OBJECTIVE: To observe the clinical efficacy of Sodium Hyaluronate Eye Drops in combination with Recombinant Bovine Basic Fibroblast Growth Factor Eye Drops for dry eye after undergoing eximer laser in-situ keratomileusis(LASIK).METHODS: 62 cases(100 eyes) of patients with dry eye after undergoing LASIK were randomly divided into two groups: 32 cases(53 eyes) in the treatment group were treated with Sodium Hyaluronate Eye Drops in combination with Recombinant Bovine Basic Fibroblast Growth Factor Eye Drops,and 30 cases(47 eyes) in the control group with Sodium Hyaluronate Eye Drops alone(4 times/day,1~2 drops/time) for 2 months.The drug efficacy was observed for 1~3 months since 1 week after the initiation of medication.RESULTS: The total effective rate in the treatment group vs.the control group was 98.11% vs.89.36%(P

Objective To observe the changes of LoVo cell growth by galectin-1.Methods Eukaryotic expression vector of galectin-1 was constructed and transfected into LoVo cells using lipofectamineRM 2000.Immunochemistry was employed to detect galectin-1 expresson.LoVo cell proliferation and apoptosis were observed.Results Galectin-1 eukaryotic expression vector pEGFP-C1/GAL1 was successfully constructed.Three cell clones,p-GAL1-LoVo and p-LoVo,transfected with pEGFP-C1/GAL1 and pEGFP-C1 correspondingly,and LoVo were cultured successfully.Galectin-1 expression downregulated Bcl-2 level only occurring in p-GAL1-LoVo cells.p-GAL1-LoVo cell proliferation was similar to p-LoVo and LoVo cells,but p-GAL1-LoVo cell apoptosis was increased(9.61?0.56)%,as compared with p-LoVo and LoVo cells,[(3.56?0.53)% and(3.46?0.46)% respectively](P
5.The Expression and Clinical Significance of DKK1, SFRP4 and Wnt1 in Cervical Squamous Cell Carcinoma

Objective To investigate the expressions of DKK1,SFRP4 and Wnt1 in cervical squamous cell carcino-ma(SCC), and the clinical significance thereof. Methods There were 76 samples of cervical squamous cell carcinoma were included in SCC group and 36 benign uterine resection specimens were control group (NC). The immunohistochemical meth-od was applied to detect the expressions of DKK1,SFRP4 and Wnt1 in two groups. Results The expression of DKK1 was significantly lower in SCC group than that in NC group (P<0.05). The expression levels of SFRP4 and Wnt1 were significant-ly higher in SCC group than those of NC group (P<0.05). There were significant differences in the expressions of DKK1, SFRP4 and Wnt1 between samples of different clinical staging, differentiation, sizes of tumor and lymph node metastasis (P<0.05). The expression of DKK1 was negatively correlated with SFRP4 and Wnt1 in SCC group (P<0.05). The expression of SFRP4 was positively correlated with Wnt1 in SCC group (P<0.05). Conclusion The roles of SFRP4 and Wnt1 are syn-ergistic interactions in the development of SCC. DKK1 is an inhibiting factor of SCC.
6.Construction of RNAi recombinant vector of galectin-1 and establishment of LST-R1 cell lines stably transfected by the recombinant vector

Objective To construct the RNA interference (RNAi) eukaryotic expression vectors of galectin-1, and establish the LST-R1 cell lines stably transfected by RNAi vectors. Methods Two pairs of small interfering RNAs (siRNA) targeting to galectin-1 mRNA (GenBank: NM002305) were designed. The corresponding single-strand short hairpin interfering RNAs (shRNA), containing BamH Ⅰ, Hind Ⅲ sites and a 9nt hairpin structure, was synthesized and annealed. The annealed product and the linear eukaryotic expression plasmid pSuperior-puro, which were digested with Bgl Ⅱ and Hind Ⅲ, were ligated by T4 ligase to set up the interfering system. The recombined plasmid was identified with EcoR Ⅰand Hind Ⅲ enzyme digestion and sequencing, and co-transfected to LST-R1 cells with pcDNA6/TR with Lipofectamine2000. Positive clones were selected with 0.8?g/ml puromycin. After incubated with 4?g/ml tetracyclin for 48 hours, RT-PCR, Western blotting and immunochemistry were employed to determine galectin-1 mRNA and protein levels. Results Sequencing results suggested that the nucleotide sequence and read frame of RNAi eukaryotic exprssion vector of galectin-1, p-shRNA1 and p-shRNA2 were perfect. Stably transfected LST-R1 cell lines of p-shRNA1-LST and p-shRNA2-LST were established. The relative values of galectin-1 expression in LST-R1 cells, p-shRNA1-LST cells, p-shRNA2-LST cells and p-LST cells by RT-PCR were 0.616, 0.298, 0.373 and 0.641, respectively, and 1.00, 0.07, 0.38 and 0.97 by Western blotting. The p-shRNA1 gave the best interfering effect, which was in conformity with the results of immunochemistry measurement. Conclusions RNAi eukaryotic expression vector of galectin-1 mRNA has been successfully constructed. Establishment of the stably transfected LST-R1 cell lines may lay a foundation to explore the roles of galectin-1 in laterally spreading tumor.
7.Effects of Group Psychotherapy on Breast Cancer Patients

