1.Effects of proliferation of rat hepatic stellate cells and collagen-Ⅰ production by antisense oligodeoxynucleotides against platelet-derived growth factor B-chain

AIM: To investigate the effect of antisense oligodeoxynucleotides against platelet-derived growth factor B-chain(PDGF-B) in the proliferation of cultured rat hepatic stellate cells and collagen-Ⅰ production.METHODS: The phosphorothioate antisense oligodeoxynucleotides and missense oligodeoxynucleotides were synthesized.The rat hepatic stellate cells(HSCs) were treated with these oligodeoxynucleotides at different concentrations,respectively.The inhibition of the proliferating of cultured rat hepatic stellate cells was assayed by MTT method.The expressions of PDGF-B and collagenⅠ mRNA were detected by means of reverse transcription-polymerase chain reaction(RT-PCR).The expression of endogenous PDGF-B was determined by flow cytometry(FCM).Collagen-Ⅰin the supernatant was determined by ELISA.RESULTS: The results showed that the ASODN against the PDGF-B at a final concentration of 10 ?mol/L inhibited the HSCs proliferation,the expressions of PDGF-B and collagen-ⅠmRNA.FCM and ELISA demonstrated that the expression of endogenous PDGF-B and the production of collagen-Ⅰin HSCs were significantly lower than those in controls,whereas the controls(missense oligodeoxynucleotides) had no effect at the same concentration.CONCLUSION:PDGF-B ASODN inhibits the proliferation of cultured rat hepatic stellate cells,the endogenous expression of PDGF-B and the production of collagen.PDGF-B ASON might be useful for gene therapy in liver fibrosis.
2.Activation of adenosine A_1 receptor inhibits glucose-induced cardiomyocyte hypertrophy

Aim To demonstrate the effects and mechanism of adenosine A1 receptor agonist R(-)-N6-(2-phenylisopropyl) adenosine(R-PIA) on high glucose(HG)-induced myocardial hypertrophy by in vitro cultured myocardial cells from neonatal rats.Methods The protein content was assayed by the method of Lowry. The expression of p-ERK1/2 and ERK1/2 was determined by Western blot.The [Ca~(2+)]I transient changes of cell loaded Fura-2/AM were measured by Till image system.Results 1 μmol·L~(-1) R-PIA and U0126 inhibited similarly HG-induced increase of the protein content and [Ca~(2+)]I transient along with the relative expression of p-ERK1/2.These responses were completely abolished by adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(CPDPX).Conclusion Adenosine A1 receptor stimulation significantly inhibits HG-induced myocardial hypertrophy by mediating ERK1/2 pathway and Ca~(2+).
3.Effectiveness of application of case management for hypertension in the elderly

Objective To explore application of case management for hypertension in the elderly.Methods In total, 106 elderly patients with hypertension treated at outpatient and inpatient departments of the First Affiliated Hospital of Xian Jiaotong University were recruited during January to October 2007, and divided randomly into two groups, one with conventional management and the other with case management.Patients with conventional treatment accepted routine diagnostic and therapeutic procedures and were followed-up passively, and patients with case management accepted integrated therapy and active interview provided by a team with a cardiovascular specialist as core, were followed-up actively and their medical records were kept in files. Percentage of the patients with blood pressure lowered to meet criteria of effectiveness and their improvement in depression and anxiety status and quality of life were evaluated two years after management for the two groups. Results Percentage of patients with their blood pressure lowered to meet effective criteria increased significantly in both of the groups, from 32. 8% to 67.5% in patients with conventional management and from 33.7% to 83.4% in those with case management ( P = 0. 007 ),respectively. Scores of self-rating anxiety scale ( SAS) and self-rating depression scale (SDS) decreased in patients of the both groups, from 57 ± 10 and 56 ± 11 before management to 40 ±9 and 48 ± 10 after it,respectively in patients of conventional management ( P = 0. 032 ), and from 56 ± 10 and 57 ± 10 before management to 35 ±9 and 40 ± 9 after it ( P = 0. 008 ), respectively in those with case management; and scores of quality of life improved in patients of the two group, from 65 ± 13 before management to 85 ± 14 after it in those with conventional management, and from 64 ± 13 before management to 98 ± 14 after it in those with case management ( P = 0.017 ), respectively. Conclusions Case management is better for chronic diseases and recommended in management for the elderly with hypertension, which can improve their targets of blood pressure lowering, decrease their scores of SAS and SDS and improve their quality of life,with an integrated, in-depth, continuous management.
4.The value of diagnosis and treatment of conization of cervix to cervical intraepithelial neoplasia Ⅱ-Ⅲ grade

