1.The healing ability of left and right colonic anastomoses in rats

Objective To compare the healing ability differences between left colonic anastomosis and right colonic anastomosis in rats, in order to find a theoretical basis for the surgical management of colon resection. Methods Forty male Wistar rats were randomly divided into two groups: group A (end-to-end anastomosis of right colon, n=20), and group B (end-to-end anastomosis of left colon, n=20). Half of the rats in each group were sacrificed on the fourth day after operation, and the rest on the seventh day. The expressions of matrix metalloproteinase-1 (MMP-1), MMP-13 and collagen type Ⅰ were shown by immunohistochemical staining, and the histopathologic changes at the anastomosis were also demonstrated. Results The expression of MMP-1, MMP-13 at left colonic anastomosis was higher than that of MMP-1, MMP-13 at right colonic anastomosis, the expression of collagen type Ⅰ was lower than that of collagen type Ⅰ at right colonic anastomosis on the fourth day (P<0.05); The expression of MMP-1 at left colonic anastomosis was higher than that of MMP-1 at right colonic anastomosis, the expression of collagen type Ⅰ was lower than that of collagen type Ⅰ at right colonic anastomosis on the seventh day (P<0.05). Conclusions The healing ability of right colonic anastomosis is higher than that of left colonic anastomosis.

Anastomotic leakage is a serious complication in colorectal surgery;The healing of intestine is still the hot spot of current research.Methods of estimating intestinal healing mainly include mechanical,biochemical,immunohistochemical and hitological Method.The estimating parameters mainly Comprise bursting pressure,breaking strength,the amount of collangen and matrix metalloproteinase(MMP),and the scale for histology.The measurement of collagen and MMP is the nost import ant index for estimating healing of intestinal anastomoses.Collagen is the most abundant extracellular matrix protein that determines the mechanical stability of the connective tissue during wound healing.Collagen degradation is usually the result of the synergistic action of several MMPs.
4.Toxic Effects of Hematoporphyrin Monoether——PsD-044 on Ocular Tissue

The histopathological changs of the dog eyes caused by acute and subacute toxic effectsof hematoporphyrin monoether--PsD-044 were observed in this research. The results showed thatPsD-044 could lead to keratoscleritis and scleral perivasculitis. Phototoxic cyclitis and inflammatory edema of the retina were more serious in deferred killed animals, suggesting that it is necessary to avoid exposure to light after the administration of PsD-044.
5.Interaction between nimodipine and growth factors during formation of new retinal vessels

Objective: To study the interaction between the calcium channel antagonist nimodipine and growth factors (vascular endothelial growth factor [VEGF] and platelet-derived growth factor[PDGF]) during the formation of new retinal vessels. Methods: The hyperoxia model was induced by proliferative retinopathy (OIR) in newborn Sprague-Dawley (SD) rats. SD rats (2 d after birth) were randomized into 5 groups: normal control group, pure OIR group and group 3, 4 and 5, where the animals received retrobulbar injection of nimodipine 10 ?l, 5 ?l and 2 ?l once every 2 d for 3 times, respectively. Both eyeballs of newborn rats were made into common pathological sections and detected by immunohistochemistry method to count the nuclei of proliferative retinal vessel cells and to investigate the expression of VEGF and PDGF in retina. Pharmacology plays an important role to bridge the transla-tional gap between the basic medicine and clinical medicine. In this article, the projects in pharmacology funded by NSFC during 2004~2013 were reviewed. The new features and new problems in the projects of pharmacology were briefly analyzed.
7.Chemical constituents of Buddlejae officinalis and their bioactivities

Object To investigate the chemical constituents of Buddlejae officinalis Maxim. , the relationship between their bioactivities, and the medicinal application of the herb. Methods Various chro-matographic techniques were employed for the isolation and purification of the constituents, including silica gel, Sephadex LH-20, and pre-HPLC. The structures of 11 isolated chemical compounds were elucidated by chemical and spectral analysis (NMR, IR, UV, and MS). The aldose reductase inhibiting test and test on DNA topoisomerase Ⅳ inhibitory effect were applied to evaluate the bioactivity of both the purified compounds and the crude fraction. Results Eleven compounds were isolated from the extracts of B. offic-inalis. They are four flavones and their glycosides; apigenin (Ⅰ), linarin (Ⅱ), apigenin-7-O-?-L-rhamnopyranosyl (1-6)-?-D-glucopyranoside (Ⅲ), and luteolin-7-O-?-D-glucoside (Ⅶ); four phenylen-thanoid glycosides: verbascoside (Ⅴ), isoacteoside (Ⅵ), cistanoside F, a and b (Ⅶ-1 and 2), campneo-side Ⅱ , a and b (Ⅷ-1 and 2); three triferpenoid saponins: mimengoside A (Ⅸ), mimengoside B (Ⅹ), songaroside A (Ⅺ). Some of the them (such as Ⅱ ,Ⅲ , and Ⅵ) showed the aldose reductase inhibitory activities with a higher inhibitory rate than that of quercetin (the positive control), others (such as Ⅰ,Ⅱ , Ⅴ, Ⅵ, and Ⅺ) displayed the inhibititory activities against the DNA topoisomerase Ⅳ . Conclusion Among all the isolated compounds, Ⅲ , Ⅶ-1 and 2, Ⅷ-1 and 2, and Ⅺ are separated for the first time both from the plant and the genus. Their bioactivities of bacteriostatic and against aldose reductase with DNA topoisomerase Ⅳ as target are tested for the first time and are related to the medicinal application of B. officinalis.
8.Molecular biological research on Borna disease virus infection in depressive patients of Chongqing