Objective To explore the effects of group psychotherapy on breast cancer patients. Methods 80 outpatients with breast cancerwho did not participate in the Rukang Salon (control group) and 100 outpatients participated (intervention group) were surveyed withquestionaire. Results The incidence of psychological distress was 36.8% in the control group and 21.1% in the intervention group ( χ2=5.02,P=0.03). In the intervention group, the efficacy of those without psychological distress (236.86±35.17) was significantly higher than thosewith psychological distress (214.21±26.52), (t=2.61, P=0.01). There were significant differences in those 5 factors: group cohesion, universality,interpersonal learning, guide instructions, and hope remodeling. Top 5 factors in scores reported were interpersonal learning, hopingremodeling, group cohesion, guide instructions, and exist consciousness. Conclusion Group psychotherapy can significantly reduce the psychologicaldistress in rehabilitating breast cancer patients. Group cohesion, universality, interpersonal learning, guide instructions, hope remodelingand exist consciousness may play important roles.
8.Investigation of Life Satisfaction and Related Factors in Primary Caregivers of Breast Cancer Patients

Objective To investigate the life satisfaction and its related factors in primary caregivers of breast cancer patients. Methods The Life Satisfactory Index A (LSIA) and self-administrated questionnaire were used. The life satisfaction between breast cancer patients'primary caregivers and breast benign patients' primary caregivers were compared. The factors of the life satisfaction in primary caregivers of breast cancer patients were analyzed. Results The life satisfaction was poorer in the breast cancer patients' primary caregivers than in the breast benign patients' primary caregivers (P<0.01). The life satisfaction in the breast cancer patients' primary caregivers was correlated with the stage of breast cancer, course of disease, primary caregivers' age and family monthly income (P<0.05). Conclusion The life satisfaction in breast cancer patients' primary caregivers is worse and is related with stage of breast cancer, course of disease, primary caregivers' age and family monthly income.
9.Effects of Radix Hedysari Flavonoid on Microvessel Density of Pulmonary Interstitial Fibrosis Model Rats

Objective To discuss the effects of Radix Hedysari flavonoids on microvessel density (MVD) of pulmonary interstitial fibrosis model rats, and provide evidence for its development. Methods SPF Wistar rats were divided into blank group, model group, prednisone group, and Radix Hedysari flavonoids high-, medium- and low-dosage group. Pulmonary interstitial fibrosis model was established by intratracheal instillation of bleomycin. From the second day after model established, each drug treatment group was administered with corresponding drugs intragastrically at different points in time for 14 and 28 days. MVD was detected by immunohistochemistry. Results Neomicrovessel was increased significantly in pulmonary tissue of model rats of 14 days treatment, but slightly decreased in 28 days. MVD in 14, 28 days model group was significantly more than blank group (P<0.01). MVD in 14, 28 days Radix Hedysari flavonoids high dosage group was significantly less than model group (P<0.01). MVD in Radix Hedysari flavonoids medium-dosage group was between high-and low-dosage group, and MVD in medium-dosage group was similar with that in model group. Conclusion Radix Hedysari flavonoids could inhibit the formation of neomicrovessel in a dose dependent manner.
10.The prevalence and charateristics of carotid atherosclerosis among healthy middle-aged people in community

Objective To investigate the prevalence and charateristics of carotid atherosclerosis among healthy middle-aged people of community. Methods Totally 1930 subjects aged 50 years and above were selected randomly from 10027 participants recruited during Phase 3 of the Guangzhou Biobank Cohort Study (GBCS).Common carotid artery intima-media thickness (CCA-IMT) and carotid plaques (CP) were measured by B-mode ultrasonography using ALT HDI 3000 mainframe with a high-resolution. Results The prevalences of CCA-IMT and CP in men were higher than women [41.5%(400/963)vs.16.2 (157/967),34.1% (328/967) vs.13.8% (133/967),both P<0.001].The prevalence of CCA-1MT was higher with age increasing in either men or women in groups of 50-yrs,60-yrs and 70-79 yrs (all P<0.001).The most of CP was cankerous spot (37.8%),secondly hard spot (24.9%),soft spot (22.1%) and the prevalence of flat spot was lowest (15.2%).The number of CP was not significantly different between the left and right side (51.0% and 49.0%).The most common location of CP was bifurcations of carotid arteries (61.7 %),next the common carotid artery (21.0%) and internal carotid artery (ICA) was lowest (17.3 %).The prevalence of CP was 44.4% (83/87) in women aged 50- yrs,it was higher than that in men 20.9% (108/517).However,the prevalence of CP was 79.1% (409/517) in men aged more than 60 yrs,higher than that in women (55.6%). Conclusions The carotid atherosclerosis is commonly encountered disease in the male elderly,and the most of CP was cankerous spot in bifurcations of carotid arteries