Objective To explore the value of diagnosis and treatment of conization of cervix to cervical intraepithelial neoplasia(CIN)Ⅱ-Ⅲ grade.Methods The data of 76 patients diagnosed as CIN Ⅱ-Ⅲ grade through pre-operative multiple-punch biopsies from July 1st 2006 to June 30th 2007 was retrospectively analyzed.Compared the pathohistology and treatment methods before and after conization of cervix.Results Accordance rate of diagnosis in pre-operative cervical biopsies under vaginoscopy was 48.68%,pathological up-grade were 24 cases(31.58%),among invasive carcinomas were 8 cases(10.53%),pathological down-grade were 15 cases(19.74%).Conclusion Diagnostic cervical conization could enhance the rate of diagnosis,and prevent the omission of invasive carcinomas for the patients with CIN Ⅱ-Ⅲ grade.
5.Influence of montelukast sodium chewable tablets in children with allergic asthma

Objective To investigate the effect of montelukast sodium chewable tablets on pulmonary function in children with allergic asthma . Methods 86 cases of children with allergic asthma were randomLy divided into 2 groups, 43 cases in the control group were treated with routine clinical treatment, 43 cases in the experiment group received more with montelukast sodium chewable tablets.Serum T lymphocyte subsets, IL-6, TNF-α, IL-4 concentration levels, as well as FVC, FEV1, PEF levels and clinical efficacy were compared of two groups pre-and post-treatment.Results Compared with pre-treatment, Serum CD4 +, CD4 +/CD8 +, FVC, FEV1 and PEF levels in 2 groups increased, serum CD8 +, IL-4, IL-6, TNF-a levels decreased, compared with the control group, serum CD4 +, CD4 +/CD8 +, FVC, FEV1, PEF level and the effective rate in the experiment group post-treatment were higher, serum CD8 +, IL-4, IL-6, TNF-αlevels and the ineffective rate were lower, the differences were statistically significant (P<0.05).Conclusion Montelukast sodium chewable tablets can effectively improve the imbalance of immune function in children with allergic asthma , improve lung function, and has good clinical efficacy.

Paroxysmal nocturnal haemoglobinuria ( PNH) is a clonal hematopoietic stem cell disease characterized by intravascular hemolytic anemia , pancytopenia and thrombosis .Although PNH is an non-malignant disease , its complications have very negative impacts on patient's quality of life .The most common serious complication is thrombosis formation .
7.The influence of anti-sense hTERT on the expression of VEGF and Flt in SGC7901 cells

Objective To investigate the effects of anti-sense hTERT on the expression of VEGF and the receptor in SGC7901 cells, and study the influence in angiogenesis and progression in gastric carcinoma. Methods SGC7901 cell line was transfected by the recombinant virus containing sense and anti-sense hTERT cDNA. Then the expression of VEGF-C and Flt-4 was observed with RT-PCR, distribution of cell cycles was determined with flow cytometry. The expression of VEGF-C and Flt-4 protein was assessed with immunohistochemistry. Result The distribution of cell cycle of antisense hTERT transfected SGC7901 cells was changed, and the mRNA and protein expression of VEGF-C and Flt-4 was significantly down-regulated. Conclusion Anti-sense hTERT can act as an agent for inhibiting VEGF-C and Flt-4 mRNA and protein level, and it blocks tumor angiogenesis and lymphogenous metastasis.
8.RNA interference directed by small interference RNA of hTERT expressed in lung carcinoma cell A549