Background Evidences, from recent studies, suggested that Borna disease virus (BDV) infection might be associated with human neuropsychosis, especially psychiatric disorders including depressive disorder(DD). However, controversy existed about the association between BDV infection and pathogenesis of DD. This study was to explore further whether the infection of Borna disease virus (BDV) is associated with the pathogenesis of depressive disorder (DD).Methods The p 24 fragment of BDV RNA in peripheral blood mononuclear cells (PBMCs) from 60DD patients and 120 healthy volunteers was detected by nested reverse transcriptase polymerase chain reaction (nRT-PCR) combined with fluorescence quantitative polymerase chain reaction (FQPCR). Positive products were cloned and sequenced before being compared with Strain V and strain He/80, from humans and animals.Results The positive rate (5%, 3/60) of BDV p 24 in PBMCs from the DD patients was significantly higher than that (0%, 0/120) from healthy volunteers ( P<0. 05). The gene sequence for the positive products showed BDV p 24 in PBMCs from DD patients in Chongqing was most homophylic with H1766 strain detected from iii horses (97.68%), with 2 situs mutations (nt 1675 T→C, nt 1678 C→T), and also similar to the standard strain V(96. 51%)and He/80(95.35 %), with basic exchanges limited to T- C and A→G.Conclusions There was BDV infection in the DD patients in China, which indicated that the pathogenesis of DD in human beings in Chongqing might be associated with the infection of BDV.
9.Ligaturing pericardial blood vessel combined splenectomy for treating children with cavernous transformation of portal vein

Objective To retrospectively analyze the therapeutic effect of ligaturing pericardial blood vessel combined splenectomy for treating children with gastrointestinal bleeding and hypersplenism secondary to cavernous transformation of portal vein (CTPV).Methods Retrospectively analyzed 30 cases of children with CTPV admitted by Beijing Children's Hospital in General Surgery Department from Jan.2005 to Dec.2012,and evaluated the clinical efficacy of ligaturing pericardial blood vessel combined splenectomy for treating children with gastrointestinal bleeding and hypersplenism secondary to CTPV.Results All of 30 cases admitted operation without serious complication.All children admitted follow-up from 6 to 60 months.Seven cases of 22 patients with gastrointestinal bleeding relapsed bleeding post operation and were cured by conservative treatment.Four cases of 7 relapsing bleeding children had NSAID.All cases of 8 children with simple hypersplenism suggested normal results of blood routine post operation,2 children occurred bloody stools who were cured by conservative treatment.None of patients had serious infection correlating with splenectomy.The size of spleen of patients with gastrointestinal bleeding is larger than patients with simple hypersplenism.Conclusions (1) Ligaturing pericardial blood vessel combined splenectomy is a good choice for children with gastrointestinal bleeding and hypersplenism secondary to CTPV.(2) NSAID may increase the recurrence rate of gastrointestinal bleeding post operation.(3) There may be a negative correlation between the size of spleen and the risk of gastrointestinal bleeding for children with CTPV.
10.Effects of Liuwei Dihuang Decoction on Expressions of Transforming Growth Factor β1 and Complement Regulatory Protein CR1 of IgA Nephropathy Rats

Objective To discuss the effects of Liuwei Dihuang Decoction on expressions of TGF-β1 and complement regulatory protein CR1 of the renal on IgA nephropathy rats.Methods Forty SD rats were randomized into normal group, model group, tripterygium glycosides group and Liuwei Dihuang Decoction group after adaptive feeding for one week. Complex method was used to establish IgA nephropathy model. All administration groups were given relevant medicine for gavage for 4 weeks;urine routine, the number of urine erythrocyte and 24 h urine protein were detected;creatinine, urea nitrogen, total serum protein and serum alburin were detected from blood;the renal pathological changes were observed under light microscope by HE and Masson staining;the expressions of TGF-β1 and CR1 were detected by immunohistochemistry.Results Compared with model group,Liuwei Dihuang Decoction could significantly decrease the proteinuria and hematuria levels and renal fibrosis changes of IgA nephropathy model rats, reduce the expression of TGF-β1, and raise the expression of CR1 (P<0.05).Conclusion Liuwei Dihuang Decoction can delay the renal fibrosis in IgA nephropathy rats through reducing the degree of TGF-β1 and promoting the expression of CR1.
