ObjectiveTo discuss the mechanism of the growth inhibitory effects of berberine on human osteosarcoma cell MG63.MethodsMG63 cells were divided into a control group and a processing group.The inhibition ratio of two groups was measured by CCK-8 assay,and the mechanism of the growth inhibitory effect was studied by Western Blot assay.ResultsThe inhibition ratio of MG63 cells,which were respectively treated with berberine at 0 (control group),1,5,10,20,50 μg/ml,were (99.37± 1.13) ％,(91.37±4.25)％,(72.44 ± 2.89)％,(57.33 ± 1.37)％,(50.14 ± 4.05)％ and(37.53 ± 4.14)％ after 48h.The semiquantitative expression of Bcl-2 in MG63 treated with berberine at 0 (control group),20,50 μg/ml after 48h was (0.4167±0.0115),(0.3458±0.0146),(0.2987±0.0207),while the Bax and caspase-9 was (0.4327±0.0206),(0.5007±0.0089),(0.7115±0.0214) and (0.1459±0.0107),(0.2416±0.0098),(0.2584±0.0330).Compared with the control group there was statistically significance (P＜0.05).ConclusionBerberine had inhibitory effects of on MG63.The mechanism lied in its regulating the expression of apoptosis gene Bcl-2 and Bax,activating caspases-9,and inducing cell apoptosis.

Objective To investigate the clinical efficacy of penicillin combined with ceftriaxone in early syphilis patients.Methods60 patients with early syphilis treated from February 2014 to February 2016 were included in the study,according to the random number table method,the patients were divided into control group (simple penicillin treatment) and treatment group (Penicillin and ceftriaxone sodium), 30 cases in each group.The skin lesions subsided within one week of two groups were recorded.The toluidine red blood serum test (TRUST) was carried out at 3 months, 6 months, 9 months and 12 months after treatment, and the serum fixation rate of the patients in the two groups were observed.ResultsThe rate of pain relief within one week in the treatment group was 90.0% (27/30), which was significantly higher than that in the control group (66.7%, 20/30), (χ2=4.81, P=0.02);TRUST negative rates of the treatment group after treatment for 3 months, 6 months, 9 months and 12 months were 36.7%(11/30), 70.0% (21/30), 86.7% (26/30), 90.0% (27/30),the control group was 13.3%(4/30), 26.7% (8/30), 50.0% (15/30), 63.3% (19/30), [χ2=(4.35,11.27,9.31,5.96), P=(0.00,0.00,0.00, 0.01)].The serum fixation rate in the treatment group was 3.3%(1/30), which was significantly lower than that in the control group (20.0%, 6/30),(χ2=4.04, P=0.04).ConclusionPenicillin combined with ceftriaxone sodium in the treatment of early syphilis patients can help to promote the disappearance of skin lesions, accelerate the patients with syphilis serum negative and reduce serum fixation rate, the effect is significant, to be further studied to promote.

ObjectiveTo evaluate the efficacy of modified ultrafiltration (MUF) combined with conventional ultrafiltration (CUF) for cardiac valve replacement in patients with severe valve disease.MethodsOne hundred and eight NYHA Ⅲ or Ⅳ patients with severe valve disease,aged≥ 18 yr,weighing 50-80 kg,scheduled for elective cardiac valve replacement under cardiopulmonary bypass (CPB) were randomized into CUF group ( n =56) and CUF combined with MUF group (CMUF group,n =52).MUF was performed at the end of CPB,and the flow and time was 400 ml/min and 15-20 min respectively.Arterial blood samples were taken for blood gas analysis and for measurement of the plasma IL-6 and IL-8 concentrations after induction of anesthesia (T1),at the beginning of CUF (T2),at the end of GUF (T3),at the beginning of MUF (T4),at the end of MUF (T5),and at 2,8 and 24 h after the termination of CPB (T5-8).The oxygenation index was calculated and the airway pressure was recorded at T5 8.The amount of urine output during operation,the amount of urine output and volume of chest tube drainage within 24 h after operation,extubation time,packed red blood cell (PRBC) transfusion and duration of stay in ICU were recorded.ResultsCompared with CUF group,the hematocrit at T5,6 and oxygenation index at T7,8 were significantly increased,extuhation time was significantly shortened,and the amount of urine output and volume of chest tube drainage within 24 h after operation and PRBC transfusion were significantly reduced ( P ＜0.05),and no significant change was found in the plasma IL-6 and IL-8 concentrations,airway pressurc,amount of urine output during operation,and the duration of stay in ICU in group CMUF ( P ＞ 0.05).ConclusionCombination of MUF and CUF during CPB can be used for cardiac valve replacement in patients with severe valve disease,improve the function of organs after operation and reduce homologous blood transfusion.

