1.Insulin-like growth factor-1 generation test in diagnosis and treatment of short stature

Short stature accounts for a large proportion of endocrine diseases in children,with a complex and cross-cutting etiology. It is one of the risk factors for physical and psychological problems in children,and needs proper diagnosis and timely treatment. The insulin-like growth factor-1 ( IGF-1 ) generation test is via re-combinant human growth hormone(rhGH)stimulating peripheral target cells to secrete IGF-1. The effects of rh-GH stimulation are then evaluated according to the responsiveness of IGF-1,which is not only helpful for further determination of clinical classification of short stature,but also for therapy assessment of rhGH in time and a-chievement of personalized treatment.
2. Inhibitory action of dithiothreitol on selenite-induced cataract in rats

Objective: To study the inhibitory action of self-designed dithiothreitol(DTT) eye drops on selenite-induced rat cataract(simulating the senile cataract). Methods: Twelve-day-old SD rats (body weight about 25 g) were injected with Na2SeO3 subcutaneouly, quaque die alterna, for 3 times (0.2 ml/time, with a total amount of 210 μg). Six days later over 90% of rats developed nuclear cataracts. The right eyes of the rats were treated with 25 mmol/L DTT eye drops (4 times/day, 1 drop/ time) for 3 weeks. The left eyes were taken as control and were treated with normal saline at the same dosage. The lens were examined using slit lamp and the diameter of the cataract plaques were measured using vernier cursor. The eyes of cataract models were also treated with 25 mmol/L DTT (n = 89), pirenoxine eye drops (PDE, Japan, n = 22), pirenoxine sodium eye drops (PSED, China, n = 24, 2 died during the experiment), and normal saline (n = 45). Each eye was administered with 1 drop/time, 4 times/day for 3 weeks. Then slit lamp was used to examine the cortical lesions of both eye and the lens opacity. Results: The DTT eye drops significantly reduced the diameter of the cataract plaques and significantly reduced the rate of the cortical opacity after the nuclear cataract. The inhibitory action of DTT eye drops on the selenite-induced cataract was superior to pirenoxine eye drops(Japan) and pirenoxine sodium eye drops(China). Conclusion: Our study shows that the DTT eye drops has satisfactory inhibitory action on selenite-induced cataract in rats.
3.Inhibitory effect and mechanism of PSD-007 photodynamic therapy on human nasopharyngeal carcinoma transplanted tumors in nude mice

