1.Clinical analysis of silent paraganglioma and the literature review:21 cases

Objective To study the clinical characteristics of silent paraganglioma ( PGL) .Methods A total of 21 patients with silent PGL confirmed by surgical pathology and hospitalized in the First Affiliated Hospital of chongqing Medical University from Mar .2008 to Dec.2013 were enrolled in this study .The clinical manifesta-tions, hormone level, imaging findings, pathologic characteristic, treatment and follow-up data were reviewed and analyzed retrospectively .Results Among the 21 cases of silent PGL , 8 were male and 13 were female, with 47.05 years as the mean age(ranging from 26 to 67 years old).PGL tumors in silent group were mainly located in the right side, and left to right ratio was 2∶5.Silent PGL with tumor diameter <3 cm, 3-5 cm, >5 cm were 5, 7,and 9 cases respectively.Its constituent ratio was 23.8%, 33.3% and 42.9% respectively, significantly higher compared with that of symptomatic PGL (9.8%,43.9%,and 46.3%respectively).Silent PGL can be i-dentified from other adrenal incidentaloma based on enhanced CT value in each period .Biochemical examination or functional imaging further could facilitate the diagnosis of silent PGL .All the 21 cases were conducted with surgical treatment , and hypertensive crisis occurred to 4 cases during operation .Conclusions Silent PGL is not easy to be detected due to its non-specific clinical manifestations .The diagnosis needs the combined analysis on anatomical imaging characteristics , biochemical examination , functional imaging and pathological findings .Sur-gery is the most effective treatment .Postoperative pathological tips and long-term follow-up are both required for PGL, no matter benign or malignant .

DNA replication and RNA biogenesis happen in the cell nucleus,while protein synthesis occurs in the cytoplasm. Integration of these activities depends on function proteins′ selective transport between the two sub-dimensions. It is a signal mediated process, which needs energy and the participation of soluble factors. By introduction of progress on function protein regulated nuclear translocation, its potential medical application has been explored. With deeper investigation in this field, it will significantly promote the design of anti-virus and gene vector therapy.
3. Spent culture supernatant of Lactobacillus acidophilus influences contraction of isolated intestinal smooth muscle of rabbits

Objective: To observe the influence of spent culture supernatant (SCS) of Lactobacillus acidophilus strain LA14 on the contraction of isolated intestinal smooth muscle and to discuss the related mechanism. Methods: The ileum samples of rabbits were prepared and the contraction frequency and amplitude of intestinal smooth muscle were observed as the normal control. Then the SCS, bacterium suspension, and SCS with bacterium suspension were added by an accumulative dose to the culture media (0.3 ml per times, at an interval of 6 min), respectively. Four minutes after each administration, the contractive curves were recorded for 2 min. The influences of various groups of Lactobacillus acidophilus on the contraction of isolated intestinal smooth muscle were observed. The effect of SCS on M cholinoceptor was observed by adding in order pilocarpine, atropine or SCS, and pilocarpine. Results: After continuous administration of SCS or SCS with bacterium suspension (0.6-1.5 ml), the contraction frequency of the intestinal smooth muscle was significantly lowered compared with before administration (P<0.05 or P<0.01), and there were no significant differences between before administration and other volume groups (P>0.05). Within the range of 0.3-1.5 ml, the SCS, bacterium suspension, and SCS with bacterium suspension resulted in no significant difference in reducing the contraction amplitude, except for SCS with bacterium suspension at 1.5 ml(P<0.05). SCS or atropine significantly inhibited pilocarpine-induced increase of contraction amplitude(P<0.05 or P<0.01). SCS also reduced the contraction frequency of the intestinal smooth muscle(P<0.01). Conclusion: SCS of Lactobacillus acidophilus may inhibit the peristalsis of the intestinal smooth muscle of rabbits by blocking M cholinoceptor.

6.Preliminary study on new parameter of myocardial elasticity

Objective To discuss the value of the new parameter of myocardial elasticity derived from circumferential strain.Methods Right ventrieular apical pacing was established on the beagle dogs.Before and after pacing,the peak of early diastolic flow velocity (E peak),peak of atrial flow velocity (A peak),and the ratio of A/E were measured.The peak pressure-rise rate (+ dp/dtmax),peak pressure-fall rate (-dp/dt max),left ventricular systolic pressure(LVSP),LV diastolic pressure(LVDP)were measured by cardiac catheterization.The circumferential strain at the levels of base (CSRA),papillary (CSPM) and cardiac apex (CSAP) were measured,and the parameter of myocardial clasticity at the levels of base (EBA),papillary (EPM) and cardiac apex (EAP) were measured and calculated,respectively.Results ①After pacing,the + dp/dt and LVDP were increased,but the-dp/dtmax was decreased (P <0.05).② After pacing,the CSPM and CSAP were all decreased,but EBA,EPM,EA were all increased(P <0.05).③ The areas under the receiver operating characteristic curves (ROC) of CSAP,CSPM,EAP,EPM and EBA were all 1,and the area under the curve of CSBA was 0.687.EAB,EPM and EAP were significant negative correlation with CSPM and CSAP,but are positive correlated with A/E and LVDP (P < 0.05).Conclusions The parameter of myocardial elasticity derived from circumferential strain includes useful information about myocardial deformation and function.The new parameter can be applied to assess the diastolic function of left ventricle.
7.Preliminary study on ventricular-arterial coupling of patients with hypertension by ultrasonography