Objective To investigate the inhibitory effect of siRNA human telomerase transcriptase (hTERT) on activity of telomerase and proliferation of lung carcinoma cell A549. Methods A plasmid including U6 promoter and siRNA of hTERT was designed and constructed. The plasmid was transfected into A549 cell line. The telomerase activity was tested by telomerase repeat amplification protocol ELISA (TRAP-ELISA). MTT assay was used to assay the cell proliferation activity,and hTERT expression was assessed by Western blot. Result The U6 expression plasmid that was constructed for hTERT gene 745 showed obvious interfering effect. hTERT-siRNA could down-regulate the expression of hTERT protein,inhibit telomerase activity and proliferation of A549 cells. Conclusion siRNA of hTERT can inhibit the expression of human telomerase and proliferation of A549 cells. It may open a new approach to the use of siRNA as a new tool to study gene function in cancer cell lines,and may be developed to be a new gene therapeutic agent for cancer.
9.Study on the antithrombotic effect of fibrinolytic proteins from Scolopendra subspinipes mutilans

Aim The aim is to purify fibrinolytic enzyme from Scolopendra subspinipes mutilans L.Koch and to study the thrombolytic and anticoagulant effect.Methods Scolopendra subspinipes mutilans L.Koch fibrinolytic enzyme(SSFE) was purified by ammonium sulphate precipitation,DEAE-cellulose and SephadexG-75 column chromatography from Scolopendra subspinipes mutilans L.Kochby.And fibrinolytic activity was determined by fibrin plate.The anticoagulant effect was measured on mice with haemolytic test and hemorrhagic test.The thrombolytic effect was measured with rats In vitro and in vivo,and the activated partial thromboplastin time(APTT),plasma prothrombin time(PT),thrombin time(TT) were measured.Results SSFE was single component with fibrinolytic activity and without any hemolyzation and hemorrhagic activity.All doses of SSFE(2,5,10 mg?kg-1) could obviously prolong activated partial thromboplastin time(APTT) and thrombin time(TT) ;Middle dose of SSFE(5 mg?kg-1) could prolong plasma prothrombin time(PT) while high dose of SSFE(10 mg?kg-1) didn't prolong obviously.Conclusion SSFE has obvious thrombolytic effect and anticoagulant effect.
10.The inhibitory effects and immunoregulatory activity of the fibrinolytic protein from Eupolyphaga sinensis Walker (EFP) in vivo on line H22 in mice

Aim To explore the inhibitory effects and immunoregulatory activity of the extract from Eupolyphaga sinensis Walker (EFP) on the growth of liver cancer H22 in mice.Methods The chemical composition of extract from Eupolyphaga sinensis Walker(EFP)was separated with water extraction methods.The tumor bearing mouse model was constructed by injecting tumor cells subcutaneously.The EFP was given to the mouse by oral and hypodermic injection to observe its antitumor activity and immunoregulatory activity in vivo.The antibody level of anti-H22 in serum was measured with ELISA methods.The weight,tumor weight,spleen index,liver index were noted,and series of enzymes,such as,MDA,SOD,GPT and GOT activity of mouse serum and liver were measured.Results The extracts from Eupolyphaga sinensis Walker markedly inhibited the proliferation of tumor H22.The EFP constituent purified might be the main antitumor active part of Eupolyphaga sinensis Walker as shown by antitumor test in vitro.After treatment with EFP,the general condition of the test mice was much better than that of control mice,with spleen index,liver index increasing more obviously,antibody level of anti-H22,and MDA,SOD,GPT and GOT activity increasing more,and tumor growing more slowly.Conclusion EFP may be the main antitumor active part of Eupolyphaga sinensis Walker. EFP also has antitumor effect in vivo.EFP can exert antitumor effect in vivo by enhancing antibody level of anti-H22 and the series enzymes to be related to immunity activity.