Objective:To explore the inhibitory effect and mechanism of photocarcinorin (PSD-007) photodynamic therapy (PDT) on human nasopharyngeal carcinoma transplanted tumors in nude mice.Methods:A total of 50 transplanted tumor nude mice models of human nasopharyngeal carcinoma were established and randomly divided into 5 groups: control group (group A), simple PSD-007 group (group B), simple light group (group C), local injection of PSD-007 + light group (group D), intraperitoneal injection of PSD-007 + light group (group E) using the method of random number table, 10 mice in each group. After 7 days of treatment, the tumor mass and tumor volume of nude mice in each group were measured and the tumor inhibition rate was calculated. HE staining was used to detect the histopathological changes of tumors in nude mice. Western blotting was used to detect the expressions of autophagy-related gene Beclin 1, microtubules associated protein 1 light chain 3-β (LC3) and phosphatidylinositol 3-kinase/protein kinase B/mammalian target of rapamycin (PI3K/Akt/mTOR) signaling pathway related proteins. Real-time fluorescent quantitative PCR was used to detect LC3, Beclin 1 mRNA expressions in tumor tissues.Results:After treatment, the tumor mass of nude mice bearing human nasopharyngeal carcinoma in group A, B, C, D and E were (2.05±0.18) g, (2.02±0.20) g, (2.04±0.15) g, (0.43±0.11) g and (0.94±0.12) g, and the tumor volumes were (1.11±0.13) cm 3, (1.18±0.16) cm 3, (1.13±0.14) cm 3, (0.51±0.07) cm 3and (0.65±0.10) cm 3, and there were statistically significant differences among the 5 groups ( F=236.749, P<0.001; F=62.418, P<0.001). Compared with group A, B and C, the tumor mass and tumor volumes of nude mice in group D and E were significantly reduced, and there were statistically significant differences (all P<0.001); compared with group E, the tumor mass and tumor volume of nude mice in group D were significantly reduced ( P<0.001; P=0.023). The tumor inhibition rates of group B, C, D and E were (1.07±0.11)%, (0.55±0.06)%, (79.11±0.06)% and (54.05±0.08)%, with a statistically significant difference ( F=235.987, P<0.001), compared with group B, C and E, group D had the most obvious tumor suppressing effect (all P<0.05). HE staining results showed that compared with group A, B and C, group D and E had larger tumor necrosis areas, more inflammatory cell infiltration, more vacuolar degeneration, and obvious nuclear shrinkage. The tumor necrosis degree in group D was higher than that in group E. The relative expressions of PI3K protein of group A, B, C, D and E were 1.01±0.06, 1.00±0.05, 1.01±0.05, 0.23±0.02, 0.48±0.04, p-Akt/Akt protein relative expressions were 0.66±0.06, 0.65±0.05, 0.64±0.05, 0.06±0.02, 0.17±0.02, p-mTOR/mTOR protein relative expressions were 1.06±0.06, 1.01±0.06, 1.01±0.06, 0.30±0.02, 0.45±0.04. The protein relative expression ratios of LC3 Ⅱ/LC3 Ⅰ were 0.85±0.05, 0.83±0.05, 0.83±0.06, 0.22±0.02, 0.41±0.04, and Beclin 1 protein relative expressions were 0.66±0.06, 0.64±0.05, 0.64±0.06, 1.67±0.07, 1.02±0.05, LC3 mRNA relative expressions were 0.98±0.29, 0.92±0.25, 1.02±0.26, 3.76±0.28, 2.38±0.28, and Beclin 1 mRNA relative expressions of were 1.11±0.40, 1.19±0.29, 1.16±0.24, 6.84±0.54, 2.94±0.48. There were statistically significant differences ( F=190.160, P<0.001; F=160.014, P<0.001; F=160.183, P<0.001; F=119.964, P<0.001; F=186.257, P<0.001; F=211.089, P<0.001; F=374.835, P<0.001). Compared with group A, B and C, PI3K, p-Akt/Akt and p-mTOR/mTOR protein relative expression levels and protein relative expression ratios of LC3 Ⅱ/LC3 Ⅰ in nude mice tumors of group D and E were significantly reduced, and LC3 mRNA, Beclin 1 protein and mRNA relative expression levels were significantly increased, with statistically significant differences (all P<0.001); PI3K, p-Akt/Akt and p-mTOR/mTOR protein relative expression levels and protein relative expression ratio of LC3 Ⅱ/LC3 Ⅰ in nude mice tumors of group E were significantly higher than those of group D, while Beclin 1 protein relative expression levels, LC3 and Beclin 1 mRNA relative expression levels were reduced (all P<0.05). Conclusion:PSD-007 PDT has an inhibitory effect on human nasopharyngeal carcinoma transplanted tumors in nude mice, which may be related to the inhibition of PI3K/Akt/mTOR signaling pathway activation and the promotion of autophagy. Compared with intraperitoneal injection, local injection of PSD-007 PDT is more effective.
4.Effect of Tai Ji Quan Training on Strength and Function of Lower Limbs in the Aged.

OBJECTIVETo observe the effect of Tai Ji Quan (TJQ) training on strength and function of lower limbs in the aged.
METHODSSixty senile subjects were recruited and assigned to the TJQ group and the control group (imparting health knowledge)by random digit table. Patients in the TJQ group received 24-style TJQ training for 18 months (60 min each time, 5 times per week), while those in the control group were imparted with sarcopenia related causes, pathogeneses, prevention and control measures. The maximum isometric strength of bilateral iliopsoas, quadriceps femoris, tibialis anterior muscle, hamstrings; the time for 5 sitting-up tests and Time Up and Go Test (TUGT), one-leg standing time with closed eyes test; and the score of Berg balance scale were compared between the two groups.
RESULTSCompared with before treatment, muscle strength increased in the TJQ group with an average increased capacity (rate) as follows, bilateral iliopsoas 5.5 kg (16.9%), quadriceps femoris 5.5 kg (26.2%), and tibialis anterior muscle 8.5 kg (36.2%) (all P < 0.05). The time for TUGT and 5 sitting-up tests was shortened by 1.3 s (16.7%) and 0.9 s (14.5%) respectively in the TJQ group. The time for one-leg standing time with closed eyes test was increased by 8.4 s (left) and 9.1 s (right) respectively. The score of Berg balance scale increased by 4.3% (P < 0.05). Compared with the control group, bilateral quadriceps femoris and tibialis anterior muscle strength increased significantly (P < 0.01); the time for TUGT and 5 sitting-up tests, the time for one-leg standing time with closed eyes test, scores of Berg balance scale were all improved in the TJQ group after intervention (P < 0.05).
CONCLUSIONTJQ training could improve strength of iliopsoas, quadriceps femoris, tibialis anterior muscle in the aged, elevate their balance and locomotor activities, and possibly prevent and treat sarcopenia.