Objective To evaluate the cardiovascular stiffness and its coupling of patients with hypertension by ultrasound.Methods Fifty patients with essential hypertension and 30 age- and gendermatched subjects without hypertension,diabetes and other cardiovascular diseases as control group were enrolled in this study.The parameters of structure and function of left ventricle,blood flow were measured by echocardiography.The blood pressure and carotid-femoral pulse wave velocity (CFPWV) were also measured.The derived corresponding parameter:end-systolic pressure(ESP),effective arterial elastance (Ea),end-diastolic (Ed),end-systolic ventricular elastance (Ees) and Ea/ Ees were calculated respectively.Results Ees was correlated positively with ejection fraction (r =0.378,P =0.005),while Ea was correlated positively with CFPWV( r =0.289,P <0.001).Ed was correlated negatively with e/a ( r =- 0.333,P =0.027).Posterior wall of left ventricle was correlated positively with Ed and Ea( r =0.388,P =0.016; r =0.336,P =0.026).Ea and Ed in patients with hypertension were significantly higher than those in control group( P <0.05),but there was no significant difference of Ees and Ea/ Ees between two groups( P > 0.05).Conclusions Arterial stiffness is associated with ventricular stiffness,and their matching relation can be applied to evaluate ventricular-arterial coupling.
8. Accumulation dynamics and distribution characters of vitexin in different parts of Vitex negundo var. cannabifolia

OBJECTIVE: To study the accumulation dynamics and distribution law of vitexin during the whole year period in different parts of Vitex negundo var.cannabifolia (Sieb. et Zucc.). METHODS: Vitexin was obtained by ultrasonic extraction, and its content was determined by HPLC. Filodoor column (4.6 mm×150 mm, 5 μm) was used. Methanol-water (volume ratio of 4;6) was used as the mobile phase at a flow rate of 1 mL·min-1. The UV detective wavelength was set at 340 nm, injection volume was 5 μL, and the column temperature was set at 35°C. RESULTS: The content of vitexin was low in vitex negundo branches and flowers, and had obvious regularity in different growth periods of Vitex negundo leaves. The content of vitexin was the highest (1.380%-1.465%) in June and July. Along with the blooming and formation of negundo chastetree to their maturation, the content of vitexin in Vitex negundo leaves decreased. Its content was the lowest (0.720%-0.751%) in September and October. The content of vitexin in Vitex negundo seeds increased(0.435%-1.231%)with seed maturation. CONCLUSION: The content of vitexin in different parts of Vitex negundo presents a regular change in different periods. The detection of vitexin in different growth periods provides experimental basis for the selection of best collection phase and medical parts.
9.Signal transduction pathways of 17?-estradiol-induced MT1-MMP expression in MG63 cells

Objective To investigate the signal transduction pathways related to 17?-estradiol-induced membrane type-1 matrix metalloproteinase (MT1-MMP) expression in human osteosarcoma cell line MG63. Methods MG63 cells were incubated with ICI 182 780 for 48 h, then exposed to different concentrations of ICI 182780, 17?-estradiol, and specific antagonists H-89, H-7, PD98059, SB203580, genistein and PTDC of protein kinase (PK) A、PKC、extracellular signal-regulated kinase (ERK)、p38MAPK、protein tyrosine kinase (PTK) and nuclear factor (NF)-?B. The MT1-MMP level was determined by Western blotting and immunohistochemistry. Results Western blotting showed that MT1-MMP level was increased in 17?-E_2 group and decreased in PDTC and genistein groups, and immunohistochemistry revealed that MT1-MMP was located in cytoplasm and cellular membrane, but not in nucleus. Conclusion 17?-estradiol regulates MT1-MMP expression mainly through NF-?B and PTK pathways in MG63 cells.
10.Construction of prokaryotic expression vector of mouse CD25 extracellular domain

Objective To construct the prokaryotic expression vector of mouse CD25 extracellular domain and to express it in E coli.Methods Total RNA was isolated from splenocytes of Balb/c mice.The CD25 extracellular domain gene was amplified by RT-PCR and cloned into the PET-32a vector.A positive recombinant,PET-32a-CD25e,was identified by enzyme cleaving and sequencing before its expression in E.coli,and transferred into E.coli BL21(DE3) plysS for its expression.After purification with Ni+ resin and renaturation in vitro,a relative molecular mass(Mr) of the interesting protein was detected by SDS-PAGE and Wes-tern blotting.Effect of the purified interesting protein on the proliferation of splenocytes from T cell vaccine-immunized syngeneic mice was detected by MTT assay.Results The cloned CD25 extracellular domain gene was identified to be functional by sequencing and expression.The purified interesting protein could significantly induce the proliferation and IL-4 secretion of splenocytes from T cell vaccine-immunized mice in vitro.Conclusion Mouse CD25 extracellular domain gene can be successfully expressed in prokaryotic cells with biological activity,which lays a foundation for further relative studies.
