OBJECTIVE:To investigate the effect of Shuxuening on interleukin 18 (IL-18) in patients with early diabetic nephropathy.METHODS:68 patients with early diabetic nephropathy were randomly divided into 2 groups (n=34).Both groups were given conventional diabetes therapy and oral dose of irbesartan (150 mg,qd).Treatment group were additionally treated with Shuxuening injection 20 mL and sodium chloride injection 250 mL (i.v.qd) for 3 weeks.The levels of IL-18 of 2 groups before and after treatment were determined by enzyme-linked immunosorbent assay,and the albumin excretion rate (UAER),fasting plasma glucose (FPG),serum creatinine (Scr) and other indicators were also detected.RESULTS:Compared with before treatment,the levels of IL-18 of 2 groups decreased after treatment (P0.05).CONCLUSION:Shuxuening can inhibit inflammatory cytokines IL-18 in peripheral blood of patients with diabetic nephropathy and have protective effects on kidney,which may be adjuvant therapy for diabetic nephropathy.

OBJECTIVE:To explore the efficacy and safety of leflunomide combined with prednison in the treatment of membra-nous nephropathy. METHODS:43 patients with primary membranous nephropathy were randomly divided into control group(21 cas-es) and test group (22 cases). Control group given cyclophosphamide interrupted shocks combined with prednison,test group re-ceived leflunomide combined with prednison,the treatment course for both groups was 12 months. 6 months after the treatment,clini-cal efficacy,changes in renal function and serum lipids indexes before and after treatment,and the incidence of adverse reactions in 2 groups were observed. RESULTS:The total effective rate in test group was 63.64%,which was significantly higher than control group (23.81%),the difference was statistically significant (P<0.05). After treatment,urine protein in 2 groups significantly de-creased,plasma albumin significantly increased,serum creatinine,serum cholesterol,triglycerides and low-density lipoprotein signifi-cantly decreased,the differences were statistically significant(P<0.05);and the changes of above-mentioned indexes(except blood cholesterol)in test group were more significantly than control group,the differences were statistically significant(P<0.05). The inci-dence of adverse reactions in test group was 9.09%,control group was 9.52%,there was no significant difference between 2 groups (P>0.05). CONCLUSIONS:Compared with cyclophosphamide interrupted shocks combined with prednison,the efficacy of lefluno-mide combined with prednison in the treatment of membranous nephropathy is more definite,and the improvement of renal function and serum lipids indexes is more obvious.

Objective To study the gait footprint parameters of the normal children and the cerebral palsy (CP) children, and to explore its clinical value. Methods A total of 2 800 normal children aged 3 to 10 years and 139 spastic CP children aged 3 to 5 years were recruited in this study. The normal children were divided into seven groups with one year interval, and were measured with regard to the length of foot and step, step width and foot angle of footprint of every age group with self made oil printed carpet. Objective To compare the advantages and disadvantages of the four DNA extraction methods according to the endophytic diversity in the roots, stems, and leaves of mulberry analyzed by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) , and by taking the yield, purity and PCR amplification as indexes. Methods Four common methods, i.e., cetyltrimethyl ammonium bromide ( CTAB) , sterile phosphate buffered saline (SPBS) vibration, liquid nitrogen grinding (LNG) , and KIT methods, were used to extract the total DNA from different tissues of mulberry, and then were compared based on the diversity analysis results for endophyte by PCR-DGGE. Results From the roots and stems of mulberry, we got the highest concentration of DNA by LNG extraction method, and got the lowest concentration by SPBS extraction method. But for the leaves of mulberry, the results of the four extraction methods were completely opposite to those for the roots and stems. For different tissues of mulberry, the purity of DNA extracted by KIT method was the best. According to the endophytic bacteria diversity analyzed by 16S rDNA PCR-DGGE, the appropriate method for extraction of DNA was LNG or CTAB, but was not KIT. And according to the results of endophytic fungi diversity analyzed by ITS PCR-DGGE, the best extraction method was KIT, and the unsuited methods varied from the tissues of mulberry. Conclusion The optimum DNA extraction method for mulberry varies from the tissues of mulberry and endophtic bacteria.

