1. Preparation and quality evaluation of self-emulsifying drug delivery system of total saponins of Sanguisorba officinalis

Objective: To optimize the formulation of the self-emulsifying drug delivery system of total saponins of Sanguisorba officinalis and evaluate its characteristics. Methods: The formulation and its proportion of the self-emulsifying drug delivery system of total saponins of S. officinalis were optimized based on the solubility tests, formula compatibility, microemulsion area in the ternary phase diagram and D-optimal mixing experiment design. The dosage of oil phase, surfactant and co-surfactant were investigated by drug loading, particle size, and polydispersity index. The appearance, particle size, polydispersity index, Zeta potential, and in vitro release of preparation were finally evaluated. Results: The ratio of oil phase, surfactant, and co-surfactant was 0.25: 0.45:0.30. The drug loading was 23.93 mg/g, the average particle size was (207.92 ± 2.13) nm and the zeta potential was (38.84 ± 0.18) mV. The release of SEDDS was superior to the bulk drug apparently. Conclusion: The self-emulsifying drug delivery system of total saponins of S. officinalis was established. The technology was feasible and the quality was stable.

3.Circadian rhythm changes of PGE2 levels in blood of growing rats after functional mandibular protrusion

Objective:To explore the relationship between wearing of functional appliance and systemic metabolism of PGE2 in the blood of growing rats.Methods:54 male Wistar rats aged 4-weeks were randomly divided into 3 groups(n =18):Daytime group,all-day group and control group.Self-guided functional appliance was made for mandible advancement of the rats in the 2 experimental groups.One week after appliance wearing,rats were sacrificed at the corresponding time points and blood in femoral artery was taken. PGE2 levels in the blood samples were measured,data were analysed by cosine equation.Results:Hypothesis was true in the control group(P <0.05),and was not true in both 2 experimental groups(P >0.05).Conclusion:PGE2 level in rat blood is circadian rhyth-mic,wearing of functional appliance made may disturb the rhythm.

Objective To investigate the chemopreventive potential of salvianolic acid B against 7,12-dimethylbenz(a)anthracene(DMBA)-induced hamster buccal pouch carcinogenesis with respect to angiogenesis. Methods(Sixty) male Syrian golden hamsters were randomly divided into 4 groups(group A and B,n=20;group C and D,(n=10).) DMBA solution(0.5% in acetone) was applied topically to the right cheek pouch of male Syrian golden hamsters in group A and B,while animals in group C were painted with acetone,3 times a week for 6 weeks.Animals in group D received neither DMBA nor acetone in the first 6 weeks.Two days after the last treatment of(DMBA,) animals in group B and D received salvianolic acid B daily(10 mg/kg) by gavage,while animals in group A and C received same volume of saline,for the next 12 weeks.At the end of the experiment,animals were sacrificed and tissue samples were collected for histopathological and immunohistochemical examination. Objective To compare the effects of two head holders on the setup errors during head and neck neoplasms radiotherapy.Methods Totally 30 patients were divided into groups A and B randomly and equally.Group A applied TypeSTM Extension for setup,and group B used Type-S Overlay Board,with thermoplastic film applied to fixation.Varian23IX image guided linear accelerator was involved in the therapy,and the patients underwent CBCT-guided therapy once a week in 6 weeks.The setup errors at left-right,head-foot and anterior-posterior directions were obtained by bone registration and manual checking.Results The absolute setup error at anterior-posterior direction of group A,was (2.31 ± 1.35) mm,which was significantly higher than (1.88±1.79) mm of group B (P=0.032).There were no obvious differences between the setup errors of group A and those of group B (P>0.05),and the weekly absolute errors of group A were not statistically different from those of group B (P>0.05).Conclusion There are no significant differences between the effects of the two head holders on the setup errors during head and neck neoplasms radiotherapy,and proper head holder can be selected according to clinical requirements.
7.Improvement and optimization of performance verification on enzyme linked immunosorbent assay for determination of hepatitis B markers

Objective To evaluate the performance of enzyme linked immunosorbent assay (ELISA) kit in detection of Hepatitis B virus(HBV) markers by using improved and optimized method ,so as to provide a practical and feasible method and reagents for clinical laboratory .Methods ELISA test was used for the detection of HBV markers .The gradient dilution method was used to e‐valuate the lower limit .The verifiation of cut off value was carried out based on clinical and laboratory standards institute (CLSI) EP12‐A2 document .Samples with cut off values were collected to evaluate the precision ,including repeatability and intermediate precision .The coincidence rates were counted through comparing the results of ELISA with those of external quality assessment and those detected by using Abbott i4000SR chemiluminescence instrument .Results The lower detection limit of HBsAg ,HBsAb , HBeAg ,HBeAb and HBcAb were 0 .2 IU/mL ,20 mIU/mL ,1 NCU/mL ,0 .75 NCU/mL and 0 .05 NCU/mL respectively .The cut‐off value(C50 )± 20% concentration included the concentration range between C5 and C95 .The within‐run coefficient of variation (CV)≤15% ,in sandwich method the between‐run CV≤25% ,in competition method the between‐run CV≤35% .The positive and negative coincidence rates in accuracy and comparing with i 4000SR all were more than 95% and all κ>0 .75 .Conclusion ELISA tests for HBV markers in our laboratory could meet the requirements of the detection performance and clinical needs .

9.Platelet-rich plasma versus corticoid in the treatment of chronic lateral elbow epicondylitis

A total of 52 patients with chronic lateral epicondylitis were randomly assigned into platelet-rich plasma (PRP) (n =28) or corticosteroid (n =24) group.The visual analog pain scale (VAS) and Mayo elbow score had no significant difference between two groups within 1 week post-treatment.However,VAS and Mayo elbow score improved more significantly in PRP group versus corticosteroid group at Month 1 and 6 post-treatment (P < 0.05).The PRP trigger point injection treatment of elbow lateral epicondylitis achieved clinical outcomes and it was superior to glucocorticoid treatment.