Objective To evaluate the effect and safety of use of tirofiban in percutaneous coronary interventional therapy (PCI) in elder patients with acute ST segment elevation myocardial infarction (STEMI). 7.Efficacy of ultra-early endovascular embolization for ruptured intracranial aneurysms:a Meta-analysis

Objective To compare the effectiveness and safety of ultra-early (< 24 h)and delayed (≥ 24 h ) endovascular embolization of ruptured intracranial aneurysms with systematic review. Methods PubMed,Embase,the Cochrane Library,VIP,Wanfang Data,and China National Knowledge Internet (CNKI)were retrieved according to inclusion,exclusion criteria and retrieval strategies,and the clinical literature of ultra-early and delayed endovascular embolization for the treatment of ruptured intracranial aneurysms were obtained. The Review Manager 5. 3 software was used to conduct Meta-analysis for good prognosis rate,postoperative mortality,and postoperative rebleeding rate. Results A total of 10 articles were included,9 of them were retrospective control studies and 1 was prospective control study. A total of 2 021 patients were enrolled,including 970 patients treated with ultra-early treatment;1 051 patients treated with delayed treatment. There was significant difference in the good prognosis rate (OR,2. 67,95% CI 2. 07 -3. 44,P < 0. 01)and the postoperative rebleeding rate (OR,0. 23,95% CI 0. 11 -0. 47)between the ultra - early embolization group and the delayed embolization group (all P < 0. 01). There were no significant difference in the mortality between the two groups (OR,0. 76,95% CI 0. 51 -1. 13,P = 0. 17). The subgroup analysis showed that there were significant differences in the good prognosis rate in the ultra-early group compared with the early group (< 3 d,OR,1. 98,95% CI 1. 33 -2. 95)and the middle and late group (≥3 d,OR,4. 66,95% CI 2. 21 -9. 81,all P < 0. 01). Conclusion Compared with the delayed group, ultra-early embolization of ruptured intracranial aneurysms may improve the good prognosis rate,reduce the rebleeding rate,and not increase the mortality after procedure at the same time. However,more high quality and large sample randomized controlled trials are needed to confirm them.

9.Dense embolization of intracranial aneurysms

Objective To investigate technique methods of dense emolization of intracranial aneurysms, factors resulting incomplete embolization. Methods 58 patients with intracranial aneurysms, using different embolization technique treated narrow and wide aneurysms with GDC. Dense emolization rate of aneurysms was elevated. Results dense emolization 50 aneurysms (86.2%), 95% in 3 aneurysms (5.2%), 90% in 3 aneurysms (5.2%), 80% in 2 aneurysms (3.4%). All embolizated aneurysms showed no enlarging recurrence or rebleeding.Conclusions Dense embolization of intracranial aneurysms should try to obtain.
10.Effect of recombinant human growth hormone on the immune function and prognosis in patients with liver neoplasms

Objective To investigate the long-term effect of recombinate human growth hormone(rhGH) on the immune function in patients with liver cancer. Methods Fifty patients with hepatoma who underwent hepatectomy were randomly divided into two groups:(1)Control group;(2)rhGH treatment(GH) group. All patients were treated with TPN for five days postoperatively,and additionally the patients in the GH group received rhGH(8U/d IH) in the evening for seven days. Results IgG,IgM,CD3,CD4 in the control group and CD8 in the GH group were significantly lower on day 10 postoperatively than that on preoperative days. IgG in the GH group significantly increased on day 15. The 1 year and 3 year survival rate and recurrent rate were not different between the two groups. Conclusions rhGH therapy can improve the immune function of patients with hepatoma who underwent hepatectomy and has no side effect on the long term survival and recurrence.