Objective To study the feasibility, indications and clinical value of primary suture after the common bile duet exploration(CBDE).Methods One hundred and fimr cases of choledocholithiasis were chosen for primary suture by 5-Oviger after CBDE. Post-operative complications and the length of hospital stay were compared with those with T-tube drainage. Results Compared with T-tube drainage group, the complication rate was reduced and the length of hospital stay in primary suture group was shortened. And also the mortality and operation time were decreased significantly. Conclusion The primary suture after CBDE in selected oases is safe and reliable.

Objective To investigate the influence of Siraitia Grosvenori and Rehmannia Glutinosa on the Hematopoietic stem cells proliferation and function .Methods Cells from the peripheral blood , spleen and bone marrow of mice were stained with indicated antibodies , and analyzed by flow cytometry .Mice were divided 3 groups:control group, Siraitia Grosvenori treatment group and Rehmannia Glutinosa treatment group .After 4.5Gy IR treatment, mice divided 4 groups: control group, 4.5Gy IR treatment and feed with normal food, 4.5Gy IR treatment and feed with Siraitia Grosvenori and 4.5Gy IR treatment and feed with Rehmannia Glutinosa for 1 month.Results Mice fed with Siraitia Grosvenori and Rehmannia Glutinosa decreased the percentage of B cells and increased the percentage of M cell .For HSCs, the number of HSCs was increased , especially the number of LT-HSCs.After 4.5Gy IR treatment, mice fed with Siraitia Grosvenori and Rehmannia Glutinosa increase the number of HSCs , and increased the percentage of M cells . Conclusion Siraitia Grosvenori and Rehmannia Glutinosa promote the hematopoietic stem cells and progenitor cells proliferation and function and recover the damage that caused by IR treatment .

Objective Type around the HRCT manifestations of pulmonary hamartoma , in order to improve the level of diag-nosis of the disease .Methods Last 14 years retrospective analysis confirmed by surgery and biopsy of peripheral pulmonary hamar -toma 23 cases of CT performance.Results Nine cases of right lung (for 3 cases of the upper lobe of right lung middle in 2 cases, 4 cases were characterized by lower lobe ), 14 cases had left lung (6 cases left upper lobe, left lower lobe 8 cases), 12 cases round, round 8 cases, shallow points Ye Zheng 3 cases, border and clear, no deep lobulated, burr and satellite lesions.1.0~5.4 cm in di-ameter, the average is about 2.6 cm in diameter, less than 3 cm17 cases.5 cases of lesions containing fat , 7 cases there are both fat and calcification;8 cases of lesions with calcification , typical popcorn calcification in 3 cases;All 11 cases in 15 cases of calcified lesions located in the middle and lower lobe basal segment .3 cases of homogeneous soft tissue density with increase in mild to moderate strengthening , soft tissue density partial reinforcement degree about 5~16 HU.Conclusions HRCT can show pulmonary hamartoma characteristic calcification and fat density with help in the diagnosis of peripheral hamartoma lungs .Fat or there are both fat and calcifi-cation isolated pulmonary nodules or masses , can consider hamartoma;Instead of n/med tuberculosis good hair part containing calcium isolated pulmonary nodules , should think of hamartoma diagnosis .

