2.Determination of chloramphenicol and metronidazolel in honey by ultra performance liquid chromatography-mass spectrometry with isotope-labelled internal standards

Objective To determine chloramphenicol and metronidazolel in honey by isotope-labelled internal standards ultra performance liquid chromatography-mass spectrometry.Methods Samples were extracted with ethyl acetate solution,and cleaned up on a MCS cartridge.The target analytes were separated on a ZORBAX SB-C18column with gradient elution using a mobile phase made up of methanol and 5 mmol/L ammonium acetate solution (containing 0.05% formic acid).Detection was carried out using positive and negative electrospray ionization and multiple reaction monitoring (MRM),and quantified with isotope internal standardmethod.Results The chloramphenicol and metronidazolel showed good linearity in the range of 0.05-5.00 ng/ml.The recovery at three spiked levels of 0.5,2.0 and 5.0 μg/kg were in the range of 79.3%-96.7%.The relative standard deviation (RSD) was 5.5%-14.8%.The limits of quantitation were 0.15 μg/kg,the limits of detection were 0.05 μg/kg.Conclusion The method is sensitive and accurate.It could be applied to the high-throughput analysis of chloramphenicol and metronidazolel.
3.Effect of Shexiang Baoxin pill on coronary vasodilation by analysis of coronary angiography.

OBJECTIVETo evaluate the effect of Shexiang Baoxin Pill (SBP) on coronary vasodilation by analysis of coronary angiography (CAG).
METHODSA consecutive cohort of 300 patients who underwent CAG between January 2013 and July 2013 were recruited and randomly assigned to 2 groups before operation. Patients in the SBP group sublingually took SBP, while those in the control group sublingually took placebos. All patients repeatedly underwent CAG 5 min after administration. The vascular diameter was calculated by quantitative angiography analysis method. The diameter of the left anterior descending coronary artery was measured in patients whose coronary arteries had no stenosis. The narrowest vascular diameter was measured in patients whose coronary arteries had stenosis. The heart rate, blood pressure, and the vascular diameter were compared between before and after administration in the two groups.
RESULTSIn the two groups, there was no significant difference in changes of heart rate, systolic pressure, or diastolic pressure between before and after administration (all P > 0.05). There were 64 patients with normal CAG in the two groups, 30 in the control group and 34 in the SBP group. CAG showed there were 236 patients with stenotic coronary artery, 110 in the control group and 126 in the SBP group. The vascular diameter was obviously larger in patients in the SBP group with normal or abnormal CAG after administration (all P < 0.01). It was also obviously larger than that of the control group after administration (P < 0.05, P < 0.01).
CONCLUSIONSBP could dilate both normal coronary artery and lesioned coronary arteries, but did not lead to fastened heart rate and decreased blood pressure.

4.Expression of Heat-Shock Protein 70 in Kidney of Acute Injury Rats Caused by Gentamicin

Objective To illustrate the expressional distribution of heat shock protein 70(HSP 70) in the kidney of acute renal injury rats caused by gentamicin.Methods One hundred and fifty rats were randomly divided into 3 groups: group A (normal group), group B [80 mg/(kg?d) gentamicin to be injected], group C [160 mg/(kg?d) gentamicin to be given]. The variations of renal pathology were observed at different time phasess by light scope and electromicroscope. Simultaneously, the expression of HSP 70 in kidney was evaluated by immunohistochemistry.Results HSP 70 distributed in epithelial cells of renal tubular in acute injury rats. 5.Assessment of Oxygen Uptake Efficiency Slope on Cardiopulmonary Functional Reserve for Patients with Chronic Obstructive Pulmonary Disease

Objective To investigate the efficacy of oxygen uptake efficiency slope (OUES) on evaluation the cardiopulmonary function of patients with chronic obstructive pulmonary disease (COPD). Methods The cardiopulmonary function of 54 stable COPD patients with the cardiopulmonary function of Ⅱ~Ⅳ were evaluated, following a symptom-limited Steep protocol with simultaneous respiratory gas measurement,they were performed exercise tests on a treadmill, simultaneously the oxygen uptake (VO2), carbon dioxide production (VCO2),peak oxygen uptake (VO2peak), minute ventilation (VE), and respiratory gas exchange rate (RER) were measured. OUES was derived from the relation between VO2 and VE during incremental exercise and was determined by VO2=algVE+b, where a=OUES, to measure anaerobic threshold (VAT) meanwhile. Results OUES correlated with the VO2peak (P<0.001). 75% OUES, 90% OUES and 100% OUES were not significantly different (F=0.239, P=0.830). Conclusion OUES can respond the cardiopulmonary function in patients with COPD, 75% OUES from sub-maximal exercise can be an index for cardiopulmonary function.
6.Analysis laboratory results of three cases of COVID-19 complicated with falciparum malaria

