1.Lentivirus PARG-shRNA transfection decreases matrix adhesion,migration and invasion potencies of colon carcinoma lovo cells

Objective To explore the effects of poly(ADP-ribose) glycohydrolase (PARG) gene silencing on colon carcinoma lovo cells matrix adhesion,migration and invasion.Methods Lentivirus PARG-shRNA was transfected into colon carcinoma lovo cells and the lovo cell strain with PARG gene silencing perpetually was selected.Western blot was used to test the expression of PARG,poly(ADP-ribose) polymerase(PARP) and NF-κB.The lovo cell matrix adhesion,migration and invasion potencies were observed by cell matrix adhesion,migration and invasion assay.Results The expression of PARP and NF-κB in experiment groups was weaker than that in the control groups (P<0.05).PARG gene silencing of lovo cells decreased the cell matrix adhesion,migration and invasion potencies.The inhibitory rates of lovo cell matrix adhesion,migration and invasion were 25.22%,38.71%,35.29% respectively.Conclusion PARG gene silencing of lovo cells could inhibite cell matrix adhesion,migration and invasion potencies,which may relate to that PARG gene silencing and decreasing the activity of PARP and NF-κB.PARG probably plays an important role in the infiltration and metastasis of tumor.

3.Effect of Aortic Valve Regurgitation on Pharmacodynamics of Cisatracurium

[Objective]The purpose of this study was to investigate the effect of severe aortic valve incompetence on the pharmacodynamics of cisatracurium.[Methods]Thirty patients were enrolled in this study:15 patients with severe aortic valve incompetence scheduled for aortic valve replacement were included in the study group(AI-group)and 15 patients without heart disease undergoing general surgical operations were allocated into the control group(C-group).Anesthesia was induced with fentanyl 5 μg/kg.propofol 1.5~2 mg/kg and cisatracurium 0.1 mg/kg and maintained by total intravenous anesthesia with propofol-remifentanil infusion.Degree of neuromuscular block was measured by train-of-four ratio using TOF-WATCH acceleragraph monitor.Onset time(from administration of cisatracurium to T1=0),total twitch suppression time,the time for spontaneous recovery of T1 to 25%and 75%.and the recovery index were recorded respectively.[Results]The onset time of cisatracurium was significantly longer in the AI group compared to the control group(5.6±0.8 min vs 3.4 ±0.4 min.P<0.001.However.there was no difference in the recovery time between the two groups.[Conclusion]We demonstrated that the onset of cisatracurium was delayed in the patients with severe aortic regurgitation in comparison to those without heart disease.

5.Expression and clinical significance of Wnt5a in gastric carcinoma

Objective To study the significance of Wnt5a gene expression in gastric carcinoma (GC). Methods The Wnt5a mRNA expressions in SGC7901 cell line and normal gastric mucosa GES-1 cell line were detected by RT-PCR. The Wnt5a protein expressions in 60 GC and 30 matched tumor-adjacent normal tissue samples were examined by immunohistochemistry using the streptavidin-peroxidase method.χ2 test was used to analyze Wnt5a expression on the clinical pathologic features of gastric carcinoma. Results Both Wnt5a mRNA and Wnt5a protein were significantly higher in SGC7901 cells than those in GES-1 cells. The positive rate of Wnt5a protein expression was also significantly higher in GC than that in tumor-adjacent normal tissue (P < 0.05). Positive expression of Wnt5a was associated with higher degree of regional lymph node metastasis and the later TNM stage (P < 0.05). Conclusion Higher expression of Wnt5a seems to promote invasion and metastasis of gastric cancer, and Wnt5a might play an oncogene-like role in the development of gastric carcinoma.

Objective To investigate the effect of poly (ADP-ribose)polymerase (PARP) expression inhibited by 5-AIQ on the liver metastasis in colorectal carcinoma CT26 cell lines. Methods Liver metastasis model of colorectal carcinoma was established by intrasplenic inoculation of colorectal carcinoma CT26 cell in BALB/c mice. Treated by 5-AIQ (100 or 500 ?mol/L),CT26 cells were inoculated to the spleen of the mice,or the rats were first inoculated of CT26 cells,then treated by 5-AIQ (3 or 10 mg/kg) ip. The changes spleen primary carcinoma and liver metastasis carcinoma were observed. The PARP expression in spleen primary carcinoma was detected by Western blotting. Objective To explore the clinical application value of flow -cytometry combined with colpo-scope ultra high frequency electric knife for diagnosis of uterine cervix carcinoma ( UCC) and cervical intraepitheli-al neoplasia(CIN).Methods A total of 705 cases was included in this study who were screened from April 2014 to April 2016 and accepted colposcopy biopsy .DNA flow cytometry was performed to determine DNA index ( DI) and proliferative index ( PI) .Results Colposcopy diagnosis results showed that the detection rate of CINⅠ,CINⅡ,CINⅢ/CIS and UCC were 62.2%,78.0%,62.2% and 78.0% respectively.The sensitivity and specificity were 87.7%(213/243)and 90.2%(417/462)for DNA ploidy test using flow-cytometry.DI and PI of CINⅠ, CINⅡ,CINⅢ/CIS and UCC increased gradually with significant differences (P<0.05).Conclusion Flow-cytometry combined with colposcopy biopsy may be a useful tool for cervical cancer screening in developing coun -tries and has a competitive sensitivity and specificity ,which is worthy of popularization and application .

10.Patterns of renal injury in type 2 diabetic patients with macroalbuminuria

Objective To find out a more rational pathological classification criteria for renal injury in patients with type 2 diabetes mellitus. Methods The renal clinicopathological features of forty-nine type 2 diabetic patients with maeroalbuminuria were collected and were compared retrospectively. The patients without diabetic renal disease were excluded. According to the pathological features, the patients were divided into two groups: typical diabetic glomerulopathy (DG) and atypical diabetes-related renal disease (ADRD). Results The renal biopsy revealed DG accounted for 59.2% of the patients, while the remaining 40.8% presented atypical renal injury defined as ADRD. In DG group, volume fraction of mesangium per glomerulus, glomerular basement membrane width, atrophic tubules index, intersititium injury index and prevalence of hyalinization of renal arteriole were higher; podocyte density per glomerulus was lower; duration of type 2 diabetes was longer; the level of fast blood glucose, systolic blood pressure, mean arterial pressure, proteinuria and prevalence of diabetic retinopathy (DR) were higher; glomerular filtration rate (GFR) was lower. In ADRD group, body mass index and prevalence of obesity were higher; dyslipidemia was more severe. GFR was negatively correlated with glomerular global sclerosis rate in both DG and ADRD group. Proteinuria was positively correlated with volume fraction of mesangium per glomerulus in DG. No correlation between proteinuria and pathological features was found in ADRD. DR (94.8%) and duration of type 2 diabetes over five years (90.7%) had high negative predictive value for DG. Conclusions Renal injuries in type 2 diabetes patients are heterogeneous. ADRD is an atypical renal injury in type 2 diabetes patients whieh is different from DG. DR and duration of diabetes are more helpful in predicting DG separating from ADRD.