Objective To investigate therapeutic effect of the low dose clomiphene treatment of idiopathic oligoasthenospermia and effect on sperm epididymal protein level 4.Methods183 cases of idiopathic oligoasthenospermia were divided into three groups, vitamin group of 61 patients were given vitamin E,clomiphene group(61 cases)treated with low dose clomiphene, combined group of 61 patients were given low dose clomiphene and vitamin E, lasted for 12 weeks.Semen volume, sperm density, sperm motility, sperm survival rate and sperm reactive oxygen species (ROS), malondialdehyde (MDA), superoxide dismutase (SOD), human epididymis protein 4 (HE4), follicle stimulating hormone, luteinizing hormone, testosterone concentration were observed before and after treatment.After treatment, the patients were followed up for 1 year, the observation of the natural pregnancy rate after treatment.ResultsSperm density, sperm survival rate and sperm and a A+B grade sperm in the clomiphene group were significantly higher comparedwith the vitamin group (P< 0.05), sperm density, sperm survival rate and sperm and a A+B grade sperm in the combined group were significantly increasedcompared with clomiphene group and vitamin group (P< 0.05),levels of HE4 after treatment was significantly lower compared with before treatment (P< 0.05), And the difference between the three groups were statistically significant (P< 0.05).Follicle stimulating hormone, luteinizing hormone, testosterone in clomiphene group and combined group after treatment, were significantly increased compared with before treatment (P< 0.05), and the combined group had no statistically difference compared with clomiphene group.The pregnancy rate in 1 year of vitamin group, clomiphene group and combined group were 24.59%, 42.62% and 62.29% (P< 0.05).ConclusionClomiphene treatment of idiopathic asthenospermia is better than vitamin E, the combination of the two has a synergistic effect.

Objective:To investigate the clinical diagnosis and treatment of idiopathic subglottic stenosis.Methods:From May 2012 to January 2014,four patients with idiopathic subglottic stenosis admitted in the Department of Otolaryngology,the Affiliated Drum Tower Hospital of Nanjing University Medical School,Nanjing,Jiangsu,China were selected for review.Results:All the four patients with idiopathic subglottic stenosis were resolved after the resection of the pathological tissues and subsequent hormone treatment.Conclusions:The confirmed diagnosis of idiopathic subglottic stenosis mainly relied on the medical history and pathological examination.Favorable treatment effects might be obtained by combining surgical resection and hormone treatment.

AIM: To investigate the effect of probucol on proliferation of rat vascular smooth muscle cells(VSMC) stimulated by basic fibroblast growth factor (bFGF) and/or hydrogen peroxide(H 2O 2). METHODS: Effects of probucol on VSMC proliferation and DNA synthesis stimulated by bFGF and/or H 2O 2 were observed by means of MTT test, cell number count and [ 3H]-TdR incorporation. RESULTS: ①Probucol significantly inhibited proliferation and DNA synthesis in VSMC stimulated by bFGF and/or H 2O 2, with dosage-dependent manner. Cell number, A value and [ 3H]-TdR incorporation in group probucol+bFGF and group probucol+H 2O 2 were reduced by 40.0%, 39.1%, 45 5% and 46 9%, 45 0%, 39 5%, respectively, compared with group bFGF and group H 2O 2 ( P 0.05). CONCLUSION: Probucol dramatically inhibited proliferation and DNA synthesis in VSMC stimulated by bFGF and/or H 2O 2, but had no inhibitory effect on the cell proliferation prestimulated by bFGF and /or H 2O 2. Objective To explore the p27 gene expression and clinical significance in acute leukemia (AL).Methods RT-PCR was used for testing the mRNA levels of p27 gene in bone marrow mononuclear cell of 65 cases of newly diagnosed AL patients and 41 controls.The correlation between the p27 gene expression positive rate and age, sex, peripheral blood white blood cell count and extramedullary infiltration were analyzed simultaneously in AL patients.Results The positive rate of p27 gene expression (36.9 ％,24/65)was significantly lower in AL group than that in the control group (87.8 ％,36/41) (P ＜0.05),and which was lower than the control group (P ＜0.05) in the AML group (34.2 ％,13/38) and the ALL group (37.5 ％,9/24).However,comparing AML group with ALL group,the difference was not statistically significant (P ＞0.05).At the same time,compared the p27 gene expression positive rate of ≥ 14 years old group (52 cases) and ＜14 years old group (13 cases),and also compared in male group (34 cases) and in female group (31 cases),there was no statistically difference (P ＞0.05); Compared the p27 gene expression positive rate of peripheral blood white blood cell count ≥20×109/L group (31 cases) and ＜20×109/L group (34 cases),also compared with in extramedullary infiltration group (37 cases) and in no extramedullary infiltration group (28 cases),the relationship of p27 gene expression positive rate with peripheral blood white blood cell count as well as extramedullary infiltration was a negative correlation (r =-0.284,P ＜ 0.05,r =-0.300,P ＜ 0.05).Conclusion There is p27 mRNA expression deletion in AL patients,and the deletion is present in the AML and ALL at the same time.p27 mRNA expression is negatively correlated with peripheral blood white blood cell count or extramedullary infiltration,which might be one of the mechanisms of occurrence and development of AL.