Abstract: Objective To analyze the laboratory indexes of patients infected with malaria patients and COVID-19, so as to provide reliable evidence for the diagnosis of mixed infection of both. Methods The routine clinical laboratory items such as routine blood, biochemistry and lymphocyte subsets were tested in three cases of COVID-19 complicated with falciparum malaria who admitted to Guangzhou Eighth People's Hospital Affiliated to Guangzhou Medical University from July to December 2020 were tested. Laboratory data were stage-wise analyzed in conjunction with changes in the course of disease. Results Three patients confirmed COVID-19 infection recruited all had malaria infection history. Fever, headache, and other symptoms emerged on the 4rd to 11th day after admission. Malaria parasite was detected by malaria parasite antigen testing and blood smear testing, and all three patients had re-ignition of malaria after being confirmed COVID-19 infection. In the early stage of malaria relapse, lymphocytes decreased, CRP and SAA increased, and gradually returned to normal level after antimalarial treatment. Interestingly, we only found one patient at the initial stage of malaria detection showed PLT decreased, no other unnormal changes in other routine blood results (WBC, ESO) and liver function results (ALT, AST, GGT, TBIL, DBIL, CG) were found from the beginning to end course of the disease. Conclusion COVID-19 infection may promote the resurgence of malaria, so the relapse of malaria should be monitored especially for the patient with malaria infection history who begin to develop fever and other symptoms a few days after the diagnosis of COVID-19. The inflammatory indicators would be worth able as an auxiliary judgment basis for the effective treatment of the two combined infection.
7.Effect of total flavonoids of astragalus on endoplasmic reticulum chaperone, calumenin and connecxin 43 in suckling mouse myocardium with myocarditis caused by coxsackievirus B3.

OBJECTIVETo investigate the effect of total flavonoids of astragalus on the expression of endoplasmic reticulum chaperone, calumenin and connecxin 43 (CX43) in suckling mouse myocardium with myocarditis caused by coxsackievirus B3 (CVB3).
METHODSThe primary culture of suckling mouse myocardium cells were randomly divided into control group, CVB3 infected group and total flavonoids of astragalus group. Firstly, to confirm the identity of the suckling mouse myocardium, α-SMA was monitored by immunohistochemistry method. Then the protein expression changes of endoplasmic reticulum chaperone-glucose regulatory protein 78 ( GRP78), calumenin and CX43 were detected by Western blot.
RESULTS(1) Compared with that of the control group, the GRP78 expression level in CVB3 infected group was improved, the expression levels of calumenin and CX43 were all reduced. (2) Compared with that of CVB3 infected group, GRP78 expression level was decreased, and the expression levels of calumenin and CX43 were increased in total flavonoids of astragalus group.
CONCLUSIONCVB3 infection may cause endoplasmic reticulum stress of rat myocardium cells by increasing the expression of GRP78 and decreasing the expression of calumenin and CX43. On the other hand, total flavonoids of astragalus can reduce the expression of GRP78 and increase the expression of calumenin and CX43.The results of this experiment may be closely related to the effects of anti-arrhythmia with viral myocarditis caused by CVB3.

9.Evaluation of alveolar bone defect in chronic periodontitis by cone-beam computed tomography.

OBJECTIVETo evaluate the morphological characteristics of alveolar bone defects of the patients with chronic periodontitis using cone-beam CT (CBCT).
METHODSSixty patients with chronic periodontitis were included in this study. CBCT was used to scan the alveolar bone and NNT software to measure the alveolar bone defects and bone loss types in different regions.
RESULTSSeventy-five percent (45/60) of the alveolar bone defect was the generalized type, 25% (15/60) was the localized type. In incisor and canine area, the defect of the mandibular alveolar bone was more severe than in the same sites of maxilla. There was less bone loss in the premolar area of mandible than in the same site of maxilla. In the mesial and buccal sites of mandibular molars and in the lingual site of maxillary molars, the most severe alveolar bone loss was found.
CONCLUSIONSThe obvious alveolar bone defect areas in chronic periodontitis were the palatal side of maxillary molars and the lingual side of mandibular incisors. CBCT can clearly demonstrate the degree of alveolar bone defects in different regions of chronic periodontitis.
