, and the loading capacity was 0.8%. With the increase of initial BSA dosage, the loading capacity increased to 2.6%, while the encapsulation efficiency decreased to 74.1%. The CMs can be uniformly distributed in PLGA scaffolds to form CMs/nHA/PLGA scaffolds, which had 100-400 μm pore diameter and over 80% porosity, with 1.1-2.3 pMPa compressive strength, and 26.5% cumulative degradation at 10 weeks. The cumulative release of BSA from nHA/PLGA scaffolds was above 85% at 36 hours, which from CMs was 33.6% at 10 days, and that from CMs/nHA/PLGA scaffolds was 81.5% at 40 days. The results demonstrated that CMs/nHA/PLGA scaffolds have an excellent releasing efficiency for protein drugs with suitable compressive strength and degradation, which would be used as delivery system and tissue engineering scaffolds.

Objective:To establish the quality standard for Kechuanning granules. Methods: A TLC method was used for the qualitative study on the main ingredients, Radix stemonae,ephedra and bitter almond. An HPLC method was used to determine the con-tents of ephedrine hydrochloride,pseudoephedrine hydrochloride and amygdalin. The separation of targeted compounds was performed on a Tech Mate C18-ST column (250 mm × 4. 6 mm,5 μm) . The mobile phase consisted of methanol(A) and 0. 1% phosphoric acid (B) with gradient elution at a flow rate of 1. 0 ml·min-1. The detection wavelength was 207nm and the column temperature was 35℃. Results:The spots in TLC were quite clear without any interference from negative control. The linear relationship of ephedrine hydrochloride,pseudoephedrine hydrochloride and amygdalin was 1. 826-182. 600μg·ml-1( r=0. 9997),1. 848-184. 815μg·ml-1 (r=0. 9997) and 2. 981-298. 069 μg·ml-1(r=0. 9996),respectively. The average recovery was 97. 00%,97. 40% and 98. 30%(RSD=3.7%,2.5%,1.6%)(n=9),respectively. Conclusion: The method is sensitive,accurate,simple and reproducible,which can be used for the quality control of Kechuanning granules.

OBJECTIVE:To observe clinical efficacy and safety of Modified xiaohuang paste in the treatment of paralytic ileus after thoraeolumbar fractures.METHODS:One hundred and thirty-eight patients with paralytic ileus after thoracolumbar fractures were divided into control group (group A,43 cases),Xiaohuang paste group (group B,47 cases) and Modified xiaohuang paste group (group C,48 cases).Group A was given routine treatment as fasting,gastrointestinal decompression,fluid replacement,nutritional support;group B was additionally given Xiaohuang paste on the basis of group A;group C was additionally given Modified xiaohuang paste on the basis of group A.Group B and C were given relevant paste every 12 h until intestinal peristalsis was recovered and gas exhausted from anus,at the most for 5 days.The improvement time of clinical symptom improvement,VAS score and CRP level were compared among 3 groups as well as the occurrence of ADR.RESULTS:After treatment,the time of gastrointestinal decompression,bowel sound recovery and passage of gas by anus in group B and C were significantly shorter than group A,and group C was significantly shorter than group B,with statistical significance (P＜0.05).Before treatment,there was no statistical significance in VAS score and CRP level among 3 groups (P＞0.05).After treatment,VAS score of abdominal pain and distension,CRP level of 3 groups were decreased significantly compared to before treatment;those of group C were significantly lower than group A and B,and CRP level of group B was significantly lower than that of group A,with statistical significance (P＜0.05).There was no statistical significance in VAS score between group A and B (P＞0.05).No significant ADR was found in 3 groups.CONCLUSIONS:For paralytic ileus after thoracolumbar fracture,Modified xiaohuang paste can significantly shorten treatment duration,relieve abdominal distension and pain,inflammation with good safety.