ABSTRACT:Objective To investigate the differential gene expression of nasopharyngeal carcinoma cell line CNE-2 and CNE-2R exposed to X-rays.Methods We used neutral comet assay to detect the intrinsic radiosensitiv-ity of tumor cells,immunofluorescence to detect the phosphorylated histone γH2AX and Rad5 1 foci formation and to assay the time-dose effects on cell radiation injury and radiation radiosensitivity change after irradiation.The dif-ferential gene expression was screened with cDNA microarray by DNA damage repair related gene chip.Differenti-ally expressed proteins screened were confirmed by Western blot.Results After 2 h of 4 Gy 9MeV-β-ray irradia-tion,CNE-2R cells had aggravated DNA damage compared to CNE-2 cells and the damage was more obvious with the prolongation of time.It suggested that cell DNA damage severity was more obvious.The immunofluorescence showed the positive rates ofγH2AX cells in CNE-2 cell line were higher than those in CNE-2R cell line.There were 37 different genes related to DNA damage repair in CNE-2 cell line compared with CNE-2R cell line,while 24 up-regulated and 13 down-regulated genes were found in SiHaR cells.In them,there were 6 genes whose ratios were 6 times higher and 3 genes whose ratios were lower than 0.1.Western blot verified the results of four differentially expressed proteins compared with CNE-2 cells,in which GADD45a and RRAD1 expression levels were significantly lowered in CNE-2R while the RAD9a and RCF2 protein significantly increased.Conclusion The radioresistance of CNE-2R cells is significantly related to DNA damage repair capacity in certain mutations at the gene level.It is possible to regulate cell radiosensitivity by regulating the related gene.

Objective To investigate the distribution of five indicators of hepatitis B(HBV-M)and its relationship with liver function parameters and HBV-DNA load in patients with chronic HBV infection in different stages.Methods The serum samples were collected from 456 patients infected with HBV.The HBV-M,liver functional parameters and HBV-DNA level were quantita-tively detected.According to the stages of disease,the patients were divided into 3 groups including chronic hepatitis B group(inclu-ding mild subgroup,moderate subgroup and severe subgroup),liver cirrhosis group(including compensatory subgroup,decompen-sated subgroup)and hepatocellular carcinoma goup.Results The ratio of HBsAg,HBeAg,HBcAb positive pattern(135 pattern) and HBsAg,HBeAb,HBcAb positive pattern(145 pattern)in the three groups were statistically different(P <0.05).In each chron-ic hepatitis B subgroup,both ALT and AST levels of 135 pattern were significantly higher than the other two patterns(P <0.05). In each liver cirrhosis subgroup and hepatocellular carcinoma group,there were statistically significant differences in ALB and TBIL levels between the three patterns(P <0.05 ).In each group,the HBV-DNA level of 135 positive pattern were significantly higher than the other two patterns(P <0.05).Conclusion With the advancement of chronic hepatitis B,there is a downtrend in the ratio of 135 pattern and increasing trend in the ratio of 145 pattern.If the stage of hepatitis B is discriminated,ALT,AST,ALB TBIL and HBV-DNA level were closely related to HBV-M pattern.

OBJECTIVE To analyze the effect of nanometer light catalysis decontamination machine on air disinfection in the hyperbaric oxygen(HBO) chamber in the hospital.METHODS The nanometer light catalysis decontamination machine was used to disinfect the HBO chamber air at the time of thirty minutes after patients leaving the chamber when the HBO therapy was finished,then the amount of bacteria in the air was monitored before and 1,2 and 3 hours after disinfection,respectively.RESULTS After disinfection using nanometer light catalysis decontamination machine,the amount of bacteria in the chamber air decreased from 764-1989 CFU/m3 before disinfection to 31-268 CFU/m3.The mean ratio of natural bacteria dissolution was more than 80%.CONCLUSIONS Using nanometer light catalysis decontamination machine is a safe and effective method for air disinfection in the HBO chamber.

OBJECTIVE To study the changes in air bacteria content under gradual circuit model and ordinary circuit model in operating room.METHODS Altogether 80 cases with thyroid gland surgery and gastrointestinal surgery were selected,and divided them randomly into two groups: the gradual circuit model and the ordinary circuit model,each group had 40 cases.The period of early morning to the end of operation was divided into 10 time spots to carry on the air bacilliculture.RESULTS Under the ordinary circuit model,air colony showed straight up with time during the two types of surgical operations,and there was no significant difference between the two types of surgery.Under the gradual circuit model,air colony began to increase with time,followed by steady,or even decline,no differences between the two types of surgery. OBJECTIVE: To study the sedative and antistress action of Shenqiwuweizi capsule. METHODS: The sedative and hypnotic experiments were made to observe the action of Shenqiwuweizi capsule on the mice's sleep induced by pentobarbital sodium at subthreshold dosage or hypnotic dosage. The swimming and hypoxia tolerance experiment were performed to observe the effects of Shenqiwuweizi capsule on the swimming time and hypoxia tolerance time in mice. RESULTS: Shenqiwuweizi capsule could obviously prolong the sleeping time of mice induced by pentobarbital sodium, increase the number of sleeping animals caused by pentobarbital sodium at subthreshold dosage, and significantly prolong the time of swimming and hypoxia tolerance in mice. CONCLUSION: Shenqiwuweizi capsule had sedative and anti-stress action.
