1.The construction of operating room nursing quality sensitive indicator system

Objective To construct nursing sensitive indicators applicable to operating room,in order to provide standard and quantitative basis for nursing quality monitoring in operating room.Methods Based on American's National Database of Nursing Quality Indicators (NDNQI) and literature review,the key content of indicators and scientific measurement methods were determined by one round of internal inquiry and two rounds of expert consultation using Delphi technique.Results Operating room nursing quality sensitive indicators system was constructed,which contained 14 indicators.The expert authority coefficients were greater than 0.80 and the variation coefficients of each index were less than 0.3.The results were reliable.Conclusion A set of specialized,sensitive and practical nursing sensitive indicators were needed to facilitate sustainable development of operating room.
2.Novel cationic liposome loading siRNA inhibits the expression of hepatitis B virus HBx gene.

In order to solve the problem of selection and in vivo delivery problem in siRNA treatment, hepatitis B virus (HBV) HBx gene which could be targeted by siRNA was studied. The siRNA expression plasmid which specific inhibits HBx expression was obtained by in vitro selection via a dual-luciferase plasmid including HBx-Fluc fusion protein expression domain. The selected siRNA expression plasmid was then encapsulated in PEG-modified cationic liposome, which was devoted into pharmacodynamic studies at both cellular and animal level. The results illustrated that the cationic liposome which encapsulated siRNA expression plasmid could effectively inhibit HBx gene expression both in vitro and in vivo.
3.Improvement of cardiac function and reversal of gap junction remodeling by Neuregulin-1β in volume-overloaded rats with heart failure

Objective We performed experiments using Neuregulin-1β (NRG-1β) treatment to determine a mechanism for the protective role derived from its beneficial effects by remodeling gap junctions (GJs) during heart failure (HF). Methods Rat models of HF were established by aortocaval fistula. Forty-eight rats were divided randomly into the HF (HF, n = 16), NRG-1β treatment (NRG, n = 16), and sham operation (S, n = 16) group. The rats in the NRG group were administered NRG-1β (10 μg/kg per day) for 7 days via the tail vein, whereas the other groups were injected with the same doses of saline. Twelve weeks after operation, Connexin 43 (Cx43) expression in single myocytes obtained from the left ventricle was determined by immunocytochemistry. Total protein was extracted from frozen left ventricular tissues for immunoblotting assay, and the ultrastructure of myocytes was observed by transmission electron microscopy. Results Compared with the HF group, the cardiac function of rats in the NRG group was markedly improved, irregular distribution and deceased Cx43 expression were relieved. The ultrastructure of myocytes was seriously damaged in HF rats, and NRG-1β reduced these pathological damages. Conclusions Short-term NRG-1β treatment can rescue pump failure in experimental models of volume overload-induced HF, which is related to the recovery of GJs structure and the improvement of Cx43 expression.
4. Multidisciplinary team-based FOCUS-PDCA reduces the adverse event rate caused by junior nurses

Objective:
To investigate the effect of FOCUS(F:find; O:organize; C:clarify; U:understand; S:select)-PDCA on reducing the adverse event rate caused by junior nurses.
Methods:
A historical control design was used, traditional method to manage adverse events among junior nurses from January to June 2017. Interdisciplinary team combined with FOCUS-PDCA courses was established from July to December 2017. In the course problems were found and analyzed, the present procedures and standard were issued and improved. And these data were summarized and analyzed to improve the quality of management continuously.
Results:
Before applying the FOCUS-PDCA model, the total incidence of adverse events caused by junior nurses, bachelor′s degree or above, nurse practitioners, and working career of 1-3 years were respectively as follows: 42.66%(285/668), 46.67%(112/240), 40.54%(105/259), 51.61%(176/341). After using this model, the above four indicators were 33.04%(229/693), 33.33%(84/252), 27.04%(73/270), 38.97%(136/349) respectively. 5.Echinacoside promotes mitochondrial biosynthesis and inhibition of myocardial apoptosis by up-regulating the PGC-1/NFR signaling pathway

【Objective】 To investigate the effects of echinacoside (ECH) on mitochondrial biosynthesis and cardiomyocytes’ apoptosis in heart failure (HF) and to explore its related mechanisms. 【Methods】 The experimental animals were divided into three groups: the rat model of HF (HF) was induced by intraperitoneal injection of ISO, and pre-treated with ECH by intraperitoneal injection (ECH) and nomal control (ctrl group). Cardiac function was detected by echocardiography after 2 weeks of treatment. The ultrastructure of myocardium was observed by transmission electron microscopy and the mitochondrial density and vacuolation rate were analyzed. The expressions of apoptosis-associated proteins were evaluated by Western blotting, and genes related to mitochondrial biosynthesis were examined by Real-time PCR. 【Results】 ECH increased 1eft ventricular ejection fraction (LVEF) and 1eft ventricular fraction shortening (LVFS), but decreased 1eft ventricular end-systolic diameter (LVEDs) and 1eft ventrieular end-diastolic diameter (LVEDd) when compared to HF group (P<0.01) and improved cardiac function. The myocardial ultrastructure was significantly improved by ECH, the density of regular shapes of mitochondria was increased, and the percentage of vacuolated rate was reduced by ECH (P<0.01). The expression of anti-apoptotic protein Bcl-2 was upregulated and that of pro-apoptotic protein Bax was downregulated in ECH group. The mRNA of mitochondrial biosynthesis related genes PGC-1, NFR-1, NFR-2 and TFAM was significantly upregulated in ECH group. 【Conclusion】 ECH promotes mitochondrial biosynthesis and inhibits cardiomyocytes’ apoptosis by up-regulating PGC-1/NFR signaling pathway, thus improving cardiac function.
